1.Effect of Panax notoginseng on gastric mucosal injury, superoxide dismutase activity, malondialdehyde and nitric oxide contents in rats with water immersion restraint stress

BACKGROUND: Panax notoginseng is an effective medicine for curing gastric mucosal ulcer. There are many clinical reports that Panax notoginseng protects gastric mucosa.OBJECTIVE: To investigate the protective mechanism of Panax notoginseng on the gastric mucosal injury induced by water immersion restraint stress in rats.DESIGN: A randomized controlled animal trial.SETTING: School of Basic Medical Sciences, Daqing Branch of Harbin Medical University.MATERIALS: The study was conducted in the laboratory of Department of Pathophysiology of Harbin Medical University from September 2004 to October 2005. Forty-eight Wistar rats were used, either male or female, weighing 180-230 g.METHODS: The 48 Wistar rats were randomly divided into six groups with 8 rats in each group: normal control group, stress model group, cimetidine treatment group, Panax notoginseng of low, middle and high-dose groups (4, 8, 12 mg/time). In the cimetidine treatment group, cimetidine tablets were grinded into powders, then mixed with distilled water to prepare into suspension (1 tablet:10 mL), which was perfused intragastrically (5 mL), 3 times a day; In the Panax notoginseng groups, the powders in Panax notoginseng capsules were mixed with distilled water to prepare into suspension of corresponding concentrations (0.8, 1.6, 2.4 g/L), then administrated the same as those in the cimetidine treatment group. Stress models in rats were established by means of water immersion restraint stress. The gross and pathohistological changes of gastric mucosa were observed, and the activity of superoxide oxidase (SOD) and contents of malondialdehyde (MDA) and nitric oxide (NO) were determined.MAIN OUTCOME MEASURES:①Gross and pathohistological changes of gastric mucosa;②Changes of MDA and NO contents and SOD activity in the homogenate of gastric mucosa.RESULTS: All the 48 rats were involved in the analysis of results. The gastric mucosal hemorrhage and erosion in the cimetidine treatment group were reduced obviously as compared with those in the stress model group, SOD activity was obviously decreased [(12.61±0.87), (1.03±0.60) mkat/g], whereas the NO content was a little higher [(5.76±1.35), (0.97±0.58) nmol/g]. The MDA content was obviously higher in the stress model group than in the normal control group [(3.10±1.13), (0.09±0.02) μmol/g, P<0.01]. There were no obvious differences between the Panax notoginseng groups and the cimetidine treatment group except that the NO contents were decreased in the in Panax notoginseng groups.CONCLUSION: Gastric mucosal injury induced by water immersion restraint stress can be significantly protected by Panax notoginseng, which is not dose-dependent. The protective mechanism may be associated with that Panax notoginseng can eliminate the product of oxygen-derived free radicals, and it is not totally the same as that of cimetidine
2.Effects of advanced glycosylation end products on interactions between cis-elements and trans-acting factors of hepatic insulin receptor gene

Objective To explore the effects of advanced glyeosylation end products (AGEs) on the binding of hepatic nuclear factors to the human insulin receptor (hlR) gene promoter. Methods The oligonueleotides with hlR gene promoter activity, 42 bp (spanning -1 441 to -1 400, US1) or 146 bp (spanning -638 to -493), were artificially synthesized, with point mutations at 5 key G residues in 42 bp US1 m5 oligonucleotides. US1 and rat hepatic nuclear extracts (HNE) were incubated with glucose 6-phosphate, prior to non-competition and competition gel retardation electrophoresis. Results Competition gel retardation electrophoresis showed that the binding capacity of 32p-labeled US1 probe to HNE could be signifficantly decreased with increased US1. US1-AGEs and US1m5 decreased the binding of probe to HNE as well, but only partly affected the electrophoretie bands [1,5,10 ng US1-AGEs: (41.08±2.86)%, (27.64±2.92)%, (15.35±1.81%) vs (52.05±1.79)%]; 5,10 ng US1m5: (5.20± 1.03)%, (1.81±0.21)% vs (52.05±1.79)%]. AGEs formed on HNE could increase the binding of HNE to probe, along with nonspecifie binding increasing. Conclusion The impact of AGEs on hlR gene expression seems to be related to the effects on cis-elements and trans-acting factors.
3.Study of gastroenteric mucosa immune responses in Balb/c mice by oral immunization with helicobacter pylori

Objective To study the mucosa immune responses of gastric and intestinal mucosa in Balb/c mice administered orally with Hp sonicate and mucosal adjuvant(LT).Methods The changes of antigen specific AFC in gastric and intestinal mucosa were detected by ELISPOT assay. Results The numbers of sIgA and IgG AFC rise significantly in PP and gastric mucosa, especially the numbers of sIgA-AFC, significant differences were observed between two immunized groups and the control. Conclusions Locally synthesized specific sIgA antibodies contribute to immunity against gastric helicobacter infection.
4.Cyclic tensile stress effects on the expression of connective tissue growth factor in human periodontal ligament fibroblasts

BACKGROUND:Previous studies have found that cyclic tensile stress can induce the proliferation of human periodontal ligament fibroblasts in a certain time. OBJECTIVE: To observe the effect of cyclic tensile stress on the expression of connective tissue growth factor in human periodontal ligament fibroblasts, and to clarify the role of JNK, p38MAPK, PI3K pathway in the cyclic tensile stress-induced expression of connective tissue growth factor in human periodontal ligament fibroblasts. METHODS:Human periodontal ligament fibroblasts culturedin vitro were given stimulation of cyclic tensile stress in vitro for 1, 6, 12, 24 hours using multi-channel cel stretch stress loading system (experimental group). Control group was set up and not given any stimulation at the same time. The cels under stress application for 12 hours were given specific inhibitors of JNK, p38MAPK, PI3K respectively at the beginning, and were compared with those with no inhibitors. The concentrations of connective tissue growth factor in the culture supernatant were measured by ELISA. The mRNA expression of connective tissue growth factor in human periodontal ligament fibroblasts was detected by real-time RT-PCR. RESULTS AND CONCLUSION:Compared with the control group, the expression of connective tissue growthfactor in human periodontal ligament fibroblasts under cyclic tensile stress began to increase at 1 hour, increased manifestly at 6 hours, reached the peak at 12 hours, and then decreased at 24 hours. The expression of connective tissue growth factor was reduced by the specific inhibitors of JNK, while the specific inhibitors of p38 MAPK, PI3K had not the same effect. In a certain time, the cyclic tensile stress can induce a time-dependent increase in mRNA and protein levels of connective tissue growth factor. Along with the extending of time, the expression of connective tissue growth factor begins to decrease. Cyclic tensile stress can regulate expression of connective tissue growth factor in human periodontal ligament fibroblasts via the JNK pathway.

Objective To access the relationship between the polymorphism of G protein-coupled receptor kinases 4(GRK4,including R65L,A486V and A142V) and essential hypertension.Methods All the literature on the subjects was searched by keywords such as essential hypertension,hypertension,G protein-coupled receptor kinases 4 gene,and polymorphism in the electronic databases,Medline of Year 1997 to 2006.RevMan 4.2 software was used for Meta analysis.Results A total of 857 cases and 913 controls from 4 studies were included.The ORs of genotypes of R65L,A142V and A486V were 0.97(0.75 to 1.25,P=0.81),0.81(0.30 to 2.08,P=0.65) and 1.48(1.16 to 1.8,P=0.001) respectively.Conclusion GRK4 gene polymorphism A486V is significantly associated with susceptibility of essential hypertension.People who carries A486V have a higher risk to be subject to essential hypertension.

Objective To study the influence of continous ven ovenuous hemofiltration on patients with multiple organ dysfunction syndrome.Methods 64 patients with MODS were divided into CVVH group(34) and IHD group(30)between 1999-12~2003-12.The serum BUN,Cr,iron,blood pressure,ar rhythmia were monitored during treatment and KT/V was also calculated after each therapy.APACHE-Ⅱ score was assessed before and after the treatment and the pr ognosis of each patient was recorded as well.Results CVVH group had more stable hemodynamics than IHD group; there was no significant difference in the removal of BUN and Cr in the two grou ps.The survival time of CVVH group was longer than IHD,and APACHE-Ⅱ score of C VVH group was lower than that in IHD group,17.23?6.86 verse 20.91?5.77 ( P
9.Pure Word Deafness after Cerebral Hemorrhage in the Right Temporal and Parietal Lobe: a Psychophysical Analysis

Objective:To study the neuropsychological features and explore possible mechanisms in pure word deafness.Methods:A case of a 55 year-old right-handed woman was analysed,who presented with a long lasting pure word deafness following subcortical and cortical hemorrhage in right temporal and parietal lobe.Results:There was no sign of aphasia. The auditory deficit was sepecific for language, while recognition of music and environmental sounds was normal. CT examinations showed that the lesion was due to right cerebral hemisphere hemorrhage, mainly the white matter of temporal and parietal lobe. Conclusion:It is proposed that pure word deafness was consequent to the isolation of Wernicke's area from incoming auditory information due to the interruption both of the association fibers from the right auditory area traveling across the corpus callosum and of the left auditory radiations.
10.Augmentation index's change and its influencing factor in patients on continuous ambulatory peritoneal dialysis

Objective To analyze the factors influencing augmentation index (AI) by examining the AI changes in patients who received continuous ambulatory peritoneal dialysis (CAPD) therapy, and evaluate the artery elastic function situation. Methods The height, weight, blood pressure, blood indicators were detected in 46 CAPD patients (CAPD group) and 42 controls (control group). AI was managed by OMRON HEM-90OOAI, and the influencing factor was analyzed. Results Systolic blood pressure(SBP),diastolic blood pressure(DBP), pulse pressure, central aortic pressure and AIp75 (AI value, when pulse was 75 beats/min) in CAPD group were significantly higher than those in control group. Within CAPD patients,the AIp75, in female was significantly higher than that in nude [(96.67±8.95 )% vs (89.89±6.46)% ,P =0.022]. Simple correlation showed positive relationship in gender, age, SBP, DBP, pulse pressure and central aortic pressure with AI; and negative relationship in height, weight and weight index with AI; while there was no relationship between AI and all kinds of chemical examination indicators or dialysis years.Multiple linear regression analysis evidenced that gender,age, height, DBP and central aortic pressure of CAPD patients had linear regression relation with AI. Conclusions It indicates a decreased artery elasticity and an increased stiffness occurs in CAPD patients. AI has tight correlation with age, height, DBP and central aortic pressure, and is an non-invasive factor thai partially reflects the arterial elasticity. Central aortic pressure is relevant with aortic elasticity and rigidity closely. Measuring central aortic pressure is very imporrant for preventing cardiovascular accident.
