1.Effect of human umbilical cord WJ-MHCs on TNF-α and NT-proBNP in the rats with heart failure of acute myocardial infarction

Objective To observe the influence of human umbilical cord wharton′s jelly‐mesenchymal stem cells(WJ‐MHCs) on the tumor necrosis factorα (TNF‐α) and N‐terminal pro‐brain natriuretic peptide(NT‐proBNP) in rats with heart failure of a‐cute myocardial infarction .Methods Totally 80 male rat models of heart failure of acute myocardial infarction were made by isopre‐naline(ISO) 200 mg/kg injected subcutaneously twice at an interval of 24 hours .After one week ,24 survival rats were randomly di‐vided into WJ‐M HCs transplantation group and normal control group .Sham group was made of 12 health rats ,and then each of the three groups was subdivided into pre‐transplantation group and post‐transplantation group 4 weeks later .WJ‐MHCs transplantation group was transplanted with WJ‐MHCs with DAPI labeled after ISO injected one week .Sham group and normal group were un‐treated and normally bred .The left ventricular ejection fraction(LVEF) measured by before transplantation and post‐transplantation 4 weeks later .The injected cells and the expression of TNF‐αwas measured .Results Compared to pre‐transplantation group ,WJ‐M HCs transplantation group increased the LVEF(P<0 .05);compared to pre‐transplantation and normal control ,WJ‐M HCs trans‐plantation group reduced the TNF‐αand NT‐proBNP in the serum(P<0 .05)and the expression of TNF‐α from the heart tissue (P<0 .05);compared to normal transplantation ,WJ‐M HCs transplantation group reduced the mortality from 33 .3% to 16 .7% ;immunofluorescence demonstrated that transplanted cells were still found alive in the heart after transplantation 4 weeks later .Con‐clusion Transplantation of WJ‐MHCS down‐regulates TNF‐α and NT‐proBNP in the serum in the serum and the expression of TNF‐αfrom the heart tissue and up‐regulates the LVEF in rats with heart failure of acute myocardial infarction .
2.The correlation between hepatitis B virus DNA replication and large envelope protein in serum

Objective To study the significance of serum hepatitis B virus large envelope protein (HBV-LP),HBV DNA and HBeAg in the diagnosis of viral replication.Methods In 623 chronic HBV patients,HBV-LP and HBeAg were detected by ELISA and HBV DNA was quantitaively detected by realtime polymerase chain reaction(PCR).Results The positive rate of HBV-LP and HBV DNA were higher than that of HBeAg in all chronic HBV patients(x2 =22.3 and 9.4,P <0.01),and the positive rate of HBVLP and HBV DNA was no significant difference (x2 =2.8,P > 0.05),The positive rate of HBV-LP and HBV DNA detected in HBeAg(+) or HBeAg(-) were not significantly different(x2 =3.2 and 0.6,P > 0.05).HBV DNA copies and A vaule of HBV-LP was a positive correlation(r =0.874,P <0.01),HBV DNA copies of different groups was significantly different from HBV-LP A values(F =6.987,P <0.01).In 100 healthy people,the detecting results of HBV DNA and HBV-LP are negetive.Conclusion HBV-LP was better than HBeAg for detection of HBV replication.There is a good correlation between the copies of HBV DNA and the levels of HBV-LP,HBV-LP expression can reflect the replication of HBV.
3.Application of dual fluorescence laparoscopy in the repair of complex ureteral stricture with lingual mucosa graft

Objective: To evaluate the feasibility and effectiveness of dual fluorescence laparoscopy in the localization of ureteral stricture and its blood supply,and to provide a new idea for the treatment of complex ureteral stenosis,thus helping doctors to improve the efficiency of ureteral reconstruction surgery. Methods: Our team developed a dual fluorescence laparoscopic system,which could simultaneously identify the ureter stricture by intra-ureteral injection of methylene blue (MB) and assess the blood supply of the ureteral stumps by intravenous injection of indocyanine green (ICG). Results: The clinical data of 3 patients who underwent lingual mucosa ureteroplasty using dual fluorescence laparoscopy in Zhongnan Hospital of Wuhan University were retrospectively analyzed.All operations were successful,without conversion to open surgery.The operation time was 144,132 and 163 minutes,respectively.The length of harvested lingual mucosa graft was 2.0,2.8 and 3.5 cm,respectively.No intraoperative or postoperative complications occurred.Eight weeks after operation,ureterography showed that the ureter was unobstructed. Conclusion: Dual fluorescence laparoscopy is safe and feasible in the repair of complex ureteral stricture with lingual mucosa graft,which provides a new idea for complex ureteral reconstruction.
