1.The effect of artesunate on heat shock protein 47 expression in rats of pulmonary fibrosis

Objective To investigate the effect of artesunate on the expression of heat shock protein 47(Hsp47)in pulmonary fi-brosis in rats.Methods Thirty SD male rats were randomly divided into control group,model group and artesunate intervention group (intervention group).Rats in control group were intrachacheally instilled with saline (0.4 mL),while rats in other two groups 5 mg/kg bleomycin.Rats in both control group and model group were intraperitoneal injected with 1 mL saline per day sub-sequently.And the rats in intervented group were intraperitoneal injected with 10 mg/100 g artesunate per day.The body weight of day 0,day 14 and day 28 were recorded.All the rats were sacrificed on day 28.The pulmonary coefficient and the hydroxyproline content were observed.The expression of Hsp47mRNA and CollagenⅠmRNA were evaluated by RT-PCR.The Hsp47 expression was also detected by Western Blot.The pathological changes were analyzed by hamatoxylin-eosin (HE)stain and Masson stain.Re-sults (1)The body weight of control group grew much faster than other groups (P <0.05).(2)Pulmonary coefficient:model group (12.31±1.89)mg/g>intervention group (8.54±0.67)mg/g>control group(4.81±0.38)mg/g (P <0.05).(3)Ashcroft Scores:model group(5.70±1.09 )>control group (3.08±0.56),model group(5.70± 1.09)>intervention group (4.01 ± 1.25).There was no significant difference between control group and intervention group(P >0.05).(4)Hydroxyproline content:model group (620.33±66.16)μg/g>control group(379.00±35.51)μg/g,model group (620.33±66.16)μg/g >intervention group(429.00± 36.51)μg/g (P <0.05),there was no significant difference between control group and intervention group.(5)Hsp47mRNA expres-sion:model group and intervention group > control group (P <0.05),there was no difference between the first two groups(P >0.05).CollagenⅠmRNA:model group > intervention group>control group (P <0.05).(6)Western blot showed that Hsp47 ex-pression of model group was the highest in all three groups.Conclusion These results indicate that artesunate might inhibit pulmo-nary fibrosis and depress CollagenⅠproduction,and it′s probably a result of Hsp47 down regulation.
2.Changes of oxygen metabolism and neutrophil cellular rheological characteristics in canine with respiratory distress syndrom induced with oleic acid

To observe the alterations of oxygen delivery/consumpation, the deformability and adherence of neutrophil in canine with respiratory distress syndrom(RDS), Method:The 15 selected healthy mongrel dogs were randomly allocated to receiving intravenous oleic acid 0.06 ml. kg~(-1) (test group, n= 8)or equivalent volume of saline (control group, n=7) respectively following anesthesia induction. The hemodynamics was monitored with Swan-Ganz catheter technique, the neutrophils were collected from arterial blood with centrifugal way to measure the deformability and adherence at 1st, 2nd, 4th and 6th hour after the administration of oleic acid separately. The pulmonary pathological examination underwent at 6th hour after the administration. Result: The pathological findings of test group were compatible with the RDS pathological characteristics. 3.Early profile and clinical significance of serum delta bilirubin after orthotopic liver transplantation

Objective To investigate the early variations and clinical value of serum delta bilirubin after orthotopic liver transplantation. Methods Orthotopic liver transplantation was performed in 27 cases.Of them,10 cases were uneventful;8 patients suffered from acute rejection.According to the outcome,18 patients (including 10 uneventful cases and 8 cases with acute rejection) survived while other 9 died.Daily determination of serum delta bilirubin was examined in all patients. Results The changing patterns of serum delta bilirubin were different between uneventful patients and those with acute rejection.For those uneventful patients,?B% tended to increase after operation.On the 21st day after transplantation,it reached(37.5?11.2)%.To the contrary,after acute rejection occurred,?B% decreased from basal level(26.4?9.8)% to(20.2?12.7)%(n=8,t=2.12,P=0.07).Serum concentration of total bilirubin (TB) and conjugated bilirubin (CB) significantly increased after acute rejection arose.The difference between peak and basal level of TB and CB was (150.3?113.2)?mol/L(n=8)and(118.5?93.4)?mol/L(n=8)respectively(t=3.75,P=0.007;t=3.59,P=0.009).On the 14th day after operation,?B% in patients with good outcome(n=18) significantly higher than that in patients who died(n=9)(33.4%?12.2% vs 22.0%?10.0%,t=2.42,P=0.02).Conclusions Two weeks after operations,?B% is correlative to the outcome of the patients.But as for the diagnosis of the acute rejection,?B% shows no advantages over serum TB or CB.
4.Role of Fas, FasL and Caspase-3 in artesunate-induced apoptosis of human embryonic lung fibroblasts

BACKGROUND: Artesunate can relieve pulmonary fibrosis, but its mechanisms are rarely reported.OBJECTIVE: To explore the effect of artesunate on apoptosis of HFL-I cells and the role of Fas, FasL and Caspase-3 in the artesunate-mediated apoptosis of HFL-I cells. METHODS: Cell counting kit-8 (CCK-8) assay was used to determine the effects of artesunate at 1, 10, 100 mg/L on the growth of HFL-I cells in vitro. Apoptosis ratio was examined by flow cytometry (FCM). The mRNA level of Fas, FasL and Caspase-3, were assessed by reverse transcription-polymerase chain reaction (RT-PCR). RESULTS AND CONCLUSION: Artesunate had a significantly inhibitory effect on the proliferation of HFL-I cells in a dose-dependent manner in vitro. The apoptosis rate of HFL-I cells was significantly increased in the artesunate-treated group compared with the control group (P < 0.05 or P < 0.01). The mRNA levels of Fas, FasL and Caspase-3 were significantly higher in the artesunate-treated group than the control group (P < 0.05). The findings of this study demonstrate that artesunate can exert a marked anti-pulmonary fibrosis effect by up-regulating mRNA levels of Fas, FasL and Caspase-3, which can induce the growth inhibition and apoptosis in HFL-I cells.
5.Study of relationship between levels of hs-CRP and IL-6 with restenosis after stent angioplasty in cerebral arterial stenosis

Objective To investigate the relationship between the dynamic changes of hs-CRP and IL-6 with restenosis after stent angioplasty in cerebral arterial stenosis .Methods 65 patients with cerebral artery stenosis stent angioplasty in Nanyang Mu-nicipal Central Hospital from March 2011 to March 2013 were retrospectively analyzed .The changes of hs-CRP and IL-6 levels be-fore operation and at different postoperative time points were observed and their correlation with vascular restenosis was anlyzed . Results (1)The cerebral artery stenosis degree and serum hs-CRP and IL-6 levels were positively correlated(P<0 .05) .(2)Com-paring the hs-CRP and IL-6 levels at different time points showed that the postoperative hs-CRP and IL-6 levels were significantly increased ,reached the highest at postoperative 12 h and then gradually declined .The hs-CRP and IL-6 levels on postoperative 7 d in the patients with restenosis were significantly higher than those in the patients without restenosis (P<0 .05) .(3)The hs-CRP and IL-6 levels at postoperative 6 months in the patients without restenosis were significantly decreased compared with before opera-tion ,but which in the patients with restenosis had no statistically significant differences (P>0 .05);△hs-CRP and △IL-6 had sta-tistically significant differences between the patients with restenosis and the patients without restenosis (P<0 .05) .Conclusion hs-CRP and IL-6 may play an important role in the process of restenosis after stent angioplasty in cerebral arterial stenosis .Monitoring the dynamic changes of hs-CRP and IL-6 has certain value for evaluating vascular restenosis .
6.The effects of intracarotid transplantation of bone marrow stem cells on learning memory abilities in the brain of VD rats

Objective To observe the survival and migration characteristics after intracarotid transplantation of bone marrow stem cells(BMSCs) and its effect on learning-memory abilities across the blood-brain barrier(BBB) in the vascular dementia(VD) rats .Methods Bone mononuclear cells(BMNCs) were isolated from bone marrow in vitro by standard Ficoll-Hypaque technique , then cells were enriched and expanded by using bone marrow adherent culture .72 Wistar rats were meanly divided into control group ,model group and treatment group .The VD rat model was established by modified pulsinellis 4-vessel occlusion(4 VO) .The treatment group received intracarotid infusion of 0 .5 mL 1 .2 × 107/mL BMSCs which were labeled with BrdU in vitro after opera-tion .Their survival ,migration and the learning-memory abilities were observed at 4th and 8th week .Results BMSCs transplanted by intracarotid transplantation survived and had been found throughout the brain tissue .They major migrated and localized in the is-chemic zone of injury such as hippocampus and cerebral cortex .Compared with the control group ,active avoidance response(AAR) ratio in the model group(42 .1 ± 4 .5) ,group(43 .6 ± 3 .6)showed significantly decrease compared with the control group (90 .0 ± 4 .3) ,(92 .5 ± 5 .0)(P<0 .01) ,and the treatment group(69 .2 ± 4 .7) ,(70 .8 ± 4 .7)was significant higher than the model group(P<0 .01) .Conclusion Intracarotid transplantation of BMSCs could enter the VD rats cerebra parenchyma via BBB ,migrate into the damaged brain tissue to gather and survive .Learning-memory abilities can be improved significantly by transplanted BMSCs .
7.Percutaneous kyphoplasty with bone cement injection rapidly eases pain in patients with multi-segmental thoracolumbar vertebral osteoporotic fractures

BACKGROUND:Percutaneous kyphoplasty with bone cement injection for repair of multi-segmental thoracolumbar vertebral osteoporotic fractures can restore the compressed vertebral height, correct deformity and reduce complications. OBJECTIVE: To explore the therapeutic effects of percutaneous kyphoplasty with bone cement in patients with multi-segmental thoracolumbar vertebral osteoporotic fractures. METHODS:Eighty patients with multi-segmental thoracolumbar vertebral osteoporotic fractures were enroled, 29 males and 51 females, aged 61-78 year, including 52 case of two-segmental vertebral fractures, 22 cases of three-segmental vertebral fractures, 6 cases of four-segmental vertebral fractures. Al the patients were divided into experimental group and control group randomly. Patients in the control group were given active basic treatment and anti-osteoporosis treatment and stayed in bed. Patients in the experimental group were given percutaneous kyphoplasty with bone cement injection on the above basis. Al the patients were folowed-up 12 months after treatment. Then the Oswesty dysfunction index, Barthel index, visual analog scale score, and theanterior vertebral height and midline height, Cobb angle were compared between the two groups. RESULTS AND CONCLUSION: In the experimental group, the Oswesty dysfunction index, Barthel index, visual analog scale score, and the anterior vertebral height and midline height, Cobb angle were al significantly improved at 3 days, 3 months and 12 months after treatment than before (P < 0.05). In the control group, the Oswesty dysfunction index, Barthel index and visual analog scale score were improved significantly at 3 and 12 months after treatment (P < 0.05), while the anterior vertebral height and midline height were improve dramaticaly at 12 months after treatment (P < 0.05). Compared with the control group, the experimental group showed better the Oswesty dysfunction index and Cobb angle than the control group at 12 months after treatment (P < 0.05), but there were no difference in the other indicators between the two groups. No adverse reactions associated with bone cement material occurred in the experimental group. Taken together, the percutaneous kyphoplasty with bone cement injection can rapidly relieve pain in patients with multi-segmental thoracolumbar vertebral osteoporotic fractures, which can correct kyphosis and improve patient's quality of life.
8.Application of anatomical isometric technique in arthroscopic reconstruction of anterior cruciate ligament and quadruple hamstring tendon autograft

[Objective]To study the feasibility and short-term effects of the arthroscopic reconstruction of ACL with LARS artificial ligaments and quadruple hamstring tendon autograft usirg anatomical isometric technique.[Method]Arthroscopic ACL reconstruction was carried out in 20 patients with ACL injuries by using the LARS artificial ligaments and quadruple hamstring tendon autograft.Bone tunnels of the femur and the tibia were prepared with ACL anatomical isometric technique.20 patients were divided into LARS artificial ligaments group(11 patients) and quadruple hamstring tendon group(9 patients).The artificial tendon group was inserted into the tunnels and fixed with two interface screws after being tightened.Quadruple hamstring tendon group was fixed with endobutton plate.[Result]The operation time were 30~80 min(mean,56 min) in LARS artificial ligaments group and 80~120 min(mean,100 min) in quadruple hamstring tendon group.No postoperative complications such as synovitis and ligament rupture and movement restriction occurred iF two groups.A follow-up check up was made in the 20 patients for 6~12 months(mean,9 months).The Lysholm scores improved from a preoperative score of 22~65(36.72~15.54) to 80~98(90.45~4.68) at follow-up(t=10.535,P
9.Arthroscopic suture fixation with cannulated screw and washer for bony avulsion of the posterior cruciate ligament

[Objective]To investigate the feasibility and short-term effects of arthroscopic suture fixation with cannulated screw and washer for bony avulsion of the posterior cruciate ligament.[Method]Arthroscopic suture fixation with cannulated screw and washer was carried out in 30 patients with bony avulsion of the posterior cruciate ligament.The posterior cruciate ligament and avulsion bone fragment were held together with two Ethibond 5 polyester threads,which were first wrapped around the posterior cruciate ligament from the anterior to the posterior,and then tied behind the posterior cruciate ligament and over the bone fragment.The threads were pulled out through tibia tunnels,tighted and fixed on cannulated screw and washer under the tibia tunnels 2 cm.[Result]The operation time were 60-90 min.A follow-up was made in 10 patients for 6-12 months.All fractures were united without displacement at 6 weeks after operation.No postoperative complications occurred.The Lysholm scores were 83-95(89.6?4.03).[Conclusion]Arthroscopic suture fixation with cannulated screw and washer for bony avulsion of the posterior cruciate ligament technique is simple to perform and gives a satisfactory short-term results.
10.Effects of IGF-1 on PRNP expression and APP metabolism of PC12 cells

Objective·To investigate the effects of insulin-like growth factor 1 on prion encoding gene (PRNP) expression and amyloid precursor protein (APP) metabolism of PC12 cells. Methods·After PC12 cells were treated with 20, 40, 80 ng/mL IGF-1 for 24 h, real-time PCR was used to detect the mRNA expression levels of PRNP, and Western blotting was used to detect the protein levels of AKT, phosphorylation of AKT (pAKT), ERK and phosphorylation of AKT (pERK). The level of β-amyloid 42 (Aβ42) in supernatant fluid was detected by ELISA. Results·Compared with the blank control group, the expression of PRNP mRNA in Alzheimer's disease (AD) model group was increased significantly (P<0.01). The expression of PRNP mRNA was significantly increased after cells were treated with 40 and 80 ng/mL IGF-1 for 24 h (P<0.01). There was no significant difference in APP protein expression among AD model group and three IGF-1 treatment groups (P>0.05). Compared with the blank control group, the level of Aβ42 in supernatant fluid of model group was decreased significantly with the increasing of IGF-1 concentration. The expression level of Aβ42 was decreased significantly in 40 and 80 ng/ml IGF-1 treatment group (P<0.05). The expression of AKT/pAKT, ERK/pERK in AD model group was significantly increased along with the increasing of IGF-1 concentration (P<0.05). Conclusion·IGF-1 could regulate the expression of PRNP gene and effect the metabolism of APP, which may be associated with PI3K/AKT, MAPK/ERK1/2 signaling pathway.
