1.Discussion on magnesium ions in children with asthma exacerbation

Objective:To determine blood magnesium and urinary magnesium in children with asthma exacerbation and discuss the relationship between the magnesium level and children with asthma attack. Methods:The observation group include 90 children who were 0~6 years old with asthma exacerbation that admission to our hospital since May 2008 to May 2012. The control group was 100 children with the same age to the observation group to accept the health examination. Every child in two groups were taking venous blood of 2ml and 24h urine samples. Record the amount of urine at the same time. Using automatic biochemical analyzer to detect the serum magnesium concentration and urinary magnesium concentration. Results: The urinary magnesium concentration of observation group was obviously lower than control group (t=8.52, P<0.001);the serum magnesium concentration of two groups had no significant difference (t=2.21, P>0.05). Conclusion:Children with asthma in acute attack stage with magnesium deficiency. Urinary magnesium concentration determination is simple, and it is a effective method that can be used to judge whether there is magnesium deficiency. Serum magnesium concentration cannot be used as a reliable indicator of magnesium ion balance.
2.Analysis of total grastrectomy and proximal subtotal gastrectomy in gastric cardia and fundus carcinoma

Objective To explore the value of total grastrectomy in gastric cardia and fundus carcinoma. Methods 118 cases with cardiac cancer were treated by surgical treatment, those patients were admitted between May 1998 and May 2007 in Affiliated Baiyin Hospital of Lanzhou University. Of them,65 cases were treated by total gastractomy (group A),and 53 cases were treated by proximal gastractomy (group B). The degree of invasion, lymph node dissection, postoperative complication rate and survival rate were compared. Results The average num-ber of lymph node were 19.4±3.9 and 15.2±2.6 (P<0.05), respectively. The occurrence of lymph node metasta-sis were 40.8% (514/1261) and 54.8% (441/806),respectively. Of them, the occurrence of metastasis of lymph node of No. 5、No. 6 in group A were 15.4% (10/65)and 12.3% (8/65),respectively. The postoperative complica-tion rate were 7.7% (5/65) and 13.2% (7/53). The 1,3, and 5 year survival rates were 63.1%,46.2%,30.8% in group A and 66.0%,36.9%, 18.5% in group B, with significant differences beteen two groups in 3, 5 year sur-vival rates(P<0.05). Conclusion The way of total grastrectomy is better in cleaning lymph node than that of proximal gastrectomy, and dosen't increase the postoperative complication rate but improves 3-year and 5-year sur-vival rates in cases with cardiac cancer of Ⅰ, Ⅱ, Ⅲ stage.

Objective:To investigate the effect of Yisui Jiedu prescription on hippocampal neuron damage in vascular dementia (VD) rats and to regulate phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt)/recombinant Bcl-2 associated death promoter (Bad) mechanisms of signaling pathways of neuronal apoptosis. Method:The 40 SD rats were divided into sham operation group, model group, donepezil hydrochloride group and Yisui Jiedu prescription group, with 10 rats in each group.VD animal model was prepared by bilateral carotid artery permanent ligation (2-VO) method.The sham operation group and the model group were intragastrically administered with normal saline, the donepezil hydrochloride group was intragastrically administered with donepezil hydrochloride 0.52 mg·kg-1. The Yisui Jiedu prescription group was administered with Yisui Jiedu prescription (11.11 g·kg-1), 1 time/d . After 30 days, Morris water maze was used to test the learning and memory ability of rats, hematoxylin-eosin (HE) staining was used to observe the histomorphological structure of hippocampal CA1 region. Ultrasound of neuron in rat hippocampal CA1 region was observed by transmission electron microscopy (TEM).Real-time fluorescent quantitative(Real-time PCR) was used to detect the Akt, Bad mRNA expression.Western blot was used to detect the Akt, p-Akt and Bad protein expression in hippocampus. 7.Expansion technique for palatine mucoperiosteum: an experimental study

Objective: To establish the model of palatine mucoperiosteal expansion and observe a new possible treatment approach to soft tissue defect of cleft palate. Methods: Expanders of 10ml were implanted beneath the palatine mucoperiosteum in 4 dogs and injected 1.0ml 0.9% NaCl at the same time. 20 days after the placement of the expander, the expander was expanded by 1.0-1.5ml every 7 days again. The dogs were killed after the expanders reached their capacity. All the specimens were performed histomorphometric and histological examinations. Results: Histological results showed the increase in the layer of cell and the number of collagen fibra and blood vessel. All the specimens of expanded tissue showed the formation of extra palatine mucoperiosteal tissue. Conclusion: Expansion technique is able to form extra palatine mucoperiosteal tissue,which could be a new restoration approach to soft tissue defect of cleft palate and others.
8. Involvement of mitochondria-mediated pathway and Akt/GSK3β signaling in protective action of albi-florin against MPP+-induced PC12 cells apoptosis

OBJECTIVE: To determine the neuro-protective effects of albiflorin against MPP+-induced PC12 cells death and related molecular mechanisms. METHODS: PC12 cells were treated with 4 mmol·L-1 MPP+ to establish apoptotic cell models. MTT method, DCFH-DA staining, JC-1 staining and Western Blot were used to determine the changes of cell viability, intracellular ROS concentration, mitochondrial membrane potential and the expression of Bel-2 and Bcl-X1, and the phosphoration of Akt/GSK3β in PC12 cells. RESULTS: Albiflorin showed significantly neuro-protective effects against MPP+-induced cell damage. Results showed that albiflorin enhanced the cell viability, reduced intracellular ROS level and caspase 3 activation, restored the mitochondrial membrane potential, increased the expression of Bcl-2 and Bcl-X1, and enhanced the phosphoration of Akt/GSK3β. CONCLUSION: Albiflorin shows neuro-protective effects against MPP+-induced PC12 cells damage via mitochondrial-dependent pathway and Akt/GSK3β signaling.
9. Anti-hepatocellular carcinoma effects of Grifola frondosa extract and its underlying mechanisms

OBJECTIVE: To investigate the anti-hepatocellular carcinoma effect and underlying mechanisms. METHODS: In PLC/PRF/5 and HepG2, after treatment with Grifola frondosa extract, MTT method, chemical method, JC-1 staining and Western Blot were applied to determine cell viability, caspase 3 activity, mitochondrial membrane potential, the expression of Bcl-2 and Bax, and the phosphorylation of Akt/GSK3β. The anti-tumor activity of Grifola frondosa extract was further confirmed in PLC/PRL/5-xengrafted mice model. RESULTS: Grifola frondosa extract significantly reduced cell viability, mitochondrial membrane potential, the expression of Bcl-2 and the phosphorylation of Akt/GSK3β, and enhanced LDH release, caspase 3 activity and the expression of Bax in both PLC/PRF/5 and HepG2 cells. 12-day Grifola frondosa extract treatment significantly inhibited the PLC/PRF/5-xenografted tumor growth without influence the body weight of mouse. CONCLUSION: All these data indicate that Grifola frondosa extract-mediated anti-hepatocellular carcinoma effects are related to its modulation of the activations of Akt/GSK3β and mitochondrial pathway.
10. Structures of ginseng glycopeptides and anti-apoptotic effect on PC12 cells treated with Aβ25-35

Objective: To study the structures of ginseng glycopeptides, and to explore the protective effect on the apoptosis of PCI 2 cells treated with amyloid beta25-35 (Aβ25-35), and to lay a theoretical foundation for the development of ginseng anti-Alzheimer' s disease (AD) drugs. Methods: The structures of ginseng glycopeptides were analyzed by reversed-phase high performance liquid chromatography coupled with Q Exactive Orbitrap mass spectrometry. The PCI2 cells were divided into 6 groups, and treated with the medium including different concentrations 0, 6. 25, 12. 50, 25. 00, 50. 00, and 100. 00 mol • L_ 1 ) of Aβ25-35 , and the survival rates of PC12 cells were measured by cell counting kit-8 (CCK-8) method. The PC12 cells were divided into blank control group, model group, and ginseng glycopeptides administration group. The medium including 50. 00 jumol • L_ 1 Aβ25-35, was added in model group, and different concentrations 0. 03, 0.10, 0. 30, and of 1. 00 g • L_ 1 ) ginseng glycopeptides+ 50 jumol • L_ 1 Aβ25-35 were added in administration groups. The survival rates of PC12 cells were measured by CCK-8 method. The apoptotic rates of PCI 2 cells after treated with glycopeptides and Aβ25-35, were detected by Annexin V-FITC/PI method. The PCI 2 cells were divided into blank control group, model group (50.00 jumol • L_ 1 Aβ25-35) and different concentrations (0 . 1 0 and 0.30 g • L_ 1 ) of ginseng glycopeptides administration groups; the percentages of PCI 2 cells in different cell cycles were tested by flow cytometry. Results: More than 20 glycopeptide structures were obtained, such as the peptide chain was NLSHYHSGSS, the glycosyl group was Nl-HexNAc, and the peptide chain was SGSSSSSSSEDDGMGR, the glycosyl group was S6-HexNAc. When the concentration of Aβ25-35 was 50 jumol • L_ 1 , the survival rate of PC12 was (55. 45 + 2. 3 4) % and the survival rate was significantly lower than that in blank control group (P < 0 . 0 1) .
