Aim To investigate the effects of PGF_(2?) upon glucose-stimulated insulin secretion and the calcium response in NIT-1 beta cells.Methods Using the radioimmunoassay(RIA),the amount of PGF_(2?) augmentation of glucose-stimulated insulin secretion was determined in different conditions and the confocal laser scanning methods by Fluo-3AM as a fluorescent probe were used to analyze the NIT-1 beta cell intracellular calcium response in correlated various terms.Results In the presence of 16.5 mmol?L~(-1) glucose,PGF_(2?)(0.1,1,5 ?mol?L~(-1)) dose-dependently augmented glucose-induced insulin secretion in NIT-1 beta cells,especially at 5 ?mol?L~(-1)(P0.05).Meanwhile,Exposure of the NIT-1 cells to 5 ?mol?L~(-1) PGF_(2?) induced a rapid increase of intracellular calcium(P
2.Determination of emodin and chrysophnol in Shierwei Chiyinkang Powder by HPLC

AIM: To develop a HPLC method to determine emodin and chrysophanol in Shierwei Chiyinkang Powder (Gypsumseu Calcitum, Radix et Rhizoma Rhei, Herba Asari, Rhizoma Chuanxiong, Rhizoma et Radix Notopterygii, Radix Gentianae, Pericarpium Zanthoxyli, etc.). METHODS: The sample was made referring to China Pharmacopoeia(2000) and determined by HPLC. The separation was performed on a Shim pack CIC ODS column (150mm?5mm, 4.5?m) with a mobile phase of methanol 0.1% H 3PO 4 solution (40∶9). The flow rate was 1.3mL?min -1 and the detective wavelength was set at 254nm. RESULTS: The emodin and chrysophanol in sample were separated. The square linear ranges were 0.52~5.28?m?mL -1 ( r =0.9999) and 0.96~9.60?m?mL -1 ( r=0.9999 ), respectively, and the average recoveries were 98.62% and 98.57% for emodin and chrysophanol, respectively. CONCLUSION: This method is sensitive and accurate for the determination of emodin and chrysophanol in the sample.
3.Mice autoimmune hepatitis treated by bone marrow mesenchymal stem cell

Objective To explore the therapeutic effects and mechanism of bone marrow mesenchymal stem cells (MSC) transplantation in mice autoimmune hepatitis (AIH).Methods AIH model was established in 44 C57BL/6 mice,which were induced by homologous series liver-specific antigen S-100 and Freund's complete adjuvant.After modeling,six mice were collected for AIH model confirming.The other 38 mice were divided into three groups.Fourteen mice of MSC transplantation group (group A) were treated by MSC tail vein injection,12 mice of dexamethasone (DXM) group (group B) were treated by DXM intraperitoneal injection,and 12 mice of PBS control group (group C) received phosphate buffer saline (PBS) intraperitoneal injection.Eighteen mice of healthy control group (group D) weren't modeled and received no treatment.At the 5th and 9th week,the mice weights and serum alanine aminotransferase (ALT) level were tested,mice liver tissues of each group were estimated by pathological examination and Knodell scoring,and spleen T lymphocytes of mice were isolated for proliferation-inhibition examination.The data were analyzed by rank sum test,ANOVA and t test.Results After treatment,mice weights of both group A and B showed upward trend (F=15.678,P<0.01; F=3.730,P=0.037).Before and after treatment,there was no significant difference in group C (P>0.05).At the 5th and 9th week,the ALT level of group A and B gradually decreased,there was statistical significance between the time points (F=20.267,P<0.01; F=4.277,P=0.034).Before and after treatment,there was no significant difference in ALT level of group C (P>0.05).At the 5th and 9th week,the degree of mice serum ALT reduction of group A was larger than that of group B,and the difference was statistically significant (t=3.566 and 3.218,both P<0.05).At the 5th and 9th week,the Kondell scores of group A and B gradually decreased,there was statistical significance between the time points (F=8.070,P=0.003; F=6.547,P=0.009).Before and after treatment,there was no significant difference in Kondell scores of group C (P>0.05).At the 9th week,there was statistical significance in Kondell scores among group A,group B and group C (F =4.477,P =0.029).The in vitro spleen lymphocytes proliferation-inhibition experiment demonstrated that the supernatant of MSC could significantly inhibit the proliferation of T lymphocytes stimulated by S-100 antigen and concanavalin A,the absorbance values were0.267±0.167 vs.0.217±0.128 and0.165±0.187 vs.0.082±0.051 respectively,the differences were statistically significant (t =7.187 and 4.602,both P< 0.01).Conclusion MSC transplantation may play a therapeutic role in mice AIH through inhibiting T lymphocyte proliferation.
4.Relationship between muscular fitness and quality of life as well as psychological distress in radiotherapy patients with cervical cancer.

OBJECTIVES:
Patients with cervical cancer who have received radiotherapy often suffer from systemic muscle volume reduction and quality of life decline due to systemic effects of tumor and side effects of radiotherapy. The purpose of this study was to investigate the status of muscle fitness, quality of life, and psychological pain in patients with cervical cancer who received radiotherapy, and to explore the correlation between muscle fitness, quality of life, and psychological pain.
METHODS:
A total of 202 cervical cancer patients aged 19-71, who received radiotherapy in Hunan Cancer Hospital from July 2020 to February 2021, were selected by convenience sampling method. Functional Assessment of Cancer Therapy Cervix (FACT-CX) and Distress Thermometer (DT) were used for the survey. The patient's grip strength was assessed by a handgrip meter and compared with that of healthy Chinese women of the same age. The correlation between muscle fitness and quality of life and psychological pain was analyzed.
RESULTS:
The grip strength of cervical cancer patients receiving radiotherapy was significantly lower than that of healthy Chinese women at the same age (
CONCLUSIONS
The grip strength of patients with cervical cancer undergoing radiotherapy is generally decreased, which is affected by many factors, and is closely related to the quality of life and psychological pain of patients. In the future, dynamic attention should be paid to the changes of grip strength and related functions in patients receiving radiotherapy for cervical cancer, the potential risks in the treatment should be identified early, and targeted intervention should be taken.

5.Congenital renal diabetes insipidus caused by aquaporin 2 gene mutation in siblings: 2 cases report and literature review

Objective:To improve clinicians' understanding of congenital nephrogenital diabetes insipidus (CNDI) and to reduce missed and misdiagnosis. Methords Based on the literature, the clinical data and gene mutation of 2 patients with CNDI who were admitted to the Department of Endocrinology and Metabolism of the First Affiliated Hospital of Henan University of Science and Technology on July 30, 2020 were analyzed retrospectively. Results:(1) The presentee, 4 years old, had irritable thirst, polydipsia and polyuria for more than 3 years. The sister, 2.5 years old, had irritable thirst, polydipsia and polyuria for more than 2 years. The clinical diagnosis was “CNDI”, and the symptoms improved after treatment with hydrochlorothiazide. (2) The genetic test revealed that the congenital nephrogenic uremia and her sister had a heterozygous mutation of c.170A>C (p.Q57P) and c.211G>A (p.Vl71M) in the aquaporin-2 gene, and the mother carried the AQP2 gene. c.170A>C(p.Q57P) mutation.Conclusion:CNDI is a rare disease. Early diagnosis and treatment can improve the prognosis of patients to the greatest extent, and prenatal diagnosis can guide eugenics.

Familial acne inversa (AI) is an autoinflammatory disorder that affects hair follicles and is caused by loss-of-function mutations in γ-secretase component genes. We and other researchers showed that nicastrin (NCSTN) is the most frequently mutated gene in familial AI. In this study, we generated a keratin 5-Cre-driven epidermis-specific Ncstn conditional knockout mutant in mice. We determined that this mutant recapitulated the major phenotypes of AI, including hyperkeratosis of hair follicles and inflammation. In Ncstn;K5-Cre mice, the IL-36a expression level markedly increased starting from postnatal day 0 (P0), and this increase occurred much earlier than those of TNF-α, IL-23A, IL-1β, and TLR4. RNA-Seq analysis indicated that Sprr2d, a member of the small proline-rich protein 2 family, in the skin tissues of the Ncstn;K5-Cre mice was also upregulated on P0. Quantitative reverse-transcription polymerase chain reaction showed that other Sprr2 genes had a similar expression pattern. Our findings suggested that IL-36a might be a key inflammatory cytokine in the pathophysiology of AI and involved in the malfunction of the skin barrier in the pathogenesis of AI.
