Objective To evaluate the value of pre-S1 Ag in combined detection of hepatitis B virus. Methods 288 patients' serums with chronicity type B hepatitis (CHB)were collected when they had been detected with HBV-DNA,HBVM or ALT/AST,and then ELISA assay were applied to detect pre-S1Ag. Results A total of 288 patients ,including 175 males and 113 females( their meso-age is 36.89 years old) were recruited into this study. 161 patients were pre-S1 Ag positive(68.4%), 197 patients' HBV DNA contents exceeded 5.0E + 2 copy/ml(55.9%) ,71patients were H BeAg positive (24.6%), 186 patients were HBeAg negative (64.58%). The rate of 64 patients' ALT and AST exceeded 0. 84, and their' ALT exceeded 40 u/ml ( 22. 2% ). 60 patients' AST exceeded 50 u/ml (20. 8% ). These patients were divided into pre-S1 Ag positive group and pre-S1 Ag group. In pre-S1 Ag positive group, 124 patients' HBV DNA contents exceeded 5.0E + 2 copy/ml( 62.9% ), 118 patients were HBeAg negative (59.8%). When HBV DNA content was less than 5.0E+2 copy/ml,pre-S1 Ag positive rate was 35.2% and pre-S1 Ag negative rate was 63.6%. when HBV DNA content was less than 5.0E + 3 copy/ml, pre-S1 Ag positive rate was 56.9% and pre-S1 Ag negative rate was 81.7%. When HBV DNA content exceeded 5.0E + 5 copy/ml but less than 5.0E + 8 copy/ml ,pre-S1 Ag positive rate was 33% and pre-S1 Ag negative rate was 14.3% ,so two groups had significant difference. Conclusion Pre-S1 Ag positive and HBeAg negative in the CHB patients serums signify virus replication, and it can help us decide prognosis.

Objective To study the effect of flee radical on kidney repedusion injury caused by infra-renal abdominal aorta occlusion in rat model and its possible mechanism. Methods Forty-two healthy Wis-ter rats were randondy divided into 6 groups as following ( n = 7) : the control group( sham group) ; simply isehemia 5 h without reperfusion( group I) ; 2 hours reperfusinn following ischemia 5 h ( I/R 2 h), 4 honrs reperfusion following isebemia 5 h ( I/R 4 h), 8 hours reperfusinn following ischemia 5 h( I/R 8 h) and 12 hours reperfusion following ischemia 5 h ( I/R 12 h). In each group the rats were killed to obtain samples of blood and kidney at the specified time points. The contents of BUN, Cr, MDA, SOD in blood and in renal homogenate were measured in each group. We observed the morphological changes of kidney and muscles of lower limb by light microscope. Results BUN level of serum in model group I, I/R 2 h, I/R 4 h, I/R 12 h were higher obvionsly than those of control group, which were maximal in I/R 4 h, then decreased. MDA level of plasma in model group I, I/R 2 h, I/R 4 h, I/R 8 h, I/R 12 h groups were higher obvionsly than those of control group, which were maximal in I/R4 h group, then decreased. SOD level of plasma in model I/R 4 h, I/R 8 h groups were lower obviously than those of control group; SOD level of renal homogenate in model group I, I/R 2 h, I/R 8 h, I/R 12 h groups were higher obviously than those of group I/R 4 h, which were minimal in I/R 4 h group, then increased. By light microscope: The injury degree of kidney and muscles of lower limb in ischemia group was slight, the injury degree of repedusion group was severer than ischemia group. Conclusion The kidney repedusion injury caused by infrarenal abdominal aorta occlusion in rat model might be concerned with the increase of lipid peroxidatian damage after ischemia-repedusion in-jury of lower limbs.

OBJECTIVETo explore the biological characteristics of fibroblasts in adult granulation tissue in vitro, including cell viability, CD phenotypic, factor and protein expression, and differentiation, so as to facilitate further research of the role of fibroblasts in early wound healing.
METHODSFibroblasts were isolated from human granulation tissue and cultured by mechanical and enzymatic digestion method. The cell morphology and proliferation were observed under inverted phase contrast microscope. The first and third passages of cells' growth curve were drawn respectively. The surface markers (CD105, CD73, CD90, CD44, CD34, CD45, CD19, CD11b, HLA-DR) of the first and third passage fibroblasts were identified by Flow Cytometry. And the expression of Vimentin, CK19, CD31 and Factor VIII were detected by immunocytochemistry.
RESULTSPrimary cultured fibroblasts were short spindle, polygonal and irregular in shape. The morphology of fibroblasts were uniform by repeatedly passage cultured in vitro and showed spindle-shaped. The proliferative capacity of the fibroblasts were not significantly different, with logarithmic growth phase. From 1 to 4 days, The primary and third passage cells' proliferation was no difference (P > 0.05), After the five days, the proliferative ability of third generation was better than the primary passage (P < 0.01). All fibroblasts highly expressed mesenchymal stem cells' surface markers CD105, CD73, CD90 and CD44, and didnt express hematopoietic stem cells' surface markers CD34, CD45, CD19, CD11b and HLA-DR. The expression of mesenchymal stem cells' surface markers in third generation of cells were increased significantly. Immunocytochemistry showed positive expression of Vimentin, CD31, and negative expression of CK19 and Factor VIII.
CONCLUSIONSFibroblasts in human granulation tissue show the biology characteristics of Mesenchymal Stem Cells. Some biological markers of endothelial cells are expressed in fibroblasts in granulation tissue. The fibroblasts may play an important role during the process of endothelial to mesenchymal transformation in early wound healing.
