2.CT characteristics of acinar cell carcinoma of the pancreas

Background and purpose:Acinar cell carcinoma of the pancreas (ACCP) is a rare malignant tumor and a few radiologic reports have been published. This study aimed to evaluate the CT characteristics of ACCP. Methods:CT signs of 9 cases of pathologically conifrmed ACCP were analyzed retrospectively.Results:The mean value of longest diameter of the 9 cases of ACCP was 52 mm. Among the 9 cases, 6 cases (66.7%) had ill-deifned bor-der, 6 cases (66.7%) showed exophytic type, 8 cases (88.9%) showed enhancement degree less than normal pancreatic tissue, 6 cases (66.7%) represented heterogeneous enhancement, 7 cases (77.8%) showed invaded vessel, 5 cases (55.6%) had lymph node metastasis, and none had hepatic metastasis. Dilated pancreatic duct was observed in only 1 case. Conclusion:When pancreatic mass is large, heterogeneous, exophytic and without dilated pancreatic duct, ACCP is suggested.
3.Determination of prim-o-glucasylcimifugin,5-o-methylvisammiosode and gardenoside in Huoxiang Qingwei Granules by HPLC

AIM: To establish a method of determing prim-o-glucasylcimifugin,5-o-methylvisammiosode,gardenoside quantitatively in Huoxiang Gingwei Granules(Herba Pogostemonis,Fructus Gardeniae,Radix Saposhnikoviae,etc.). METHODS: Three components were determined by HPLC in the same chromatographic condition(Column: Shimadzu VP-ODS 250 mm?4.60 mm,5 ?m;mobile phase: acetonitrile-0.1% phosphoric acid gradient elution;flow rate: 1.0 mL/min: column temperature: at 30 ℃). RESULTS: Prim-o-glucasylcimifugin showed a good linear relationship at the range of 26.0-390 ng,r=(0.999 9),the average recovery was 101.7% and RSD was 1.1%.5-o-methylvisammiosode showed a good linear relationship at the range of 21.5-322.2 ng,r=(0.999 9),the average recovery was 101.9% and RSD was 1.1%.Gardenoside showed a good linear relationship at the range of 47.1-706.5 ng,r=(0.999 9),the average recovery was 101.6% and RSD was 1.1%.(CONCLUSION):() The established method can provide the content determination of 3 ingriedents at the same test condition and be used for the quality control of Huoxiang Qingwei Granules.
4.Effect of eradication of Helicobacter pylori on the expression of telomerase-associated genes and c-myc protein

Objective To investigate the expression of telomerase-associated genes and c-myc protein before and after eradication of Helicobacter pylori(H.pylori) and to elucidate the possible correlation between human telemerase catalytic subunit(hTERT) and human telomerase RNA(hTR) and c-myc protein. Methods Thirty nine H.pylori positive and 21 negative patients were enrolled. The expression of hTR,hTERT and c-myc protein before and after H.pylori eradication were compared. The expression of hTR was determined by in situ RNA hybridization whereas hTERT and c-myc protein were detected by immunohistochemical staining. Results Before treatment,the positive expression of hTR,hTERT and c-myc protein in H.pylori -positive group were significantly higher than those in H.pylori -negative group(51.3% vs. 19.0%,53.8% vs. 23.8% ,53.8% vs. 28.6%,P 0.05). Conclusions H.pylori infection may upregulate the expression of hTERT by inducing the overexpression of c-myc protein,which can cause telomerase activation and gastric carcinogenesis. The expression of telomerase-associated genes and c-myc protein may decrease or disappear after H.pylori eradication,which can lower the risk of gastric carcinogenesis.
5.The expression of ET-1 and ETA/B receptors mRNA in benign prostatic hyperplasia and its significance

Objective To investigate the expression of endothelin 1 (ET 1) and ETA/B receptors mRNA in prostatic tissues and its clinical significance in patients with benign prostatic hyperplasia (BPH). Methods Immunohistochemical staining and RT PCR were used to detect the expression level of ET 1,ETAR mRNA and ETBR mRNA,respectively.The detection results were analyzed in correlation with the clinical parameters of BPH patients. Results The expression level of ET 1 and ETA/BR mRNA in BPH tissue(integral optical densities,0.94?0.08,0.64?0.08,0.97?0.08,respectively)was higher than that in normal prostatic tissue(0.57?0.06,0.37?0.05,0.51?0.04,respectively).In BPH group,the expressed quantities of ET 1 and ETAR mRNA showed positive correlation with IPSS,prostatic volume,prostatic urethral length,prostatic urethral pressure and maximum urethral pressure of BPH patients,but it showed negative correlation with maximum flow rate and average flow rate of them. Conclusions Increased expression of ET 1 and ETAR might play a role in bladder outflow obstruction(BOO) of the patients with BPH.
6.Determination of Dracorhodin in Diedaqili Tablets by HPLC

OBJECTIVE: To determine the content of dracorhodin in Diedaqili Tablets by HPLC. METHODS: The separation was performed on Diamonsil TM C18 column. The mobile phase consisted of acetonitril-0. 05mol? L-1 sodium biphosphate solution ( 45∶ 55) with detection wavelength at 440nm and flow rate at 1. 0mL? min-1. RESULTS: The calibration curve of dracorhodin was linear within the range of 0. 122~ 0. 854? g ( r=0. 999 9) , with average recovery at 101. 4% ( RSD=0. 91% ) . CONCLUSION: This method is simple, reliable and reproducible, and suitable for the quality control of Diedaqili Tablets.
7.Human umbilical cord blood plasma can replace fetal bovine serum for primary culture, proliferation and cryopreservation of umbilical cord mesenchymal stem cells

BACKGROUND:Fetal bovine serum based media used for expanding and cryopreserving human mesenchymal stem cells raise safety concerns in the clinical setting. OBJECTIVE:To investigate the feasibility of human umbilical cord blood plasma as a replacement for fetal bovine serum in culture and cryopreservation of human mesenchymal stem cells derived from umbilical cord. METHODS:Umbilical cord blood units were suitable for this research if they fulfil ed the donor selection criteria of the Guangzhou Cord Blood Bank strictly. Cord blood plasma was ready to use after col ected from the plasma reduction during the suitable cord blood units processing and pooling. Umbilical cord mesenchymal stem cells were harvested from the umbilical cord tissue of health ful-term newborns after delivery by enzyme digestion and were cultured in the presence of Dulbecco’s modified Eagle’s medium/Ham’s nutrient mixture F-12 containing either fetal bovine serum or pooled cord blood plasma. Morphology, proliferation, immunophenotype detected by flow cytometry and differentiation toward adipogenic and osteogenic lineages were utilized for investigating the effect of media on umbilical cord mesenchymal stem cells after 3-5 passages. Then cells were cryopreserved in media containing 10%dimethyl sulfoxide, 20%fetal bovine serum or 20%pooled cord blood plasma for at least 6 months. Viability, adhesion, proliferation, immunophenotype and osteogenic differentiation of the cells were assessed after thawing. RESULTS AND CONCLUSION:The morphology (spindle-shaped and plastic-adherent), phenotype and differentiation potential (osteogenic and adipogenic) were almost indistinguishable between cells cultured in fetal bovine serum or cord blood plasma medium, while cells grown in cord blood plasma medium demonstrated significantly higher proliferation rates than those in medium containing fetal bovine serum. After thawing, the cells maintained their adherence to the culture surface and differentiation potential to osteoblasts, but cells from cord blood plasma cryopreservation medium showed significantly better plastic attachment and produced greater cellnumbers than fetal bovine serum for the first three post-thaw passages. The results demonstrate that cord blood plasma can sever as an effective substitute to fetal bovine serum for growth, maintenance and differentiation of umbilical cord mesenchymal stem cells, and thus it wil be a safe choice for clinical-scale production of human mesenchymal stem cells.
8.The expressions of nuclear factor-κB and activator protein-1 in oxazolone induced colitis in mice

Objective To investigate the expression changes of nuclear factor(NF)-κB and activator protein (AP)-1 in oxazolone induced colitis in mice and their mechanisms. Methods Twenty-four mice were randomly divided into normal group and model group with 12 each. Experimental colitis was induced with skin sensitization of 3% oxazolone for 5 days, then rectal administration of 0.15 ml of 0. 5% oxazolone solution in mice. All mice were sacrificed on day 3. Peripheral blood mononuclear cells (PBMC), spleen mononuclear cells (SMC) and lamina propria mononuclear cells (LPMC) were isolated from the colon tissues. Expression of NF-κB and AP-1 in SMC, LPMC and PBMC were determined by fluorescence quantitative polymerase chain reaction (PCR). The colitis was evaluated histologically. Results The expressions of NF-κB and AP-1 in SMC, LPMC,PBMC of model groupwere significantly higher than those in normal group(NF-κB : 5.62±0.78 vs. 3.16±0.59,5.46±0.38 vs. 3.18±0.58, 5.65±0.56 vs. 3.36±0.59, P<0.01; AP-1; 5.61±0.54 vs. 3.22±0.50, 5.50±0.69 vs. 3.19± 0.40,5.67±0.44 vs. 3. 27±0.41, P<0.01). Conclusion The activation of NF-κB and AP-1 are involved in the mechanisms of ulcerative colitis.
9.Retrospective analysis of the clinical outcomes in 12 recurrent borderline ovarian tumor patients with second fertility-sparing surgery

Objective To analyse the clinical practicability and significance of fertility-sparing surgery for women with recurrent borderline ovarian tumors ( BOT).Methods The clinical and pathological recordings of 12 patients suffered from recurrent BOT in the Obstetrics and Gynecology Hospital of Fudan University from Jan.2002 to Dec.2012 were analyzed retrospectively.The menstruation situation , fertility function , recurrence rate and survival after second conservative surgery were collected and analyzed .Results The average onset age of these 12 patients was 26 years, and none of them had procreation before the second operation.The pathological diagnosis were still BOT and the recurrent sites of 10 patients were still restricted to the ovaries.Of the 12 cases, 8 cases of them had normal menstruation functions and 4 cases had less menstrual volumes or longer menstrual periods.Among 6 cases desired for pregnancy after the second operation,4 cases of them were successful.The spontaneous pregnancy rate was 4/6.The mean duration of follow-up was 53 months(range:23-97 months).All recurrent BOT, three cases developed recurrence once more,two cases of them were just be hinted by B ultrasounds and only one cases was approved to be BOT again by a once more surgical pathology.But no disease related deaths occurred.Conclusions The second fertility-sparing surgery may be feasible for women with recurrent BOT who are young and strongly desired for preservation of fertility.The spontaneous pregnancy rate is satisfactory.However, carefully follow-up is still needed.
10.Role of EGFR-STAT3 signal pathway in carcinogenesis of experimental hepatoma in rats

Background and purpose:It has be reported that the activation of EGFR-STAT3 signal transduction pathway is involved in oncogenesis of many cancers.This study was to investigate whether EGFR-STAT3 pathway plays a role in the carcinogenesis of hepatoma in rats.Methods:Hepatoma induced by 3'Me-DAB was used as a model.EGFR,TGF?,STAT3,p-STAT3 in different stages of carcinogenesis were detected by immunohistochemistry and Western blot.In situ hybridization was applied to investigate the expression of STAT3 mRNA.The slides were assessed by Carl Zeiss Image Analysis system.The data were statistically evaluated.Results:EGFR,TGF?,STAT3 were highly expressed at the stages of liver necrosis and repair.the expression of EGFR,TGFa,STAT3 and p-STAT3 has been found in all hepatomas and the levels of EGFR and TGFa were statistically higher than that in normal tissue,similarlly the STAT3 mRNA and protein level in hepatoma was much higher than in normal tissue(P
