1.Effects of Sec-O-Glucosylhamaudol on expressions of nitric oxide and interleukin-6 in Lipopolysaccharide stimulated murine macrophage

Objective:To investigate the effect of Sec-O-Glucosylhamaudol on inflammatory mediator and cytokine( NO and IL-6)production in Lipopolysaccharide(LPS) stimulated murine macrophage(RAW264.7).Methods: Macrophages were induced with LPS,and incubated with different concentrations of Sec-O-Glucosylhamaudol(20,40,80 μg/ml),the quantity of NO production was measured by Griess reagent;the IL-6 production were measured by enzyme linked immunosorbent assay(ELISA),the expression of nitric oxide synthase( iNOS) in cells were detected by Western blot;the expression of iNOS and IL-6 mRNA were analyzed by real-time PCR.Results:Each concentrations of Sec-O-Glucosylhamaudol(20,40,80 μg/ml) inhibited the production of NO and IL-6 in LPS-stimulated RAW264.7 cells(P<0.01).This compound also reduced the mRNA expression of iNOS and IL-6.Conclusion:Sec-O-Glu-cosylhamaudol exhibited anti-inflammatory activity by inhibited the NO and IL-6 production in LPS stimulated RAW264.7 cells.
2.Construction and Characteristic of Human Multidrug-Resistant Hepatocellular Carcinoma Cell Line

Objective: To establish human multidrug-resistant hepatocellular carcinoma cell line(QGY7703/DDP) to analyze its different resistance and sensitivity. Methods: The human hepatocellular carcinoma cell line QGY7703 was continuously grown in the presence of cisplatin(DDP) for up to 6 months. Cell proliferation and cytotoxicity were serially monitored. Results: The chemosensitivities to 5-FU,DDP,adramycin (ADM) and mitomycin(MMC) were different between QGY7703 and QGY7703/DDP. The exhibitions were 1.533 fold to 5-FU,2.181 fold to DDP,7.08 fold to ADM, 9.461 fold to MMC in QGY7703/DDP. Conclusion: The drug resistance in cancer cell lines can be established by exposing to antitumor drugs in cell culture system.
3.Design of transfusion monitoring system and nursing flow optimization under field conditions

Objective To introduce transfusion monitoring system to optimize nursing flow under field conditions.Methods The organization and arrangement of nursing staffs were optimized with high-precision droplet sensor and transfusion monitoring system.A post of caring nurse was added to execute mental healthcare of the patient.Results Humanistic care was carried out by the caring nurse with daily nursing completed,and there's no need for additional nursing staff.Conclusion The transfusion monitoring system is rapid in deployment and withdrawal,optimizes nursing flow and enhances nursing quality,and thus is worthy promoting widely.
4.Clinical characteristics of retinal detachment after vitrectomy

?AIM:To investigate the causes and characteristics of retinal detachment after vitrectomy.?METHODS: Retrospective analysis of patients in our hospital in recent 4a who underwent primary, standard, three -port pars plana vitrectomy ( PPV ) . Retinal detachment occurred in 24 eyes ( 23 cases ) , 24-67 years old, average 49.0±12.0 years old.There were male in 17 cases, female in 6 cases, 6 right eyes, 18 left eyes.?RESULTS:Eighteen (75%) eyes without any symptoms were found during a routine check, 6 eyes ( 25%) with decreased visual acuity or in front of shadow.It took 1-223d from PPV to retinal detachment, average 40.7 ± 54.2d, of which retinal detachment occurred in 10 (42%) eyes within 1wk, 14 (58%) eyes within 1mo.The direct cause of retinal detachment was new retinal break ( a total of 27 holes ) in 19 eyes, proliferative vitreoretinopathy ( PVR) in 4 eyes, 1 eyes with primary hole not closed.All retinal detachment were ultimately completely reset.?CONCLUSION: The occurrence of new retinal breaks is the main cause of retinal detachment after vitrectomy, attention should be paid to the early postoperative peripheral retinal examination;after a timely surgery, the retina can be reset.
5.Advances in pathogenetic mechanisms of diabetic nephropathy

This review is intended to present the latest information on the pathogenesis of diabetic nephropathy based on the studies of recent years,involving potential therapeutic targets of diabetic nephropathy,such as hemodynamic dysfunctions,advanced glycation end-product,lipid metabolism disorders,inflammatory cytokines,and oxidative stress. Further studies are mandatory to investigate the effect of various interventions that modulate the pathogenesis in the treatment of diabetic nephropathy.
6.Scientific research management promotes the development of translational medicine

7.Study on the relationship between BBC3 expression and prognosis of patients with squamous cell lung neoplasms and its mechanism

Objective To explore the correlation between BCL2 binding component 3 (BBC3) expression and postoperative survival time of lung squamous cell carcinoma (LUSC),and investigate its molecular mechanism.Methods The expression of BBC3 in 39 patients was detected by using qRT-PCR assay.Meanwhile,clinical data of all patients were collected by follow-up visit.Then,the survival analysis was performed by using Kaplan-Meier and log rank tests.Moreover,multiple factors analysis was performed using COX proportional hazard model.At last,BBC3 was over-expressed in NCI-H226 cell lines,then detected the effects of BBC3 expression on cell proliferation and apoptosis by using MTT assay and flow cytometry.Results BBC3 expression were significantly correlated with the tumor metastasis (r=0.556,P=0.023),tumor size (r=0.532,P =0.042),T staging (r =0.551,P=0.021) and TNM staging (r=0.524,P=0.047).Meanwhile,the results of Kaplan-Meier and log rank tests found that BBC3 expression was significantly correlated with survival time of patients with LUSC,and the length of survival time in patients with high BBC3 expression was longer than that in patients with low BBC3 expression (x2 =7.542,P=0.006).The COX proportional hazard model indicated that tumor metastasis,T staging,TNM staging and BBC3 expression were independent factors which significantly affected survival time.Moreover,the proportion of apoptotic cells in the recombinant plasmid BBCs group was higher than that in the empty plasmid group and control group (P<0.05).Conclusion BBC3 expression could suppress the proliferation of tumor cells and promote apoptosis,and are significantly correlated with survival time of patients,so which may be assistant biomarkers for prognosis of LUSC.
8.Fusion and non-fusion fixation for cervical spondylotic myelopathy:which is more appropriate for maintaining range-of-motion and stability of vertebrae?

BACKGROUND:In recent years, many scholars adopted a joint non-fusion and fusion spinal fixation for multisegmental cervical spondylotic myelopathy, and achieved good clinical results. However, long-term clinical efficacy and possible related complications also require long-term fol ow-up of more in-depth study.
OBJECTIVE:To review the research and application progress of anterior fusion and non-fusion fixation surgical operation for cervical myelopathy.
METHODS:Computer-based search was conducted in China Journal Ful-text Database and PubMed database by the first author for articles related to anterior fusion and non-fusion fixation surgical operation for cervical myelopathy published between January 2004 and January 2014. The key words were“implant material;cervical spondylotic myelopathy;anterior;surgery;fusion;non-fusion;progress;hybrid;summary”in Chinese and“implant material;cervical spondylotic myelopathy;anterior;surgery/operation;fusion;non-fusion;research progress;hybrid;summary”in English. Final y, 35 articles were included for review.
RESULTS AND CONCLUSION:Fusion and non-fusion fixation methods for cervical spondylotic myelopathy had their own advantages. From the view point of indications, the non-fusion fixation was relatively limited. Currently, the fusion fixation was stil the main method in treatment of cervical myelopathy. Non-fusion method as an emerging fixation method also achieved good short-period results, but it needs a long fol ow-up study. Hybrid fixation integrates the advantages of both methods, and can reduce the pressure. Simultaneously, Hybrid fixation also can reduce fusion segments, and retain the range-of-motion of the cervical spine as much as possible. Hybrid fixation becomes a hot topic in recent studies. However, most current researches on Hybrid fixation are retrospective study, lack of control. The overal fol ow-up time is short. Thus, Hybrid fixation needs to be researched more deeply.
9.K-ras mutations in colorectal cancer at different stages

Objective:To study the alteration of K-ras mutations in different stages of colorectal cancer(CRC) and its influence on the progression of CRC. Methods:The 20 paraffin-embedded tissues, including primary foci, metastatic lymph nodes, remoter metastatic foci, colorectal adenoma, and normal colorectal tissues, were collected from 20 patients with colorectal cancer. The sequence of PCR-amplified products were analyzed. Results:The wild K-ras gene was expressed in normal colorectal tissues. The mutation frequency of K-ras gene was 20.0% (4/20) in colorectal adenoma, 30.0% (6/20) in primary foci, 25.0% (5/20) in metastatic lymph nodes, and 30% (6/20) in remote metastatic lesions. In the samples with K-ras mutations, the consistency of the types of K-ras mutations between primary foci and colorectal carcinoma, lymph node metastatic lesions, remote metastatic lesions was 0.0%(0/4), 40.0%(2/5), and 50.0%(3/6), respectively.Conclusion:The colorectal adenoma, metastatic lymph nodes and remote metastatic lesions were not suited for K-ras analysis as routine samples in clinical practice. If the samples of primary lesions were not available, the detection results of metastatic lymph nodes and remote metastatic remote lesions will provide some reference values. K-ras gene had several different mutations in the progression of CRC.
10.Imaging findings and clinical manifestations of pelvic lipomatosis:a report of 3 cases

The imaging findings of pelvic lipomatosis as confirmed by operation and pathology were examined in 1 case and two follow-up asymptomatic cases retrospectively analyzed.The imaging findings included a compressed and deformed bladder with a superior displacement.Its shape was like an inverted tear and pear in coronal view and a banana in sagittal view.Bilateral ureters were both compressed with a medial deviation.And bilateral ureters were dilated with hydronephrosis in 1 case.Rectum and sigmoid were both compressed and became narrowed.The clinical manifestations included frequent urination , urgent urination , urination pain, dysuria, constipation, nausea, vomit and fever in 1 case while another 2 cases stayed asymptomatic.
