Objective To investigate the mechanism of Smad ubiquitination-related factor 2 (Smurf2)neddylation. Methods The Smurf2 protein level was tested by overexpression of Nedd8,while the method of immunoprecipitation(IP) and Western blotting were used to analyz Smurf2-Nedd8 modification. The GST-pulldown experiment was conducted to prove protein interactions. The protein was obtained by grinding mouse tissue and Western blotting was used to test the protein expression level. Results Over expression of Nedd8 could lead to the down regulation of the Smurf2′s protein level.Smurf1 and Smurf2 could interact in the GST-pulldown experiment. Smurf1 could enhance Smurf2-Nedd8 modification. The Smurf2′s protein level was up-regulated in mouse tissue of Smurf1 C426A.Conclusion There is some relationship and also difference between Smurf1 and Smurf2.Smurf1 can enhance Smurf2-Nedd8 modification.

Objective To establish experimental radioactive wound model in miniature pig and observe its pathological changes in order to carry out in vivo study of radioactive wound-related diseases. Methods According to the random number table, eight adult Bama miniature pigs were divided into four groups after being anesthetized:normal control and experiment groups exposed to 0,10,20 and 30 Gy respectively using the X-ray linear accelerator. Then a puncher was use to prepare full-thickness skin defects (4 cm in length) in the irradiation site.Wound samples were collected every other week to observe wound healing.RT-PCR was used to measure expressions of tumor necrosis factor (TNF)-α and transforming growth factor (TGF)-β in wound tissues 35 days after irradiation. At the same time,Western blot was utilized to detect expressions of CD31 and collagen Ⅰ.HE staining was applied to observe histopathological changes of the radioactive wound. Results Skin wounds in normal control group healed in 35 days.Whereas in 20 and 30 Gy experiment groups, the wounds were not healed in 63 days and the healing rate was gradually weakened with the irradiation dose increasing.Expression ratio of TNF-α mRNA was 0.315 ±0.007 in normal control group,0.984 ±0.211 in 10 Gy experiment group,1.140 ±0.088 in 20 Gy experiment group, and 0.471 ±0.030 in 30 Gy experiment group.Expression ratio of TGF-β mRNA was 0.402 ± 0.018 in normal control group,1.707 ± 0.147 in 10 Gy experiment group,1.933 ± 0.078 in 20 Gy experiment group, and 0.968 ± 0.084 in 30 Gy experiment group.Expressions of TNF-α and TGF-β mRNA were significantly increased in 20 Gy experiment group compared to normal control group (P < 0.05).Expressions of CD 31 and Collagen Ⅰ protein were inhibited in 20 Gy experiment group compared to normal control group (P<0.05).Epithelialization and granulation tissue regeneration in 20 Gy experiment group were impaired. Conclusion An irradiation wound model in Bama miniature pigs can be efficiently and effectively simulated using the 20 Gy X-ray irradiation combined with full-thickness skin defect, and can be used to study the pathophysiology and therapeutic intervention of radioactive wound-related diseases in vivo.

Journal of Applied Clinical Pediatrics 2006;0(14):-
Objective To explore the perioperative experiences of congenital heart disease in infants. Methods From Jan.2000 to Aug.2006,109 patients with congenital heart disease were operated in our department, their clinical data were retrospectively collected and analyzed. The patients′ age ranged from 31 days to 3 years old (13.6 months). The body weight ranged from 2.1 to 16 kg (8.6 kg).Ninety-three patients were operated under hypothermic anaesthesia with cardiopulmonary bypass(CPB).Sixteen patients underwent deep thermal and low flow CPB.Ultrafiltration was used in 62 patients. Results There were 8 deaths and the operative mortality was 7.3%,4 cases caused by low output syndromeclos(LOS),3 cases caused by pulmonary hypertension and 1 case caused by lung intection. The morbidity was in 25 cases(22.9%),the main complications were LOS in 6 patients and respiratory complications in 18 patients, hydropericardium in 1 case, respectively. Conclusion To improve the operative and CPB technique, and to improve the skills of the postoperative managements of LOS and respiratory complications are the main points in the success of the cardiac operation in infants.

Mechanosensitive ion channels are mechanoreceptors that convert mechanical stimulation on the cell surface into intracellular electrical signal or chemical signal. In recent years, Piezo protein as a mechanically activated cation channel in eukaryotes has gradually become a research hotspot. Piezo1 and Piezo2 proteins are expressed in different organs and tissues of mammals, and tissues such as bones and joints are directly related to mechanical conduction. The intracellular effects caused by the conduction of mechanical force play a very important role in the treatment and prevention of orthopedic diseases. In this paper, the researches on Piezo protein mechanically activated cation channels in bone tissue, cartilage tissue, bone marrow tissue, intervertebral disc tissue and orthopedic diseases were reviewed.

To analysis the differences between Tripterygium wilfordii and T. hypoglaucum, specimens of their leaves were collected from five production regions and analyzed by ultra performance liquid chromatography coupled with quadrupole time of flight mass spectrometry (UPLC-Q-TOF-MS). The data were analyzed by multivariate statistical method, such as hierarchical cluster analysis (HCA) principal component analysis (PCA) and orthogonal signal correction partial least square discrimination (OPLS-DA). Potential markers with VIP values above 5.0 and corresponding r values above 0.85, were selected and further tested by combining mann-Whitney nonparametric. Those with P < 0.001 and AUC = 1 were confirmed as metabolite markers to discriminate them from each other. Results revealed that the two species were obviously different in their leaf metabolites. Based on their mass spectra, 23 potential metabolite markers were identified to distinguish T. wilfordii from T. hypoglaucum.
