And the y were as follows: the underlayer of CHCl3-CH3OH-H2O(65∶35∶10) below 10 ℃was developed at 15~18℃ and sprayed with 10%H2SO4-C2H5OH,the heating tempe rature was 105℃ for 3~5 min,and λS=400 nm, λR =700 nm.

Objective To evaluate the feasibility of ~ 125 Iodine -labeled IL-2 scintigraphic methods for the imaging of lymphocyte infiltration of the Langerhans′ islets. Methods ~ 125 I-IL2 was injected intravenously to pre-diabetic female NOD mice (aged 16-17 weeks)and female Balb/c mice as control. 4.Immunotoxin BAC5-CT treated nasopharyngeal carcinoma

In this study, immunotoxin (IT) was prepared by conjugating BAC5 and CT with SPDP. The effects of IT on NPC and its mechanisms were explored using double labeled with radioactive nuclides, immunography and electron microscope technique in vivo and in vitro. The specific concentration of BAC5 in the tumor area showed. The radioactivity rate of tumor/nontumor (T/NT) was up to 10.26. IT had cytotoxic effects both on the cultured CNE-2 cell line and tumor multicell spheroides. In vivo, the preliminary result indicated that IT also had a inhibitory action on the nude mice models bearing human NPC (Reported in another article). Under electron microscope, the necrosis and apoptosis of tumor cells were found. The membranes of most tumor cells were found intacted not or corrosined, some of them had the character of apoptosis, including reduce of tumor cells membrane villi, condensation of cytoplasm and pyknosis or cleavage of nuclear. There were many of apoptosis bodies, which were occasionally phagocytosed by tumor cells. The infiltration of immunocytoes in tumor tissue could be seen. The results indicated that BAC5 can specifically combine with NPC cells and BAC5-CT has the inhibitory effect on NPC in vitro and in vivo, mechanism of which may be related to the effects that ‘warhead' CT dissolute the membrane of tumor cells directly, or/and IT promote the infiltration of immunocytoes so as to induce the apoptosis of tumor cell.

6.Expression of CD28/B7 in the Lesional Skin and Peripheral Lymphocytes in Psoriasis

Objective To investigate the role of costimulatory molecules CD28/B7 in the pathogenesis of psoriasis. Methods The expression of CD28, CD80 and CD86 was detected in the lesional skin of 22 cases by immunohistochemical technique (ABC), and in the peripheral blood lymphocytes of 17 cases by flow cytometry respectively. As control, normal skin and lymphocytes from peripheral blood of healthy volunteers were also tested. Results The immunohistochemistry study showed that the expression of CD28, CD80 and CD86 in the psoriatic lesions was significantly higher than that in the normal controls (P .05). Conclusion CD28, CD80 and CD86 might play a certain role in the pathogenesis in psoriasis.
7.Influences of dexamethasone on expression of fas, bcl-2 mRNA and apoptosis in eosinophils in vitro

AIM: To investigate the effects of dexamethasone (DM) on expression of fas and bcl-2 mRNA and apoptosis in eosinophils (Eos) of bronchoalveolar lavage fluid (BALF) in asthmatic guinea pig. METHODS: The guineas pigs were stimulated with ovalbumin (OVA) for 48 hours. Hypodense Eos (HEos) and normodense Eos (NEos) were purified from BALF by gradients of Percoll. Eos was cultured in PRMI-1640 medium and DM (10 -10-10 -5 mol/L) was added. The mRNA of fas and bcl-2 in Eos was measured by hybridization. Apoptosis was detected by TdT-mediated dUTP nick end labeling (TUNEL). RESULTS: Eos was cultured under the condition of DM administration in vitro. Apoptotic Eos increased, fas mRNA expression increased and the bcl-2 mRNA expression decreased in a dose-dependent manner. There was significant correlation between Eos apoptosis and fas mRNA expression. The expression of bcl-2 mRNA was inversely correlated with Eos apoptosis. CONCLUSION: DM promoted Eos apoptosis, increased expression of fas and inhibited expression of bcl-2 in a dose-dependent manner in vitro. fas and bcl-2 might be one of mechanisms of DM promoting apoptosis in Eos.
8.Effects of modified fat clearance technique in the diagnosis of metastatic lymph nodes in the mesorectum of patients with rectal cancer

Objective To investigate the number, distribution and metastatic rule of lymph nodes in the mesorectum of rectal cancer specimen after processing the recta] cancer specimen with modified fat clearance technique. Methods Sixty patients with mid-low rectal cancer who had been admitted to Peking University School of Ontology from 2003 to 2008 were assigned to test group. All the 60 patients who denied neo-adjuvant therapy were treated with total mesorectal excision, and the rectums resected were processed with the modified fat clearance technique. Rectums from another 50 patients with mid-low rectal cancer in control group were fixed in formalin solution for 24 hours. The mesorectum was divided into anterior, posterior, left and right quadrants, and each quadrant was further divided into upper, middle and lower parts. The numbers of lymph nodes harvested and metastatic rate of lymph nodes between the 2 groups were analyzed by t test and chi-square test. Results (1) The numbers of lymph nodes harvested in the test and control groups were 1436 and 525, with statistical difference between the 2 groups (t =- 12. 153, P <0.05). The number of small lymph nodes(diameter≤5 mm) harvested in test group was 985. (2) The numbers of lymph nodes harvested in the anterior, posterior and bilateral mesorectum were 125,696 and 615, respectively. The numbers of lymph nodes harvested in the upper, middle and lower part of the mesorectum were 395,534 and 507, respectively. The metastatic rate of lymph nodes in the upper part of the mesorectum was 18.5% (37/200), which was significantly lower than in the middle [43.5% (87/200)] and lower [38.0% (76/200)] part of the mesoreetum (X2= 9. 414, 6.406, P < 0.05). Two hundred metastatic lymph nodes in 33 patients were harvested, 48.0% (96/200) of which with a diameter of ≤ 5 mm. Twenty percent patients had their TNM stage changed after the retrieval of metastatic small lymph nodes. Conclusions Modified fat clearance technique significandy improves the retrieval of lymph nodes, and more small lymph nodes retrieved can increase the accuracy of staging. Rectal cancer cells have strong tendency to disseminate to the distal mesorectum. Adequate excision of the distal mesorectum is key in controlling the local recurrence.
9. Clinical efficacy of famitinib malate for treatment of metastatic renal cell carcinoma—a report of 9 cases

Objective To study the clinical effect of famitinib malate for treatment of metastatic renal cell carcinoma (mRCC%. Methods Nine mRCC patients treated with famitinib malate in our center completed their follow-up from October 2011 to June 2015. The patients received famitinib malate at an initial dose of 25 mg, which was given orally before breakfast once a day. One cycle consisted of 42 days. The antitumor efficacy and the adverse event (AE% were observed. The most common treatment-related AE of famitinib malate was proteinura, and other AEs were similar to those of sunitinib. Six patients had reduction and interruption as appropriate due to AEs, and 2 patients stopped the treatment due to intolerable proteinuria. Conclusion Famitinib malate has a good antitumor activity upon mRCC, and its adverse reaction is controllable, making it a promising molecule-targeted drug for clinical application.
10. Single nucleotide polymorphism analysis on ossification of posterior longitudinal ligament in han population in china

Objective To investigate the polymorphic loci of four candidate genes of ossification of posterior longitudinal ligament (OPLL) patients in the Han population in China, so as to study the loci associated with diseases. Methods The Sequenom system was adopted to analyze the 19 SNP of the four candidate genes (namely, COL6A1, BMP-2, VDR and Runx2) in 180 Han individuals, with 78 in the pathological group and 102 in the control group. The genotype distributions and allele frequencies of each SNP were compared between the two groups, and the polymorphic loci related to OPLL and their relationship were analyzed. Results Allelic analysis found no disease related locus in the 19 SNP loci of the four candidate genes. However, the genotyping analysis found that rs1321075 and rs12333172 of Runx2 were significantly different between the two groups (P=0.0339, P=0.0428), both loci were on the No.6 chromosome and Linkage Disequilibrium (LD) analysis showed linkage disequilibrium between them. The 11 SNP loci on the No.6 chromosome formed two blocks, with the range being 51 kb and 21 kb, respectively. One of the two blocks was a haploid (CTCG) made up of rs967588, rs16873379, rs3749863 and rs6908650, with a patient/control ratio of 1.75 and a risk rate of 0.81, indicating the possibility of increased incidence rate, but with no significant difference (P=0.259). No polymorphic loci were found associated with diseases for COL6A1, BMP-2 and VDR genes. Conclusion This is the first report that SNP variation of Runx2 gene may be a reason for OPLL in the Han population in China, and there was no notable connection between the polymorphic loci of COLA1, BMP-2 and VDR and OPLL.
