1.Effects of heat shock protein 70 activator SW02 on lipopolysaccharide-induced expression of inducible nitric oxide synthase in macrophages

Aim To observe the effects of heat shock protein 70 ( Hsp70 ) activator SW02 on lipopolysaccha-ride( LPS)-induced expression of inducible nitric oxide synthase ( iNOS ) and LPS-induced production of nitric oxide ( NO ) in macrophages. Methods RAW264. 7 cells were stimulated by LPS, and were divided into DMSO,DMSO+LPS(1 mg·L-1),SW02,and SW02+LPS ( 1 mg · L-1 ) groups. The protein expression was detected by Western blot. NO concentration was measured by Griess kit. The iNOS mRNA was detected by real-time PCR. The NF-κB binding to iNOS promot-ers was measured by chromatin immunoprecipitation ( ChIP ) assays. Results SW02 significantly blocked the protein and mRNA expression of iNOS as well as the production of NO in LPS-stimulated RAW264 . 7 cells(P<0. 01 or P<0. 05,SW02+LPS group vs DM-SO+LPS group) . SW02 did not affect the LPS-induced degradation of IκB-α( P>0. 05 , SW02 +LPS group vs DMSO+LPS group ) and nuclear translocation of NF-κB ( P > 0. 05 , SW02 + LPS group vs DMSO + LPS group) . However,SW02 reduced the NF-κB binding to iNOS promoters inside the cell( P<0. 05,SW02+LPS group vs DMSO+LPS group) . Conclusion These re-sults show that SW02 prevents iNOS expression and NO induction likely through attenuation of the NF-κB bind-ing to iNOS promoters in macrophages.

Plasma aldosterene/renin ratio (ARR) is a sensitive screening test and parameter for primary aldosteronism(PA).The use of ARR leads to a marked increase in the detection rate of PA in the hypertensive population.However,ARR remains a nonstandardized test,and the cutoff value of ARR used in the different studies is varied.Further and systematical studies are needed to improve the accuracy of the test.
5.Influencing Factors for Genomic DNA Extraction of Paeonia Suffruticosa

OBJECTIVE:To study the influencing factors for the extraction of the genomic DNA from Paeonia Suffruticosa.METHODS:Taking Paeonia Suffruticosa(root bark of Chinese medicinal herb) as material to investigate the influencing factors including concentrations of the NaCl and beta-mercaptoethanol,temperature and time of water bath,RNaseA,PCR(polymerase chain reaction) system etc in the buffer solution on the basis of modified CTAB method.RESULTS:The DNA obtained by modified CTAB method was pure,integrated,with the value of A260/A280 ranged from 1.8 to 2.0,the ampl-ified bands of PCR were clear and bright,which lay a solid foundation for the following molecular biology experiments.CONCLUSION:The modified CTAB method is economical,rapid and efficient,and it can be served as an extraction of genomic DNA from root bark Chinese medicinal herb as well as a theoretical basis for full scale production.
6.The relationship between highly sensitive C-reactive protein and acute cerebrovascular disease

Objective To investigate the changes of the serum level of highly sensitive C-reactive protein (hs-CRP)in patients with cerebrovascular disease and its clinical significance. Methods The serum level of hs-CRP in 60 patients with acute cerebral infarction and 60 patients with acute cerebral hemorrhage and 40 healthy persons were determined.All the patients were scored by clinic neurological deficit scale(NDS),and were compared in groups. Results The serum level of hs-CRP in patients with acute cerebral infarction and with acute cerebral hemorrhage were significantly higher than that in normal controls(t=19.712 13.217 P <0.01).There was no obvious difference between the patients with acute cerebral infarction and with acute cerebral hemorrhage. Conclusion The serum level of hs-CRP was closely correlated with the severity of cerebrovascular disease.
