1.Study on Quality Standard for Baji Bushen Pills

Objective To improve the quality standard for quality control of Baji Bushen Pills.Methods Radix Rehmanniae Praeparata and Radix Angelicae Sinensis in this medicine were identified by TLC,the 2,3,5,4’-Tetrahydroxystilbene-2-O-?-D-glucoside content in this medicine was determined by HPLC.The acetonitril-water(18:82)solution was used as the mobile phase,the column was C18,the flow rate was 1.0 mL/min,and the detective wavelength was at 320 nm.Results Radix Rehmanniae Praeparata and Radix Angelicae Sinensis could be identified by TLC.2,3,5,4’-Tetrahydroxystilbene-2-O-?-D-glucoside showed a good linear relationship at the range of 11.28 ?g/mL~282.00 ?g/mL(r=0.999 9).The average recovery was 96.1 %with RSD of 0.8 %(n=6).Conclusion The method is simple,reliable,accurate and can be applied to the quality control of the preparation.
2.Determination of Content and Related Substances of Cyproheptadine Hydyochloride Tablets by HPLC

Objective:To establish a determination method for the content of cyproheptadine hydyochloride tablets and the related substances in the tablets by HPLC. Methods:The assay was performed on a CAPCELL PAK C18(Shiseido)(250 mm ×4.6 mm, 5μm) column with methanol-0. 002 5mol·L-1 sodium heptanesulfonate (adjusting pH to 3 with phosphoric acid)(60: 40) as the mo-bile phase. The detection wavelength was 225 nm, the flow rate was 1. 0 ml·min-1 , the column temperature was 30℃ and the sample size was 10 μl. Results: Cyproheptadine hydyochloride had good linear relationship within the range of 4. 12-82. 40 μg·ml-1 ( r=1. 000 0), and the average recovery was 99. 2%(RSD=0. 8%, n=9). The peaks of the related substances were well separated from that of cyproheptadine hydrochloride. Conclusion:The method is simple, fast and accurate, and can be used for the quality control of cyproheptadine hydyochloride tablets.
3.Analysis on Content Determination of 5 Components in Vitamin C Yinqiao Tablet from Multiple Manufactur-ers

OBJECTIVE:To improve the quality standard of Vitamin C yinqiao tablet,and to provide a theoretical basis for im-proving the standards and quality of medicines. METHODS:HPLC in Chinese Pharmacopoeia(2010 edition)was conducted to de-termine the content of chlorogenic acid,arctiin,vitamin C,acetaminophen,chlorphenamine maleate in Vitamin C yinqiao tablet from multiple manufacturers. RESULTS:There were big differences of the 5 components in Vitamin C yinqiao tablet among differ-ent manufacturers. CONCLUSIONS:The content determination method for Vitamin C yinqiao tablet contained in Chinese Pharmaco-poeia remains to be further improved;and the manufacturers should pay attention to the process of production and quality of tradi-tional Chinese medicinal materials of the preparation.
4.A Correlative Study on Multiplanar Coronal Reconstructed and Direct Coronal Images of Orbit and Paranasal Sinuses with Multi-slice Helical CT

Objective To assess the quality of coronal multi-planar reconstructed versus direct coronal scan images and to confirm whether the multiplanar reconstructed image can replace coronal scan.Methods One hundred and sixteen patients with orbital and paranasalsinuses disease were examined with GE Lightspeed plus 4 slices helical CT.All patients had transvers scan,and the coronal multiplannarreconstructed images were reformatted basis on transverse scan.All reformatted and scanned images were evaluated by the senior radiologists.Results In 116 cases,simultaneously multiplanar reconstructed images were performed in 60 cases,the quality of reformatted coronal imageswas similar to direct coronal scan in revealing normal anatomy and lesions of orbit and paranasal sinuses,all images were excellent andimaging quality was grade Ⅰ.54 cases couldn't be undergone direct coronal scan but reformatted coronal images also acquired the effect of diagnosis(32 cases were grade Ⅰ and 3 cases were grade Ⅱ on quality).Conclusion Direct coronal scan can be substitued by multiplanar reconstruction of multi-slice helical CT scan for orbit and paranasal sinuses.
5.Effects of palm pressing PICC and mandible-pressing-on-shoulder on PICC′s misplacement in jugular veins

Objective To study effects of palm pressing peripherally inserted untral catheter(PICC) and mandible-pressing-on-shoulder on PICC′s misplacement in jugular veins during intubation. Methods Seventy-six patients undergoing intubation with PICC were randomized into observation group and control group equally by random digital table. The palm pressing PICC method was used in the observation group and the mandible-pressing-on-shoulder method was used in the control group. The two groups were compared in terms of the incidence of PICC misplacement. Result The misplacement rate of PICC in the experiment group was significantly lower than that in control group (P<0.01). Conclusion Palm pressing method is much effective in prevention of PICC misplacement in jugular veins and worth promoting especially in hospitals of basic level.
6.Effects of benazepril on cardiac function, free oxygen radicals, sarcoplasmic reticulum Ca~(2+)- ATPase following cardiac ischemia- reperfusion in spontaneously hypertensive rats

AIM: To investigate the effects of angiotensin converting enzyme inhibitor (ACEI), benazepril(B), on cardiac function, free oxygen radicals, sarcoplasmic reticulum(SR) Ca~(2+)-ATPase following ischemia-reper-fusion in sportaneously hypertensive rats (SHRs). METHODS: Thirty 10-week-old female SHRs were randomly assigned into two groups: group SHR was control; The animal in group SHR+B was given with 10 mg/kg of benazepril perday. Another 15 Wistar rats with the same age and sex were normal control (group Wistar). After 12 weeks of pretreatment, all rats in each group were subjected to 30 min of left anterior descending coronary artery occlusion and 30 min of reperfusion. Hemodynamic parameters, left heart-to-body weight ratio(LVW/BW), myocardial malondialdehyde (MDA) concentration, superoxide dismutase (SOD) activity, and SR Ca~(2+)-ATPase activity were measured. RESULTS: Compared to group Wistar, the rats in group SHR had higher blood pressure, LVW/BW and myocardial MDA concentration, more serious left cardiac function injury and lower myocardial SOD activity and SR Ca~(2+)-ATPase activity; group SHR+B had lower myocardial MDA concentration, higher myocardial SOD activity, but no difference in blood pressure, LVW/BW, the degree of left cardiac function injury and myocardial SR Ca~(2+)-ATPase activity. CONCLUSION: Benazepril can attenuate ischemia-reperfusion-induced cardiac function injury by regression of left ventricular hypertrophy (LVH), improving SR Ca~(2+)-ATPase activity and decreasing oxygen free radicals injury in SHRs.
7.Density and Affinity of IL-6 Receptors in Human Leukemic Cells

Objective: To make a study of density and affinity of IL-6R in human leukemic cell lines, and discuss the affection of high affinity IL-6R to the targeted treatment of leukemia with IL-6-PE40 fusion protein. Methods: Radial binding assay with scatchard plot and FACS were used to analysis the density and affinity of IL-6R and protein expression of IL-6Rα and β subunits in totally 8 representative human leukemic cell lines. Results: Myelocytie, monocytic and erythrocytic leukemic cell lines U937, HL-60, KG1 and TF1 express high affinity IL-6R, whose average density per cell is 2 502,2 874, 2 319 and 9 329 respectively, however no 125I-IL-6 binding was detected on chronic myelocytic leukemic cell line K562 and lymphoblastic leukemic cell lines such as Raji, CEM and HUT28. These results correlate with those of FACS highly. Conclusion:These observations suggest that acute nonlymphoblastic leukemic cells may be more suitable for targeted treatment with IL-6-PFA0 fusion protein.
8.The Comparison between the Killing Effects of Two Anti-T Immunotoxins on Target Cells

The key to killing target cells by immunotoxin depends on the specific recognition of antibody to target cell and the cytotoxic effect of toxin. The comparative study of the killing effects of two anti-T immunotoxins, CD5:Ricin and CD5:rRA, on target cells was performed. The elimination rate of immunotoxins was analysed by flow cytometry and MLR. The effect of immunotoxins on the proliferation of hematopoiesis was evaluted by CFU-GM. The results showed that (1) CD5(+) T cells were eliminated and CD25(+) CD3(+) activated T cells were concentration-dependently inhibited by the two immunotoxins in the range of 10(-9) - 10(-11) mol/L; (2) both immunotoxins significantly inhibited the mixed lymphocyte reaction, and the inhibiting effect of CD5:rRA to T cell proliferation was markedly lower than that of CD5:Ricin in the range of 10(-10) - 10(-11) mol/L; (3) the combination of CD5:rRA with 10 mmol/L NH(4)Cl increased the T cell elimination rate; and (4) the two immunotoxins and the combination of NH(4)Cl and CD5:rRA did not suppressed proliferation of granulocyte-macrophage progenitors in the range of concentrations with killing effect. It was concluded that T cell and activated T cell could be eliminated effectively by immunotoxins, the proliferation of granulocyte-macrophage progenitor was not inhibited significantly.
9. Analysis of the impact of job characteristics and organizational support for workplace violence

Objective:
To analyze the effect of job characteristics and organizational support for workplace violence, explore the influence path and the theoretical model, and provide a theoretical basis for reducing workplace violence.
Methods:
Stratified random sampling was used to select 813 medical staff, conductors and bus drivers in Chongqing with a self-made questionnaire to investigate job characteristics, organization attitude toward workplace violence, workplace violence, fear of violence, workplace violence, etc from February to October, 2014. Amos 21.0 was used to analyze the path and to establish a theoretical model of workplace violence.
Results:
The odds ratio of work characteristics and organizational attitude to workplace violence were 6.033 and 0.669, respectively, and the path coefficients were 0.41 and-0.14, respectively (
10.Spatiotemporal expression and analysis of responding consecutive monoculture genes in Rehmannia glutinosa.

OBJECTIVEBased on previous study, authors used the suppression subtractive hybridization (SSH) technique to construct the forward and reverse subtractive cDNA libraries of consecutive monoulture Rehmannia glutinosa. Five genes related with consecutive monoculture problem of R. glutinosa were chosen from the each of two subtractive libraries. And their spatiotemporal expression was measured in order to explore the functions in consecutive monoculture problem of R. glutinosa.
METHODUsing the real-time quantitative PCR, we tested the relative expression values of the genes in different development stages and tissues of normal growth (one-year culturing) and consecutive monoculture (two-year culturing) R. glutinosa.
RESULTThe five genes (calcium-dependent protein kinase, s-adenosyl-methionine synthetase, Aminocyclopropane-1-carboxylate oxidase, methyltransferase, calpain), which were chosen from the forward library had high expression in consecutive monoculture R. glutinosa, especially in root, and were hardly expression in normal growth R. glutinosa. On the contrary, the other five genes (RNA-dependent RNA polymerase, RNA replicase, DNA-directed RNA polymerase IIa, cyclin D, RNA binding protein) chosen from the reverse library had high expression in one-year R. glutinosa, but were down regulated or shut down in consecutive monoculture R. glutinosa.
CONCLUSIONThe key genes, which regulate inessential metabolism parthway (such as cyclin D, DNA-directed RNA polymerase IIa), were restrained or shut down in consecutive monoculture R. glutinosa. Calcium and ethylene signaling might played key roles in the formation of consecutive monoculture problem, resulting in disturbing normal metabolic process and syndrome of disease in R. glutinosa appeared in turn.
