AIM To investigate the effects of apoptosis and the expression of telomerase hTERT mRNA activity of HL-60 cells treated with As 2O 3 in different concentrations. METHODS HL-60 cells was cultured with As 2O 3 in different concentrations for 48 hours, then, cell apoptosis was detected with flow cytometry and the expression of telomerase hTERT mRNA was determined with RT-PCR respectively. RESULTS The percentage of the apoptosis in HL-60 cells were 2 28%?0 40%?2 55%?0 22% and 47 57%?2 79% respectively, as the cells were treated with 0 1 ?mol?L -1,1 0 ?mol?L -1,5 0 ?mol?L -1 As 2O 3 for 48 hours. The difference was significant between 5 0 ?mol?L -1 and 0 1~1 0 ?mol?L -1 groups(P

3.Effects of pulsed electromagnetic fields on mechanical allodynia after acute skeletal muscle contusion in rats

Objective To observe the effects of pulsed electromagnetic fields (PEMFs) on behavior and mechanical paw withdrawal thresholds (MPWTs) after acute skeletal muscle contusion (ASMC) in rats, and to in-vestigate the application of PEMFs in rats with ASMC during the early stage. Methods Forty-two Sprague-Dawley rats were randomly divided into a PEMF group (P group) , control group (C group) and blank control group ( BC group). ASMC models were set up in groups P and C, and no intervention was applied in the BC group. A PEMF was administered to animals in the P group immediately after the ASMC was inflicted. The behavior of the rats in each group was then observed. The MPWT of each rat was tested 2 days before and 0, 12, and 18 hours after the ASMC was inflicted). Results In the P and C groups, MPWT of the left hind paw at the 12th and 18th hour after ASMC was significantly lower than the baseline pain threshold 2 days before the ASMC. At 18 hours, the MPWT was signifi-cantly higher than at 12 hours in the P group. MPWT at 12 hours in the P group and at both 12 and 18 hours in the C group were significantly lower than in the BC group. MPWT in the P group at 18 hours was significantly higher than in the C group. Conclusions The behavior of rats treated with PEMF immediately after ASMC was improved, and their pain threshold was still elevated 18 hours later.
4.Effects of pulsed electromagnetic fields on the expression of MyoD after acute skeletal muscle contusion

Objective To observe the effects of pulsed electromagnetic fields (PEMFs) on histological changes and myogenic differentiation factor D (MyoD) expression in rats with acute skeletal muscle contusion ( ASMC), and to explore the effects of PEMF therapy on rats with ASMC in its very early stages. Methods Forty-two rats were randomly divided into three groups : a treatment group, a control group and a blank control group. ASMC models were established with all the animals in the treatment and control groups. PEMF treatment was admin-istered to the treatment group immediately after the establishment of the ASMC model. Seven rats in each group were sacrificed at the 12th and 18th h after the models were set up. Their triceps surae muscles were sampled and treated with haematoxylin-eosin staining for study using immunofluorescence techniques and a fluorescence microscope. Re-suits In the control group at the 12th h and 18th h, HE staining showed pale cytoplasm and polymorphism in the cell nuclei ; in the treatment group these effects were significantly lighter, but in both groups it was more serious than in the blank control group. In the treatment and control groups, the fluorescence intensity of MyoD at the 18th h was higher than at the 12th h, and at each time point in both groups it was higher than in the blank control group. At the 18th h, fluorescence in the treatment group was stronger than in the control group. Conclusion MyoD expression in rats with ASMC is upregulated by thel8th h after early PEMF treatment. This might be one of the mechanisms ac-celerating the regeneration of skeletal muscles after trauma.
5.Bacteriological analysis and treatment strategy in patients with biliary sepsis

Objectives To access the bacteriology in patients with sepsis due to biliary tract infection to provide a basis for empirical selection of proper antibiotic treatment.Methods This is a single-center retrospective study on 214 patients with biliary tract infection admitted from August 2014 to July 2016 to the surgical intensive care units (ICU) of The First Affiliated Hospital of Sun Yat-sen University.To study the demographic information,sequential organ failure assessment (SOFA),usage of antibiotics before ICU and duration of ICU were analyzed.Bile,peritoneal drainage and blood samples were collected.Results 47 septic shock patients and 25 septic patients due to biliary tract infection were enrolled in the trial.The two groups (the shock group vs.the sepsis group) had a significant difference in the duration of ICU stay [(6.4 ± 4.6) d vs.(2.3 ± 1.8) d,P < 0.05].48 strains of pathogens were isolated from the bile samples.The major pathogens were Escherichia coli (E.coli) (n =23,47.9%),Enterococcus faecalis (n =8,16.7%) and Enterococcus faecium (n =2,4.2%).80 strains of pathogens were isolated from the peritoneal drainage culture samples.E.coli,pseudomonas aeruginosa,and Klebsiella pneumoniae ranked the top 3 species,accounting for 26.3%,11.3% and 7.5%,respectively.The sensitivity of E.coli isolated from bile to amikacin,imipenem and panipenem were all over 90.0%.Conclusions E.coli was the principal gram-negative bacterium in biliary infection induced sepsis.Early administration of carbapenemes may reduce the occurrence of septic shock in these patients.

ABSTRACT:Objective To observe the effect of acupoint plaster based on the theory of meridian stream of time-choosing in patients with bronchial asthma.Methods 200 bronchial asthma patients were randomly divided into experimental group and control group with 100 patients in each group.The experimental group were applied acupoint plaster on Shen time of 15:00-17:00 of the hottest time of summer while the control group were applied plasters on different time of Shen time on the early,middle and Mofu time in the summer for three times in total.Effective rate,the quality of life and psychological status of patients in two groups were compared in the summer.Re-sults Efficacy of anti-recurrence,quality of life and mental state of patients in experimental group were significantly better than that in control group,which showed statistically significant difference (P <0.05).Conclusion Meridian stream of acupoint plaster can significantly improve the thera-peutic efficacy of bronchial asthma,so it is worthy of clinical popularization and application.

8.Preliminary study on expression profiles of plasma circulating microRNAs in patients with sepsis and healthy control people

Objective To identify the circulating miRNAs which can be used to evaluate the diagnosis and prognosis of sepsis by microarray and quantitative real-time PCR,and to predict target genes of miR-519c-5p by bioinformatics analysis. Methods Three sepsis patients,3 septic shock patients and 3 normal controls were enrolled in this study. Plasma RNA was extracted,and was used for hybridized by miRCURY LNATMmicroRNA Array.The signals were scanned and used to conduct differential expression profilings and cluster analysis.Further-more,we performed qRT-PCR to confirm the expression of miRNAs chosen from microarray screening. We used the miRanda and Targetscan databases to predict target genes of the concerned miRNAs and used KEGG database to analyze the related pathways. Results Fifty-seven and 11 miRNAs were observed significantly upregulated in sepsis and septic shock patients,respectively(fold change≥2.0;P<0.05).qRT-PCR results showed that miR-519c-5p was significantly upregulated in patients with sepsis or patients with septic shock compared with the healthy normal controls(P<0.05).Twenty-nine target genes of miR-519c-5p were predicted by the bioinformatics analysis,and 7 potential target genes participate in the sepsis-related pathways.MiR-519c-5p might be a potential positive regulator for the critical cell cycle control gene of MAP2K4,contributing to the vascular endothelial cells apoptosis via MAPK signaling pathway. Conclusions We demonstrated that the plasma level of miR-519c-5p can be used for the diagnosis of sepsis and miR-519c-5p may be a potential therapeutic target for sepsis.
