1.Application of AgNO3-Processed Silica Gel Chromatography in Isolation and Purification of Active Components of Herbal Medicine

[Objectivs] To investigate the principle of AgNO3-processed silica gel chromatography in isolation and purification of mixtures with carbon-carbon double bonds (CCDB) from herbal medicine. [Methods] AgN03-processed gel chromatography was used for the isolation of mixtures with carbon-carbon double bonds in the volatile oils and active components from Pteris semipinnata L. ; silver-ion complexion chromatography was applied to isolate arachidom'c acid from lipids. The principle of AgNO3-processed silica gel chromatography was analyzed by coordination theory. [Results] Stable AgNO3 coordinate complexes were obtained. [ Conclusion ] The principle of the formation of stable AgNO3 coordinate complexes may be: (1) the stability will increase with CCDB in free space; (2) exocyclic CCDB is more stable than that of intra-cyclic CCDB; (3) cis-form CCDB is more stable than that of trans-form CCDB; (4) terminal CCDB is more stable than that of cis-form CCDB; (5) aromatic hydrocarbon with multiple benzene rings is more stable than that with single ber zene ring. The possible principle will benefit to the isolation and purification of the mixtures with CCDB, which have similar structure and physiochemical property and are difficult to be isolated and purified by common chromatography and extriction method.
2.Study on the Polymorphism of Intron 22 (CA)n Repeat within FVIII Gene in Dai, Yi and Han Populations of Yunnan Province

This study was designed to investigate the polymorphism of intron 22 (CA)n repeat within FVIII gene in Dai, Yi and Han populations of Yunnan province. PCR, DNA sequencing technique and PAGE were used in this study. The results showed that three different alleles corresponding to 24, 25 and 26 dinucleotides were found at this locus and the allele frequency ranged from 1.20% to 57.7% in Dai and from 16.43% to 63.00% in Yi populations. In Han population, five different alleles with 24, 25, 26, 27 and 28 (CA)s were found, the allele frequency ranged from 8.97% to 32.00%. Heterozygotes of intron 22 (CA) repeat within FVIII gene were not found in the three populations. In conclusion, it was obvious that the locus cannot be acted as DNA genetic marker of FVIII gene in the three populations in Yunnan.
3.Clinical studies of 162 patients with von Willebrand disease.

OBJECTIVETo analyse the epidemiological feature, clinical characters and therapeutic regimens for von Willebrand disease(VWD).
METHODSThe clinical data and laboratory tests results of 162 VWD patients in our center were analyzed.
RESULTSThere were 76 males and 86 female among these patients with the mean age of 7.2(2.0-41.0) and 20.7(5.0-48.0) years, respectively. 86 patients (53.1%) were identified to be type 1 VWD, 34 patients (21.0%) type 3 VWD and 42 patients (25.9%) type 2 VWD. Among type 2 VWD patients, 33 patients were type 2A, 4 patients type 2M, 5 patients type 2B. Eighty-seven patients (53.7%) had a definite family history of bleeding tendency. The most common and specific bleeding symptoms were easy bruising (61.7%), epistaxis (53.7%), prolonged bleeding after surgery or minor injury (53.1%). Menorrhagia (66.3%) was common in female patients. The analysis of Vicenza bleeding scores in all patients showed that only 56(34.6%) patients had abnormal bleeding scores. FVIII/VWF concentrates and cryoprecipitate were applied to 45 patients (27.8% ), Desmopressin (DDAVP) to 8 patients. Eight female patients need oral contraceptives jointly to control menorrhagia. Hysterectomy had to be performed in 2 female patients with VWD.
CONCLUSIONVWD was a common congenital bleeding disorder with heterogeneous characters, it was necessary to screen, identify, classify accurately this disease in order to supply to effectively individualized treatment.
