2.Comparative Research on Middle School Student's Self-concept between Complete and Divorced Family

Objective: To compare the difference of self-concept between children of complete and divorced family. Methods: 42 subjects in divorced family and 45 controls were tested by Piers-Harris Self-concept Scale for Children. Rusults: The students in divorced family reported significantly lower scores on self-concept than the controls on behavior,anxiety, gregariousness, satisfaction and total score. Conclusion: The self-concept of divorced family students is lower than that of the control group.
3.Expressions of Rock2 and Wnt11 in esophageal cancer cell lines and their implications

Objective Early clinical symptoms of esophageal cancer are often not obvious , so a deeper insight into tumor markers is very important for the early diagnosis of esophageal cancer .This study was to investigate the expressions of Rock 2 and Wnt11 in the specific esophageal cancer cell lines Eca-109 and HEEC and the relationship of the signal transduction pathway of proteins with the development of esophageal cancer . Methods Esophageal cancer cell lines Eca-109 and HEEC were cultured and the expression levels of Rock2 and Wnt11 in the cell lines were determined by real-time quantitative PCR and Western blot . Results The mRNA expressions of Rock2 and Wnt11 were significantly increased in the Eca-109 as compared with those in the HEEC cell line (4.955± 0.539 vs 1.000±0.000, P<0.01;2.925±0.230 vs 1.000±0.000, P<0.01), and so were the protein expressions of Rock 2 and Wnt11 (955.000±21.628 vs 778.844±102.193, P<0.05;2175.316±145.623 vs 1312.233±50.734, P<0.05). Conclusion The up-regula-ted expressions of Rock 2 and Wint11 may be the markers of the metastasis of esophageal squamous cancer .
4.Epidemiological Survey of Anxiety Disorders in Urban and Rural Residents of Liaoning Province

Objective To describe the prevalence and distribution of anxiety disorders among people aged 18~65 yrs in Liaoning Province. Methods 15 518 subjects in 7 200 families aged 18~65 yrs were randomly selected from 3 cities and 3 counties by multi-stage stratified random cluster sampling.They were assessed for DSMⅢ-R diagnoses by face to face interview with Composite International Diagnostic Interview(CIDI-1.0). Results There were 13 359 subjects had finished the survey,and the response rate was 86.09%.The 2-week,12-month and lifetime prevalence of anxiety disorders were 2.22%,4.60% and 7.21% regulated by Liaoning provincial population in 2000,and1.91%,3.97%and6.32% regulated by countrywide population in 2000.The major disorders were specific phobia and agoraphobia,and their lifetime prevalence were 3.80% and 3.04%,respectively.The people with higher prevalence were females,aged 25~55,with lower educational and economic level. Conclusion Anxiety disorders become a serious public health problem in Liaoning province.Prevention measures should be taken among people with high risk.
5.Cloning and expression of urease B subunit (UreB) of Helicobacter pylori

Objective To obtain the recombinant urease B subunit (ureB) of Helicobacter pylori (Hp) and apply it in serological detection of Hp infected patients. Methods Urease B subunit gene was amplified from the complete genome of Helicobacter pylori by PCR, and cloned into the PinPoint TM Xa Ⅲ fusion expression vector, then was sequenced. The protein of urease B subunit was expressed in E.coli JM109 and purified with affinity chromatography. 113 serum samples of peptic ulcer patients were detected by ELISA combined with purified UreB protein. Results DNA sequence analysis showed the nucleic acid sequence homology of ureB gene was 96.44% and the putative amino acid sequence homology was 99.65%. The recombinant UreB protein was composed of 571 amino acid residues and kept original immunologic reaction with corresponding antibody, and its purity was over 90% after affinity chromatography. The results of ELISA associated with recombination UreB antigen showed the sensitivity and specificity was 92%, 98.5%. Conclusion The recombinant UreB protein will be of value for clinical serodiagnosis and epidemiological study of Helicobacter pylori.
6.Effect of overexpressing isocitrate lyase on succinate production in ldh(-1) Corynebacterium glutamicum.

Corynebacterium glutamicum SA001 is a mutant with lactate dehydrogenase (ldhA) deletion. In order to increase metabolic flux from isocitrate to succinate, and to improve the production of succinate under anaerobic conditions,we transducted the gene aceA coding isocitrate lyase (ICL) from Escherichia coli K12 into Corynebacterium glutamicum SA001 (SA001/pXMJ19-aceA). After 12 h aerobic induction by adding 0.8 mmol/L of IPTG, the recombinant strain was transferred to anaerobic fermentation for 16 h. Succinate reached 14.84 g/L, with a productivity of 0.83 g/(L x h). Compared to C. glutamicum SA001, the activity of ICL of the recombinant strain was increased 5.8-fold, and the succinate productivity was increased 48%. Overexpression of isocitrate lyase will increase the metabolic flux of glyoxylate bypass flowing to succinate.

7.Clinical analysis of lung infection in patients with traumatic brain injury

Objective To determine the incidence of lung infection and associated factors in patients with traumatic brain injury for the sake of improving the clinical outcomes.Methods A retrospective analysis was made on records of 325 patients who hospitalized between January 2014 and June 2014.There were 198 male and 127 female patients,aged 11-78 years [(38.4 ±8.3) years].A total of 172 patients were injured in traffic accidents,80 in high falls,56 in blow accidents,and 17 in others.Lung infection status was documented and related risk factors were analyzed.Results Thirty-two patients (9.8%) had lung infection.Pseudomonas aeruginosa amounting to 12 strains was the most common pathogenic bacteria.Univariate analysis showed mechanical ventilation,airway open,and aspiration were significantly related to lung infection.Logistic regression identified aspiration (OR =2.891,P < 0.05) and mechanical ventilation (OR =1.323,P < 0.05) as the independent risk factors for lung infection.Conclusions Lung infection is a serious complication of traumatic brain injury,affected largely by aspiration and mechanical ventilation.Active preventions,reductions of risk factors,and early treatments should be done to get the best efficacy.

9.Study of crown and framework fitness of DA9-4 alloys

10.Inhibitory effects of amiodarone on simvastatin metabolism in human liver microsomes

Objective To investigate the effects ofamiodarone (AMD) on simvastatin (SV) in human liver microsomes and the possible underlying mechanisms. Methods Time-, NADPH- and concentration-dependent inhibitions were tested in HLM. The logarithm of relative inhibition values was plotted versus preincubation time (0, 5, 10, 15, 20min) for a series concentration of AMD used (0, 2, 5,25, 50 μ mol/L), and the slopes determined by linear regression. These slope values represente the observed inactivation rate constants (kobs). A double-reciprocal plot was then constructed using the reciprocal of the ko~ (y-axis) and the reciprocal of the associated inhibitor concentration (x-axis) to estimate the values ofkinact and K, which were two principal kinetic constants that were specific for mechanism-based inhibition (MBI).drug-drug interactions (DDI) potential was predicted based on in vitro data and by using the in vitro-in vivo extrapolation. Results The time-, concentration- and NADPH-dependent charactga'istics confirmed that when SV was the substrate of CYP3A4, the inhibition of AMD to CYP3A4 is MBI. Kj and kinact value were calculated to be 5.1 μ mol/L and 0.018min-1 The Clint of SV was reduced 2.96-5.63 fold when it was administrated with AMD. Conclusion Based on the results, AMD would inhibit SV metabolism via the mechanism-based manner, which would lead to DDI when they are taken together. Careful clinical observation is recommended when AMD and SV have to be simultaneously prescribed.
