1.The effect of irbesartan on the expression of angiotensin-converting enzyme 2 mRNA in kidney of spontaneously hypertensive rats

Objective To investigate the regulation effect of irbesartan on the expression of angiotensin-converting enzyme 2 (ACE2) mRNA in the kidney of spontaneously hypertensive rats (SHR). Methods Twenty 14-week-old male SHR were randomly divided to SHR group (n=10) and irbesartan group (n=10); ten 14-week-old male WKY group were served as control. Irbesartan group was given a daily dose of 50 mg/kg irbesartan for 12 weeks by gavage and the other two groups were fed with a normal diet. ACE2 in kidney was examined by immunohistochemistry and RT-PCR. Plasma angiotensin II in each group were estimated by radioimmunoassay (RIA). Results The ACE2 mRNA expression was significantly down-regulated in the kidney of SHR group compared with WKY group (0. 72?0. 11 vs 1. 11?0. 15). After 12 weeks treatment with irbesartan, the attenuated ACE2 expression in irbesartan group was markedly enhanced compared with SHR group (1. 03?0. 13 vs 0. 72?. 0. 11). 2.Polymorphism of 17 Y-STR loci in three minority populations in Guangxi of China

Objective To investigate the polymorphism of seventeen Y chromosomal short tandem repeat(STR) loci in Dong,Miao and Zhuang nationality in Guangxi,China.Then to evaluate their forensic application.MethodsDNA was extracted from oral swab of minority population with magnetic beads method and then seventeen Y-STR loci were amplified with AmpFlSTR○R YfilerTM PCR Amplification Kit System.The PCR products were detected with 3l30XL Genetic Analyzer.The allele frequencies,diversity and genetic distance were calculated.Results The information of allele distribution and gene diversity of the 17 Y-STR loci were obtained from three minority population.Total 275 haplotypes were found in three minority population.The haplotype diversity of Dong nationality is 0.9972,Miao is 0.9966,Zhuang is 0.9964.The genetic distance between Dong and Miao nationality is most close.Conclusion The haplotype diversity of 17 Y-STR loci is quite sensitive in racial identification,paternity identification and human population genetics.
3.Genetic polymorphic studies on 15 loci of 3 populations in Guangxi province

Objective To study the genetic polymorphism of 15 STR loci of 3 populations in Guangxi province. Methords DNA samples of unrelated individuals from 314 Gelao population,332 Mulao population,238 Yao population were analyzed using AmpFlSTR IdentifilerTM PCR Amplification Kit and 3100 Genetic Analyzer. Results The matching probability of the 15 STR loci was 1.839 ?10-16 ~ 5.073 ? 10 -17 and combined paternity of exclusion was 0.9999983 ~ 0.9999991 in the 3 populations. Conclusion The results showed that the 15 STR loci in Identifiler?PCR Amplification systems were useful for forensic case works in Gelao population, Mulao population and Yao population .

Objective To investigate the level changes of plasma soluble intercellular adhesion molecule-1(sICAM-1), endothelin(ET) and Nitric oxide(NO) and the effects of perindopril on their levels in elderly patients with essential hypertension. 5.Effects of angiotensin type-1 receptor blockade on expression of angiotensin-converting enzyme 2 in renal of spontaneously hypertensive rats

AIM: To investigate the expression of angiotensin-converting enzyme 2 (ACE_2) in the renal of spontaneously hypertensive rats and the effects of angiotensin type-1 receptor blockade. METHODS: Twenty of 14-week-old male SHR were randomly divided into two groups (n=10 in each): SHR group and irbesartan group. Ten of 14-week-old male WKY group were served as control. Irbesartan group were given a daily dose of 50 mg?kg -1 irbesartan for 12 weeks by gavage and the other two groups were fed with a normal diet. Angiotensin-converting enzyme 2 was examined by immunohistochemistry and RT-PCR. RESULTS: ACE_2 in the renal was significantly down-regulated in the SHR group compared with the WKY group ( 0.72? 0.11 vs 1.11? 0.15). After 12 weeks treatment with irbesartan, the attenuated ACE_2 expression in irbesartan group was markedly enhanced compared with the SHR group ( 1.03? 0.13 vs 0.72? 0.11). CONCLUSION: Hypertension induces the reduction of ACE_2 in the renal in rats and irbesartan markedly increases ACE_2 expression. It suggests that up-regulation ACE_2 which is one of counter-regulatory particular components in the renin-angiotensin system may be a new mechanism of curing hypertension beyond blockade angiotensin type-1 receptor.
6.Effects of losartan on expression of TGF-?_1 in nephric tubule of spontaneous hypertensive rats

AIM: To study the expression of transforming growth factor ? 1(TGF ? 1) in the nephric tubule of spontaneous hypertensive rats and the relation of the TGF ? 1 and the renal tubulointerstitial fibrosis, and to investigate the effect of losartan on this expression and the protection effects of it on the kidney. METHODS: Same age male WKYs and SHRs were involved in this experiment. SHRs were divided into positive control (SHR C) group and losartan treatment (SHR L) group. The artenia pressure, renal function and ? 2 MG of mice was monitored before and after experiment and the expression of TGF ? 1 in nephric tubule were measured by immuonohistochemitry assay. RESULTS: Compared with the WKY group, the contents of ? 2 MG markedly increased in SHR C group (P 0.01 ). No or a few expression of TGF ? 1 occurred in WKY group. The expression of TGF ? 1 significantly increased in the positive control group with the progression of the hypertensive course, and meantime, the expression of TGF ? 1 significantly decreased in the losartan treatment group and the blood pressure also decreased. CONCLUSION: As the expression of TGF ? 1 intensively increased in hypertensive course, TGF ? 1 may be the important factor leaded to the renal tubulointerstitial fibrosis. Losartan has the effects on lowering blood pressure, reducing albuminuria, protecting renal function and delaying the renal tubulointerstitial fibrosis.
7.In-Vitro Culture of Different Explants from Andrographis paniculate (Burm. f.) Nees

[ Objective ] To supply a technological evidence for the rapid reproduction of Andrographis paniculata (Burm. f.) Nees (APN). [Methods] Different explants from sterile seedling of Andrographis paniculata (Burm. f.) Nees were cultured in vitro to induce the production of fascicular buds. [ Results ] Cotyledon with nodes and half cotyledon with nodes were the most suitable explants for culture, and the budding rate was 100% and over 90% respectively. 6-Benzylaminopurine (BA) in the concentrations of 0.5 ~ 1.0mg/L were beneficial for the reproduction and growth of buds. The age of explants had no correlation with budding. The optimal rooting medium was MT (Murashige and Tucher) medium added with Img/L NAA (naphthaleneacetic acid) or IBA (indolebutyric acid). A highest rooting rate as much as 61.9% was achieved after 10 days of culture. [Conclusion] An effective plant regeneration system has been established for explants of Andrographis paniculata (Burm. f.) Nees.
8.Study on Extraction Method for Assaying Jinyou Tankeqing Tablets

Objective To optimize the extraction method for assaying Jinyou Tankeqing Tablets(JTT).Methods With the total flavonoids content as the index,Orthogonal Experimental Design was used to optimize the extraction condition for assaying JTT.Results The optimized ultrasonic extraction method was as follows: adding 0.2 mL?mg-1 powder to 95 % ethanol,extracting for 30 min;the optimized Sohlex extraction method was: adding 1.2 mL?mg-1 powder to 95 % ethanol,extracting for 2 h,the former method being better than the later.Conclusion The optimized extraction method for assaying JTT is adding 0.2 mL?mg-1 powder to 95 % ethanol and extracting for 30 min with ultrasonic wave.
9.Study on Quality Standard of Jinyou Tankeqing Tablets

Objective To study the quality standard of Jinyou Tankeqing Tablets (JTT). Methods Total flavonoids in JTT were identified by TLC. Naringin content in JTT was determined by HPLC. Results The relevant spots of total flavonoids in JTT can be identified by TLC. The content of naringin in JTT can be determined by HPLC. The linearity of naringin was good in the range of 2~20 ?g (r= 0.999 9). The average recovery of naringin was 101.29 %with RSD = 2.58 %. Conclusion The established quality standard is simple,feasible and repeatable,and can be used for the quality supervisory of JTT.
10.Effects of valsartan on phosphatase and tensin homologue deleted on chromosome ten (PTEN) in spontaneously hypertensive rats

AIM: To observe the effects of valsartan on the cardiac hypertrophy and the expression of PTEN in spontaneously hypertensive rats (SHR). METHODS: Twenty 12-week-old male SHR were randomly divided into 2 groups:SHR positive control group and valsartan treating group ( 30 mg?kg -1?d -1). 10 homogenous Wistar-kyoto (WKY) rats were served as normal control group. Blood pressure and the ratio of left ventricular weight to body weight(LVW/BW)of rats were monitored periodically during the 8-weeks studies. Expression of PTEN in cardiac myocytes was determined by immunohistochemistry. RESULTS: Blood pressure and LVW/BW increased in the valsartan group more than those in the SHR control group, and the expression of PTEN in cardiac myocytes in valsartan group increased more than that in the SHR control group, but the indexes were lower than those in the WKY group. CONCLUSION: Valsartan can not only inhibit the progression of cardiac hypertrophy in spontaneously hypertensive rats, but also increase the expression of PTEN. PTEN may play a role in cardiac hypertrophy.
