2.Expression and clinical significance of ribosomal protein L6 in prostate cancer

Objective To explore the expression of ribosomal protein L6 (RPL6) in prostate cancer and its clinical sig-nificance. Methods RT-qPCR and Western blot assay were used to measure the mRNA transcription and protein expres-sion levels of RPL6 in prostate cancer tissues (n=80) and adjacent non-cancerous tissues (n=62). The relationship between RPL6 mRNA expression level and clinicopathological factors of prostate cancer was statistically analyzed. Results The mRNA and protein expression levels of RPL6 were significantly higher in prostate cancer tissues compared with those of non-cancerous tissues (P<0.05). There were higher serum expression levels of prostate specific antigen (PSA) and higher Gleason score in prostate cancer tissues. The expression level of RPL6 mRNA was significantly higher in patients with lymph node metastasis and late clinical stage (P<0.05). There were no significant differences in PSA levels between different ages, with or without seminal vesicle invasion and different surgical margin status (P>0.05). Kaplan-Meier survival analysis of biochemical recurrence (BCR)-free survival time showed the significantly lower recurrent rate in patients with high RPL 6 mRNA expression(χ2=4.530,P=0.033). Conclusion The elevated expression of RPL6 may play a role in the development of prostate cancer, and which can be used as a tumor marker to assess the prognosis of prostate cancer.
3.Long-term effect of direct percutaneous coronary intervention on left ventricular remodeling in patients aged 75 years and over with acute myocardial infarction

Objective To investigate the long-term effect of direct percutaneous coronary intervention (PCI) on left ventricular remodeling in patients aged ≥75 years with acute myocardial infarction (AMI).Methods 108 cases of patients with acute myocardial infarction were collected into the study.Direct PCI were completed in patients in PCI group within 12 hours of onset.Patients in non-PCI group received conventional conservative treatment with drugs.Patients were followed up for 0.5-8.0 years,and cardiac function (NYHA) and the detected ultrasonic cardiogram were evaluated during follow-up.Results The 106 patients received follow-up.Two female patients who were not treated with PCI died during follow-up,one patient died of sudden cardiac death and another patient died of severe pneumonia.Compared with non-PCI group,direct PCI group showed that cardiac function (NYHA) grade was lower(t=3.17,P<0.05),the end-systolic and end-diastolic volume of left ventricle were less(t=3.50、3.90,all P<0.01),the left ventricular ejection fraction was increased (t=2.00,P<0.05),the ventricular wall motion index was smaller (t=2.96,P<0.01).E/A ratio was higher,E wave deceleration time was shorter,and left ventricular mass index was smaller (t=4.04,4.29,4.70,respectively,all P<0.01),the left ventricular long and short axis diameter were decreased (t=2.30,t=5.53,P<0.05 or 0.01),and Spherical index was increased (t=2.97,P<0.01).Conclusions Direct PCI treatment improves chronic ventricular remodeling in elderly patients with AMI and contributes to long-term improvement in cardiac function.
4.Disinfection Experimentation of a Stable Peracetic Acid

3.00 and exposed to the solution containing 1500 mg/L peracetic acid for 30 min could completely killing the spores.Aerosol spraying with solution containing 1000 mg/L peracetic acid at a dose of 10 ml/m3 for 30 min caused over 90.0% decay rate of natural bacteria in indoor air.CONCLUSIONS Weikangshi peracetic acid has good stability and its germicidal efficacy is not affected.

The number average molecular weights of five fractionated samples of pomelo pectin were determined osmometrically in aqueous solution. The values of Mn ranged from 4.74?104 to 1.83?144 for different fractions.From the data of osmotic pressure and intrinsic viscosity, the [?]-M relation obtained is[?]3.23?10-7M1.75in 0.9% NaCl solution of pH 4.83 at 37℃.
6.Expression of the nod-like receptor protein 3 inflammasome in calcium oxalate stone kidney

Objective To explore the expression level and significance of the nod-like receptor protein 3 (NLRP3) inflammasome in renal tissue with calcium oxalate stone.Methods 20 kidney specimens were collected as the experimental group from patients with calcium oxalate stone who underwent nephrectomy because of stones in our hospital between January 2008 and December 2014;another 20 renal specimens were get as the control group from patients with renal carcinoma,the renal tissues were obtained 2cm far from the tumor and proved as normal tissue.Immunohistochemical detection was carried out to analyze the expression level of NLRP3,Caspase-1,and IL-1β in the 40 renal samples.Animal experiment:fourteen male SD rats were randomly divided into calcium oxalate stone group and control group.For calcium oxalate stone group we established an ethylene glycol method induced hyperoxaluric rat model featured by crystalline material within tubule lumens;for control group normal feeding was performed.After 6 weeks,all rats were sacrificed,and the kidneys were harvested for further experiments.HE staining and Pizzolato staining were used to detect calcium oxalate crystals within tubule lumens.Western boltting and RT-PCR was applied to detect protein level and mRNA quantity of NLRP3,Caspase-1,and IL-1β from tissue lysates in rat model.Results In renal tissue samples obtained from patients with calcium oxalate stone disease,we demonstrated that the expression level of NLRP3,Caspase-1,and IL-1β were above to the normal renal tissue samples.We established a hyperoxaluric rat model character with crystalline material within tubule lumens examined by renal histology with HE staining and Pizzolato staining.And we detected that the protein and mRNA levels of NLRP3,Caspase-1 and IL-1β were remarkably increased in the lysates from the hyperoxaluric rat model (P < 0.05).Conclusions The NLRP3 inflammasome has overexpression in the renal tissue of patients with calcium oxalate stone as well as in the renal tissue of hyperoxaluric rat,and it provides a new thought to reveal the formation of calcium oxalate stone.
7.Clinical efficiency of deferred limited TURP in the treatment of urinary retention after 125I seed implantation

Objective To evaluate the clinical efficiency and complications of deferred limited TURP for treating urinary retention after 125I seed implantation.Methods From Jan.2006 to Jan.2014,36 prostate cancer patients with severely dysuria or retention were performed with deferred limited TURP 6 months after 125I seed implatation.The average age was 66 (57 to 82) years.The average PSA was 8.5 (3.5 to 25.6) μg/L before seed implantation.The average prostate volume was 78 (45 to 110) ml.Before limited TURP,the average IPSS was 16.5 (13 to 32),the average QOL score was 5.5 (5 to 6),the Qave was 5.6 (2 to 9) ml/s,the average PVR was 285 (120 to 550) ml.The urination state,QOL and complications were evaluated the second day after catheter removal and one,three and six months after limited TURP.Results Limited TURP was successfully performed in all 36 patients.The average operation time was 45 (35 to 60) min.The average fellow-up time was 42 (6 to 84) mon.The second day after catheter removal,the average IPSS was 4.5 (3 to 6),the average QOL score was 2 (1 to 3),the Qave was 14.5 (12 to 21) ml/s,the average PVR was 35 (20 to 50) ml.One month later,the average IPSS was 3.5 (2 to 5),the average QOL score was 2.0 (1 to 3),the Qave was 15.5 (12 to 23) ml/s,the average PVR was 30 (20 to 40) ml.Three months later,the parameters continued to improve and stabilized.The second day after catheter removal and one,three and six months after limited TURP,all parameters had significant improvement compared with those before limited TURP with statistical significance (P < 0.05).Four cases had mild incontinence,no case had urethral ischemia and necrosis.Conclusions 6 months after 125I seed implatation,prostate cancer patients with severely dysuria or retention can be safely and effectively treated with limited TURP.
8.The complete sequence analysis of Enterovirus 71 strain from the fatal case of the hand, foot and mouth disease during an epidemic of Guangdong province in 2008

Objective To understand the genetic characteristics of Enterovirus 71 ( EVT1 ) circu-lating strains of Guangdong province in 2008. Methods We isolated an EV71 strain from the fatal case of the hand, foot and mouth disease during an epidemic of Guangdong in 2008. Its complete genome was se-quenced and analyzed comparatively. Results The results showed that the full length of EV71 GDFS-3 ge-nome( not including poly A tail ) is 7405 bp. No insertion or deletion is detected in the coding region. There are several insertions and deletions in 5'and 3'UTR. Phylogenetic analysis of GDFS-3 and reference strains showed GDFS-3 strain shares the highest nueleotide homology with TW984 strain(96.0% ) but low homology with SIN5865, MS and BrCr( about 81.0% ). GDFS-3 strain also shares the highest amino acid homology with TW984 strain(99.0% ). It clustered with reference strains of CA subgenotype in the phylogenetie tree. The nucleotide identity with CA reference strains is 91.0% -95.0%. Conclusion The phylogenetic analysis based on the entire genome demonstrates that GDFS-3 strain has the nearest genetic relationship with TW984 strains ( isolated in 2004). GDFS-3 may belong to the same subgenogroup ( CA ) with Taiwan predominant strains. Otherwise,Some mutations in 5'UTR of EV71 may play the very important role in heightened viru-lence.
9.Photodynamic therapy combined with cell therapies for advanced esophageal cancer

Objective To explore the clinical efficacy and immunological effects of photodynamic therapy combined with cell therapies for advanced esophageal cancer. Methods Ninety patients with advanced esophageal cancer were collected and divided into three groups by a non-randomized controlled trial according to treatment intention. Group A (30 patients) received photodynamic therapy (PDT) alone; group B (30 patients), PDT received PDT plus cytokine-induced killer (CIK) cell therapies; and group C (30 patients) received CIK cell therapy aloen. In all the patients, the efficacy was assessed, the quality of life was documented, the immune function was detected, and the 6-month and 1-year death tolls were counted. Results The total clinical effectiveness rate was much higher in groups B and A than in group C (90.0% and 86.7% vs. 63.3%, P < 0.05), and there was no a statistical difference between group B and group A (P > 0.05). The rate of an increase in quality of life was significantly higher in group B than in groups A and C (86.7%vs. 60.0%and 33.3%, P<0.05), and it was higher in group A than in group C (60.0% vs. 33.3%, P < 0.05). As compared groups A and C, the percentages of CD3+ and CD3+ CD56+ were significantly improved in B group (P < 0.05), there was no a statistical difference between group A and group C. The 6-month survival rate did not differ statistically among the three groups (P > 0.05), while the 1-year survival rate was much higher in group B than in groups A and C (73.9% vs. 55.6% and 29.4%, P < 0.05). Conclusion Photodynamic therapy combined with cell therapies has a synergistic effect, and it enhances the overall immune function, significantly improves the quality of life and prolongs the survival period, showing a better clinical prospect.
10.First isolation and identification of Zika virus in China

Objective To establish a method for the isolation of Zika virus and to gather experi-ences for viral isolation. Methods Suckling mice at age 1-3 days were inoculated with serum samples posi-tive for Zika virus through intracranial injection. All mice were sacrificed 6 days after the injection. Viral nu-cleic acids were extracted from brain, heart, liver, spleen, lung, kidney, muscle, skin and intestine tissue samples and analyzed by real-time RT-PCR. The supernatants of brain tissues positive for Zika virus were used for subculturing. Nested PCR was performed to amplify the NS5 gene of the isolated virus. The se-quences of NS5 gene were analyzed by using MEGA6. 0 software. Results All of the tissue samples were positive for Zika virus. Higher viral loads were detected in heart and brain tissue samples with cycle thresh-old (Ct) values of 24. 4 and 25. 3, respectively. The second generation of Zika virus was identified in suck-ling mice brain tissues 2 days after infection by using real-time RT-PCR. The amplified product of nested PCR was 972 bp in length. Sequencing analysis showed that the isolated Zika virus ( GDZ16002 strain) be-longed to the Asian lineage. Conclusion A strain of Zika virus was successfully isolated in China by using intracranial injection via a suckling mouse model. The isolated Zika virus belonged to the Asian lineage.
