1.Granulocyte Colony Stimulating Factor for Focal Cerebral Ischemic Injury

Granulocyte colony stimulating factor(G-CSF)can mobilize hemopoietic stem cells(HSCs)from bone marrow to peripheral circulation,stimulating HSCs and marrow stromal cells(MSCs)homing to the lesion of cerebral ischemia,contributing to the interactions of HSCs and MSCs with the cells in ischemic penumbra,and promoting the generation of neurotrophic factors.G-CSF also has antiapoptotic effect on neurons and drives neurogenesis.Mobilizing autologous HSCs and MSCs using G-CSF for the treatment of local cerebral ischemic injury may reduce the volume of cerebral infarction,and improve neurological functional recovery.

3.Application of 3D-power Doppler ultrasound in diagnosis of breast carcinoma in elderly patients

Objective To explore the diagnosis value of 3D-power Doppler ultrasound in differentiating benign and malignant breast masses in elderly patients.Methods A retrospective study of 53 lesions in 45 patients was performed,and breast masses were divided into benign and malignant group by pathological diagnosis after operation.Display of cross,vertical and coronal plane of total 53 masses were automatically acquired by 3D-power Doppler examination before operation,and 3D imaging of masses were reconstructed by virtual organ computer-aided analysis.The vascular morphology inside and beside the breast neoplasm was observed by tomographic ultrasound imagine layer by layer in different sections.Vascular distribution characteristics were divided into penetrating,central,surrounding and few vessel type.Quantitative analysis of vascularization-flow index inside the breast neoplasm was measured by 3D-power Doppler histogram.Results The difference in vascular distribution characteristics by 3D-power Doppler ultrasound was statistically significant between 15 (28.3%) benign and 38 (71.7%) malignant lesions(x2=19.52,P<0.01).The case number of presence of penetrating vessels was 26(68.4%)in the malignantuniors.The sensitivity and specificity of penetrating vessel diagnosed malignant breast tumor were 68.4% and 86.7%.Vascular distribution characteristics of benign masses was mainly of surrounding vessel type(53.3%).Vascularization-flow index was higher in malignant group than in benign group [(1.32 ± 1.58) vs.(0.63 ± 0.53),t=2.187,P<0.05].Conclusions 3D-power Doppler ultrasound completely presents the vessel stereoscopical distribution and shows quantificationally blood perfusion of breast neoplasm.The 3D-power Doppler ultrasound is superior to conventional ultrasound for enhancing differential diagnosis ability between benign and malignant breast tumors.
4.Study on establishment of golden hamster buccal squamous carcinoma model for and its biological characteristics

Objective To establish the golden hamster model of buccal squamous carcinoma and observe its biological character-istics .Methods 50 golden hamster were randomly divided into two group :experiment group (n=40) and control group(n=10) . Buccal mucosa of golden hamster were daubed by exposure 4-nitroquinoline-1-oxide (4NQO) in experiment group and tap water in control group .HE staining and immunohistochemistry were used to observe the tissue sample on the 8th and 12th week .The tissue samples of golden hamster buccal-mucosa cancer were used for the in-vitro subculture .Then flat cloning formation rate ,expression of CK and Vim ,and cell karyotype were detected .Results The observations of cell morphology and biology showed that the tissue of buccal squamous carcinoma were conformed to the basic characteristics of squamous carcinoma cell in 26 golden hamster(84 .6% ) after 12 weeks .The positive rate of CK and Vim were 96 .0% by immunohistochemical staining .The Chromosomes were tetraploid karyotype .Conclusion We successfully established the golden hamster model of buccal squamous carcinoma by the daub method of 4NQO .
5.Analysis of DMD gene mutations in 59 families in northern China

Objective To detect the DMD gene mutation sites and the regions of breakpoints in Duchenne/Becker muscular dystrophy (DMD/BMD) patients in northern China. Methods Multiplex amplifiable probe hybridization (MLPA) was used to detect the mutation in 59 cases (51 cases with DMD and 8 with BMD) from northern China and dystrophin gene mutations in their parents. Results From northern China and dystrophin gene mutations 59 families found gene deletions in 33 cases of 59 DMD/BMD patients (55.9%), duplications in 6 cases (10. 2%) and point mutation in one case (1.7%). Intron 44 was most frequently affected (n = 13, 33.3%), followed by intron 50 (n = 11, 28.2%) and intron 45 (n=8, 20.5%). The novel mutations were identified, in two patients including two independent duplications carried by patient D1 149 and a point mutation [5208del(A)] carried by patient D1 65, which were not included in Leiden database. In addition, an exon 22 deletion was found in one patient, which was the first reported case in Chinese patients. Conclusions Deletions are mostly located in the hotspot between exon 45 and 50. Duplications mostly occurred in the 5' end of the gene. Intron 44 is the most frequently affected breakpoint in northern Chinese population.
6.Effects of thoracoscopic lobectomy on early-stage non-small cell lung cancer

Objective To discuss effects of thoracoscopic lobectomy on early-stage non-small cell lung cancer . Methods 60 patients with non-small cell lung cancer patients were randomly divided into two groups of 30 cases in each group using thoracoscopic lobectomy (VATS) treatment,as the VATS group,another group using traditional open surgery(TOS) therapy,TOS as a group.(1) Operative time,blood loss,postoperative drainage time,drainage, postoperative pain,postoperative bed days,after several days of hospitalization were compared between the two groups . (2) 1,3 and 5 days after surgery,serum C-reactive protein(CRP) and interleukin-6(IL-6)levels were compared. Results (1)were operative time,postoperative drainage time,pain time,bed time and hospitalization time in VATS group were (69.50 ±15.33)min,(60.23 ±9.75)h,(46.75 ±7.36)h,(3.54 ±1.58)d,(9.50 ±2.50)d,com-pared with TOS group (86.77 ±19.56)min,(66.50 ±10.50)h,(58.36 ±9.58)h,(5.50 ±1.55)d,(13.55 ± 3.18)d short d observed group blood loss ,drainage volume(161.55 ±25.54)mL,(155.75 ±23.18)mL,compared with TOS group(186.58 ±25.35)mL,(169.65 ±26.87)mL less,the difference was statistically significant (t =39.14,28.19,54.11,6.03,7.24,98.36,76.34,all P<0.05).(2) VATS group after 1,3 and 5 days CRP serum and were(79.58 ±22.15) mg/L,(97.23 ±28.16) mg/L,(98.44 ±27.19) mg/L,IL-6,respectively (69.36 ± 17.32) ng/L, ( 96.33 ±25.67 ) ng/L, ( 103.16 ±28.89 ) ng/L, were higher than the TOS group ( 97.84 ± 25.37)mg/L,(119.17 ±31.77)mg/L,(130.81 ±33.29)mg/L,(76.83 ±21.97)ng/L,(121.15 ±32.64)ng/L, (122.08 ±30.74)ng/L,the difference was statistically significant (t=70.12,58.33,69.17,64.34,79.58,92.17,all P<0.05).Conclusion VATS lobectomy for non-small cell lung cancer has better effects and safety .
7.ISSR Analysis for genetic diversity of Coptis chinensis

Objective To discuss the genetic diversity of Coptis chinensis.Methods The genetic diversity of 78 individuals from seven populations was analyzed by inter-simple sequence repeat(ISSR).Results Twelve primers were selected to produce highly reproducible ISSR bands.Among 106 amplified bands,72 showed polymorphism,the percentage of polymorphic bands reached to 67.92%.Nei's gene diversity index(H)was 0.180 3,Shannon information index(I)was 0.283 2,Gst was 0.681 5.The genetic distance coefficient and genetic similarity were 0.089 4—0.184 6 and 0.832 1—0.912 7,respectively.ConclusionC.chinensis holds high genetic diversity and the majority of genetic variation occurs among the populations.By cluster analysis,the geographical distribution is very obvious.The ISSR marker could be used for the analysis of the genetic diversity and genetic variation of C.chinensis.
8.Effect of electroacupuncture on endoplasmic reticulum stress in lung tissues of rats with acute lung injury induced by endotoxin

Objective To evaluate the effect of electroacupuncture on endoplasmic reticulum stress in lung tissues of rats with acute lung injury (ALI) induced by endotoxin.Methods Forty healthy pathogen-free male Sprague-Dawley rats,aged 8 weeks,weighing 180-210 g,were divided into 4 groups (n=10 each) using a random number table:control group (group C),ALI group,electroacupuncture group (group E),and electroacupuncture at non-acupoint group (group NE).Lipopolysaccharide 5 mg/kg (in 0.5 ml normal saline) was injected intravenously to establish the model of endotoxin-induced ALI.Bilateral 30 min electroacupuncture stimulation of Zusanli and Neiguan acupoints was performed with the dispersedense wave (frequency 2/15 Hz,wave length 0.2-0.6 ms,intensity 1-2 mA) once a day (time for stimulation 9:30-10:30) for 4 consecutive days before and during establishment of the model in group E.Electroacupuncture was performed with the same parameters at the points 0.5 cm lateral to the acupoints of Zusanli and Neiguan in group NE.At 6 h after lipopolysaccharide injection,the rats were sacrificed,and lungs were removed for microscopic examination and for determination of wet to dry weight ratio (W/D ratio),apoptosis in alveolar epithelial cells and expression of glucose-regulated protein 78 (GRP78) and CCAAT/enhancer-binding protein homologous protein (CHOP) and caspase-12 in lung tissues (by Western blot).The pathological changes of lungs were scored.Apoptosis index (AI) was calculated.Results Compared with group C,lung injury scores,W/D ratio and AI were significantly increased,and the expression of GRP78,CHOP and caspase-12 in lung tissues was up-regulated in the other three groups (P<0.05).Compared with group ALI,lung injury scores,W/D ratio and AI were significantly decreased,and the expression of GRP78,CHOP and caspase-12 in lung tissues was down-regulated in group E (P<0.05),and no significant change was found in the paramneters mentioned above in group NE (P>0.05).Conclusion The mechanism by which electroacupuncture attenuates endotoxin-induced ALI is related to inhibition of endoplasmic reticulum stress and reduction of apoptosis in alveolar epithelial cells in rats.
9. The relationship between cholecystolithiasis or cholecystectomy and the risk of colorectal cancer: a Meta-analysis

Objective:
To analyze the correlations between cholecystolithiasis or cholecystectomy and the risk of colorectal cancer, and make a brief summary combining with the present study.
Methods:
Qualified studies about the correlations between cholecystolithiasis or cholecystectomy and the risk of colorectal cancer published in English and Chinese before April 2018 were retrieved from PubMed, Cochrane Library, China National Knowledge Infrastructure(CNKI), Wan Fang Data, and VIP. Case-control and cohort studies were selected according to the inclusion and exclusion criteria and assessed by the Newcastle-Ottowa Scale, then we chose the high-quality literature to extract the data and analyze those data by RevMan 5.3 software. Publication bias was analyzed by Stata 12.1 software.
Results:
A total of 28 articles were finally included in the systematic review, including 23 case-control studies and 5 cohort studies. The results showed that there is a significant relationship between cholecystolithiasis and the risk of colorectal cancer(
10.Use of specific antibody to detect fetal erythroblasts in maternal circulation.

OBJECTIVETo investigate the feasibility of using immunohistochemical technique to detect the presence of fetal erythroblasts in the maternal circulation for prenatal diagnosis.
METHODSMaternal blood was obtained from 30 pregnant women at 8 to 26 weeks of gestation. Nucleated red blood cells (NRBCs) were separated with Percoll using a discontinuous density gradient method, and then smeared on microscope slides using cytocentrifugation. Slides were stained with antibody against the gamma-chain of fetal hemoglobin (HbF). All positive NRBCs were collected by micromanipulator under microscopic observation, and then amplified by improved primer extension preamplification(PEP). Sex and Duchenne's musclar dystrophy (DMD) genetic diagnosis were determined from a small aliquot of the PEP reaction.
RESULTSNRBCs stained with HbF were found in all of the blood from the 30 pregnant women at 8 to 26 weeks of gestation. 17 male fetuses and 13 female fetuses were detected in the 30 cases. These results coincided with those of induced labor or amniotic fluid control, and 8 fetuses at the risk of DMD were diagnosed.
CONCLUSIONThis diagnostic method using immunohistochemical technique to mark fetal NRBC shows good application prospects.
