The study intended to evaluate the effect of high-dese atorvastafin on serum high sensitive C-reactive protein (hs-CRP) and renal function in patients with acute myocardial infarction undergoing elective pereutancous coronary intervention ( PCI ). One hundred and sixty seven patients were randomly divided into two groups: in test group (n =84) patients received oral atorvastatin 80 mg/d and in control group (n = 83) patients received atorvastatin 20 mg/d, the medication in both groups was lasted for 7 days before PCL Compared to levels at 24 h before PCI, serum hs-CRP and creatinine levels at 48 h after PCI were increased in both groups ( both P < 0. 05), and glomerular filtration rate was decreased ( P < 0. 05 ). Compared to control group, serum hs-CRP and creatinine levels 24 h before PCI and 48 h after PCI in test group were significantly lower, and glomerular filtration rate was significantly higher (P <0. 05, respectively). The incidence of contrast-induced nephropathy was lower in test group than that in control group[7% (6/84) vs.18% (15/83), P <0.05]. The results indicate that high-dose atorvastatin might be effective in protecting patients with acute myocardial infarction undergoing elective PCI from contrast-induced nephropathy via inflammatory response inhibition.

Objective To explore the effects of tumor necrosis factor-α (TNF-α) on ventricular arrhythmias resulted from acute myocardial infarction (AMI) in rats. Method Two hundred and forty male Wistar rats were randomized (random number) into sham operation group, AMI group and recombinant human tumor necrosis factor receptor (rhTNFR) fusion protein (Fc) group. Acute anterior wall myocardial infarction was produced in rats of AMI group with ligating the left anterior descending coronary artery (LAD) , and the rats were just operated without ligation of LAD in sham group. The rats of Fc group were treated with rhTNFR-Fc (10 mg/kg), a TNF-α antagonist, 24 hours before LAD ligation. The original ECG was recorded 10 min before ligation and the ECGs of ventricular arrhythmias occurred spontaneously or induced by programmed electrical stimulation were recorded 10 min, 20 min, 30 min, 60 min, 3 h, 6 h and 12 hours after ligation. The protein levels and mRNA expressions of TNF-α in rats in different groups were detected with histochemistry and real-time fluorescent quantitative PCR. Results The expressions of TNF-α mRNA and levels of TNF-α protein markedly increased 10 min after infarction, reached the climax 20-30 min later, and then gradually returned to the original level in AMI group and Fc group. The time-windows of spontaneous and induced ventricular arrhythmias were consistent with the time-window of expressions of TNF-α mRNA and levels of TNF-α protein. Compared with AMI group, there were lower levels of TNF-α protein and lower incidence of ventricular arrhythmias in Fc group ( P < 0.05) , but there was no significant difference in TNF-α mRNA between two groups. There was no obvious change in TNF-α in rats of sham operation group. Conclusions The expressions of TNF-α mRNA and levels of TNF-α protein induced by AMI could contribute the initiation of ventricular arrhythmias.

Objective To study the relationship of proliferation of neural stem cells (NSCs) in SVZ and the expressions of c-jun and c-myc in rats with middle cerebral artery occlusion/reperfusion (MCAO/R) injury model administrated byDanlong XingnaoFormula. Methods The focal cerebral ischemia reperfusion injury models were prepared by longa method. Totally 150 male SD rats were randomly divided into sham-operation group, cerebral model group,Danlong XingnaoFormula low-, medium-, and high-dose groups. The treatment groups were given corresponding dose ofDanlong XingnaoFormula, while the sham-operation group and model group were given the same amount of distilled water 24 h after modeling by gavage, once a day, 7 days in a row. 1 d, 3 d and 7 d after reperfusion, modified Neurological Severity Scores (m-NSS) was used to grade neurologic impairment. 7 d after reperfusion taken to the SVZ brain tissue of ischemia side, Brdu immunohistochemical method was used to record the BrdU positive cells number. The hippocampal c-jun, c-myc mRNA and protein expressions were determined respectively by RT-qPCR method and Western blot method. Results Grades of neurologic impairment in others groups were improved obviously than sham-operation group (P<0.01); 3 d, and 7 d after reperfusion, grades of neurologic impairment inDanlong XingnaoFormula groups were obviously lower compared with model group (P<0.05,P<0.01). Brdu positive cell rates in others groups increased obviously compared with sham-operation group; Compared with model group, Brdu positive cell rates inDanlong XingnaoFormula groups increased obviously (P<0.01). The expressions of c-jun and c-myc protein and mRNA inDanlong XingnaoFormula groups improved obviously than sham-operation group and model group (P<0.01).ConclusionDanlong Xingnao Formula can improve the neural function after cerebral ischemia and stimulate the proliferation of NSCs, and the mechanism may be related to activating the expression of c-jun and c-myc and extending the duration.
