1.Investigation on relationship between whole blood zinc and Fe elements with children pneumonia caused by respiratory syncytial virus

Objective To investigate the relationship between zinc and Fe microelements with the respiratory syncytial virus (RSV) infection and to find a clinical and practical treatment and prevention method for RSV caused pneumonia .Methods Sixty children patients with RSV pneumonia were enrolled in the observation group ,and contemporaneous sixty healthy children served as the control group .Then the concentrations of whole blood zinc and Fe microelements were detected by using atomic absorption spectroscopy .Results Compared with the healthy control group ,the levels of whole blood zinc and Fe microelements in different age periods of RSV pneumonia group were significantly decreased with statistical difference(P<0 .05) .Conclusion The decrease of whole blood zinc and Fe microelements levels in children may be associated with RSV caused pneumonia .
2.Establishment of residual ridge resorption model of rats after the mandible incisor was extracted

Objective:To establish an experimental model of residual ridge resorption after mandibular incisor extraction and to study the occurrence mechanisms.Methods:Thirty male Wistar rats were used, the edge of the right mandibular incisor was cut every three days(a total of three times) and the incisor was extracted at three days after the final cut. The animals were sacrificed 0,1,2,4,8,12 weeks respectively after the extraction and the mandibles were dissected out. The length of the alveolar bone and the ratio of extracted side length to unextracted side length were measured on soft X-ray photographs and the mandibles were processed for histomorphometry.Eighteen male Wistar rats were used to observe the morphologic alteration of periodontal tissues by hematoxylin and eosin when the incisor crown was cut at different time intervals.Results:As compared with the unextracted side, the length of alveolar ridge of the extracted side was significantly decreased at 4,8,12 weeks;the ratio of extracted side length to unextracted side length significantly decreased at 4 week after tooth extraction(P

Objective To study the anticoagulant effects of Pheretima extracts and analyze the effective constituents. Methods Antithrombotic effects of Pheretima extracts were observed by determining the coagulation time, bleeding time of mice and the recalcification time of rabbit plasma in vitro, meanwhile the content of free amino acid in the extracts were determined. Results Pheretima extracts could prolong the coagulation time, bleeding time of mice, and the recalcification time of rabbit plasma in vitro. The content of free amino acid was rich in Pheretima extracts. Conclusion Free amino acid in Pheretima extracts may be the main anticoagulant effective constituents.
4.Heterologous expression and substrate specificity of ketoreductase domain in bacillaene polyketide synthase.

The ketoreductase (KR) domain in the first extending module of the polyketide synthase (PKS) catalyzes the reductions of both an α-keto group and a β-keto group in the biosynthesis of bacillaene, suggesting the intrinsic substrate promiscuity. In order to further investigate the substrate specificity, the KR domain (BacKR1) was heterologously overexpressed in Escherichia coli. In vitro enzymatic analysis showed that only one of the four diastereomers was formed in the reduction of the racemic (±)-2-methyl-3-oxopentanoyl-N-acetylcysteamine thioester catalyzed by BacKR1. In addition, BacKR1 was revealed to catalyze the reductions of cyclohexanone and p-chloroacetophenone, indicating the potential of KR domians of PKSs as biocatalysts.

5.Effect of labor analgesia on development of postpartum depression

Objective To evaluate the effect of labor analgesia on the development of postpartum depression.Methods Seventy nulliparous parturients who were at full term with a singleton fetus in vertex presentation,aged 20-35 yr,with body mass index<27 kg/m2,at 38-41 weeks of gestation,of ASA physical status Ⅰ,were enrolled and divided into 2 groups (n =35 each) using a random number table:vaginal delivery group (group VD) and labor analgesia group (group LA).In LA group,the epidural catheter was placed at L2,3 interspace for combined spinal-epidural analgesia when their cervical dilations were in 2-3 cm.VAS score was maintained below 3 after the analgesia.Parturients completed Edinburgh Postnatal Depression Scale questionnaires 42 days after the labor.The development of depression was recorded.Results The incidence of postnatal depression was significantly lower in LA group (17%) than in VD group (40%).Conclusion Labor analgesia can decrease the development of postpartum depression.
6.Study on optimizing human acellular dermal matrix and fluorescence labeling the Co-cultured fibroblasts

Objective To optimize human acellular dermal matrix(ADM) and evaluate its biological characters. Methods Human skin was treated with hypertonic saline followed by NaOH maceration(group A), hypertonic saline followed by sodium dodecyl sulfate (SDS) detergent(group B) or Dispase Ⅱ followed by Triton X-100(group C), the resulting ADM were sectioned, and then were stained by special immunohistochemistry method. The cytotoxicity of them were evaluated by methyl thiazolyl tetrazolium (MTT) colorimetry and then cell compatibility was analyzed by cell culture;The optimized ADM resulted was choosen for use. Fibrablasts(FBs)were transfected with adenovirus vector encoding green fluorescent protein gene(Ad-GFP)and the growth of them on the optimized ADM was observed by fluorescent microscopy. Results Collagen and elastic fibers can still be observed in three kinds of ADM. The cells in dermis can be disintegrated both in group A and C, but not in group B. The cytotoxicity scores of the ADM prepared in group A and B were grade 0 or grade 1, while that of group C was more than grade 1.The ADM prepared by NaCl-NaOH maceration had good biocompatibility. There was statistical difference in adhering number of NIH3T3 cells in group A and B. NIH3T3 cells grew well in group A and the resulted ADM was optimized. FBs transfected with Ad-GFP grew well in the optimized ADM. Conclusion The ADM prepared by NaCl-NaOH maceration was a good tissue engineering biomaterial with a little cytotoxicity and rich in resouce.
7.Silence of VEGFR2 expression mediated by PEI/siRNA complexes

The aim of this paper is to report the study on gene silencing efficiency of siRNA targeted against mouse VEGFR2 (siVEGFR2) in vitro mediated by polyethyleneimine (PEI) and its anti-tumor effect in vivo. CY3-labeled siRNA was compounded into PEI and transfected into MS1 cells. Confocal microscopy was used to image the subcellular distribution of siRNA in MS1 cells. Semi-quantitative RT-PCR and Western blotting were used to evaluate VEGFR2 gene silencing induced by siVEGFR2/PEI complexes. A tumor-bearing nude mice model was established to compare the anti-tumor effect after delivered by local and systemic routes. siVEGFR2/PEI complex-transfected cells exhibited much fluorescence in cytoplasm with no evidence of nuclear accumulation. The expression levels of VEGFR2 mRNA and protein in PEI-transfected cells were significantly down-regulated compared with that in blank group, the silencing efficiency were 28.2% and 23.6% respectively. The tumor sizes in mice intratumorally injected with siVEGFR2/PEI complexes (189.429 +/- 17.562 mm3) were reduced definitely compared to that in mice injected with siVEGFR2/PEI complexes via vein route (315.507 +/- 20.491 mm3), or to saline groups (365.844 +/- 20.713 mm3). The study demonstrated that PEI could effectively transfect siRNA into cells and silence the VEGFR2 gene expression. Intratumoral delivery is more suitable for non-targeted modified PEI/siRNA complexes to inhibit the tumor growth in vivo. The present data lay a solid foundation to further study on the gene silencing mechanism for PEI-medicated RNAi and its anti-tumor efficiency in vivo.
8.Observations on the Efficacy of Electroacupuncture plus a Multi-function Cervical Rehabilitation System in Treating Cervical Spondylosis

Objective To investigate the clinical efficacy of electroacupuncture plus a multi-function cervical rehabilitation system in treating cervical spondylosis.Methods Sixty patients with cervical spondylosis were randomly allocated to treatment and control groups, 30 cases each. The treatment group received electroacupuncture plus treatment with a multi-function cervical rehabilitation system and the control group, electroacupuncture alone. After 30 days of treatment, changes in the pain score, the neck disability index, and cervical active range of motion and isometric muscle strength were observed in the two groups and the clinical therapeutic effects were compared between the two groups.Results The total efficacy rate was 96.7% in the treatment group and 93.3% in the control group; there was no statistically significant difference between the two groups (P>0.05). There were statistically significant pre-/post-treatment differences in the pain score, the neck disability index, and cervical active range of motion and isometric muscle strength in the two groups (P<0.01,P<0.05). There were statistically significant post-treatment differences in the neck disability index, and cervical active range of motion and isometric muscle strength between the treatment and control groups (P<0.05).Conclusions Electroacupuncture plus a multi-function cervical rehabilitation system is an effective way to treat cervical spondylosis. It can relieve the clinical symptoms in the patients.
9.EFFECTS OF CHROMIUM ON BLOOD GLUCOSE AND LIPID METABOLISM AND GENE EXPRESSION OF SKELETAL MUSCLES IN DIABETIC RATS

Objective: To study the effects of chromium on glucose and lipid metabolism and gene expression in diabetic rats.Methods: Male Wistar rats were assigned to three groups:normal control(NC), alloxan-induced diabetic control group(DM), and DM with chromium supplementation group(DM+Cr). Cr 200 ?g/(kg bw?d) was supplemented orally for 60 days. At the end of the treatment, the blood glucose, lipid and serum insulin were measured, and the changes in gene expression among three groups were studied by mRNA differential display technique.Results: Blood glucose in DM+Cr group decreased significantly than that before experiment. The levels of serum TG, TC, LDL-C and AI in DM+Cr group were lower than those of DM group, while the serum HDL-C levels were higher. Serum insulin was not improved obviously in DM+Cr group. 11 cDNA fragments larger than 400 bp expressed differences in skeletal muscles between DM+Cr group and DM group and were isolated, 4 of which expressed higher in DM+Cr group, while the rest expressed higher in DM group.Conclusion: Chromium supplementation could partially improve the disorders of glucose and lipid metabolism, and have an impact on some gene expression in diabetic rats, which may contribute to the regulating effects on its disorders of metabolism.
