Objective To observe the effects of mouse indoleamine 2,3-dioxygenase (IDO) gene on CD4+ T cells by transfecting an eukaryotic expression vector containing mouse IDO gene fused with enhanced green fluorescent protein (pEGFP-N1). Methods The immature dendritic cells (imDCs) derived from mouse bone marrow were cultured in vitro and morphologically identified by transmission electron microscope, scanning electron microscope and flow cytometry. The immature dendritic cells were transfected by DOTAP liposome, and the expression of green fluorescence protein was confirmed with inverted fluorescence microscope. The spleen-derived CD4+ T lymphocytes from mice were isolated and purified in vitro, and then carried to mixed lymphocyte reaction (MLR) with 3H-TdR incorporation. The effect of IDO gene on the proliferation of spleen-derived CD4+ T lymphocytes of the allergized mouse challenged by ovalbumin was assayed by 3H-TdR MLR incorporation, and the apoptosis of spleen-derived CD4+ T lymphocytes was assayed by TUNEL. 7.Transformation of endoscopic and endoscopic ultrasonic findings after endoscopic variceal sclerotherapy

Objective To investigate the rule of changes in endoscopic and endoscopic ultrsonic findings after endoscopic variceal sclerotherapy in the follow-up period. Methods Patients suffered from esophagogastric variceal bleeding and received endoscopic variceal sclerotherapy were investigated. Patients with liver cirrhosis after viral heptitis and their varices eliminated or reduced to grade Ⅰor Ⅱ after sclerotherapy entered this study for at least 5 years' follow - up. Endoscopic and endoscopic ultrsonic findings were recorded to find its course of transformation. Results Lumens of varices were occluded by thrombus and eventually replaced by white fibrous tissue after endoscopic sclerotherapy. Vessels reappeared within e-sophageal wall gradually after a lot of time in the form of formation of new venules (93. 8% ) or re-vascular-ization (6. 2% ) . Newly formed varices within gastric wall after sclerotherapy were seen in 5. 3% of the cases and some cases of duodenal varices were also found rarely in the follow-up. Endoscopic ultrasonic finding confirmed the revascularization and dilatation of vessles after sclerotherapy. Conclusion From the follow-up results it indicates that varices would reappear gradually after endoscopic sclerotherapy, regular re-examination and appropriate treatment in time are the most important measure to assure its long term effect.
8.Optimizing super pressure extracting technique for ginsenoside from Panax ginseng by orthogonal test

Objective To study the optimum procedure of extracting ginsenoside from Panax ginseng by super pressure extracting technique (SPET). Methods SPET was used to extract ginsenoside from P. ginseng at the normal temperature. The optimum extracting process was selected by the orthogonal test. The contents of ginsenoside in P. ginseng were determined by UV-spectrophotometry. Results The optimum condition of SPET was as follows: when the solid dissolved in 50% ethanol, ratio of raw P. ginseng (g) and solvent (mL) was 1∶75, the pressure was kept at 500 MPa for 2 min; the high yield of ginsenoside was up to 7.76%. Conclusion SPET has a series of advantages, such as higher efficiency, shorter extracting time, and lower exhausting energy, at the same time, the impurity is little, etc.
9.Meta-analysis of Early Postoperative Small Bowel Obstruction after Laparoscopic Colorectal Resection

Objective To compare the efficacies of laparoscopic colorectal resection and conventional open surgery by collecting and investigating related studies with Meta-analysis.Methods Randomized controlled trials(RCT) on the outcomes of laparoscopic colorectal resection and conventional open surgery that published between January 1989 and March 2008 in English were collected and analyzed.The quality of the original investigations was evaluated,and the low-level reports were excluded.Then the odds ratio(OR) for the incidence of early postoperative small bowel obstruction between the two methods were calculated.ResultsTotally 12 studies including 3032 cases were analyzed.Among which,1522 patients received laparoscopy,33 of them developed early postoperative small bowel obstruction.1510 cases underwent open surgery,and 71 of these patients showed small bower obstruction before being discharged form hospital.The pooled OR was 0.46 with a 95% confidence interval of(0.30-0.69).Conclusion Compared with open surgery,laparoscopic colorectal resection is superior in the incidence of early postoperative small bowel obstruction.
10.Wnt3a induces rat bone marrow mesenchymal stem cells differentiation into neuron-like cells

BACKGROUND: Wnt signaling pathway is a key regulator of cellular proliferation and differentiation, but its correlation with neural differentiation of bone marrow mesenchymal stem cells (BMSCs) is not very clear. OBJECTIVE: To find out the molecules of the Wnt family which are involved in differentiation of rat BMSCs into neuron-like cells. METHODS: The rat BMSCs were separated and cultured in vitro. The morphology of the BMSCs was observed. Flow cytometry analysis was performed to detect cell phenotype CD44, CD9, CD34 and CD45. Wnt3a and Wnt5a were respectively combined with basic fibroblast growth factor to induce BMSCs differentiation into neuron-like cells, and then were identified by using immunocytochemistry and RT-PCR. RESULTS AND CONCLUSION: The BMSCs were long-spindle. CD9 and CD44 were highly expressed, while CD34 and CD45 were lowly expressed. Nestin and neuron specific enolase were positive but glial fibrillary acidic protein were not obviously expressed when they were cultured with Wnt3a. In Wnt5a group, Nestin expression was weakly positive, while neuron specific enolase and glial fibrillary acidic protein were negative. RT-PCR result revealed Nestin expressed both before and after induction in the Wnt3a induced group, neuron-specific enolase exhibited apparent amplified bands 5 days after the induction, and more apparent at 10 days. A weak amplification band of glial fibrillary acidic protein could be seen at 10 days after the induction. In Wnt5a and control groups, BMSCs induced by 10 days weakly expressed Nestin, while neuron-specific enolase and glial fibrillary acidic protein were almost not expressed. It is indicated that Wnt3a molecule can promote the differentiation of BMSCs cultured in vitro to neuron-like cells.
