2.Experimental evaluation of the antihistamine effect of domestic azelastine hydrochloride

AIM: To evaluate the antihistamine effects of domestic azelastine hydrochloride. METHODS: Histamine increased skin vascular permeability and induced shock model in guinea pigs. In isolated guinea pig ileal rings, the contraction of smooth muscle induced by histamine was determined. RESULTS: Pretreament with azelastine hydrochloride( 0.05, 0.15,and 0.45 mg?kg -1,ig) significantly inhibited the increase in skin vascular permeability induced by histamine in a dose-dependent manner in guinea pigs. Pretreament with azelastine hydrochloride( 0.05, 0.1,and 0.2 mg?kg -1,ig)produced a significant improvement of shock, as shown by a decrease in reactivity degree, mortality and a prolongation of latency. In isolated guinea pig ileal rings, azelastine hydrochloride (10 -8,3?10 -8,10 -7,3? 10 -7 mol?L -1)significantly inhibited contraction of smooth muscle induced by histamine in a concentration-dependent manner and caused a parallel right shift of the dose-effect curve of histamine (pA 2= 8.55). CONCLUSION: Domestic azelastine hydrochloride shows significant the antihistamine effects.
3.Effect of initial treatment protocol on prognosis of pneumonia in the kidney transplant recipients

Objective To explore the effects of different initial treatment protocols on the prognosis of pneumonia in the kidney transplant recipients. Methods Sixty-seven cases of pneumonia following kidney transplantation were divided into case group (34 cases) and control group (33 cases).The patients in case group were treated with Imipenem and Cilastatin, SMX/TMP and ganciclovir,and those in control group received routine treatments. Mortality and length of stay in hospital (LOS)were analyzed. Results Mortality (5.9 % ) and LOS (15 days) in case group were reduced as compared with those in control group ( 18. 2 %, 23 days, respectively) (P< 0. 05). Among the patients without severe pneumonia, the mortality and incidence of LOS beyond 21 days in case group (3. 4 %, 1/29; 3. 4 %, 1/29) were lower than those in control group (17. 2 %, 5/29; 37. 9 %, 11/29, respectively) (P<0. 05). Whether the patients were admitted to hospital sooner or later, the incidence of LOS beyond 21 days in case group was lower than that in control group (P<0. 05).Whether the etiologies were determined or undetermined, the incidence of LOS beyond 21 days in case group was lower than that in control group (P<0. 05). The incidence of LOS beyond 21 days in case group was lower than that in control group among the patients with cardiopulmonary disease (P<0. 05). Conclusion The initial appropriate treatment would improve the prognosis of pneumonia in the kidney transplant recipients.

With the development of science and technology, new medical equipments is toward the direction of intelligent and portable. In order to assist medical personnel to patients with blood, developing from previous devices, a new kind of vein locating projection instrument based on two-dimensional scanning mirror is put forward. It can scan and project vein image using a scanning mirror. The related algorithm is also be improved, make vein scan projection more practical. The system finally set up can perform well in vein scan projection.

5.Influence of times of CIK cell treatment on survival of elderly patients with gastric cancer

Objective To investigate the relevance between the times of cytokine-induced killer cell (CIK cell) adoptive immunotherapy and the survival of the elderly patients with gastric cancer.Methods Lymphocyte separation medium was used to isolate mononuclear cells, and then the cultured CIK cells were infused back into the patients with gastric cancer. A retrospective cohort study was adopted by using Kaplan-Meier to estimate median survival time and survival rate, using Log-rank test to analyze the impact of clinical factors on survival rate, and using RR and 95% CI to estimate the contact intensity of death outcome, survival time and CIK cell treatment. Results There were nostatistically significant differences in gender,age, tumor site, histological type, invasion depth, lymph node metastasis, pathological grade, tumor size or tumor distribution between chemotherapy group and CIK treatment group (all P>0.05). The median survival time of patients with gastric cancer was significantly longer in CIK treatment group than in chemotherapy group (61 vs. 21, χ2=10.215, P=0.001). Compared with the patients treated by chemotherapy alone, the increased times of CIK treatment induced more survival rate and 2-5 years life spans (χ2=12. 461, P=0.006). Conclusions With the treatment that CIK cells are infused back into the elderly patients with gastric cancer, the risk of death is reduced, and the lifespan is prolonged, which is associated with the CIK cell treatment times.
6.Docetaxel combined with cisplatin and fluorouracil in the treatment of advanced gastric cancer

Background and purpose:Gastric cancer is one of the leading causes of cancer death throughout the world.When compared with optimal supportive care alone,combination chemotherapy yields a significant advantage in the management of advanced gastric cancer.However,no single regimen has emerged or been accepted as being clearly superior over another.Here we investigated the efficacy and safety of "docetaxel+ cisplatin + fluorouracil" 5-day combination chemotherapy as treatment in patients with advanced gastric cancer.Methods:Between 2004 January and 2005 July,we enrolled 30 patients [males 22,median age 53 years(range 28-72)] with advanced gastric cancer.The regimen consisted of docetaxel 75 mg/m~(2) on day 1,cisplatin 15 mg/ m~(2) on days 1-5,and fluorouracil 500 mg/ m~(2) on days 1-5,every 3 weeks.All patients received over two cycles of this regimen.Results:A total of 99 cycles were administered.Mean cycle number per patient was 3.3.The administered dose intensity of docetaxel was 23.5 mg/m~(2)/week,fluorouracil 735 mg/ m~(2)/week and cisplatin 14.7 mg/ m~(2)/week,which corresponded to 94%,97.5% and 92.3% of planned doses.Of these patients,16(53.3%) achieved a partial response,8(26.6%) stable disease,and 6 patients(20%) showed progressive disease.The median time to progression was 5.1 months.(95% CI 4.1-6.0 months).Median overall survival was 9.9 months.(95% CI 8.1-11.6 months).Leukopenia occurred during 36.7% of cycles(36 of 99 cycles);18.2% grade Ⅰ,10.1% grade Ⅱ,5.0% grade Ⅲ and 3.0% grade Ⅳ.Anemia occurred in 12.1%(12 of 99 cycles);7.1% grade Ⅰ and 5.0% grade Ⅱ.Thrombocytopenia was 15.2%(15 of 99 cycles) and all were grade Ⅰ or Ⅱ.Diarrhea,stomatitis and hypersensitivity occurred in 16.7%(5 out of 30 patients),respectively.Neutropenic fever occurred in two patients(6.7%) and myalgia in nine(30%).Conclusions:Docetaxel combined with fluorouracil and cisplatin is an active and tolerable regimen for the treatment of patients with advanced gastric cancer.
7.Effect of bacailin on the angiogenic regulating factor and migration of laryngeal cancer cells

Objective To explore the effects of bacailin on the angiogenic regulating factor and migration activity of laryngeal cancer cells.Methods Hep-2 was cultured in vitro,(1)in the groups adding phorbol ester,gelatin-zymogram and Transwell assay were used to observe the effects of bacailin on the gelatinase activity of matrix metalloproteinase (MMP) and the migration ability of Hep-2,respectively; (2)in the group without phorbol ester,enzyme-linked immunosorbent assay (ELISA) was used to detect the effects of bacailin on the protein level of endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF).Results Among the control group,the low-dose group,the middle-dose group,and the high-dose group,the activity of MMP-3 of each corresponding group was (100.0 ±0.0) %,(95.0 ±4.9) %,(93.1± 6.2)%,and (90.7 ± 7.3)% with statistically significant difference (t =2.123,2.309,2.412,P <0.05) ; the activity of MMP-9 of each corresponding group was (100.0 ± 0.0) %,(92.3 ± 7.5) %,(89.5 ± 9.3) %,and (85.6 ± 6.1) % with statistically significant difference (t =2.253,2.302,3.708,P <0.05 or P <0.01) ;the protein level of VEGF of each corresponding group was (242.7 ±9.5)ng/L,(230.6 ± 12.7)ng/L,(212.1 ± 15.9)ng/L,and (184.2 ± 23.5)ng/L with statistically significant difference (t =2.408,3.733,4.146,P <0.05 or P <0.01) ; the protein level of bFGF of each corresponding group was (190.7±8.2) ng/L,(181.2±13.0) ng/L,(169.9±17.3)ng/L,and (140.7±9.2)ng/Lwith statistically significant difference (t =2.330,2.922,5.145,P <0.05 or P <0.01) ; Cellular migration rate of each corresponding group was (38.3 ± 5.6) %,(32.9 ± 7.1) %,(27.9 ± 4.4) %,and (23.3 ± 6.0) % with statistically significant difference (t =2.141,3.365,4.027,P < 0.05 or P <0.01).Conclusions Bacailin could suppress the migration of Hep-2 through inhibiting expression of angiogenic regulating factor,which might has clinical value in the treatment of laryngeal cancer.

Objective To studyeffect of different processes for preparation ofβ- cyclodextrin inclusion pericarpium citri reticulatae - Radix Aucklandiae essential oil.Methods Extract pericarpium citri reticulatae-radix aucklandiae essential oil with steam distillation method, conductβ- cyclodextrin inclusion of pericarpium citri reticulatae-radix Aucklandiae essential oil with ultrasonic method, grinding method andsaturated aqueous solution method, and detect inclusion effect respectively by polarized light microscopy observation method and TG-DTA analysis method.Results Polarized lightphenomenon disappears under a microscope for the three inclusion preparation products, and TG-DTA curve of ultrasound method is superior to that of saturated aqueous solution method and grinding method.Conclusion Polarized light microscope observation method and TG-DTA method enjoysa good effect in determination of cyclodextrin inclusion, which provides a new method for determination of cyclodextrin inclusion results.
9.Impact of oral contrast agent for assisting in outlining small bowel on pelvic VMAT dose in patients with cervical cancer

Objective To investigate the impact of oral contrast agent for assisting in outlining the small bowel on pelvic volumetric modulated arc therapy (VMAT) dose in patients with cervical cancer.Methods Nine cervical cancer patients for postoperative radiotherapy underwent CT scans,and the target volumes and organs at risk including the small bowel were contoured.The VMAT plan was designed for each case.Then another plan was generated by re-calculating the radiation dose after changing the electron density of the small bowel.The first plan (plan A) was the conventional VMAT plan,and the second one (plan B) specified the electron density of the small bowel.Paired t-test was used to compare the dose distribution between the two plans.Results The Dg8,D5o,conformity index,and homogeneity index of plans A and B were 4 989.1 vs.5 000.1 cGy (P =0.026),5 208.6 vs.5 191.6 cGy (P =0.005),0.766 vs.0.765 (P =0.920),and 0.081 vs.0.074(P =0.055),respectively.The volumes of the small bowel receiving at least 30 Gy for plans A and B were 309.3 vs.314.3 cm3(P =0.207),while bladder V45 of the two plans was 52.4% vs.51.1% (P =0.168).To achieve the same prescribed dose,plan A and plan B needed 893.3 MU and 865.8 MU (P =0.093).Conclusions The contrast agent filling the small bowel does not lead to a significant increase in the pelvic VMAT dose in patients with cervical cancer after surgery.
10.Relationships between T and NK cells in peripheral blood and the relapse or metastasis of malignant tumors

ObjectiveTo investigate the relationships between T and NK cells in peripheral blood and the relapse or metastasis of malignant tumors.MethodsFlow cytometry was used to test the percentages ofT and NK cells in peripheral blood of 48 malignant tumor patients with relapse or metastasis (relapse or metastasis group) and 48 malignant tumor patients without relapse or metastasis(control group).ResultsCompared with control group,the ratio of Th/Ts decreased,CD4+/CD25+ raised and the activities of NK cells decreased in relapse or metastasis group (0.95 ±0.52 vs.1.35 ±0.43,7.15 ±3.81 vs.6.01 ±2.67,0.2053 ±0.1135 vs.0.2501 ±0.0745)(P<0.01 or <0.05).ConclusionsTesting the T and NK cells in peripheral blood of malignant tumor patients regularly can know their immune status and find the relapse or metastasis early.It is easy and useful in patients' follow-up.
