1.Evaluation on the quality of cryoprecipitate prepared from fresh frozen plasma via two methods

Objective To evaluate the quality of cryoprecipitates prepared from fresh frozen plasma via deepfreeze method and siphonage method thawing at 4℃ water bath,and to select a proper preparation method for cryoprecipitates.Methods Sixty samples of cryoprecipitate prepared by deepfreeze method,and 60 samples of cryoprecipitate prepared by siphonage method were selected randomly.Factor FⅧ,fibrinogen(Fg) in cryoprecipitates were measured by coagulometer,and the qualification rates were calculated.Results The concentration of FⅧ(IU/pack) in the cryoprecipitates prepared via deepfreeze method and siphonage method were(70.30?23.68),and(161.62?39.45),respectively;the qualification rate of FⅧ prepared via the two methods were 43.33%(26/60),and 93.33%(56/60),respectively(P0.05).Conclusion Siphonage method,with a higher qualification rate of FⅧ,is simple and fast,which deserves a popular application.
2.Estimate a method of automatic extracting genomic DNA from large numbers of whole blood samples

Objective To estimate an automatic DNA extracting method, and evaluate the quantity and quality of these DNA samples.Methods Genomic DNA was extracted automatically from blood samples by MiniPrep 75-Ⅱ workstation. The yield and purity of DNA samples were detected by UV-spectrophotometer, and the integrality of these DNA samples were measured by agarose gel electrophoresis.Results The yield of genomic DNA extracted from 100?l whole blood were (7.33?2.58)?g. The average purity of these DNA samples was 1.647?0.135 (A260/A280) and the molecular weight of them was about 21 kb.Conclusion High quality and intact genomic DNA can be extracted rapidly from whole blood by using MiniPrep 75-Ⅱ workstation, and these DNA samples were suitable for downstream molecular biology experiments.
3.Application of laparoscopy in acute abdominal emergency

Objective To evaluate clinical effects of laparoscopy in acute abdominal emergency. Methods A retrospective analysis was made on clinical date of 146 cases of acute abdominal emergency from January 2001 to August 2005,including 86 cases of acute cholecystitis,45 cases of acute appendicitis,6 cases of closed abdominal trauma,5 cases of perforation of gastroduodenal ulcer,3 cases of pyosalpinx,and 1 case of adhesive intestinal obstruction. Results Out of the 146 cases,134 cases(91.8%) were successfully diagnosed and treated under laparcoscope,including 7 cases which could not be confirmatively diagnosed preoperatively.Conversions to open surgery were required in 12 cases(8.2%).No complications or fatal cases were noted.(Conclusions Laparoscopic) technique is safe,reliable,and micro-invasive in the diagnosis and treatment of acute abdominal emergency.
4.Detection of HCV RNA in Plasma/Serum from Donors by NASBA

Objective To analyse the necessity and feasibility of HCV RNA detection for donor screening.Methods Plasma from 50 donors was mixed into a minipool, the nucleic acid was extracted by NucliSens Extractor and amplified by NASBA, the amplification products were detected by ECL. A performance control and a positive control were coupled with the samples through storage, nucleic acid extraction, amplification and detection.Results Two specimens were found HCV RNA positive in the 10000 anti HCV negative samples.Conclusion HCV RNA screening of plasma minipools by NASBA may prevent hepatitis C virus infection.

Wether cancer patients can re-adapt to life or not after experiencing tremendous frustration is the focus of tumor care nowadays.Since the measurement methods and outcome indexes of cancer patients adaptation level are different,adaptation related factors and predictors are controversial and there is not yet a unified opinion.Currently,it is thought that demographic information (like age,gender,education),disease-related information (like physiological indicators,treatment options,body image and sex) are closely related to cancer patients adaptation after stress.However there are huge differences among the extents and effects.
7.Ultrafiltration of Jinlida Granule with ceramic membranes

AIM:To observe the influence of the different bore diameter ceramic membranes on the processing parameters of Jinlida Granules(Radix et Rhizoma Salviae miltiorrhizae,Radix Sophorae flavescentis,Rhizoma Polygonati,etc.) and optimize the process of membrane filtration. METHODS: Three different membranes were tested to observe the changes in membrane flux and the content of effective components by Jinlida Granules extract. RESULTS: The 50 nm membrane had the great flux,the transfer rates of matrine and salvianolic acid were the highest;The optimum conditions: the operation differential pressure was 0.08~0.12MPa;the operation temperature was 50~60℃;the membrane surface flow rate was 3.0 m/s.When the volume of filtration solution was condensed to one tenth,adding the same volume aeionized water,and the effective components transfer rate reached 80%.Using the strong acid and strong alkali to wash it in turn,the flux can revivify above 90%. CONCLUSION: Applying the ceramic membrane to ultrafiltrate Jinlida Granule is good and useful to herb purification.
8.Association of C1GALT1 gene polymorphisms with susceptibility and prognosis of IgA nephropathy on Uyghur population in Xinjiang Uyghur Autonomous Region

Objective To explore the genetic association of C1GALT1 gene polymorphisms with susceptibility and prognosis of IgA nephropathy (IgAN) on Uyghur population in Xinjiang Uyghur Autonomous Region.Methods Ninety Uighur patients with IgAN and ninety geographically and age matched healthy controls were recruited.Peripheral blood was collected from recruited individuals for DNA extracting.After amplified by polymerase chain reaction (PCR),genotyping of the four single nucleotide polymorphisms (SNPs) in C1GALT1 gene,which were rs9639031,rs5882115,rs1008898,-527A/G,were detected by direct sequencing analysis.Differences of allele and genotype frequency were analyzed between IgAN and healthy controls.Moreover,the association between these SNPs and the risk and progress in IgAN patients were further analyzed.Results (1) The I allele frequency of rs5882115 in C1GALT1 gene was significantly higher in IgAN than that in healthy controls (x2 =7.788,P =0.015),no difference in allele frequencies of rs9639031,rs1008898,-527A/G between IgAN and healthy controls was found.Under the dominant mode,the DI+ Ⅱ genotype frequencies of rs5882115 was significantly higher in IgAN than that in healthy controls (x2 =8.563,P =0.009),no difference in genotype frequencies of rs9639031,rs1008898,-527A/G between IgAN and healthy controls was found.Under the hidden mode,no difference in genotype frequencies of rs5882115,rs9639031,rs1008898,-527A/G between IgAN and healthy controls was found.Logistic single factor regression analysis showed that the risk to IgAN of whom carry I allele of rs5882115 was 2.469 times than the D allele (OR =2.469),and the risk to IgAN of whom carry DI+ Ⅱ genotype was also higher (OR =2.852).(3) No association was found between these SNPs in C1GALT1 gene and serum creatinine in IgAN.Conclusion Association between rs5882115 in C1GALT1 gene and Uighur IgAN susceptibility suggests that there may be variants in C1GALT1 gene or its linked genetic region,which needs further exploration.
9.The status investigation and analysis of indwelling central venous catheter infection with internal jugular vein

Objective To explore the related infection factors of the internal jugular vein indwelling central venous catheter , and make prevention countermeasures according to the infection factors of central venous catheter infections .Methods A total of 564 patients admitted in liver and gallbladder surgical ward with external jugular vein indwelling central venous catheter were se‐lected ,extract the relevant hospital infection data in patients by the XingLin hospital infection real‐time monitoring system ,and SPSS15 .0 statistical analysis was conducted .Results The infection rate of 564 cases of patients was 4 .07% .The rate of gram‐neg‐ative bacteria infection was 43 .5% ,the gram positive bacteria infection accounted for 34 .8% ,fungi accounted for 21 .7% ,including multiple drug‐resistant bacteria infection accounted for 52 .1% .Catheter indwelling 14 d or more infection rate was 8 .5% ,14 d fol‐lowing infection rate was 2 .1% ,and infected patients for more advanced cancer and patients with severe acute pancreatitis .Pipe joint respectively with heparin cap and needle positive pressure infusion joint connections ,infection rate was statistically different (P<0 .05) .Conclusion The infection of internal jugular vein indwelling central venous catheter should not be ignored ,and the oc‐currence of catheter‐related infection of patients is closely related to state of an illness ,the time of catheter insertion ,and the joint device and so on .
