1.The clinic observation of polysaccharide nucleicacid of bacille calmette-guerin injection on bronchus asthmatic patients

Objective To observe the influence on clinic symptom and pulmonary function in asthmatic patients treated with polysaccharide nucleicacid of calmette-guerin(BCG-PSN) injection in asthmatic patients.Methods 30 cases(treatment group) with additional BCG-PSN injection and 30 cases(control group) with only normal treatment were studied in contrast.They were observed in pulmonary function and clinic symptom before and after treatment.Results Symptom and pulmonary function improved in both groups.The validity rate in treatment group was 96.7%,and the control group was 80%.Conclusion BCG-PSN could alleviate the inflammation of bronchus and excellent effects of BCG-PSN injection in treatment on clinic symptom and pulmonary function of asthmatic patients were found.
2.Epidemiological investigation of dental fluorosis and dental caries in children from binhai new area

Objective To investigate the incidence of dental fluorosis and pit caries in school-age children from binhai new area of Tianjin, and to discuss the relationship between dental fluorosis and pit caries, providing the guidance for the prevention and treatment of them. Methods Referring to WHO’s“Oral Health Surveys Basic Methods”(Fourth Edition) and protocols of the third national oral epidemiology investigation, 3 778 children aged 7 to 9 year-old with similar levels of education were investigated for their dental fluorosis and fissure caries using the cross sectional survey method. Results The data showed that the incidence of dental fluorosis was less than 10%in the samples, and the community fluorosis index (CFI) was 0.075. The caries prevalence rate and mean decayed missing filled tooth (DMFT) of first permanent molars were 15.57%and 0.46 in male students, and those were 17.41%and 0.58 in female students. There were no significant differences in the caries prevalence rate and DMFT of first molar between male and female children (χ2=2.345, P>0.05). There were statistically significant differences in the caries rate and DMFT between different age groups (χ2=172.576, P<0.05), and both increased with age. Conclusion After years of defluoridation project in Tianjin, the detection rate and index of dental fluorosis in children have showed a downward trend. The caries prevalence rate may be related to the eruption rate of the first permanent molar.
3.The clinical significance of peripheral blood IL-33 and Da-jvGlu in acute cerebral infarction patients giving Shenqi injection

Objective To explore the levels of peripheral blood IL-33 and the difference of arteriovenous fistula glucose (Da-jvGlu) after giving Shenqi glucose injection in acute cerebral infarction patients.Methods The levels of plasma IL-33 and Da-jvGlu were detected in 126 patients of acute cerebral infarction and 10 normal controls.The correlation between IL-33 and cerebral infarction was analyzed.Results Compared with those in normal control group and the pretherapy groups,the levels of IL-33 in large volume were significantly higher (P<0.05).After giving Shenqi glucose injection,the level of IL-33 in mid volume and small volume were significantly higher between groups (P<0.05).Compared with those in normal control group and the pretherapy groups,the level of Da-jvGlu in large volume was significantly higher (P<0.05).After giving Shenqi glucose injection,there was no significant difference of Da-jvGlu in statics between groups (P>0.05).Conclusion Shenqi glucose injection can affect the levels of IL-33 to protect nerve cells and stabilize blood glucose concentration.
4. Correlation Between Effective Components Content and Color Values of Gentianae Radix et Rhizoma Based on Color Difference Principle

Objective:To measure the color values of Gentianae Radix et Rhizoma and the contents of gentiopicroside,the appearance index value of the representative color was correlated with the intrinsic index value representing the quality,in order to explore the correlation between the contents of active ingredients and the color values,and provide basis for the quality evaluation of Gentianae Radix et Rhizoma. Method:The color difference values of Gentianae Radix et Rhizoma powder was measured by colorimeter.The content determination method of gentiopicroside in the 2015 edition of China Pharmacopoeia was adopted. The content of gentiopicroside was determined by HPLC,and the correlation and regression analysis was carried out by SPSS 21.0 software. Result:There was a significantly positive correlation between the contents of gentiopicroside and L* (representing colorshade)and E* ab(representing total color difference)(P<0.01),while the contents of gentiopicroside and a* (representing color red-green direction) and b* (representing color yellow-blue direction) had a significantly negative correlation (P<0.01). Conclusion:The color value of Gentianae Radix et Rhizoma has a certain correlation with the content of gentiopicroside,and yellowish Gentianae Radix et Rhizoma contain more active ingredients, with a better quality.The contents of gentiopicroside active ingredients in Gentianae Radix et Rhizoma can be quickly predicted by determining the color difference values,which can provide a new idea for quality evaluation of this herb.
5.Causal relationship between gut microbiota and vestibular dysfunction: a mendelian randomization study

Objective Previous research has indicated a discernible association between gut microbiota and vestibular dysfunction; however,further investigation is required to establish a causal relationship. This study aims to comprehensively study the causal relationship between gut microbiota and vestibular dysfunction and determine the species of related gut microbes. Methods A two-sample Mendelian randomization analysis was performed. The gut microbiome data of 18 340 individuals obtained from the genome-wide association study (GWAS) by the MiBioGen consortium in 2021 were used as the exposure samples. The GWAS data related to vestibular dysfunction were used as the outcome samples. The inverse variance weighting method was employed to analyze the gut microbiome data. The results were assessed based on the odds ratio(OR)as the effect index with 95% confidence interval(CI). The stability and reliability of the findings were assessed using the leave-one-out,heterogeneity,horizontal pleiotropy,and false discovery rate approaches. Results The results indicated a significant association between an elevated abundance of Bifidobacterium and a decreased risk of vestibular dysfunction. Furthermore,the leave-one-out method confirmed the stability of the results and the absence of instrumental variables exerting substantial influence on the findings. The causal relationship was negative. The influence of heterogeneity and horizontal pleiotropy on the causal effect could be eliminated. Conclusion Bifidobacterium may affect vestibular dysfunction by influencing 5-HT through the “gut-brain axis”,but this result still needs further verification.

Objective:To investigate the effect of Yisui Jiedu prescription on hippocampal neuron damage in vascular dementia (VD) rats and to regulate phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt)/recombinant Bcl-2 associated death promoter (Bad) mechanisms of signaling pathways of neuronal apoptosis. Method:The 40 SD rats were divided into sham operation group, model group, donepezil hydrochloride group and Yisui Jiedu prescription group, with 10 rats in each group.VD animal model was prepared by bilateral carotid artery permanent ligation (2-VO) method.The sham operation group and the model group were intragastrically administered with normal saline, the donepezil hydrochloride group was intragastrically administered with donepezil hydrochloride 0.52 mg·kg-1. The Yisui Jiedu prescription group was administered with Yisui Jiedu prescription (11.11 g·kg-1), 1 time/d . After 30 days, Morris water maze was used to test the learning and memory ability of rats, hematoxylin-eosin (HE) staining was used to observe the histomorphological structure of hippocampal CA1 region. Ultrasound of neuron in rat hippocampal CA1 region was observed by transmission electron microscopy (TEM).Real-time fluorescent quantitative(Real-time PCR) was used to detect the Akt, Bad mRNA expression.Western blot was used to detect the Akt, p-Akt and Bad protein expression in hippocampus. Immunoglobulin A nephropathy (IgAN) is the primary glomerulonephritis with the highest incidence rate in the world. It is also the main cause of end-stage renal disease (ESRD) in China, which has brought heavy economic burden to the society and patient families. Traditional Chinese medicine (TCM) has certain advantages in treating IgAN. In TCM, IgAN is classified into consumptive disease, hematuria, and edema categories, with the location in the kidney and involving the lung, liver, and spleen. Professor Ren Jixue, a master of TCM, believes that kidney deficiency and spleen deficiency are the root causes of IgAN, and the throat is the source of the disease. He proposed the theory of throat-kidney correlation and used the method of tonifying kidney and invigorating spleen as well as detoxifying and relieving sore-throat to treat IgAN, achieving significant therapeutic effects. Studies have shown that IgAN is closely related to mucosal immune defense. IgAN patients often experience recurrent and gradually worsening symptoms due to mucosal infections, and polymeric Ig receptor (PIgR) is an important component of mucosal defense function. The lack of PIgR leads to the accumulation of IgA molecules in the mucosal lamina propria, and the molecules enter the bloodstream in large quantities and ultimately deposit in the kidneys, causing kidney damage. Complement regulatory protein complement receptor type 1 (CR1) exists on red blood cells and glomeruli and has the function of inhibiting the activation and differentiation of B cells, clearing immune complexes, and inhibiting excessive activation of the complement system. Therefore, regulating the immune defense function through the mucosal-renal axis mediated by PIgR-CR1 will be an important target for preventing and treating IgAN. Based on the theory of throat-kidney correlation, this article explores the effects and molecular mechanisms of tonifying kidney and invigorating spleen as well as detoxifying and relieving sore-throat in preventing and treating IgAN by regulating the mucosal-kidney axis mediated by PIgR-CR1. It provides effective theoretical support and a scientific basis for TCM prevention and treatment of IgAN based on the theory of throat-kidney correlation.

9.Mechanism of Chaishao Liujunzi Tang in Treatment of Ulcerative Colitis Through TLR4/MyD88/NF-κB Signaling Pathway

ObjectiveTo investigate the preventive and curative effect of Chaishao Liujunzi Tang (CSLJZT) on colonic mucosal injury induced by dextran sulfate sodium (DSS) in mice with ulcerative colitis (UC) and its mechanism. MethodFifty Balb/c male mice were randomly divided into normal group, model group, CSLJZT low-dose group, CSLJZT high-dose group, and sulfasalazine group. Except for the normal group, other groups were given 2.5% DSS freely for 7 d, and were given drug intervention after successful modeling for 7 d. Bodyweight, feces, and other general physiological statuses of mice were recorded every day, and disease activity index (DAI) scores were calculated.The colon length was measured, and stained by hematoxylin-eosin (HE) staining to observe the morphological changes of the colon.The enzyme-linked immunosorbent assay (ELISA) was used to determine the content of interleukin-1β (IL-1β), myeloperoxidase (MPO), and superoxide dismutase (SOD) in the serum. Western blot was used to determine the protein expression levels of Toll-like receptor 4 (TLR4), myeloid differentiation factor 88 (MyD88), nuclear factor kappa B (NF-κB), inhibitor-kappa binding protein (IκB), Caspase-1, and nucleotide-binding oligomerization domain-like receptor 3 (NLRP3) in the colon tissues. ResultAs compared with the normal group, mice in the model group had significantly decreased body weight (P<0.01), severe diarrhea and hematochezia, and significantly increased DAI score (P<0.01). As compared with the model group, the decreasing trend of body weight was significantly alleviated in the CSLJZT groups (P<0.01), diarrhea and hematochezia were significantly improved, DAI score was significantly decreased (P<0.01), and colon length increased (P<0.05). HE staining showed that the pathological damage of colon tissue was significantly improved and the inflammatory cell infiltration was reduced in the CSLJZT groups as compared with the model group. As compared with the normal group, the serum levels of IL-1β and MPO were significantly higher (P<0.01) and SOD levels were significantly lower (P<0.01) of mice in the model group.Compared with the model group, the treated group reduced the serum IL-1β and MPO levels (P<0.01), and raised the SOD level (P<0.01). The results of Western blot showed that as compared with the normal group, the expression levels of TLR4, MyD88, NF-κB, Ccaspase-1, and NLRP3 proteins were significantly increased (P<0.01), whereas the expression level of IκB protein was significantly decreased (P<0.01) in the colonic tissue of mice in the model group. As compared with the model group, the expression levels of TLR4, MyD88, NF-κB, Caspase-1, and NLRP3 proteins were decreased (P<0.01), whereas the expression level of IκB protein was increased (P<0.01) in the colonic tissue of mice in the CSLJZT groups. ConclusionCSLJZT improves the inflammatory injury of the colon tissue in DSS-induced UC mice through TLR4/MyD88/NF-κB signaling pathway.
10.Prediction of Platycodonis Radix Quality Marker Based on Network Pharmacology and Chemometrics Methods

OBJECTIVE To preliminary predict and analyze the quality marker(Q-markers)of Platycodonis Radix decoction based on the validity and measurability of traditional Chinese medicine Q-marker. METHODS The fingerprints of 20 batches of Platycodonis Radix samples were established, and the similarity was evaluated. At the same time, a "component-target-pathway" network diagram was constructed through network pharmacology to predict the potential active components of Platycodonis Radix. Mathematical analysis methods such as principal component analysis and orthogonal partial least-squares discrimination analysis were used to screen differential components; the content of candidate components were determined. RESULTS There were 19 common peaks in the established fingerprint, and 8 common components were identified as candidate active components for network pharmacology analysis. Network pharmacology indicated that 6 compounds were screened out with a high degree of connectivity. Based on the comprehensive consideration of Q-marker principle:traceability and transmissibility, specificity, efficacy, and measurability, luteolin, syringin, chlorogen acid, platycoside D, platycoside D3 and platycoside E were preliminarily predicted as potential Q-markers of platycodon. CONCLUSION The prediction analysis of Platycodonis Radix Q-marker provides a reference for the comprehensive control of the quality of Platycodonis Radix medicinal materials, and also lays a foundation for the research and exploration of the substance basis and mechanism of action of Platycodonis Radix.
