1.Distribution of ~(125)Iodine-labeled Interleukin-2 and its relation to insulitis in NOD mice

Objective To evaluate the feasibility of ~ 125 Iodine -labeled IL-2 scintigraphic methods for the imaging of lymphocyte infiltration of the Langerhans′ islets. Methods ~ 125 I-IL2 was injected intravenously to pre-diabetic female NOD mice (aged 16-17 weeks)and female Balb/c mice as control. Animals were sacrificed at different time points and the radioactivity in different organs was measured.~ 131 I-IL-2 was injected iv to 3 pre-diabetic female NOD mice, and pancreas was imaged through SPECT after 30 minutes injection. Results The mean radio activities in the pancreas of NOD mice were significantly higher as compared to those of Balb/c mice at time points from 30 to 120 minutes (P

4.Immunotoxin BAC5-CT treated nasopharyngeal carcinoma

In this study, immunotoxin (IT) was prepared by conjugating BAC5 and CT with SPDP. The effects of IT on NPC and its mechanisms were explored using double labeled with radioactive nuclides, immunography and electron microscope technique in vivo and in vitro. The specific concentration of BAC5 in the tumor area showed. The radioactivity rate of tumor/nontumor (T/NT) was up to 10.26. IT had cytotoxic effects both on the cultured CNE-2 cell line and tumor multicell spheroides. In vivo, the preliminary result indicated that IT also had a inhibitory action on the nude mice models bearing human NPC (Reported in another article). Under electron microscope, the necrosis and apoptosis of tumor cells were found. The membranes of most tumor cells were found intacted not or corrosined, some of them had the character of apoptosis, including reduce of tumor cells membrane villi, condensation of cytoplasm and pyknosis or cleavage of nuclear. There were many of apoptosis bodies, which were occasionally phagocytosed by tumor cells. The infiltration of immunocytoes in tumor tissue could be seen. The results indicated that BAC5 can specifically combine with NPC cells and BAC5-CT has the inhibitory effect on NPC in vitro and in vivo, mechanism of which may be related to the effects that ‘warhead' CT dissolute the membrane of tumor cells directly, or/and IT promote the infiltration of immunocytoes so as to induce the apoptosis of tumor cell.

6.Influences of dexamethasone on expression of fas, bcl-2 mRNA and apoptosis in eosinophils in vitro

AIM: To investigate the effects of dexamethasone (DM) on expression of fas and bcl-2 mRNA and apoptosis in eosinophils (Eos) of bronchoalveolar lavage fluid (BALF) in asthmatic guinea pig. METHODS: The guineas pigs were stimulated with ovalbumin (OVA) for 48 hours. Hypodense Eos (HEos) and normodense Eos (NEos) were purified from BALF by gradients of Percoll. Eos was cultured in PRMI-1640 medium and DM (10 -10-10 -5 mol/L) was added. The mRNA of fas and bcl-2 in Eos was measured by hybridization. Apoptosis was detected by TdT-mediated dUTP nick end labeling (TUNEL). RESULTS: Eos was cultured under the condition of DM administration in vitro. Apoptotic Eos increased, fas mRNA expression increased and the bcl-2 mRNA expression decreased in a dose-dependent manner. There was significant correlation between Eos apoptosis and fas mRNA expression. The expression of bcl-2 mRNA was inversely correlated with Eos apoptosis. CONCLUSION: DM promoted Eos apoptosis, increased expression of fas and inhibited expression of bcl-2 in a dose-dependent manner in vitro. fas and bcl-2 might be one of mechanisms of DM promoting apoptosis in Eos.
7. Sympathetic nerves and its relationship with sudden cardiac death

Sudden cardiac death (SCD) refers to death within 1 hour after onset of cardiovascular disease or death within 24 hours from the last stable status. SCD has a high incidence and mortality rate. The relationship between heart autonomie nerves, especially sympathetic nerve and SCD, is the focus of study. The increase and/or heterogeneity of sympathetic activity and the remodeling of sympathetic innervation can lead to arrhythmogenic substrate in the heart or even SCD. Medicine treatment, implanted cardioverter defibrillator, left cardiac sympathetic denervation, renal sympathtic nerves ablation and spine stimulation have obtained encouraging outcomes. Therefore the present paper reviews the autonomie system, especially sympathetic nerves, and its relationship with cardiac arrhythmia.
8. Role of a regulating protein in the upper stream of mTOR signaling pathway Rheb in adipocyte differentiation

Objective To observe the effect of Rheb, a regulating protein in the upper stream of mTOR signaling pathway, on adipocyte differentiation. Methods (1) The recombinant plasmid pCAG-Insulator-Rheb was constructed and injected into the embryo of B6 mouse to produce transgenic mouse over-expressing Rheb. (2) The mouse embryonic fibroblast cells (MEFs) were collected from Rheb transgenic mice on pregnancy day 13. 5, and they were induced to differentiate into adipocytes after identified by PCR. (3) On differentiation day 12, the MEFs were subjected to Oil Red O staining and the contents of triglyceride in the MEFs were determined; moreover, the expression of special transcription factor PPARy and C/ EBPa was examined by real-time PCR, and all of the above parameters were used to assess the role of Rheb in the differentiation of adipocytes. Results We successfully constructed the transgenic mouse model over-expressing Rheb. We found that mouse over-expressing Rheb promoted adipogenesis in MEFs and increased the content of triglyceride in MEFs; moreover, the overexpression also greatly changed the results of Oil Red O staining and greatly promoted the expression of adipocyte specific transcript factor PPARy and C/EBPα. Conclusion Over-expression of Rheb can promote the differentiation of MEFs into adipocytes.
9.Expression of CD28/B7 in the Lesional Skin and Peripheral Lymphocytes in Psoriasis

Objective To investigate the role of costimulatory molecules CD28/B7 in the pathogenesis of psoriasis. Methods The expression of CD28, CD80 and CD86 was detected in the lesional skin of 22 cases by immunohistochemical technique (ABC), and in the peripheral blood lymphocytes of 17 cases by flow cytometry respectively. As control, normal skin and lymphocytes from peripheral blood of healthy volunteers were also tested. Results The immunohistochemistry study showed that the expression of CD28, CD80 and CD86 in the psoriatic lesions was significantly higher than that in the normal controls (P .05). Conclusion CD28, CD80 and CD86 might play a certain role in the pathogenesis in psoriasis.
10.Effects of different perfusate on the morphological structure of rabbit corneal endothelium during phacoemulsification

BACKGROUND: The association between the ingredients of perfusate and its protection on corneal endothelium is always the hot issue in ophthalmology and pharmacology.OBJECTIVE: To observe the influence of different perfusates on the structure and function of rabbit corneal endothelium during phacoemulsification.DESIGN: A randomized grouping designed and controlled animal trial.SETTINGS: Laboratory of experimental animal center of Jinzhou Medical College and the laboratory of experimental animal operation of an urban hospital.MATERIALS: The experiments were carried out in the laboratory of experimental animal center of Jinzhou Medical College and the laboratory of experimental animal operation of Jinzhou Yadong Ophthalmology Hospital from September 2004 to March 2005. Sixteen pure Japanese big-ear rabbits of 3.5 months old, clean degree, were randomly divided into four groups with 4 rabbits in each group: normal control group, saline group, shike group and balanced salt solution group. Shike was produced by Shenyang Qixing Pharmaceutical Co.,Ltd.; Balanced salt solution by Alcon Company (USA).METHODS: The rabbits were intraperitoneally anesthetized with 100 ml/L chloral hydrate (3 mL/kg), 4% oxybuprocaine hydrochloride eye drops were used for surface anesthesia of eyes, and both eyes were operated. Alcon phacoemulsification apparatus (USA) and routine microsurgical instruments were used. A 3.5-mm incision was made on sclerotic tunnel at 2 mm posterior to superior limbus of sclera, punctured into the anterior chamber, then 0.25 mL Viscoat (Alcon) was infused. Curvilinear capsulorhexis was performed with the diameter of about 5 mm. The phacoemulsification head was placed in the center to suck out the crystal nucleus and cortex, and the incision was closed after the operation. The morphology of the corneal endothelium was quantitatively determined using contact specular microscope preoperativley and 6 hours postoperatively, including the density and area of corneal endothelium,percent of hexagonal cells and the coefficient of variation. At 6 hours postoperatively, trypan blue-alizarin red active staining was performed, and the changes of slight structures of corneal endothelium were observed under light microscope.MATN OUTCOME MEASURES: ① Quantitative analysis of the forms morphology of corneal endothelium (density and area of corneal endothelium, percent of hexagonal cells and the coefficient of variation); ② Characters of forms and structures of corneal endothelium.RESULTS: All the 16 rabbits were involved in the analysis of results. ① There were no significant differences in the morphological indexes among the four groups preoperatively. ② At 6 hours postoperatively, density and areas of endothelial cells, percent of hexagonal cells and the coefficient of variation were significantly lower in the saline group,shike group and balanced salt solution group than in the normal control group (P < 0.05), and no significant difference was observed between the shike group and balanced salt solution group (P > 0.05). ③ The structural changes of corneal endothelium in the shike group and in balanced salt solution group were alleviated more significantly than those in the saline control group, no necrosis was observed.CONCLUSTON: In phacoemulsification, the damage of perfusate to corneal endothelium is a chemical one. Under the same surgerical conditions, domestic perfusate of shike is as effective as balanced salt solution in protecting endothelial cells.
