BACKGROUND: The relation of epilepsy to immunity has been investigated at cellular and molecular levels in recent years, and the results show many immunological changes in epileptic patients such as immunocytes,immune molecules and immune functions.OBJECTIVE: To investigate the immunologic pathogenesis of epilepsy and expression of MHC- Ⅰ, MHC- Ⅱ and C3b receptor of microglia. DESIGN: Randomized and controlled trial. SETTING: Medical College of Dalian University; University of Sanchong,Japan.MATERIALS: The experiment was conducted in the Immunology Laboratory of Jilin Beihua University from 2001 to 2002. Forty adult Wistar rats were randomized into model group and control group with 20 in each group.METHODS: 12 mg/kg kainic acid was administered subcutaneously into rats in model group, while no intervention was given to rats in control group. Seizure was observed within 6 hours following kainic acid administration. Rats were killed 3 days after medication. Neuronal degeneration was observed with crystal violet staining and the expression of MHC molecules and C3b receptor in the hippocampus of epileptogenic rats was observed immunnhistochemically.administration, spasm occurred within 3 hours after kainic acid administration and continued from the first stage to the fifth stage. More severe served in cone-like cell layer of hippocampus of rats in model group, but molecule, MHC- Ⅱ molecule and C3b receptor was (201.6±6.43), (493.8±7.92) and (362.5±3.18) cells per visual field respectively in the hippocampus of epileptogenic rats inmodel group, but no obvious expression was observed in control group. The differences were significant between the two groups (P ＜ 0.01).pocampal sclerosis, immunological inflammation is observed combined with the complement system, which indicates immunologic inflammatory mechanism of epilepsy.

BACKGROUND: The recent researches show that there are various kinds of autoantibodies in epilepsy patients such as anti-nuclear antibodies, anticardiolipin antibodies,anti-?2 glucoprotein antibodies and anti-glutamic acid decarboxylase antibodies,which indicate that some types of epilepsy are mediated by autoimmune mechanism.OBJECTIVE: To detect the level of anti-glutamic acid receptor (GluR1)antibody in epilepsy children.DESIGN: A controlled trial.SETTING: Medical College of Dalian University, General Hospital of Jilin Chemistry Group and Sanchong University, Japan.PARTICIPANTS: Totally 56 cases of epilepsy children were from Maternal and Child Health in Qingdao City including 30 males and 26 females and aged from 3 months to 13 years.While 48 health examinees and 12 patients with brain tumor from General Hospital of Jilin Chemistry Group were taken as controls, including 38 males and 12 females and aged from 6months to 17 years.Their guardians were all informed of the detection index and consented to join the study.METHODS: Polystyrene plates were coated with GluR1 polypeptide antigen,and optimal conditions of enzyme linked immunosorbent assay (ELISA) for GluRI antibody was ascertained with chessboard titration.MAIN OUTCOME MEASURES: The GluR1 antibody level was detected with ELISA.RESULTS:The concentration of coating GluR1 polypeptide antigen was20 mg/L; The dilution of serum and horseradish peroxidase sheep anti-hu-man (HRP-SaH) IgG were 1:50 and 1:1 500 respectively. Intra-assay andinter-assay coefficientof variability were 4.48％-8.80％ and 11.18％-16.60％ respectively; The specific absorption test demonstrated that the ab-sorbance value of positive serum decreased 2.4 times after absorption ofGluR1 polypeptide compared with that before absorption.There was obvi-ous higher positive rate of GluR1 antibody in serum of epilepsy childrenthan control group (40％, 5％, P＜0.01).CONCLUSION:①ELISA is specific and stable for GluR1 antibody in serum. ②There is the possible autoimmune response of GluR1 antibody in serum of epilepsy children.

Objective:To apply immuno-PCR for detection of anti-Sm antibodies.Methods: Immuno-PCR was established for anti-Sm antibodies in SLE sera using SaHIgG-DNA probe. The experimental conditions were optomized and methological identifieation indicated obvious in crease(×107 ) in sensitivity compared with that of ELISA. The positive rate of anti-Sm antibodies was 54.9% in SLE sera, and significantly higher than that of ELISA. Intra-and inter-CV were 2.3%～4.8% and 3.7%～6.9% respectively. Immuno-PCR was positively correlated with ELISA( p ＜0.001). Therefore, immuno-PCR for anti-Sm antibodies is specific,sensitive,reproducible and practical clinically.

Based on the medical immunology exquisite course of Dalian city,we expound the guiding thought,construction and system evaluation of medical immunology test bank,in order to normalize examination,consummate teaching quality control system and improve teaching quality.

Based on our school's specific conditions, to make full use of the laboratory, improve students' ability of practice and innovation, for the past few years, Medical College of Dalian University has intensified the soft and hard ware construction of the labs, has done some exploration in the management structure and mechanism innovation of laboratory, and has constructed various experimental and innovative platform and experimental teaching reform etc.The effect of lab construction has been em erging and the quality of students has been bettered greatly.

This study was aimed to explore the antitumor effect of volatile oil of X ihuang pill and its immune mecha-nism in order to screen the antitumor active site of Xihuang pill. Among 70 female Wistar rats, 10 rats were random-ly selected as the blank control group; and the other 60 rats was used in the establishment of walker 256 breast can-cer cell tumor-bearing rat models. The model rats were randomly divided into the negative control group (model group), high-dose volatile oil group, middle-dose volatile oil group, low-dose volatile oil group, high-dose Xihuang pill group, and lentinan group (positive control group), with 10 rats in each group. The intragastric administration was given twice a day for 14 days. Blood was taken from the abdominal aorta. Tumor tissues was removed and weighed to calculate the tumor inhibitory rate. ELISA method was used to detect the level of IL-2, IL-6, IL-10, IFN-γ and TGF-β in peripheral blood. The flow cytometry was used to detect the content of CD3+ T cell, CD4+ T cell, CD8+ T cell, and B7-1 cell (CD80). The results showed that the tumor inhibitory rates of volatile oil of high-dose group and middle-dose group were 28.4% and 24.1%, respectively. Compared with the model group, the average level of IL-2 and IFN-γ of volatile oil of high-dose group and middle-dose group and CD3+ T cell, CD8+ T cell, B7-1 cell con-tent were obviously increased (P< 0.05). It was concluded that volatile oil of Xihuang pill had certain antitumor ef-fect, which was one of the antitumor active sites of Xihuang pill. The volatile oil of Xihuang pill upregulates the lev-els of IL-2 and IFN-γ, as well as the contents of CD3+ T cell, CD8+ T cell, B7-1 cell in order to increase the im-mune clearance function of tumor-bearing rats.

Objectlve To detect the expression of CXCR3 mRNA in peripheral blood mononuclear cells (PBMC) in patients with primary biliary cirrhosis (PBC) ,and explore its relationship with activity of PBC.Methods Reverse transcription-real time quantitative polymerase chain reaction (FQ-RT-PCR) was used to examine CXCR3 mRNA expression in peripheral blood monocytes of 29 cases of PBC in active stage. 30 cases in stable stage,20 cases in CTD,and 30 healthy controls.Results The mean level of CXCR3 mRNA expression of PBc inactive stage (△Ct=5.41±2.69) Was higher than that of stable stage (△Ct=7.77±2.74,t=3.39,P＜0.01),CTD(△Ct=7.24±2.75,t=2.53,P＜0.01),and healthy controls (△Ct =7.16±2.76,t=2.45,P＜0.01).There was no significant difference of expression levels of CXCR3 mRNA among stable stage PBC patients,CTD diseases patients and healthy controls (P＞0.05).Conclusion This study indicated that the CXCR3 mRNA expression levels of PBMC is significantly elevated in patients with active PBC.and it could be implicated in pathogenesis and activity of disease.

Objective:To investigate the effects of teaching reform to strengthen the integration of basic medicine and clinical medicine by analyzing the scores of the National Medical Licensing Examination from 2014 to 2018.Methods:Through the improvement of the training program, the establishment of subject integration courses, the standardization of case teaching mode and the establishment of comprehensive medical experiment courses, the teaching reform of the combination of basic medicine and clinical medicine was carried out. The differences in the scores of the National Medical Licensing Examination between the students from Dalian University and those from the whole country were compared, including the average total scores, the passing rates, the average mastery rates of the basic medicine, and the average mastery rates of the cognitive level of the basic medicine.Results:In 2014, 2015 and 2016, the average total scores of the students from Dalian University were 384.70±53.62, 395.45±50.49, and 401.43±50.88, respectively, which were lower than those from the whole country (396.15±58.85, 400.78±56.88, and 405.15±58.06). The average mastery rates of the basic medicine of the students from Dalian University were 56.28%, 62.45%, and 64.92%, respectively, lower than those from the whole country (58.41%, 62.46%, and 65.36%). The average mastery rates of the basic medicine of the students form Dalian University from 2014 to 2016 were lower than those from the whole country, and those were higher in 2017 to 2018 than the whole country. The average total scores of the students form Dalian University in 2017 and 2018 were 397.11 and 400.26 ± 61.15, respectively, higher than those from the whole country (388.91 and 397.01±58.35). The average mastery rates of the basic medicine of the students form Dalian University were 61.53% and 59.85%, respectively, higher than those from the whole country (58.64% and 58.38%). The total passing rates of the students from Dalian University in 2014 and 2016 were 91.11% and 72.73%, respectively, lower than those from the whole country (91.18% and 73.75%), while in 2015, 2017 and 2018 those were 76.47%, 77.78%, and 77.59%, respectively, all higher than those from the whole country (73.59%, 74.29%, and 76.89%). All the data showed a trend of increase with the teaching reform process.Conclusion:Strengthening the teaching reform of the combination of basic medicine and clinical medicine during the learning stage of basic medicine could improve students' learning interest and attention to the basic medical knowledge, strengthen the understanding of the relevance of basic medicine to clinical medicine, and play a positive role in the cultivation of medical students' post competency and the achievement of the goal of training applied medical talents.

Objective :To analyze correlation among fasting (FBL) and postprandial blood lipids (PBL) ,blood lipid fluctuation (absolute value of PBL‐FBL) and severity of coronary artery disease .Methods :Cross‐sectional study was performed among 264 patients undergoing coronary angiography (CAG) in our hospital .According to percutaneous coronary intervention (PCI) or not based on CAG results ,patients were divided into plaque group (n＝128) and PCI group (n＝136).Gensini score was used to assess severity of coronary artery disease .Blood lipid levels and its fluctu‐ation were compared between two groups .Correlation among blood lipid levels ,blood lipid fluctuation and severity of coronary artery disease were analyzed .Results :Compared with plaque group ,there were significant rise in per‐centages of men and smokers ,waist circumference ,levels of postprandial‐fasting (P‐F ) serum LDL‐C (ΔLDL‐C ) and P‐F plasma apolipoprotein B (ΔApoB ) , and significant reduction in plasma level of P‐F apolipoprotein A1 (ΔApoA1) in PCI group , P＜0. 05 or ＜ 0. 01. Pearson correlation analysis indicated that serum fasting and post‐prandial HDL‐C levels ,plasma fasting and postprandial levels of ApoA1 and ΔApoA1 were significant inversely cor‐related with Gensini score ( r＝ －0. 130～ －0.218 , P＜0. 05 or ＜0. 01) ,and levels of plasma fasting lipoprotein a (Lp (a)) ,serum fasting and postprandial levels of free fatty acid (FFA) ,serum P‐F FFA (Δ FFA) were significant positively correlated with Gensini score ( r＝0. 139－0. 176 , P＜0.05 or ＜0.01).Multifactor linear regression anal‐ysis indicated that postprandial serum HDL‐C was protective factor for Gensini score (B＝ －22.274 , P＝0.002 ) , while postprandial serum FFA ,Δ FFA ,waist circumference and hyperlipidemia history were its influencing factors (B＝0. 388～24. 135 , P＜0. 05 or ＜0.01).Conclusion :Measurements of fasting and postprandial blood lipid levels and their fluctuation contribute to more comprehensively and objectively assessing blood lipid levels and severity of coronary disease in patients with coronary artery disease .
