1.Effect of three chemical molecules on adhesion molecules expression in HMECs induced by activated complement alternative pathway

Aim To investigate the effect of resveratrol ( Res) , PDTC and AG490 on adhesion molecules ex-pression induced by product of activated complement alternative pathway on human microvascular endothelial cells ( HMECs) and the possible mechanisms. Meth-ods HMECs were exposed to the product of comple-ment alternative pathway activation, then the superna-tant was removed to detect the concentration of malond-ialdehyde ( MDA ) with TBA method. ELISA method was used to detect the expression of soluble ICAM-1 , VCAM-1 ( and E-selectin) in the culture supernatant. Res, PDTC and AG490 with different concentrations were used to determine their effect on cell oxidation level and adhesion molecules expression. The phospho-rylation of NF-κB p65 was detected by Western blot, and the intervention of Res, PDTC and AG490 was as-sayed by the same way. Results The activation of complements alternative pathway resulted in the phos-phorylation of NF-κB p65 , and increased the concen-tration of MDA and up-regulated the expression of ICAM-1, VCAM-1 and E-selectin. Res reduced the concentration of MDA. Res, PDTC and AG490 inhibi-ted the phosphorylation of NF-κB p65 . Res and PDTC showed similar inhibition on expression of ICAM-1 and VCAM-1 , while exhibiting little effect on expression of E-selectin, and AG490 significantly inhibited the ex-pression of the above adhesion molecules. Conclusions Res, PDTC and AG490 could inhibit the expression of adhesion molecules induced by activated complement alternative pathway, the inhibition of NF-κB pathway activation was involved in their mechanism, and JAK2 may be a more important intervention target in regula-ting adhesion molecule expression.
2.Expression of Caspase-3 in Renal Glomerulus Cells in Rats with Sub-chronic Arsenic Poisoning

Objective To investigate the influence of caspase-3 on the apoptosis of glomerulus cells in chronic arsenic poisoning. Methods Sprague-Dawley(SD) rats in cleanliness grade were randomly divided into high-dose group,low-dose group and control group,10 in each (5 males and 5 females). The rats in high and low groups were treated with As2O3 through drinking water,10 and 0.4 mg/(kg?d) respectively,for four months. The content of arsenic in the blood and urine was determined. The expression of caspase-3 in the glomerulus cells was detected by SABC immunohistochemistry and analyzed through image patterns. Results Compared with the control group,a higher arsenic content in the blood and urine,more positive cells of caspase-3 and lower OD value the glomerulus cells were found in both of high-dose and low-dose groups. Compared with low-dose group,a higher arsenic content in the blood and urine,more positive cells of caspase-3 and the lower value of grey degree in the renal glomerulus cells were all found in high-dose group. Conclusion The obvious increase of caspase-3 in the glomerulus cells may play a role in the apoptosis of the glomerulus cells induced by chronic arsenic poisoning.
3.Effect of Chronic Arsenism on Ultra-structure of Rats' Hippocampus CA3

Objective To study effect of chronic arsenism on ultra-structure of rats’ hippocampus CA3. Methods 50 SD rats were randomly divided into 2 groups(25 rats in each group):the control group and the arsenic poisoning group. The control group drank distilled water. The arsenic poisoning group drank distilled water containing 100 mg/L AS2O3. Both groups were fed on common feed. All rats were killed after 4 months and the hippocampus tissue was observed by optic and electron microscope. Results By optic microscope,the pyramidal cells at the hippocampus CA3 region of the control group were dense and orderly. The cells bodies were pyramidal and clear,cytoplasm were abundant with Nissl bodies. While the pyramidal cells of the arsenic poisoning group were fewer and scattered unorderly and the pyramidal cell form was irregular. 4.Determination of Total Saponins in Jiu-Bi-Ying Yi-Xin Tablet with Ultraviolet Spectrophotometry

This study was aimed to establish the determination method of total saponins content in Jiu-Bi-Y ing Y i-Xin (JBYYX) tablet. The content of total saponins in JBYYX tablet was determined by ultraviolet spectrophotometry. The results showed that in the range of 2.16í10-5-6.47í10-5 mg·mL-1, the total saponins had good linear relation-ship. The average recycle rate was 98.81%. RSD was 2.06%(n=6). It was concluded that the method was accurate, reliable and repeatable, which was suitable for determining the content of total saponins in JBYYX tablet.
5.Differentiation of marrow derived mesenchymal stem cells into neurons by noggin gene transfected with recombinant adenovirus vector

Objective To explore the appropriate method of isolating,purifying and culturing the rat bone marrow mesenchymal stem cells(MSCs) in vitro,and observe the differentiation of MSCs into neuron-like cells induced by noggin gene transfected with adenovirus vector.Methods MSCs obtained from bone marrow of SD rats were isolated,cultivated and amplified by Percoll density gradient centrifugation with adherent method,the the third passage of purified MSCs was then induced to differentiate into neurocytes by using the reconstructed noggin adenovirus vector(pAdEasy-1-GFP-noggin) and control vector(pAdEasy-1-GFP),respectively.After cultivation,the differentiated cells were identified by using immunocytochemical method with neurone-specific enolase(NSE),neurofilament 200(NF-200),neuronal nuclei(NeuN) and glial fibrillary acidic protein(GFAP),and the inductivity in the respective groups were analyzed.The Nissl bodies in the induced cells were displayed by thionine-eosin staining.Results The primarily cultured MSCs were spindle in shape,adhered 24 hours after cultivation,and then grew into small clones.Forty-eight hours after transfection by noggin recombinant adenovirus vector,the MSCs started to change their shape as observed under inverted microscope,and several axon-or dendrite-like processes with branches stretched out from the cell body.The induced cells derived from bone marrow MSCs specifically expressed NSE,NF and NeuN,but not GFAP by immunocytochemistry.A lot of Nissl bodies could be seen in the cell body of induced cells shown by Nissl staining.Conclusions Highly purified MSCs can be obtained by Percoll density gradient centrifugation combined with adherent method.The bone marrow derived MSCs transfected with noggin gene can differentiate into neuron-like cells in vitro.
6.Optimization on Extraction Process of Ginseng in Shen-Qi Bu-Qi Granules by Orthogonal Design

This study was aimed to optimize the extraction process of ginseng in Shen-Qi Bu-Qi (SQBQ) granules. The orthogonal experiment method was used to optimize the extraction process of ginseng adopting ethanol concentration, ethanol volume, extraction time and extraction times as factors. The content of ginsenoside Rg1, Re, Rb1 content in the extract was determined by HPLC. And the optimum extraction conditions were determined with indexes of three kinds of saponin yield. The results showed that the optimum extraction process of ginseng for SQBZ granules was that adding 6 times amount of 60% ethanol technology for medicinal materials, extract for 2 times, and 3 h for each time. It was concluded that the optimization of ginseng extraction process was stable, reasonable and feasible with high extraction rate.
7.Laparoscopic-assisted transanal Soave pull-through procedure for Hirschsprung's disease and allied disorders

ObjectiveTo explore the feasibility and outcomes of natural orifice transanal laparoscopic Soave procedure for Hirschsprung's disease and allied disorders (HAD).MethodsFrom March 2010 to December 2011,31 cases (at the age from 3 mos to 6 yrs) with Hirschsprung's disease or allied disorders (5 cases)underwent laparoscopic-assisted Soave pull-through procedure at two tertiary hospitals.We modified this technique by mobilizing the left hemicolon or whole colon via rectal muscular sleeve approach under transanal or transumbilical laparoscopic vision,then endorectal pull-through to completearectosigmoidectomyorsubtotalcolectomy. ResultsAllprocedureswerecompleted successfully.A rectosigmoidectomy was performed in 16 cases with classic HD and subtotal colectomy in 15 cases with extended HD and HAD.The average operative time was ( 117 ± 13) min.The length of the resected segment was 35 -80 cm,and the estimated blood loss was 5 -20 ml. One infant developed postoperative intestinal obstruction that required open exploration.Follow-up of one to 20 mos found no stoma stenosis or constipation recurrence. Enterocolitis developed in 1patient.ConclusionsTransanal or transumbilical laparoscopic-assisted Soave pull-through surgery is safe,effective and with a benefit of much less invasion and almost invisible scars.
9.A prognostic model for predicting extracorporeal circuit clotting in patients with continuous renal replacement therapy.

OBJECTIVETo establish a prognostic model for predicting extracorporeal circulation clotting in patients with continuous renal replacement therapy (CRRT).
METHODS425 patients with CRRT were involved in the study. We built a predictive risk model of extracorporeal blood clotting with the 302 participants, and 103 participants were used to validate the model. The primary endpoint of CRRT was extracorporeal circulation pipe blockage.
RESULTSWe used a score of 0-5 point evaluating system to predict the risk of 24 hours CRRT integral model of cardiopulmonary bypass clogging. The area under the CRRT predictive model of cardiopulmonary bypass clogging integral system ROC curve was 0.790 (95% CI 0.719-0.826) (P<0.001). The evaluating system can determine the blockage of 24 hours CRRT extracorporeal circulation. The results showed that CRRT extracorporeal plugging prediction fitted the integral model and could predict the chance of plugging. The actual plugging rate showed no significant difference from the predicted rate (R² = 0.301, P=0.232). The cardiopulmonary pipe survival time between the 3 groups(low risk, intermediate risk, and high risk) showed a significant difference (P<0.05).
CONCLUSIONWe established a continuity extracorporeal blood purification plugging risk score model, to predict plugging risks during CRRT treatment.

10.Clinical observation of ureteroscopy and minimally invasive percutaneous nephrolithotomy in the treatment of ;124 urinary calculi patients with acute renal dysfunction

Objective To explore the clinical value of ureteroscopy and minimally invasive percutaneous nephrolithotomy ( MPCNL) in the treatment of ureteral calculi patients with acute renal dysfunction .Methods Clini-cal data of 124 ureteral calculi patients with acute renal dysfunction were retrospectively analyzed .86 cases were trea-ted with holmium-laser lithotripsy under ureteroscope .38 cases were treated with MPCNL under the guide of ultra-sound.Results Three months after operation ,the stone clearance rate was 100%,and no severe complications were observed.The renal function decreased to normal levels in 102 cases(82.3%).Conclusion The holmium laser lith-otripsy under ureteroscope and MPCNL can deal with double sites of ureteral calculi ,and offer advantages with less in-vasion,safety and efficiency ,which can be the first choice for the ureteral calculi combined with acute renal dysfunc -tion.
