1.Influence factor analysis of application of the new gray correlation analysis method to hospitalization costs

Objective By investigating the influencing factors and the tendency of devel-opment of 6-year hospitalization costs in our hospital,our hospital explores an appropriate way of analysis on the influencing factors of medical costs,in order to offer some quantifiable evidence of evaluating and meaningful references to controlling medical costs. Methods to use the new method of gray correlation analysis to make the quantitative analysis of the composing of the hospital fees from 2002 to 2007.Results the new method of gray correlation analysis is able to overcome the influence of non-dimensional treatment which affects the role of factors and simplifies the calculation steps. Conclusion It is a statistical analysis method whose result is reasonable and reliable,whose application is flexible and simple. It can also offer a quantifiable index of the tendency of the system of development and changes. It is suitable for measuring the interrelated degree among the factors and the dynamic process of analysis.
2.Study on correlation between plasma NT-pro BNP level and heart function in children with heart failure

Objective To investigate the relationship between the plasma NT-pro BNP level and the hear function in children pa-tients with heart failure.Methods 30 children patients with heart failure and 30 healthy children were selected as the patients group and the control group respectively.The plasma NT-pro BNP level was detected in the patients group before and after treatment and the control group.The changes of heart functional indexes and the clinical symptoms in the patients group were detected,the detec-tion results and the plasma NT-pro BNP levels were performed the correlation retrospective analysis.Results The plasma NT-pro BNP level before treatment in the patients group was significantly higher than that in the control group,there was the statistically significant difference between them(P <0.05 ).After treatment,the clinical symptoms in the patients group were obviously im-proved,the plasma NT-pro BNP level was significantly decreased and LVEF by ultrasound was also increased,the differences were statistically significant(P <0.05).The plasma NT-pro BNP level after treatment in the patients group was obviously higher than that in the control group with statistical difference between them(P <0.05).The plasma NT-pro BNP level before and after treat-ment in the patients group demonstrated a negative correlation with LVEF(r=-1.32,r=-1.78).Conclusion Detecting the plas-ma NT-pro BNP level in children patients with heart failure has the important clinical guidance significance for the diagnosis,disease condition evaluation,prognostic evaluation and cardiac function grading in the children patients with heart failure.
3.Mass spectrometry technology and its application in analysis of biological samples.

With the excellent merits of wide analytical range, high sensitivity, small sample size, fast analysis speed, good repeatability, simple operation, low mobile phase consumption, as well as its capability of simultaneous isolation and identification, etc, mass spectrometry techniques have become widely used in the area of environmental science, energy chemical industry, biological medicine, and so on. This article reviews the application of mass spectrometry technology in biological sample analysis in the latest three years with the focus on the new applications in pharmacokinetics and bioequivalence, toxicokinetics, pharmacokinetic-pharmacodynamic, population pharmacokinetics, identification and fragmentation pathways of drugs and their metabolites and metabonomics to provide references for further study of biological sample analysis.

Objective To explore the clinical efficacy of knee osteoarthritis treated by abdominal acupuncture combined with sodium hyaluronate injected in intraarticular.Methods 120 patients with knee osteoarthritis were randomly divided into observation group and control group,60 patients in each group.The observation group was treated by abdominal acupuncture combined with sodium hyaluronate injected in intraarticular.The control group was treated with intraarticular injection of sodium hyaluronate alone.Results After a follow up of 6 months, the total effective rate of the treatment group was 95.00%,which was higher than 78.33% of the control group, the difference was statistically significant (x2=7.21,P<0.01).Conclusion Abdominal acupuncture combined with intraarticular injection of sodium hyaluronate is a good way to treat middle and old age patients with knee osteoarthritis.
5.Expression of four-jointed box kinase 1 gene in colorectal cancer and its clinical significance

Objective:To investigate the differential expression of four-jointed box kinase 1 (FJX1) gene in colorectal cancer and its relationship with prognosis and the related mechanisms.Methods:On July 16, 2021, the transcriptome data and clinical data of colorectal cancer were downloaded from The Cancer Genome Atlas (TCGA) database to analyze the expressions of FJX1 mRNA in colorectal cancer tissues and paracancerous tissues, and the relationship between FJX1 mRNA and clinicopathological characteristics and prognosis of patients. Receiver operating characteristic (ROC) curve was drawn to evaluate the value of FJX1 mRNA in predicting the survival of patients with colorectal cancer. Cox proportional hazards model was used to evaluate whether FJX1 mRNA was an independent influencing factor for prognosis of colorectal cancer. The overall survival (OS) time and survival status of colorectal cancer patients were downloaded from the Gene Expression Omnibus (GEO) database, and the relationship between FJX1 mRNA and prognosis of patients was analyzed. The methylation data of colorectal cancer was downloaded from the University of California, Santa Cruz (UCSC xena) database to determine the degree of methylation at each site of FJX1 mRNA and the correlation between the expression of FJX1 mRNA and the degree of methylation at each site. Signaling pathways associated with FJX1 mRNA in colorectal cancer were analyzed by using the Gene Set Enrichment Analysis (GSEA) (4.1.0). The correlation between FJX1 mRNA and tumor-infiltrating immune cells was investigated by using the Tumor Immunity Evaluation Resource (TIMER) database. Spearman analysis and small molecule/drug sensitivity analysis were used to explore the correlation between FJX1 mRNA expression and drug sensitivity.Results:In the transcriptome data of 612 colorectal cancer cases in TCGA database, the expression of FJX1 mRNA in colorectal cancer tissues was higher than that in the paracancerous tissues ( P < 0.001). In 549 colorectal cancer patients with complete data, FJX1 mRNA expression was correlated with M stage ( P = 0.007), pathological stage (stage Ⅳ vs. stage Ⅰ, P = 0.016; stage Ⅳ vs. stage Ⅱ, P = 0.03; stage Ⅳ vs. stage Ⅲ, P = 0.012), but it was not correlated with age, gender, T stage and N stage (all P > 0.05). In TCGA database and GEO database, the patients were divided into high expression group and low expression group according to the median expression of FJX1 mRNA. The OS in FJX1 mRNA high expression group was worse than that in low expression group (all P<0.05). The ROC curve of FJX1 mRNA expression on the 1-, 3-, and 5-year OS rates of colorectal cancer patients was drawn by using the data in TCGA database, and the areas under the curve (AUC) were 0.595, 0.625 and 0.764, respectively. Multivariate Cox regression analysis showed that age ( HR = 1.050, 95% CI 1.028-1.073, P < 0.001), T stage ( HR = 1.787, 95% CI 1.090-2.927, P = 0.021) and high FJX1 mRNA expression ( HR = 1.160, 95% CI 1.049-1.282, P = 0.004) were independent influencing factors for poor OS in colorectal cancer. The gene set enrichment analysis found that FJX1 mRNA was related to colorectal cancer, TGF-β signaling pathway, VEGF signaling pathway, Wnt signaling pathway, etc. The expression of FJX1 mRNA in colon cancer was negatively correlated with the degree of methylation of FJX1 mRNA ( r = -0.16, P < 0.001), and the expression of FJX1 mRNA in rectal cancer was positively correlated with the degree of methylation of FJX1 mRNA ( r = 0.33, P < 0.001). The expression of FJX1 mRNA was related to the infiltration of resting memory CD4 + T cells, M0 macrophages and resting dendritic cells. FJX1 mRNA was significantly associated with the resistance of various chemotherapeutic drugs and tumor-targeted drugs such as methotrexate, 5-fluorouracil, gefitinib, etc. Conclusions:FJX1 mRNA may be a potential biomarker of colorectal cancer and is associated with the infiltration of immune cells.
6.Nutrient Requirement Pattern of Belamcanda chinensis

ObjectiveTo study the changes of dry matter accumulation, mineral element absorption and medicinal component accumulation in Belamcanda chinensis and to provide a theoretical basis for fertilization in the standardized cultivation of B. chinensis. MethodDuring the two-year growth period of B. chinensis, the samples were collected at 30-day intervals from day 90 after sowing for the measurement of dry matter accumulation and mineral element content in different organs as well as the flavonoid content in the rhizome. ResultAccording to the accumulation of dry matter in different organs, the two-year growth period of B. chinensis was divided into seven stages: seedling stage (before July in the first year), first-year vegetative growth stage (August to October in the first year), first-year late growth stage (November to January of the second year), re-growth stage (February to March in the second year), second-year vegetative growth stage (April to May in the second year), reproductive growth stage (June to October in the second year), second-year late growth stage (November of the second year to January of the third year). During the whole growth period, the late growth stage of each year witnessed the expansion of the rhizome, and the accumulation of dry matter of the whole plant peaked after the reproductive growth stage. The content of flavonoids in the rhizome varied greatly among different stages. Specifically, the content of mangiferin, tectorigenin and irigenin was higher in the second-year vegetative growth stage, and that of tectoridin and iridin was higher in the first-year late growth stage and the second-year late growth stage. The content of irisflorentin was higher in the second-year vegetative growth stage and the second-year late growth stage than that in other stages. The total content of six flavonoids peaked in the second-year vegetative growth stage (April in the second year). The correlation analysis showed that all the nine mineral elements had different degrees of positive correlations with tectorigenin, irigenin and irisflorentin. ConclusionIn the first year, B. chinensis presents slow dry matter accumulation and low demand for fertilizer, so the application of base fertilizer can be reduced. In the second year, especially after the reproductive growth, B. chinensis demands high fertilizer, so nitrogen, phosphorus and potassium fertilizers and other micronutrients should be applied as early as possible. In addition, the rhizome expands in October to November of the first year and November to December of the second year, when the supply of nitrogen and potassium to the rhizome should be ensured or beneficial elements should be supplemented to improve the quality of B. chinensis.
7.Effect of Different Nitrogen Application Rates on Growth, Yield and Quality of Two-year-old Belamcanda chinensis

ObjectiveTo investigate the effects of different nitrogen application rates on the growth, yield and quality of Belamcanda chinensis and to provide a theoretical basis for the quality and production efficiency improvement of B. chinensis. MethodA field experiment was carried out on the basis of 120 kg·hm-2 phosphate and potassium fertilizers, with five nitrogen fertilizer treatments: 0 (N0), 60 (N60), 120 (N120), 180 (N180) and 240 (N240) kg·hm-2. The effects of different nitrogen application rates on the agronomic traits, yield, mineral element accumulation and flavonoid content of B. chinensis were studied. ResultApplication of nitrogen fertilizer was beneficial to dry matter accumulation and significantly increased the yield of rhizome. After the application of nitrogen fertilizer, the yield of rhizome increased by 48.75%-86.62%. The nitrogen application at 60-180 kg·hm-2 increased the active components such as tectoridin, iridin and irisflorentin, while it was not conducive to the accumulation of tectorigenin and irigenin in the rhizome. The accumulation of Mn and Fe reached the highest when the nitrogen application rate was 120 and 60 kg·hm-2, respectively. The principal component analysis of active component content showed that the comprehensive score was ranked as N180>N120>N60>N240>N0. The correlation analysis indicated that the tectoridin content had a positive correlation with N accumulation and a negative correlation with Ca accumulation in B. chinensis. The content of tectorigenin was negatively correlated with N accumulation and positively correlated with Ca accumulation. The content of irigenin had positive correlations with Ca and Zn accumulation. ConclusionThe nitrogen application at 120-180 kg·hm-2 can increase the rhizome yield and the content of tectoridin, iridin and irisflorentin in the rhizome, so as to improve the yield and quality of this herb.
8.Effect of Phosphorus Application on Agronomic Traits, Yield and Quality of Two-year-old Belamcanda chinensis

ObjectiveTo investigate the effects of different phosphorus application rates on the growth, yield and quality of two-year-old Belamcanda chinensis and provide a basis for phosphorus application in the high-quality and high-yielding cultivation of B. chinensis. MethodIn this study, we designed five phosphorus application rates of 0 (P0), 60 (P60), 120 (P120), 180 (P180) and 240 (P240) kg·hm-2 on the basis of 120 kg·hm-2 nitrogen and potassium fertilizers to investigate the effects of different phosphorus levels on the agronomic traits, yield, mineral content, flavonoid content and phosphorus utilization efficiency of B. chinensis through field experiments. ResultThe plant height and tillering number of B. chinensis gradually increased after phosphorus fertilization. The phosphorus application increased the rhizome yield by 37.59%-61.03% compared with P0 treatment and the highest yield was achieved when phosphorus fertilizer was applied at 120 kg·hm-2. The content of mangiferin and irisflorentin in the rhizomes gradually increased with the increase in phosphorus application and the moderate application of phosphorus fertilizer increased the content of tectoridin, iridin and irigenin. The highest Mn, Cu and Fe elements were achieved at 60 and 120 kg·hm-2 of phosphorus fertilization, respectively. The overall quality scores of B. chinensis were ranked in the order of P240>P180>P120>P60>P0. With the increase in phosphorus application, the phosphorus accumulation of B. chinensis first increased and then decreased, and the partial factor productivity, apparent utilization rate and agronomic efficiency of phosphorus fertilizer gradually decreased. ConclusionConsidering the yield, quality and phosphorus utilization efficiency of B. chinensis, it is recommended to apply phosphorus fertilizer at about 120 kg·hm-2 in the production of B. chinensis in Tuanfeng county.
9.Artemisia argyi extract subfraction exerts an antifungal effect against dermatophytes by disrupting mitochondrial morphology and function.

Artemisia argyi (A. argyi), a plant with a longstanding history as a raw material for traditional medicine and functional diets in Asia, has been used traditionally to bathe and soak feet for its disinfectant and itch-relieving properties. Despite its widespread use, scientific evidence validating the antifungal efficacy of A. argyi water extract (AAWE) against dermatophytes, particularly Trichophyton rubrum, Trichophyton mentagrophytes, and Microsporum gypseum, remains limited. This study aimed to substantiate the scientific basis of the folkloric use of A. argyi by evaluating the antifungal effects and the underlying molecular mechanisms of its active subfraction against dermatophytes. The results indicated that AAWE exhibited excellent antifungal effects against the three aforementioned dermatophyte species. The subfraction AAWE6, isolated using D101 macroporous resin, emerged as the most potent subfraction. The minimum inhibitory concentrations (MICs) of AAWE6 against T. rubrum, M. gypseum, and T. mentagrophytes were 312.5, 312.5, and 625 μg·mL-1, respectively. Transmission electron microscopy (TEM) results and assays of enzymes linked to cell wall integrity and cell membrane function indicated that AAWE6 could penetrate the external protective barrier of T. rubrum, creating breaches ("small holes"), and disrupt the internal mitochondrial structure ("granary"). Furthermore, transcriptome data, quantitative real-time PCR (RT-qPCR), and biochemical assays corroborated the severe disruption of mitochondrial function, evidenced by inhibited tricarboxylic acid (TCA) cycle and energy metabolism. Additionally, chemical characterization and molecular docking analyses identified flavonoids, primarily eupatilin (131.16 ± 4.52 mg·g-1) and jaceosidin (4.17 ± 0.18 mg·g-1), as the active components of AAWE6. In conclusion, the subfraction AAWE6 from A. argyi exerts antifungal effects against dermatophytes by disrupting mitochondrial morphology and function. This research validates the traditional use of A. argyi and provides scientific support for its anti-dermatophytic applications, as recognized in the Chinese patent (No. ZL202111161301.9).
