1.Impact of Anziheji against ERK1/2 signaling pathways on the maternal-fetal interface of ACA positive abortion in mice

Objective To study the role of ERK1/2 signaling pathways on the process of ACA positive abortion and explore the effect of Anziheji on ERK1/2 signaling pathways from cell proliferation.Methods Levels of ERK1/2 on maternal-fetal interface of every mice in ACA positive abortion model group,blank control group,Anziheji low,mediate,high dose group,aspirin group were detected by immunohistochemical. Results The average optical density value of ERK1 in Anziheji low dose group(0.678 ±0.097)was close to normal group(0.727 ±0.047),there was no significant difference between them.Compared with blank control group,the ERK1 average optical density value in model group(0.434 ±0.066),Anziheji dose group(0.613 ±0.041),high dose group(0.487 ±0.061)and aspirin group(0.551 ±0.056)were significantly lower,and the differences were all statistically significant(P<0.01).Compared with blank control group(0.708 ±0.038),the ERK2 average optical density values in model group(0.430 ±0.058), Anziheji low dose group(0.621 ±0.041),mediate dose group(0.562 ±0.047),high dose group(0.449 ±0.062)and aspirin group(0.515 ±0.045) were significantly lower,and the differences were all statistically significant (P <0.01 ).Conclusion Anziheji can improve the activity of cell proliferation,improve the function of the placenta and change the outcome of pregnancy through ERK signal pathway.
2.Therapeutic Observation of Thread-embedding at Scalp Points for Insomnia

Objective To observe the clinical efficacy of thread-embedding at scalp points in treating insomnia.Method Sixty-four insomnia patients were randomized into a treatment group of 33 cases and a control group of 31 cases. The treatment group was intervened by thread-embedding at scalp points, while the control group was by orally taking Estazolam. Four weeks were taken as a treatment course, and the clinical efficacies and changes of indexes were measured after a treatment course.Result The total effective rate was 90.0% in the treatment group versus 90.3% in the control group, and the difference was statistically insignificant (P>0.05). The item scores of Pittsburgh Sleep Quality Index (PSQI) were changed significantly after treatment in both groups (P<0.01). The changes of sleep latency and daily function disturbance in the treatment group were significantly different from that in the control group (P<0.05).Conclusion Thread-embedding at scalp points and oral administration of Estazolam both can improve insomnia symptoms. Thread-embedding at scalp points is inferior to Estazolam in improving sleep latency but is superior to Estazolam in improving daily function.
3.The influence of obstetric complications and pregnancy outcome because of prenatal anxiety and depression

Objective To investigate the influence of obstetric complications and pregnancy outcome because of prenatal anxiety and depression.Me~ods One hundred primipara were selected as the research objects,their emotions were evaluated by Self-Rating Anxiety Scale(SAS),Self-Rating depression Scale(SDS).The patients' scores of SAS ≥50points or SDS ≥ 53points were selected as the observation group,the patients'scores of SAS ≥ 50points and SDS≥53points were selected as the control group.The pregnant women childbirth way,preeclampsia,hyperemesis gravidarum,gestational diabetes,perinatal infection,hydramnios,oligohydramnios,premature rupture of membranes,postpartum hemorrhage,fetal distress,premature birth,low birth quality,neonatal asphyxia of two groups were compared.ResultsThe primipara cesarean delivery rate of the control group was lower than that of the observation group(4.00% vs 22.00%,x2 =7.161,P <0.05),the obstetric complications incidence of the observation group was higher than that of the control group (32.00% vs 2.00%,x2 =15.946,P < 0.05),the birth outcomes incidence of the observation group was lower than that of the control group (4.00% vs 20.00%,x2 =6.061,P < 0.05).Conclusion Prenatal anxiety and depression significantly increase pregnant women obstetric complications,increase the perinatal fetal intrauterine distress and the probability of premature birth,low birth quality and neonatal asphyxia.
4.Expression of nitric oxide synthase in the process of cerebral ischemia/reperfusion

BACKGROUND: Nitric oxide synthase(NOS) is the key factor for the synthesis of nitric oxide (NO) . Because NO combines with oxygen, hemoglobin and other substances in vivo easily and deactivates quickly, and it is not exactly determined, so determining the activity of NOS is the important link for further studying the pathogenesis of NO in cerebral ischemia/reperfusion (I/R)injury.OBJECTIVE: To study the effect of different types of NOS in the process of cerebral I/R injury.DESIGN: Randomized and controlled animal experiment.SETTING: Instituteof Cerebrovascular Disease, Affiliated Hospital of Medical College of Qingdao University.MATERIALS: This experiment was carried out in the Shandong Key Laboratory of Prevention and Treatment for Encephalopathy from May to December 2005. Twenty-eight adult healthy male Wistar rats, of clean grade, weighing from 220 to 260 g, were provided by the Experimental Animal Center of Shandong University. The involved rats were randomly divided into sham-operation group (n =4) and cerebral ischemia group (n =24). Six time points were set in cerebral ischemia group: ischemia 1 hour reperfusion 6 hours, 12 hours, 1 day, 3 days, 7 days and 14 days, 4 rats at each time point.METHODS: Rat models of middle cerebral artery occlusion/reperfusion were established by suture-occluded method through inserting a suture into the left internal-external carotid artery. The expressions of different types of NOS at different time points after cerebral I/R were detected by immunohistochemical technique.MAIN OUTCOME MEASURES: ①Toluidine blue-stained two groups of nerve cells; ② The expression and distribution of neuronal NOS (nNOS), endothelial NOS (eNOS) and inducible NOS(iNOS) at different time points.RESULTS: ①Karyopyknosis and cell debris appeared in the nerve cells of the injured region of cerebral ischemia group,and there were no significant differences of cells among different time points. ② Six hours after reperfusion, the expressions of nNOS, eNOS and iNOS were found in the neurons of brain tissue and increased with the elongation of time of reperfusion. The regions in which different types of NOS in neurons of brain tissue were expressed were cortical area and corpora striata. nNOS and iNOS were highly expressed within 12 hours to 7 days after reperfusion in the brain, and eNOS was highly expressed within a short time period, i.e. 6 hours to 3 days after reperfusion. eNOS expression increasing and decreasing occurred earlier than nNOS and iNOS. But the expressions of three kinds of NOS all reached peak on the first day after reperfusion. The changing tendencies of the expression of three kinds of NOS in the cortical area and corpora striata were the same basically.CONCLUSION: After cerebral I/R injury, the high expression of eNOS occurs early and lasts for a short time, while that of nNOS and iNOS occurs late and lasts for a long time.
5.Meta-analysis of femoral shaft fractures treated with interlocking intramedullary nail and extramedullary plate

BACKGROUND:With deep understanding of the concept of biological fixation, more and more physicians choose interlocking intramedulary nail in the repair of femoral shaft fracture. Compared with traditional extramedulary plate, the superiority of the interlocking intramedulary nail has not been reported at present. Randomized controled clinical study is less, and lacks of systematic evaluation.
OBJECTIVE:The results of meta-analysis were used to compare the therapeutic effects of interlocking intramedulary nail and steel plate for femoral shaft fractures.
METHODS: We retrieved the MEDLINE, Embase, PubMed, Cochrane library, CNKI, Wanfang database, and Vip database from 2000 to 2015 by computer to colect randomized controled study on interlocking intramedulary nail and extramedulary plate for treatment of femoral shaft fractures. We screened the literatures that met the inclusion criteria, were strict quality evaluation of the selection. Excelent and good rate, operation time, intraoperative blood loss, hospitalization time, recovery time of knee joint function reaching 135°, knee joint function recovery time of the second operation to remove the internal fixation for reaching 135°, postoperative drainage volume, fracture healing time, nonunion or delayed union, internal fixation loosening, postoperative infection, and osteomyelitis were considered as the evaluation index of meta-analysis. Meta-analysis was performed with RevMan 5.2 software from the Cochrane Colaboration.
RESULTS AND CONCLUSION:Finaly 10 Chinese articles were included, including 915 patients with femoral shaft fractures. The results of meta-analysis showed that compared with extramedulary plate, interlocking intramedulary nails for femoral shaft fractures could effectively reduce the amount of blood loss, postoperative drainage, shorten operation time, hospitalization time, fracture healing time, reduce the incidence of postoperative infection, and obtain recovery of knee joint function. These results suggest that interlocking intramedulary nail for treating femoral shaft fractures has certain advantages. The interlocking intramedulary nail can be firstly selected in the permit of patient’s economic conditions and hospital conditions.
6.Expression and clinical significance of RNA-binding protein Musashi1 in colorectal cancer

Musashi1 (Msi1) is an evolutionary conservative RNA-binding protein (RBP),and it is a stem marker in a variety of organizations,including intestinal,neural system.Msi1 maintains the balance between self-renewal and differentiation.Recently,many researchers report that Msi1 is overexpressed in many types of tumors,especially in colorectal neoplasms,participating in the regulation of cell cycle,proliferation,apoptosis and so on.Msi1 becomes a key regulator of many cancers,which is expected to turn into a new target for cancer therapy.
7.Comparison of 3.0T MRI and SPECT-CT in the diagnosis of osteoporotic vertebral compression fractures

BACKGROUND:There have been many studies on the diagnosis of osteoporotic vertebral compression fractures. However, there are few studies on the diagnostic methods of fracture in different periods. OBJECTIVE:To comparatively analyze the application value of high-intensity magnetic resonance imaging (MRI) and single photon emission computed tomography combined with CT scan image fusion (SPECT-CT) in the diagnosis of osteoporotic vertebral compression fractures. METHODS:Clinical data of 35 patients with osteoporotic vertebral compression fractures were retrospectively analyzed, including 56 vertebrae. There were 14 males and 21 females, with an average age of 72.6 years. The course of disease was within 3 weeks in 22 patients, belonging to acute fractures. The course of the disease was between 4 and 12 weeks in 10 patients, belonging to the subacute fractures. The course of disease was 6 months in 3 patients, belonging to the fracture healing period. Al patients were tested with high-intensity MRI and SPECT-CT examination, and were treated with vertebroplasty or percutaneous kyphoplasty. The Fisher exact method was used to compare the two diagnostic methods in the diagnosis of vertebral fracture. RESULTS AND CONCLUSION:(1) In the 56 responsible vertebral bodies, 32 vertebral bodies belonged to the acute-stage fractures, and 24 vertebral bodies belonged to the non-acute-stage fractures. In al the fractures, MRI diagnosed 49 segments;SPECT-CT diagnosed 52 segments. The sensitivity of SPECT-CT testing was higher than MRI, and its specificity was less than MRI. There was a high consistency of diagnosis between fractures. (2) In the 32 acute vertebral fractures, the two tests diagnosed 29 segments. The sensitivity of SPECT-CT was higher than that of MRI, but its specificity was lower than that of the MRI. There was a high consistency between the diagnoses of fractures. (3) In the 24 non-acute vertebral fractures, MRI diagnosed 20 segments;SPECT-CT diagnosed 23 segments. SPECT-CT was more sensitive than MRI;the specificity of SPECT-CT was lower than MRI. There was a high consistency between the two diagnoses of fractures. (4) There was a high consistency in the MRI and SPECT-CT examination to determine the fracture of different periods of responsibility of the vertebral body, but the sensitivity of SPECT-CT is higher than MRI examination;SPECT-CT is an effective inspection method in the diagnosis of vertebral fractures.
8.Expression of Nupr1 in Gastric Cancer and its Clinical Significance

Background:The morbidity and mortality of gastric cancer remain high worldwide,and it is urgent to explore new targets for the diagnosis and treatment of gastric cancer. Nuclear protein 1 (Nupr1)has a variety of biological functions, especially in the tumorigenesis and development of the malignancies. Aims:To investigate the expression and clinical significance of Nupr1 in gastric cancer. Methods:Quantitative RT-PCR was used to determine the mRNA expression of Nupr1 in 72 cases of gastric cancerous tissue and the paired paracancerous tissue. Western blotting and immunohisto-chemistry were employed to detect the protein expression and cellular localization of Nupr1. The correlation of Nupr1 with the clinicopathological characteristics of gastric cancer was analyzed. Results:Expressions of Nupr1 mRNA and protein in gastric cancer were both significantly higher than those in paracancerous tissue (P < 0. 05),and their expressions were coincident. Nupr1 protein was expressed mainly in cytoplasm,and the nuclear expression was reduced. Expression of Nupr1 mRNA in gastric cancer was significantly correlated with tumor size,differentiation,pathological type,Bormann's classification,depth of invasion,lymph node and distant metastases and TNM staging (P < 0. 05),and was not correlated with gender,age and site of tumor (P > 0. 05). Conclusions:Nupr1 is overexpressed and correlated with invasion, metastasis and progression of gastric cancer. It can be used as a novel biomarker for early diagnosis and prognosis evaluation,and as a potential target for treatment of gastric cancer.
9.Clinical significance of dynamic morphological changes of circle cistern observed by computed tomography scanning in patients with severe traumatic brain injury

Objective To investigate the clinical significance of observing the dynamic morphological changes of circle cistern on CT scanning in the patients with acute severe traumatic brain injtuy (TBI).Methods The CT imaging data of circle cistern of 135 patients with acute severe TBI were analyzed retrospectively.According to the morphological changes of circle cisterns on the initial CT images,135 cases were divided into 5 groups,and 135 cases carried on prognostic evaluation with Glasgow outcome score (GOS) 6 months later.The relationship between the dynamic morphological changes of circle cistern and the prognosis of patients were analyzed statistically.Results There was a significant relationship between the state of circle cistern and the prognosis of patients.The more obvious the dynamic morphological changes of circle cistern were,the worse prognosis and the higher the fatality was,especially higher mortality in patients with exacerbation of the dynamic morphological changes of circle cistern (79.5%),and the difference was statistically significant among 5 groups (P < 0.01).Conclusions There was a significant relationship between the state of circle cistern and the prognosis of the patients with acute severe TBI.The dynamic morphological changes of circle cistern on CT scanning might be used as a promising marker to evaluate the severity of severe TBI,especially in the patients at earlier posttraumatic interval.It is necessary to observe the dynamic change of circle cistern.
10.Human immunodeficiency virus vulnerabilities and related health risk behaviors of out-of-school children of the floating population in Beijing

Objective:To explore the vulnerability and related risk behaviors of the out-of-school adolescents floating with their parents from country to city.Methods:From September to October in 2005,260 out-of-school adolescents who were 14 to 20 years old were investigated in our study.Results:The out-of-school adolescents lacked of HIV/AIDS knowledge and life skills,and their psychosocial competence was at a lower level.The adolescents were open-minded to sex,but they were not conscious of using condoms.Attitude towards people with HIV was positively correlated with a well-being family,harmonious relation between their parents,a stable job,knowledge,life skills and psychosocial competence(such as in control of emotions,empathy).Their life skills were positively correlated with their mothers'schooling satisfaction with family income,a stable job,high scores of knowledge,social responsibility,and interpersonal relationship.Competence of HIV/AIDS prevention was positively correlated with schooling,a stable job,knowledge,life skills,and psychosocial competence.Conclusion:Out-of-school adolescents were vulnerable to HIV/AIDS.Individual level intervention combined with structural intervention could effectively reduce their vulnerability.Effective and feasible health education approach to enhance their life skills and psychosocial competence was a key point to health education on HIV/AIDS prevention.
