1.Study on apoptosis of K562 cells induced by tumor-bearing herb serum of selenium-protein polysaccharide extracted from Qinba selenium-mushroom(秦巴硒菇)

Objective: To explore the effect of selenium-protein polysaccharide extracted from Qinba (selenium-mushroom)(秦巴硒菇) on apoptosis of K562 cells.Methods: Tumor-bearing mice model was established,and selenium-protein polysaccharide extracted from Qinba selenium-mushroom contained-herb serum was prepared by the method of serum pharmacology.K562 cells were conventionally cultured.Growth inhibition rate of tumor cells was detected by methyl thiazolyl tetrazolium(MTT) method.Morphological changes of apoptotic cell were observed under a fluorescent microscope.Cell apoptosis was observed by DNA electrophoresis.Caspase3 relative gene was measured by colorimetry.Results: Selenium-protein(polysaccharide) extracted from Qinba selenium-mushroom could significantly inhibit the growth of K562 cells,there was a remarkably positive correlation between drug concentration,time and inhibitory rate.Apoptotic phenomenon was certained via morphological examination and DNA electrophoresis.Compared with control group,caspase3 gene was markedly upregulated.Conclusion: Selenium-protein(polysaccharide) extracted from Qinba selenium-mushroom could induce apoptosis of K562 cells.The mechanism may be related to upregulation of caspase3.
2.Experimental Study on Eqi Compound Prescription Induced Apoptosis of Acute Myelocytic Leukemia HL-60

Objective To explore the apoptotic effect of Eqi compound prescription (contained-herb serum) on cute myelocytic leukemia HL-60 cell, which is relative to intracellular Ca2+, Caspase-3 and Bcl-2. Methods According to serum pharmacology, HL-60 cells were exposed to 10% concentrations of contained-herb serum for 24, 48, 72 and 96 hours respectively. Cells were observed under a fluore-scence microscope. SubG1 DNA was examined by flow cytometer. Intracellular Ca2+ concentration was measured by fura-2 fluorescence load method. Caspase-3 enzymatic activity were measured by colorimetry. Bcl-2 gene expression were measured by LSAB. Results The contained-herb serum could induce apoptosis of HL-60 cells. 3.Video-assisted thoracoscopic treatment of spontaneous pneumothorax

Objective To propose a treatment protocol by videothoracoscopy in spontaneous pneumothorax. Methods One hundred and three patients underwent Video assisted thoracoscopic (VATS) treatment of spontaneous peumothorax and hemothorax. Indications included recurrent pneumothorax, persistent air leakage following conservative therapy, complicated hemothorax and CT scan identified bullae formation. Results No operative deaths occurred, conversion rate was 2.91%, recurrence rate was 0.97%, complication rate was 3.81% and mean postoperative hospital stay was 5.6 days. Conclusions VATS treatment of spontaneous pneumothorax is better than open chest surgery and also superior than conservative therapy.
4.Effect of Cefradine on pharmacokinetics of baicalin in rats in vivo

Objective To study the effect of oral Cefradine on pharmacokinetics of baicalin in rats in vivo. Methods The rats were divided into two groups: one was supplied with baicalin individually and the other was supplied with the combination of Cefradine and baicalin. Plasma concentrations of two groups were detected by HPLC with electrochemical detection (ECD). The pharmacokinetic parameters were calculated by statistical analysis and compared. Results The pharmacokinetic parameters of combination administration group were C_ max (782.63?469.37) ng/mL, AUC_ 0-24 h (8 407.86?3 476.14) ng/mL?h; while the individual administration group of baicalin was C_ max (2 645.62?601.42) ng/mL, AUC_ 0-24 h (28 952.90 ?5 731.42) ng/mL?h. 5.Effect of Total Flavonoids of Hedysarum Polybotry on Inducing Differentiation of Leukemic Cells

Objective:To investigate the effect and mechanism of total flavonoids of Hedysarum polybotry on induction of differentiation in human leukemia HL-60 cells. Method After the treatment of HL-60 cells with total flavonoids of Hedysarum polybotry, the cell differentiation was detected with NBT reduction method. Cell cycle, CD11b and C-fos were analysed by the flow cytometry. Result The positive rate of NBT reduction and the expression of CD11b were significantly increased. Similar, the expression of C-fos gene was upregulated. The growth of HL-60 cells was arrested at G0/G1 and G2/M phase. Conclusion Total flavonoids of Hedysarum polybotry could induce differentiation of HL-60 cells. Its molecular mechanism might be related to the modulation of gene expressions associated with the proliferation and differentiation, which leads to the inhibition of DNA synthesis.

7.Preliminary Study on Expression of Osteopontin in Human Gliomas

Objective To investigate the expression of osteopontin(OPN) mRNA and its correlation with clinicopathologic features of glioma. Methods The expression of OPN mRNA was detected by semi-quantitive RT-PCR in 60 cases of gliomas and their correspondent normal tissues.The relationship between the relative content of OPN mRNA and clinicopathologic features of glioma was also analyzed. Results The positive rate of OPN mRNA expression was 73.3%(44/60) in our group.The OPN mRNA expression in normal brain tissue was all negative.Furthermore,the OPN mRNA expression was associated with the pathological grade of glioma.All patients were followed up for an average of 12 months.We did observed that the OPN mRNA expression positive group(44 cases) had recurrence in 36 patients and the negative group(16 cases) had only 2 case with recurrence (P
8.The Study of Blood Pressure Changing Patterns in Normal Pregnancy and Pregnancy Induced Hypertension

Objective To study the blood pressure changing patterns in normal pregnancy and pregnancy induced hypertension(PIH). Methods 24 hour ambulatory blood pressure monitoring (ABPM) were performed in 38 cases of normal pregnancy and 36 cases of PIH during 18~20,28~30 and 38~40 gestational week. Results The 24 hour mean blood pressure were increased with pregnancy advanced. In normal pregnancy, a typical circadian rhythm of blood pressure pattern was shown that it increases during the day and decreased during the night with the disparity of (16.3? 4.1)%, (14.1?3.2)% and (15.7?2.0)% in three different gestational week respectively. In moderate and severe PIH, the 24 hour mean blood pressure,blood pressure load and the frequence of abnormal rhythm were significantly higher than those in mild PIH and normal pregnancy, while the disparity between day and night blood pressure was lower. Conclusion The blood pressure patterns show a typical circadian rhythm in most normal pregnancy. The more severe the PIH is, the higher rate of abnormal blood pressure rhythm occured.
9.TRIMETHYLGLYCINE COULD OVERCOME THE DIFFICULTY IN PCR AMPLIFICATION OF G+C-RICH GENES

To explore the effects of different organic solvents including trimethylglycine on the PCR amplification of G+C rich DNA templates,two high G+C containing DNA templates (cyclin dependent kinase inhibitor gene ARF and IGFR II) were amplified using standard PCR and modified PCR with the addition of glycerin, dimethylsulfoxide (DMSO), formamide and trimethylglycine respectively. There were no specific amplified products, when standard PCR and modified PCR with the addition of glycerin, DMSO and formamide were used to amplify the G+C rich ARF and IGFR II genes. But the two genes could be specifically amplified with the addition of optimal concentration of trimethylglycine in the PCR mixture. These results indicate that trimethylglycine can abolish or reduce the formation of second structures in G+C rich DNA regions and promote the amplification of high G+C containing DNA templates.
