Objective135 case treatemet of circumgyration originated insufficiency blood of brain of Songling Xuemaikang Jiaonang combined Xuanyunning Pian.MethodsDivided 396 case patient into thee groups:song,ling Xuemaikang Jiaonang group,Xuanyunning Pian group and combined group.songling Xuemaikang Jiaonang group:songling Xuemaikang Jiaonang 1.5g,oral,tid,treatment of 15 days.Xuanyunning Pian group:Xuanyunnings Pian 2 piece,tid,treatment of 15 days.songling Xuemaikang Jiaonang combined Xuanyunning Pian group:songling Xuemaikang Jisonang 1.5g,oral,tid,treatment of 15 days and Xuanyunning Pian 2 piece,tid,treatment of 15 days.And the treatment during three months was observed.ResultsThe effective power of three group were 63.6%(82/129,songling Xuemaikang Jiaonang group),48.5% (64/132,Xuanyunning Pian group) and 83.7% ( 113/135,songling Xuemaikang Jiaonang combined Xuanyunning Pian group).Compare songling Xuemaikang Jiaonang group with Xuanyunning Pian group,the effective power were increased significant difference.Compare combined group with songling Xuemaikang Jiaonang group,the effective power were increased very significant difference.ConclusionTreatment of circumgyration originated insufficiency blood of brain,songling Xuemaikang Jiaonang combined Xuanyunning Pian were outweighed than single medication.
2.Expressions of CD44 and transforming growth factor-?_1 in endometrium and ectopic endometrial tissues in endometriosis patients

Objective To explore relationship of CD44 and transforming growth factor-? 1 (TGF-? 1) in endometriosis (EM) patients. Methods The histopathological changes and the expressions of CD44 and TGF-? 1 were observed and determined by immunohistochemistry in 17 cases of ectopic endometrium of EM patients and 17 cases of ovarian ectopic endometrium of EM patients. The other 20 cases of non-endometriosis and non-tumorous endometrial tissues were used as the control group. Results No difference in the expressions of CD44 and TGF-? 1 was found in the ectopic endometrium of EM patients and the control group, but the expressions in the ovarian ectopic endometrium of EM patients were significantly higher than those in the control group. Conclusion The abnormal expressions of CD44 and TGF-? 1 in ectopic endometrium of EM patients may be associated with the genesis and progression of EM.
3.Expression of GnRH and GnRH-R in various endometrial tissue and its significance

Objective To detect the protein expression of gonadotropin-releasing hormone (GnRH) and its receptor (GnRH-R) in various endometrial tissues, and to investigate its significance. Methods Immunohistochemistry was employed to analyse the protein expression and localization of GnRH and GnRH-R from endometriosis(EMS), endometrial hyperplasia, endometrial carcinoma and control group. Results GnRH and GnRH-R are expressed at the protein level in all eutopic endometrium, in 50% ectopic endometrium and 92.3% endometrial hyperplasia, while the expressional rate in endometrial carcinoma are 58.8% and 82.4%, respectively. The majority of GnRH and GnRH-R existed in the plasma of glandular epithelium cells and some stroma cells. Conclusions There exists GnRH and its receptor proteins in endometrial tissues, and their abundant expression in endometriosis, endometrial hyperplasia and in endometrial carcinoma may imply that they could be used in clinical diagnosis and treatment.

Objective To investigate the effects of narrow-band ultraviolet B (NB-UVB) on the expression of keratin 17 (K17) in the human keratinocyte cell line HaCaT.Methods Cultured HaCaT cells were irradiated with different doses (0,50,100,200,400 mJ/cm2) of NB-UVB followed by additional culture for 6,12 and 24 hours respectively.To explore the mechanisms underlying the effects of NB-UVB on K17 expression,some HaCaT cells were pretreated with PD98059 (an inhibitor of mitogen-activated protein kinase kinase) followed by UVB radiation and additional culture as described above.Cell counting kit-8 (CCK 8) was used to evaluate the proliferation of,real time PCR to measure the mRNA expressions of K17 in,and Western blot to determine the protein expression of K17,Erk1/2 and phosphorylated Erk1/2 in,HaCaT ceils.Results Both the proliferation of and K17 expression in HaCaT cells were promoted by NB-UVB radiation at low doses (50,100 mJ/cm2),but inhibited by NB-UVB radiation at high doses (200,400 mJ/cm2).Significant differences were observed for the proliferative activity between HaCaT cells irradiated with NB-UVB at 100 or 400 mJ/cm2 and unirradiated HaCaT ceils at 12 and 24 hours (P < 0.01 or 0.05).The phosphorylation of Erk1/2 was upregulated by NB-UVB radiation at 100 mJ/cm2,but downregulated by that at 400 mJ/cm2,and the upregulation induced by low dose NB-UVB could be suppressed by blocking the Erk 1/2 pathway.Conclusion The effects of NB-UVB radiation on K17 expression may be modulated by the Erk 1/2 pathway.
6.Transcriptome analysis of bioenergy plant Miscanthus sinensis Anderss by RNA-Seq.

Miscanthus sinensis Anderss is a perennial C4-grass. It is a promising bioenergy plant, which has been proposed as general feedstock for biomass and lignocellulosic biofuel production. In this study, the flower and leaf buds transcriptomes of Miscanthus sinensis Anderss were sequenced by the platform of Illumina HiSeq 2000. In total 98 326 Unigenes were generated by de novo assembly with an average length of 822 bp and N50 of 1 023 bp. Based on the NR, NT, Swiss-Prot, KEGG, GO and COG databases (Evalue < le-5), 74 134 (75.40%) Unigenes were annotated. A total of 45 507 Unigenes were mapped into different GO terms. In KEGG pathways identification, 36 710 sequences were assigned to 128 KEGG pathways. Sorghum bicolor (37 731, 60.86%), Zea mays (16 258, 26.22%), and Oryza sativa (3 065, 4.94%) showed high similarity to Miscanthus sinensis Anderss. And 24 photosynthesis-related enzyme genes were identified. The result provides a foundation for further characterizing the functional genes in Miscanthus sinensis Anderss.

7.Changes in transcriptome expression profiles and functional analysis of differentially expressed genes during mycelium-to-yeast transformation of Sporothrix schenckii

Objective To compare and analyze the differences in the transcriptomics between mycelium and early yeast phases of Sporothrix schenckii (S.schenckii),and to realize the changes in transcriptome expression profiles during mycelium-to-yeast transformation.Methods A standard strain of S.schenckii (ATCC 10268) was subjected to 96-hour culture with Sabouraud medium at 25 ℃ or 36-hour culture with brain-heart infusion medium at 37 ℃ to obtain the mycelium and yeast form of S.schenckii,and then,their transcriptomes were sequenced.Functional annotation was performed for screened unigenes by comparison using several databases (such as NR,Swiss-Prot,KEGG,COG,KOG,GO and Pfam),coding sequence prediction,and gene expression analysis in each sample.Finally,the differentially expressed genes were subjected to pattern clustering,functional annotation and enrichment analysis.Results A total of 14.76 Gb valid data (clean data) were obtained,and functional annotation results were acquired in 28 094 of 43 863 assembled unigene clusters.Compared with S.schenckii in mycelium phase,there were 10 969 up-regulated genes and 199 down-regulated genes in S.schenckii in yeast phase.These differentially expressed genes were involved in protein phosphorylation,intracellular protein transport,cellular protein modification,small guanosine triphosphate-mediated signal transduction,vesicle-mediated transport,translation,intracellular signal transduction,microtubule formation,adenosine triphosphate synthesis,coupled proton transport and so on.Sixteen genes in the mitogen-activated protein kinase (MAPK) pathway and two-component signaling pathway,which were two important signal transduction pathways involved in fungal morphogenesis,and 16 genes involved in chitin synthesis and metabolism were all confirmed to be up-regulated in S.schenckii in yeast phase.Conclusions Compared with S.schenckii in mycelium phase,great changes in gene expression profiles were observed in S.schenckii in yeast phase.These differentially expressed genes are involved in many functions,suggesting that the dimorphic transition of S.schenckii is regulated by a multi-gene network system.
8.Application and effect of electrocardiograhy on totally implantable venous access ports

Objective To investigate the accuracy and safety of using normal saline as a medium to guide the catheter lumen to assist the localization of the catheter tip. Methods This study included the patients with hematological malignancies in our fully implantable venous port. We enrolled 105 patients from January 2014 to December 2015 as control group, and 220 patients from January 2016 to June 2017 as the experimental group. The control group used the traditional fully implantable venous port after the chest X slice to determine the location of the catheter tip. The experimental group used the intracavitary electrocardiogram location technology to assist the complete implantable venous port catheter tip positioning catheter, then chest X film. Results No catheterization occurred in all the patients. The rate of catheter placement was 100%. In the experimental group, 179 patients (81.36%) had the best placement of catheter tip (i.e., superior vena cava right atrium junction, CAJ), and 35 patients (52.38%) in the control group had the best placement of catheter tip. The proportion of the catheter tip located in the best position (the superior vena cava auricular commissure and CAJ) in the experimental group was higher than that in the control group (χ2=29.615, P < 0.05). Conclusions By the injection of saline guided endocardial mapping real-time monitoring of totally implantable venous transfusion port catheter tip position, can guide the surgeon to grasp the real-time operational direction of totally implantable venous transfusion port catheter tip, accurately positioning the catheter tip position, improve the surgical success rate, worthy of clinical use.
9.Medium optimization for antagonistic Streptomyces S24 and its inhibition on Aspergillus flavus.

Streptomyces S24 has broad spectrum against Aspergillus spp. in food and feed, such as Aspergillus flavus, Aspergillus niger and Asperegillus alutacells. The objective of this study was to improve the production of antifungal substances produced by S24 and to test their inhibitory effects on Aspergillus flavus. By using one-factor-at-a-time experiment and orthogonal design method, we optimized the fermentation medium. The composition of an optimized medium for the production of antifungal substances contained (g/L): starch soluble, 10; Glucose, 40; yeast extract, 8; soybean powder, 24; KH2PO4 4; and CaCO3 0.8. As a result, the productivity of antifungal substances could reach to 10 235.45 microg/mL, and this value was 2.81 times higher than that of initial medium before optimization. Additionally, inhibitory effects of the products on Aspergillus flavus were analyzed. Antagonistic tests indicated that the antifungal substances greatly inhibited mycelium growth and spores germination of Aspergillus flavus. We observed through microscope that the mycelia grew abnormally, such as contorting, bulging, vacuole increasing and the cytoplasmic contents inside effusing and the spores appeared unusual, such as gathering, deforming, cytoplasmic contents inside effusing and fracturing.

10.Effect of selection techniques on chromosome and ultrastructure of human spermatozoa.

OBJECTIVETo know the effect of different selection techniques on chromosome abnormality rate and the percentage of spermatozoa with relatively intact head ultrastructure in human sperm.
METHODSThe selection techniques of swim-up, Percoll density gradient centrifugation and doule-flection tube were used to observe sperm chromosome abnormality rate, sex chromosomal ratio and the percentage of spermatozoa with relatively intact head ultrastructure.
RESULTSThe sperm chromosome abnormality rate, sex chromosomal ratio and the percentage of spermatozoa with relatively intact head ultrastructure remained unchanged after the application of the three selection techniques(P > 0.05).
CONCLUSIONThe three selection techniques used in this study neither increase the damage to sperm ultrastructure nor exert effects on human chromosome.
