1.Simultaneous Quantitative Analysis of Contents of Salidroside, Loganin and Polydatin in Buxin Ruanmai Granules by Multi-Components Assay with Single Marker

Objective To develop a method of quantitative analysis of multi-components with single marker (QAMS) for simultaneous determination of Salidroside, Loganin and Polydatin;To validate its accuracy and feasibility in Buxin Ruanmai Granules.Methods Salidroside was used as the internal reference substance, and the content of Salidroside in Buxin Ruanmai granules was determined by external standard method, with a purpose to establish its relative correction factor with Loganin and Polydatin. The contents of Loganin and Polydatin were calculated by the relative correction factor, and then multi-components assay with single marker was realized. The three contents in Buxin Ruanmai Granules were determined by both QAMS and external standard method, and their results were also compared.Results The results of 10 batches of samples by two methods did not show significant difference, which showed that relative correction factors had good reproducibility and credits.Conclusion The QAMS method can be used to quality evaluation with multiple indicators of Buxin Ruanmai Granules.

AngiotensinⅡ( AngⅡ) is the main functional bioac-tive peptide of the renin angiotensin system,and its basic function is to regulate salt-water homeostasis and blood pressure. Howev-er,recent studies have shown that AngⅡ plays a very important role in tumor formation and angiogenesis by acting on its type 1 receptor (AT1R) as a kind of potential growth factor. This arti-cle is a brief overview of the research on effects of AngⅡ-AT1R system in the process of tumor angiogenesis in recent years.
3.The study of the biodegradable biliary duct stent in vivo.

The present paper is aimed to evaluate safety and efficiency of a biodegradable biliary stent used in the Roux-en-Y cholangiojejunostomy of dogs in vivo. We separated 18 hybrid dogs into control group and experimental group randomly, with each group 9 dogs. The Roux-en-Y cholangiojejunostomy was carried out in the two groups. We placed the biodegradable stents into the anastomotic stoma of the dogs in the experimental group during the operation. After 3 months' close observation, we evaluated the degradable property and biocompatibility of the stents. We found that the morbidity rate of bile leakage in the dogs in the control group was much higher than that in the experimental group (P > 0.05). The X-ray photograph showed that the figures of the stents were complete and the stents were fixed at right place throughout the experimental period. The diameter of the dogs' common bile ducts of control group narrowed down obviously compared to those in the experimental group (P < 0.05). There were no impure materials adhered to the inside walls of the stents. The pathological test showed that there were no inflammation signs in mucous membrane and abnormal epithelium hyperplasia of the bile tract and jejunum. It can be concluded that the biliary duct stent, which was made with a mixture of poly-L-lactic acid (PLLA) and poly lactide-co-glycolide (PLGA), possesses qualities of relatively long degradable period (> 3 months) and good biocompatibility. Moreover, using the stent can effectively prevent bile leakage and anastomotic stom stenosis in the cholangio-intestinal anastomosis from happening.

4.Experimental study on the anti-tumor effect of monocytes/macrophages against Tca8113 cells and the secretion of vascular endothelial growth factor in acid microenvironment.

OBJECTIVETo investigate the anti-tumor effect of monocytes/macrophages against Tca8113 cells and the secretion of vascular endothelial growth factor (VEGF) in acid microenvironment in vitro.
METHODSPeripheral blood mononuclear cells were extracted from healthy person's blood and cultivated to transform into monocytes/macrophages. The monocytes/macrophages were cultured with Tca8113 in acid microenvironment (pH6.6 and pH6.8) and in normal microenvironment (pH7.2). The anti-tumor effect of monocytes/macrophages against Tca8113 cells were examined by MTT assay. The expression of VEGF was detected by enzyme-link immunoassay (ELISA).
RESULTSThe anti-tumor effect of monocytes/macrophages against Tca8113 cells in acid environment was lower than in normal environment (P < 0.05). VEGF excreted by mononuclear/macrophage was significantly higher in acid microenvironment than in normal microenvironment.
CONCLUSIONDue to acid microenvironment inside tumor, the anti-tumor effect of monocytes/macrophages against tumor cell was decreased, but the secretion of VEGF was gradually increased. However, the function of monocytes/macrophages on anti-tumor need more research.

5.Application value of magnetic compression anastomosis in digestive tract reconstruction.

Magnetic compression anastomosis can compress tissues together and restore the continuity. Magnetic compression anastomosis mainly experienced three stages: magnetic ring, magnetic ring and column, and smart self-assembling magnets for endoscopy (SAMSEN). Nowadays, the magnetic compression anastomosis has been applied in vascular and different digestive tract surgeries, especially for complex surgery, such as anastomotic stenosis of biliary ducts after liver transplantation or congenital esophageal stenosis. Although only case reports are available at present, the advantages of the magnetic compression anastomosis includes lower cost, simplicity, individualization, good efficacy, safety, and minimally invasiveness. We are building a better technical platform to make magnetic compression anastomosis more advanced and popularized.

METHODWe made use of the methods of stratified sampling and random sampling to investigate 38 736 broth prescriptions of pediatrics of TCM in 21 various hospitals, 18 provinces, and constructed the database, then applied Weka KDD software to analyze.
RESULTWe identified the frequency and clinical dosage intervals of 300 drugs, and the practical dosage of 155 drugs. Most of drug's dosage centralized the four intervals: (3-6], (6-9], (9-12], (12-15], and the concrete dosage were 6, 9, 10, 12, 15 g, etc.
CONCLUSIONThe dosages in the pharmacopoeia are not coincided with the practical dosages in pediatrics, and demand to revise. This paper will offer the consummate data for formulating the new dosage standard.

8.Effects of rabbit limbs ischemia/ reperfusion on myocardial necrosis and apoptosis.

OBJECTIVETo investigate the effects of rabbit limbs ischemia/reperfusion on myocardial necrosis and apoptosis in vivo.
METHODSThirty-six healthy new zealand rabbits were randomly divided into 3 groups: (1) Sham group; (2) I/R(Ischemia/reperfusion) group; (3) RPostC (remote postconditioning) group. The activity of blood serum creatine kinase (CK) and lactate dehydrogenase (LDH) were measured at baseline, the end of ischemia after 60 min and 120 min of reperfusion respectively. The extent of myocardial ischemia and the scope of myocardial infarction were assessed by evans blue and Triphenyl tetrazolium chloride (TTC). The myocardial cell's apoptosis at the area of myocardial ischemia was estimated by Tunel. Protein expression of caspase-3, Bcl-2 and Bax in myocardial ischemic area were analyzed with the method of immunohistochemistry.
RESULTSCompared with I/R group, the myocardial infarct size and the CK activity were significantly reduced in RPostC group. The Tunel positive index of RPostC group in ischemic myocardium was significantly lower than that in I/R group (21.79% +/- 1.07% vs 35.81% +/- 1.10%, P < 0.05). Caspase-3 positive cells index was calculated with randomly selected five regions in each slide and then the positive cells in per hundred cells were calculated. The RPostC group of caspase-3 positive cells was significantly lower than that in I/ R group(25.03% +/- 1.16% as 39% +/- 2.43%, P < 0.05). Compared with the sham group, the Bax protein expression index and the Bcl-2 protein expression index of I/R group and RPostC group were increased. The Bax/Bcl-2 ratio of RPostC group decreased, while it was increased in I/R. Compared with the I/R group, the Bax protein expression and Bax/Bcl-2 ratio of RPostC group significantly reduced, but the expression index of Bcl-2 ratio was significantly increased, the differences were statistically significant.
CONCLUSIONLimbs ischemia/postconditioning could significantly reduce necrosis and apoptosis of ischemia/reperfusion myocardium. The mechanism of reducing the myocardial cell apoptosis may have relation to inhibiting the activation of pro-apoptotic gene caspase-3 and increased expression of Bcl-2.
