1.Study of MRI findings and clinicopathological characteristics of DPT (report of 5 cases and literature review)

Objective To explore the characteristics of MRI images and pathological features of demyelinating pseudotumor (DPT).Methods Five patients with demyelinating pseudotumor were histologically confirmed.Their MR imaging were re-viewed and the enhancement patterns were analyzed.Results Among the five cases with demyelinating pseudotumor,single le-sion was found in 4 cases,and multiple lesions in 1 case.Most lesions had a clear boundary and these space occupying lesions were irregular (3 cases).The signal intensity on T1 WI was lower than edema,and it had a specific “core and halo”feature,du-al-high signal with different intensity on T2 WI,slightly higher signal intensity on DWI,and on enhanced MRI,nodular or patch-y enhancement and open ring enhancement with edema around the lesions were observed.The pathological results showed that the demyelination occurred but the neuraxon survived,lymphocytes were infiltrated,glial cells were proliferated,megakaryo-cytes also could be found,and multiple monocyte-macrophages were all around the lesions.Conclusion The MRI of DPT has some characteristic features.
2.Current LC-MS-based strategies for characterization and quantification of antibody-drug conjugates

The past few years have witnessed enormous progresses in the development of antibody-drug conju-gates (ADCs). Consequently, comprehensive analysis of ADCs in biological systems is critical in sup-porting discovery, development and evaluation of these agents. Liquid chromatography-mass spectrometry (LC-MS) has emerged as a promising and versatile tool for ADC analysis across a wide range of scenarios, owing to its multiplexing ability, rapid method development, as well as the capability of analyzing a variety of targets ranging from small-molecule payloads to the intact protein with a high, molecular resolution. However, despite this tremendous potential, challenges persist due to the high complexity in both the ADC molecules and the related biological systems. This review summarizes the up-to-date LC-MS-based strategies in ADC analysis and discusses the challenges and opportunities in this rapidly-evolving field.
4.Epidemiological analysis of imported malaria in Yancheng City from 2011 to 2015

Objective To understand the epidemiological trend of imported malaria and its monitoring and control effect in Yancheng City from 2011 to 2015,so as to provide the evidence for adjusting the prevention and control strategy. Methods The data of malaria surveillance,epidemic,prevention and control were collected and analyzed in the districts and counties of Yancheng City from 2011 to 2015. Results From 2011 to 2015,there were 104 imported malaria cases reported,and the most cases(52 cases,50%)were reported from Jianhu County. Most of the cases were males(only 2 females),the average age was 38.8 years,and the occupation was mainly labor service worker abroad(94 cases,97%). The infection source of these cases mainly came from Africa. From 2011 to 2015,151980 fever patients received malaria blood tests,and the positive rate was 0.07%. Falciparum malaria cases were the most(90 cases,86.5%). The confirmed diagnostic rate of malaria within 24 hours in-creased year by year. All the 104 patients received the standardized treatment. Conclusion There are no local malaria cases in Yancheng City from 2011 to2015,and in the next stage,the monitoring and prevention should be focused on imported malaria, including shortening the confirmed diagnostic time and improving the diagnostic efficiency.

Cerebral ischemia is due to cerebral blood supply disorders caused by ischemia and hypoxia resulting in localized ischemic brain necrosis or brain softening of the disease, leading to irreversible brain damage and subsequent loss of neuronal function is a serious threat to human health One of several diseases. For patients with cerebral ischemia, often the lack of effective and extensive treatment. In addition, cerebral ischemia with morbidity, morbidity and mortality are characterized by high, so by the medical profession at home and abroad attention. As a traditional Chinese medicine, cordyceps sinensis (CS) is a complex of ergot fungus, which is parasitized on the larvae of the bat-moth family. The compound is composed of cordycepin, cordyceps polysaccharide, cordyceps sinensis peptides, ergosterol, mannitol, fatty acids and trace elements such as a variety of ingredients, with a wide range of pharmacological effects. Over the years, domestic and foreign scholars on the pharmacological effects of cordyceps sinensis were more comprehensive study of its prevention and treatment of cerebral ischemia is also deepening, found that cordyceps sinensis on cerebral ischemia with anti-inflammatory, reduce oxidative damage and neuronal ischemia damage, reduce neuronal apoptosis, improve memory cognition, reduce thrombosis, inhibit NO production, enhance mitochondrial energy supply, scavenging free radicals and other prevention and treatment. But no relevant review. In this paper, the domestic and foreign literatures on the prevention and treatment of cerebral ischemia by cordyceps sinensis were summarized, analyzed and summarized in order to provide useful information for the research and further development of cordyceps sinensis.
6.Effect of JAK2/STAT3/vimentin signaling pathway on proliferation and migration of human colon cancer cells.

OBJECTIVETo investigate the regulatory role of JAK2/STAT3/vimentin signaling pathway on the proliferation and migration of human colon cancer cells.
METHODSThe human colon cancer cell Lovo was treated with Janus kinase inhibitor AG490. The proliferation of Lovo cells was quantified by MTT assay. The migration of Lovo cells was measured with scratch assay. The intracellular phosphorylation STAT3 (P-STAT3) and vimentin protein was detected by Western blot and immunofluorescence.
RESULTSAfter AG490 treatment, the proliferative ability of Lovo cells decreased as compared with control group (P<0.05). This suppression was dose-independent and time-independent. At 24 hrs after scratching, scratch width in the AG490 group recovered to 20%, lower than that in the control group (60%, P<0.05). After AG490 treatment, the expression of P-STAT3 and vimentin in Lovo cells decreased significantly (P<0.05).
CONCLUSIONJAK2/STAT3/vimentin signaling pathway participates in regulating the proliferation and migration of human colon cancer cells.

7.Effect of free hemoglobin on hemorrhagic shock in pigs: TNF-alpha, IL-6 expressions in serum and rates of MODS after the blood transfusion.

This study was aimed to investigate the expressions of tumor necrosis factor-alpha (TNF-alpha), Interleukin-6 (IL-6) in serum and the incidence of multiple organ dysfunction syndrome (MODS) in pigs with hemorrhagic shock after the blood transfusion simultaneously combined with different doses of free hemoglobin (FHb) so as to provide guidance of banked blood with high concentration of FHb during war injury through understanding effect of FHb on the animals. The different doses of FHb were given intravenously during the recovery of pig from shock, the vital signs and functional changes of vital organs were monitored and the incidence of MODS was determined, as well as the serum specimens were collected and the TNF-alpha, IL-6 levels in serum were detected by ELISA. The results showed that there were statistical differences of serum levels of TNF-alpha and IL-6 in pigs after FHb 10 mg/kg infusion, as compared to shock control group. There was significantly difference of the serum levels of TNF-alpha, IL-6 after FHb 15 mg/kg infusion, compared to the control group. The incidence of MODS increased significantly. It is concluded that the blood infusion containing high dose (more than 10 mg/kg) of FHb influences significantly on the cytokines in pigs with hemorrhagic shock, and increases damage of cytokines to vital organs and the incidence of MODS. The tolerance dose of the pigs to free hemoglobin is about 10 mg/kg or so. The infusion of blood with less than 10 mg/kg is relatively safe for pig in hemorrhagic shock.

8.Advance in studies on chemical constitutions, pharmacological mechanism and pharmacokinetic profile of dalbergiae odoriferae lignum.

Dalbergiae Odoriferae Lignum is a traditional Chinese medicine (TCM) commonly used for promoting blood circulation, relieving pain and removing blood stasis. Volatile oil and flavonoid compounds are two main chemical constituents of Dalbergiae Odoriferae Lignum. Modern pharmacological studies show that Dalbergiae Odoriferae Lignum has many effects, such as relaxing blood, increasing blood flow of coronary, anti-oxidation, anti-inflammation and antitumor. Dalbergiae Odoriferae Lignum, as a characteristic TCM with the potential of further development, is generally compatible with other TCMs to treat cardio-cerebral vascular diseases. This article summarizes studies on chemical composition, pharmacological action, pharmacokinetic procfile in vivo and TCM compatibility in recent years, in order to provide references for further studies.

9.Effect of HMGB1-siRNA on proliferation and apoptosis of HepG2 cells.

OBJECTIVETo investigate the effect of decreased expression of high mobility group Box-1 on the proliferation and apoptosis of HepG2 cells.
METHODSThree specific siRNAs of HMGB1 were designed and synthesized, and were transiently transfected into HepG2 cells by Lipofectamine 2000. The HMGB1 expression in HepG2 cells was detected by RT-PCR and Western blotting respectively. The proliferation activity in vitro was assessed by MTT assay. In situ apoptosis was evaluated by terminal deoxynucleotidyl transferase-deoxyuridine triphosphate nick end labeling (TUNEL) assay.
RESULTSAll of these specific HMGB1-siRNAs (1, 2, 3) efficiently and specifically inhibited the expression of the HMGB1 gene, and the levels of HMGB1 mRNA were 1.147+/-0.024, 1.014+/-0.042, 0.435+/-0.055, respectively, in HMGB1-siRNAs transfection group, which were significantly lower than that in Lipofectamine 2000 alone group (1.411+/-0.065, P < 0.01). Correspondingly, all of these specific HMGB1-siRNAs (1, 2, 3) could efficiently and specifically inhibit the expression of the HMGB1 protein, and the levels of HMGB1 protein were 0.369+/-0.035, 0.340+/-0.028, 0.097+/-0.020, respectively, in HMGB1-siRNAs transfection group, which were significantly lower than that in Lipofectamine 2000 alone group (0.553+/-0.051, P < 0.01). Of the 3 specific HMGB1-siRNAs, HMGB1-siRNA-3 (siRNAH3) had the highest inhibition rate (80%). The proliferation of HepG2 cells was markedly inhibited by siRNAH3 transfection. Compared to mock-transfection, siRNAH3 transfection dramatically suppressed the proliferation of HepG2 cells (P < 0.01). Moreover, siRNAH3 can induce apoptosis (P < 0.01).
CONCLUSIONsiRNA targeting HMGB1 mRNA can specifically reduce HMGB1 gene and protein expression. siRNAH3 can effectively suppress the proliferation and induce apoptosis of HepG2 cells.

10.Effects of TGF-β2 on mechanical behavior of posterior scleral flbroblasts in experimental myopia

Background There have been an abundance of literature on the analysis of the mechanical characteristic of the sclera at the entire seleral level in high myopia.However,some recent studies on high myopia are focused on the mechanical changes of the sclera on a cellular level. Objective This experiment was purposed to study how transforming growth factor-β2(TGF-β2)affected sclerotic desmocytes and the mechanical behaviors of scleral fibroblasts in the posterior part of the eyes in guinea pigs with experimental myopia. Methods Induced myopic animal models were established by wearing-10.00 D concave lens for 30 days in lateral eyes of 2-week-old guinea pigs.The fellow eyes were used as control group.Another 5 matched animals served as normal controls.The scleral fibroblasts of each group were purified with the tissue explant method and passaged for 2 generations in vitro.Cultured cells were identified by immunochemistry with vimentin,desmin,keratin and S-100 antibodies.Different concentrations of TGF-β2(0,1,10,100mg/L)were added into serum-free DMEM for 24 hours,and the viscoelastic properties of scleral fibroblasts were measured by micropipette aspiration technique. Results Compared with the fellow,eyes and normal control eyes,the equilibrium modulus and apparent viscosity in model eyes were significantly higher(P<0.05).After treatment of TGF-β2,the equilibrium modulus and apparent viscosity in the model group and fellow eyes were positively correlated with the concentrations of TGF-β2(r=0.743,r=0.533,r=0.654,r=0.576,P<0.05).Following the addition of 1 mg/L TGF-β2 and 10 mg/L TGF-β2,the equilibrium modulus and apparent viscosity of scleral fibroblasts were significantly reduced in model eyes compared with fellow eyes(P<0.05).No significant difference was found in the equilibrium modulus and apparent viscosity of scleral fibroblasts between model eyes and fellow eyes after treatment with 100 mg/L TGF-β2(P>0.05). Conclusion TGF-β2 car increase the mechanic indexes in a concentration.dependent manner.1 mg/L,10 mg/L TGF-β2 can lower the equilibrium modulus and apparent viscosity of scleral fibroblasts of normal eye and thus cause more changes in the mechanical behavior of scleral fibroblasts.
