1.Influence of Zhibitai Capsule Combined with Atorvastatin Calcium Tablet on Blood Lipid and Inflammatory Factors in Elder Patients with Dyslipidemia

Objective To observe the influence of Zhibitai capsule combined with atorvastatin calcium tablet on blood lipid and inflammatory factors in elder patients with dyslipidemia. Methods Sixty-four elder patients with dyslipidemia were randomly divided into control group and treatment group, with 32 cases in each group. Patients of the control group were treated with atorvastatin calcium tablet (10 mg, qd, bedtime administration), while patients in the treatment group were given Zhibitai capsule (240 mg, bid) for 6 weeks. Serum levels of TC, TG, HDL-C, LDL-C, tumor necrosis factor-alpha ( TNF-α) and interleukin-6 (IL-6) were measured before and after treatment. Results After treatment, compared with the control group, TC [(5. 18±0. 57) mmol·L-1 vs. (5. 73±0. 65)mmol·L-1], TG [(1. 52±0. 43) mmol·L-1 vs. (1. 86±0. 48) mmol·L-1], LDL-C [(3. 36±0. 48) mmol·L-1 vs. (3. 85±0. 53)mmol·L-1], TNF-α[(5. 37±2. 21) ng·L-1 vs. (7. 63±2. 59) ng·L-1] and IL-6 [(12.27±2.75) ng·L-1 vs. (15. 63±2. 92) ng·L-1] were significantly decreased (P<0. 01 for all); however, HDL-C [(1. 26±0. 25) mmol·L-1 vs. (1. 13±0. 23) mmol·L-1] was significantly increased in the patients of treatment group (P<0. 05). Conclusion Zhibitai capsule combined with atorvastatin calcium tablet may not only regulate blood lipid level, but also decrease serum tumor necrosis factor-alpha and interleukin-6 so as to inhibit inflammation reaction in elder patients with dyslipidemia.
2.Research on anti-cancer active component in red wine

Objective:To detect the anti-cancer component, trans-resveratrol in red wine by high performance liquid chromatography, and to determine the influence of environmental factors on its content. Methods: A method for determining trans-resveratrol in red wine by HPLC is described. The operating conditions were ZORBAX-C18 column(4.6 mm×250 mm, 5μm) at room temperature, 30%acetonitrile as mobile phase at a flow of 1ml/min and UV detection at 306 nm. Results:The content of trans-resveratrol in Changyu dry red wine and Great Wall Claret Cabernet are (1.01±0.03)mg/L and (2.36±0.02)mg/L, respectively. The results show that, sunlight, high temperature and poor seal condition can decrease the content of trans-resveratrol. Conclusion:The content of trans-resveratrol varies in different brands of red wine. Red wine should be stored in cool, dark and sealed conditions.
3.Clinical characteristics of pancreas damage in children with Henoch-Sch(o)nlein purpura

Objective To explore the clinical characteristics of pancreas damage in children with Henoch-Sch(o)nlein purpura (HSP).Methods The clinical data of 206 HSP patients was analyzed from February 2008 to July 2013.The serum and urine example was detected by the automatic biochemical analyzer and the clinical characteristics of them was analyzed.The pancreatic morphology of patients was observed by type-B ultrasonic,and the clinical feature of skin,joints,digestive tract and kidney damage were observed.The relationship between pancreas damage and other systems damage was analyzed.Results There was 58 cases suffering from pancreas damage in 206 HSP children:31 cases were male,27 cases were female.There was no patients with pancreatic edema and deformation of structure in 58 patients.The incidence of pancreas damage with HSP combined with abdominalgia patients was 39.7% (50/126),which was obviously higher than that in the patients without abdominalgia 10.0% (8/80) (x2 =14.355,P < 0.01).The incidence of pancreas damage in mixed type of HSP [31.8 % (55/173)] was significantly higher than that in simple type of HSP [9.1% (3/33)] (x2 =18.292,P < 0.01).The mean hospital stay of the HSP patients with pancreas damage was 6-38 (10.39± 6.80) d,while without pancreas damage was 4-18 (8.51 ± 3.42) d,and there was no significant difference (t =5.790,P > 0.05).Conclusions The HSP children usually combine with pancreas damage.If they have abdominalgia and multi-system damage,they are more likely to be accompanied with pancreas damage,which should be paid more attention.
4.Clinical analysis of three internal fixation devices for intertrochanteric fractures of femur

Objective To compare the clinical effects of 3i nternal fixation devices,130?comp res-sion-plate,Richards screw-plate system and Gamma locking nail,in tre atment of intertrochanteric fractu res of femur in order to provide evidence fo r clinical choice.Methods The 3different internal fixation devices were statistically compared in terms of t heir surgery time,volume of operative bleeding,hospitalization time,postop-erative complications,as well as excellent and good rate of function in t reatment of intertrochanteric frac tures of femur.Results Compared with the other two treatmen t groups,the Gamma locking nail grou p presented the best therapeutic effects.Its posto perative complications were significantly lower and its excellent and f ine rate of function was significantly higher t han those of the other two groups.Conclusion Gamma locking nails are a kind of effective internal fixation device for the treatment of intertro chanteric fractures in elderly patients,because it has better biomechanical properties which facilitate fracture healing and recovery of hip joint function.[
5.Mechanism of nerve root stretch injury

BACKGROUND:Under physiological conditions, nerve roots can move along with the movement of limbs and spine. However, the mechanisms of nerve root stretch injury under physiological conditions and the neurological dysfunction after injury are unclear.
OBJECTIVE:To review the reason of nerve root stretch injury, and to analyze the mechanism of nerve function from biomechanics, pathology and neurophysiology.
METHODS:A computer-based online search of PubMed database was undertaken by the first author to identify the articles related to the research of nerve root injury and nerve stretch injury between 1990 and 2012, with the key words of“nerve root, nerve, stretch injury”. A total of 391 articles were screened out. The articles on the anatomy and biomechanics research of nerve root were included, as wel as the pathology and neurophysiology research after nerve root stretch injury. Final y, 44 articles were included for review.
RESULTS AND CONCLUSION:Nerve root can be stretched along with limbs and spine movement. However, diseases or trauma may cause pathological nerve root stretch, and thus leading neurological dysfunction. The nerve root is often influenced stretch because of its anatomical and tissue structure. Mechanical injury mechanism of stretch composes of peripheral (peripheral nerve conduction stress) and central mechanism (displacement of spinal cord content). Pathological studies found that the local fibrosis is caused by the extracted serum protein that difficult to remove after injury, and this is because of the shortage of lymphatic system in nerve roots. Fibrosis can cause nerve root ischemia, affect the nerve function, and change the biomechanical properties of nerve root. Nerve root injury can cause primary and secondary injury of internal axons, and this is the main reason for neurological dysfunction after injury.
6.Exploration Study of the osteosarcoma devitalization technique by vacuum dehydra-tion at room temperature

Objective:To demonstrate the feasibility of the osteosarcoma devitalization method by vacuum dehydration at room tem-perature. Methods:For the in vivo study, the VX2 tumor mass was treated by vacuum dehydration, rehydrated in ice water, and im-planted in the rabbit to determine the safety time to deactivate the tumor. For the in vitro study, the osteosarcoma mass was devital-ized by vacuum dehydration, and the dehydration rate and ATPase activity were determined. Histopathological changes in the tumor were also observed. The change in the biomechanical strength of rabbit bone and tendon after vacuum devitalization treatment was detected. Results:At room temperature, the safety time to deactivate the VX2 tumor was 60 min, and the dehydration rate was 93.8%at this time point. After vacuum dehydration, the tumor mass evidently shrunk, presenting a porous structure. The osteosarcoma cell became small, and cell structure damage was observed under light microscope. Disrupted cell membrane and organelles were seen un-der transmission electron microscope as well as broken down chromosomes. The activity of ATPase was evidently lower than in the control group. The strength of bone and tendon did not decrease significantly after vacuum dehydration. Conclusion:Treatment by vacuum dehydration at room temperature for 60 min does not result in differences of the bone and tendon strength. However, it can inactivate both soft tissue and bone tumor mass completely.
7.Inhibition of combined application of arsenic trioxide and cisplatin in the HSQ-89 cells

Objective To discuss the anticancer role of arsenic trioxide (ATO) with cisplatin on human oral carcinoma HSQ-89 cells. Methods The human oral epidermoid HSQ-89 cells were chosen as the subjects. Different concentrations of ATO were added into Cisplatin(DDP)-treated cells. The inhibition rate of tumor cells was detected by MTT assay. Results Different concentrations of ATO (0,2.5,5,7.5,10,12.5 μmol/mL) were added into oral cancer HSQ-89 cells which have been treated with DDP (15 μg/mL). The inhibition rate of tumor cells were 26.9%, 67.5%, 73.0%, 88.5%, 90.4%, 98.7%respectively; The combined application of ATO with cisplatin could improve the inhibition rate of HSQ-89 cells in a dose-dependent relation. Conclusion The combined application of ATO and DDP can produce a synergistic action of inhibition on oral cancer cell.
8.Sunitinib malate-induced high expression of NKG2D ligands in nasopharyngeal carcinoma cell ABCG2~(high) CNE2/DDP

Objective:To investigate the inducing effects of sunitinib malate on expression of NKG2D ligands in nasopharyngeal carcinoma cell ABCG2high CNE2/DDP.Methods:ABCG2highCNE2/DDP cells and Allo-NK cells were isolated by magnetic activated cell sorting(MACS).Flow cytometry was used to evaluate the purity of isolated cells and the expression of NKG2D-ligands on target cells before and after incubation with sunitinib malate.Then the cytotoxic sensitivity of treated and un-treated ABCG2high CNE2/DDP cells to Allo-NK cells were measured by LDH releasing assay.Results:The positive rate of ABCG2 in ABCG2highCNE2/DDP cells was(91.40?2.32)%.More than 90% of isolated Allo-NK cells were proven to be CD3-CD16+CD56+ cells.The expression of MICA,MICB,ULBP1,ULBP2 and ULBP3 on ABCG2high CNE2/DDP cells incubated with sunitinib malate increased from(2.92?0.33)%,(4.27?0.33)%,(5.80?0.62)%,(11.10?3.15)%,and(7.75?1.14)% to(89.12?4.56)%,(66.10?2.22)%,(67.56?4.19)%,(69.37?8.83)%,and(63.28?3.31)%,respectively.At the E ∶T ratios of 10 ∶1 and 20 ∶1,the cytotoxic sensitivities of ABCG2high CNE2/DDP cells to Allo-NK cells increased from(15.32?13.86)% and(27.26?6.81)% to(41.12?4.12)% and(57.25?2.37)%,respectively,after treatment with sunitinib malate,with significantly difference found in the cytotoxic sensitivities of target cells in each group before and after sunitinib malate treatment(F=15.58,P=0.000).Conclusion:Sunitinib malate can up-regulate expression of NKG2D-ligands(MICA/B,ULBP1-3)in ABCG2high nasopharyngeal carcinoma cells,which results in higher cytotoxic sensitivity to Allo-NK cells.
9.Inhibitory effects of Co-SZ eye drops on apoptosis of lens epithelial cells and their signal transduction mechanisms

AIM: To investigate the inhibitory effect of Co-SZ eye drops on apoptosis of lens epithelial cells(LEC) induced by H_2O_2 and to study the cellular and molecular mechanisms.METHODS:(1) All lenses of Sprague-Dawley rats were incubated with H_2O_2 and Co-SZ eye drops.The apoptosis rates of LEC were determined by TUNEL method.The changes of LEC ultrastructure and the formation of apoptotic body were observed by electron microscopy.(2) Bovine LEC were incubated with H_2O_2 and Co-SZ eye drops.The inhibitory of LEC apoptosis was detected by MTT after incubation.The changes of fractional DNA content in LEC were detected by flow cytometry(FCM).[Ca~(2+)]i,cAMP and cGMP of LEC were determined by spectrofluoremeter and radioimmunoassay,respectively.RESULTS: The LEC apoptosis rates in Co-SZ eye drops group were decreased significantly compared with H_2O_2 group by TUNEL.The ultrastructure changes in LEC of Co-SZ eye drops group were lighter than that in H_2O_2 group.The LEC apoptosis rates of Co-SZ eye drops group were dose-dependently decreased significantly compared with H_2O_2 groups via MTT assay.LEC apoptosis induced by H_2O_2 was inhibited by Co-SZ eye drops,and showing dose-dependent.The DNA contents in LEC of Co-SZ group were increased.The [Ca~(2+)]i and cAMP in Co-SZ group were decreased obviously.The cGMP was increased.CONCLUSION: The LEC apoptosis induced by H_2O_2 was inhibited by Co-SZ eye drops.The mechanism of apoptosis inhibition by Co-SZ eye drops maybe contribute to the increase in DNA content.The signal transduction mechanisms are related to the decrease in [Ca~(2+)]i and cAMP and the increase in cGMP.
10.Clinical characterization and genetic testing for a patient with creatine deficiency syndrome 1.

OBJECTIVE:
To explore the genetic basis for a child affected with cerebral creatine deficiency syndrome 1 (CCDS1).
METHODS:
High-throughput sequencing was carried out to screen pathogenic variant associated with the clinical phenotype of the proband. The candidate variant was verified by Sanger sequencing.
RESULTS:
High-throughput sequencing revealed that the proband has carried heterozygous c.327delG variant of the SLC6A8 gene, which was verified by Sanger sequencing.Neither parent was found to carry the same variant.
CONCLUSION
The de novo heterozygous c.327delG variant of the SLC6A8 gene probably underlay the CCDS1 in this child.
