1.Autosomal dominant polycystic kidney disease in children's early identification and outcome

Autosomal dominant polycystic kidney disease( ADPKD)is the most common hereditary renal disease,which has an incidence of 1 / 1 000 to 1 / 500 in live births,was characterized by renal enlargement along with renal tubular cystic dilatation,approximately half of affected individuals reach end stage renal disease(ESRD)by 60 years of age,accounting for 7% - 10% of patients with ESRD. ADPKD could be diagnosed in children and even in ute-ro,early identification and intervention in children would ameliorate the progression of ADPKD to ESRD. Therefore,it is imperative for pediatricians to recognize the early clinical manifestations and outcome of the disease. The aim of this re-view will focus on the early identification,management and outcome in childhood.

3.A Modified Method for Transfection to SKOV3 Cells with Protamine-DOT-AP/Chol Liposome-Oligodeoxynuleotides Complex

Objective: To observe the transfer efficiency of protamine-DOTAP/Chol liposome-oligodeoxynuleotides complex to SKOV3 cells. Methods; Synthesized NF-?B decoy ODNs or scrambled ODNs labeled by FITC on the 5'ends was mixed with rationic DOTAP/cholesteral liposome and protamine, making the charge ratio of positive: negative 4. SK-OV3 cells were transfected by the complex (1 ?mol/L) and the efficiency were measured by flow cytometry ( FCM) and fluorescence meter. The direct effect of NF-KB decoy ODNs transfer on the growth of SKOV3 cell line in vitro was measured by MTT method. Results: The efficiency of LPD transfer to SKOV3 cell line was 96. 41% , significantly higher than that of naked DNA transfer which was 12. 86%. The growth of SKOV3 cell line was not affected by 48h incubation after NF-KB decoy ODNs or scrambled ODNs transfection. Conclusion: LPD method is the one with high transfer efficiency not affected by NBS( newborn bovine serum) with possibility of use in vivo.
4.Abnormal DNA-binding abilities of NF-?B and glucocorticoid receptor in cortex of kindey from rats with Adriamycin-induced nephrosis

Objective To explore the role of transcription factors such as nuclear factor-kappa B(NF-?B) and glucocorticoid receptor(GR) in the pathogenesis of Adriamycin(ADM)-induced nephrosis in rats and the therapeutic effects of dexamethasone(Dex) and cyclosporin(CsA) on these animals.Methods The DNA-binding abilities of NF-?B and GR in cortex of kindey were examined with electrophoretic mobility shift assay(EMSA) and isotopic radioautography on the 7th,14th,21th and 28th day after a single intravenous injection of ADM,and the therapeutic effects of Dex and CsA were estimated.The biochemistry parameters from blood and urine of rats and the urine protein excretion were also measured.Results The NF-?B DNA-binding ability was significantly increased after 7 days and achieved maximum after 28 days(P0.05).Conclusion The DNA-binding ability of NF-?B is abnormally increased and that of GR is decreased in cells from cortex of kindey in Adriamycin-induced nephrotic rats.The NF-?B DNA-binding ability can be inhibited and the urine protein excretion is decreased by the treatment of CsA.
5.Study on physiological characteristics and the association with low prevalence of metabolic syndrome in Keriyans, a nomadic tribe, in Taklimakan Desert

Objective To investigate the physiological characteristics and the association with low prevalence of metabolic syndrome in Keriyans,a nomadic tribe,in the Taklimakan Desert.Methods An epidemiological survey focusing on diet and standardized physical examinations were made in a randomly selected natural population of 508 Keriyans individuals residing in the Taklimakan desert.In addition,237 of Uyghur subjects from Yutian county were enrolled as controls.Results Between these two populations,statistical significances were observed in body mass index (BMI) [(21.25 ± 3.10 vs 22.66 ± 4.18) kg/m2,P<0.05],blood pressure (P<0.01),triglyceride [TG,(0.98 ± 0.77 vs 1.50 ± 1.29) mmol/L,P<0.01],total cholesterol [TC,(2.23 ± 1.02 vs 3.26 ± 0.85) mmol/L,P<0.01],low density lipoprotein-cholesterol[LDL-C,(1.41 ± 0.68) mmol/L,P<0.01],fasting blood glucose [(4.43 ± 1.19 vs 5.56 ± 1.39) mmol/L,P<0.01],fat and salt intake (P<0.05).The Keriyans had a low incidence of hypertension(9.94% vs 29.11%,P<0.01),raised blood cholesterol and diabetes mellitus(2.2% vs 11.91%,P<0.01) compared with the control group.The prevalence of metabolic syndrome was 0.6% (3/500) in Keriyans and 13.62% (32/235) in the control group.The prevalence of metabolic syndrome in Keriyans was much lower than that in the control group (P<0.01).The caloric intake in keriyans group was lower tha than in the control group [(1 502.341 vs 2106.870) kcal,P < 0.01].Conclusions The preliminary results showed that the physiological characteristics with low BMI,blood glucose,and the low levels of TG,TC,LDL-C,as well as they were inured to low salt loading,little smoking and no alcohol drinking,and natural living environment free from pollution,these factors might be associated with the low prevalence of metabolic syndrome in Keriyans.And their low calorie intake might be the most important factor of their low prevalence of metabolic syndrome.
6.Effect of Different Compatibility Ratio of Gardenia jasminoides to Fermented Soybean on the Content of Flavo- noids in Zhizichi Decoction

OBJECTIVE:To investigate t he effects of different compatibility ratio of Gardenia jasminoides to fermented soybean on the content of genistein and total flavonoids ,and to investigate the compatibility regularity of Zhizichi decoction. METHODS:The decoction method was used to prepare the mixed decoction with different compatibility ratio of G. jasminoides to fermented soybean (2∶1,1∶1,1∶2,1∶4,m/m,the same hereinafter ). UPLC-MS/MS method was used to determine the content of genistein in Zhizichi decoction with different compatibility ratio and corresponding fermented soybean single decoction. UV method was used to determine the content of total flavonoids in Zhizichi decoction with different compatibility ratio and corresponding gardenia single decoction and fermented soybean single decoction. RESULTS :The established method had good linearity , precision,repeatability,stability and accuracy. Compared with single decoction ,the content of genistein in the mixed decoction with different compatibility ratio of G. jasminoides to fermented soybean (2∶1,1∶1,1∶2,1∶4)was decreased to different extents , while the content of total flavonoids was increased to different extents. With the increase of fermented soybean ,the content of genistein in the decoction increased at first and then decreased. When the compatibility ratios of G. jasminoides to fermented soybean were 1 ∶ 1 and 1 ∶ 2,the content of genistein in the decoction was the highest (all 0.071 μg/mL). With the increase of fermented soybean ,the content of total flavonoids in the decoction did not change regularly ;when the ratio of G. jasminoides to fermented soybean was 1 ∶ 1,the content of total flavonoids in the decoction was the highest (1.861 μg/mL). CONCLUSIONS : When the compatibility ratio of G. jasminoides to fermented soybean was 1 ∶ 1,the content of flavonoids in the decoction is the highest.
7.Traditional Chinese Medicine Regulation of MAPK Pathway for Treatment of Obesity: A Review

With a global rise in morbidity rates, obesity has become a pressing public health issue. With increased adipocyte number and volume as the main characteristics, obesity is also manifested by metabolic disorders to varying degrees. At the same time, obesity is a risk factor for diabetes, hypertension, stroke, cancer, and cardiovascular diseases, imposing burdens on society and families. Influenced by lifestyle, environment, behavior, and genetics, obesity is caused by the interaction of many factors, and its pathological process is complex, involving inflammation, autophagy, and intestinal dysbiosis. The mitogen-activated protein kinase (MAPK) cascade reaction, a pivotal signaling pathway, plays a crucial role in cellular processes such as proliferation, differentiation, apoptosis, and stress responses. Both Chinese and international studies indicate that the MAPK signaling pathway can effectively regulate obesity through various pathways, including the modulation of adipocyte differentiation and apoptosis, appetite control, and inflammation improvement. Moreover, traditional Chinese medicine (TCM) has demonstrated significant efficacy in preventing and treating obesity, leveraging advantages such as multiple targets, diverse components, and minimal adverse effects. Research indicates that the MAPK signaling pathway is a primary focus of TCM regulation in this context, although a systematic review in this field is currently lacking. Therefore, this paper, by reviewing the latest Chinese and international research, provided a concise overview of the basic structure of the MAPK pathway, with a specific emphasis on recent progress in TCM interventions targeting the MAPK pathway for obesity treatment. The results indicate that regulating adipose tissue formation, differentiation, and thermogenesis, reducing inflammation and oxidative stress levels, and improving insulin sensitivity and metabolic disorders seem to be the main ways for TCM to regulate the MAPK pathway to prevent and treat obesity. However, it is necessary to find more research methods and explore potential mechanisms underlying TCM formulations based on the MAPK pathway for obesity prevention and treatment.
8. Modern Pharmacological Effect and Mechanism of Shengmai San

Shengmai San was first recorded in Medical Origin and Insights, a medical book written by ZHANG Yuan\|su. Shengmai San composed of Ginseng Radix et Rhizoma, Ophiopogonis Radix and Schiandrae Chinensisin Fructus. Ginseng Radix et Rhizoma as the monarch drug in the prescription has effects in invigorating vigour potently, and promoting production of body fluid to quench thirst. Ophiopogonis Radix as a sweet-cold minister drug has effects in nourishing yin, clearing heat and generating fluid and moistening lung to stop cough in the prescription. Both medicines are combined to have a good effect in replenishing Qi. Schiandrae Chinensisin Fructus is an adjuvant medicine, with effects in acid astringency, retaining Yin with astringent and hidroschesis. Ginseng Radix et Rhizoma has effects in strengthening the healthy energy, and Ophiopogonis Radix has a effect in retaining yin with astringent. It is a classic prescription for treating deficiency of Qi and Yin. With the in-depth research of modern medical experts, it is found that Shengmai San not only has a significant therapeutic effect on cardiovascular diseases, central nervous system diseases and endocrine system diseases, but also a good effect on digestive system diseases, immune system and hematopoietic system diseases and shock. As China's aging population, irregular diet and other problems become increasingly serious, the incidences of cardiovascular diseases, senile dementia, cerebral infarction, diabetes, liver dysfunction and other diseases have become higher and younger, which is a serious threat to human health. Therefore, by consulting a large number of domestic literatures, the authors respectively elaborated the therapeutic effect and mechanism of Shengmai San and its modified prescriptions on the above diseases, with the aim to reveal the target and mechanism of this prescription on the above diseases, provide theoretical basis for better treatment of the above diseases and promote the clinical application of this prescription. In addition, it provides reference ideas for the research of other classical famous formula.
9. Standard Decoction of Saposhnikoviae Radix Pieces

Objective:To prepare standard decoction of Saposhnikoviae Radix pieces,and conduct a study on the quality standards, in order to provide the scientific reference for the study of clinical application and other decoction standard decoctions. Method:By the traditional water decocting method,11 batches of standard decoction of Saposhnikoviae Radix pieces were prepared to determine pH,prim-O-glucosylcimifugin and 4'-O-β-D-glucosyl-5-O-methylvisamminol content,calculate the paste rate and transfer rate,and investigate the feasibility of the measurement method. An analytical method for HPLC fingerprint was established. Hypersil ODS2 column (4.6 mm×250 mm,5 μm) was adopted and eluted with methanol-water as the mobile phase in a gradient mode. The detection wavelength was 254 nm,the flow rate was 1 mL·min-1,and the column temperature was 25℃. The fingerprint analysis method was used to evaluate the similarity of the 11 batches of standard decoction of Saposhnikoviae Radix pieces. Result:Through the determination of 11 batches of standard decoctions of Saposhnikoviae Radix pieces,the average pH value was 5.5,the paste-out rates were between 34.3% and 46.3%,and the average paste-out rate was 41.4%,the standard deviation was 3.7%, the transfer rate of prim-O-glucosylcimifugin was 66.8%-93.5%,the average metastasis rate was 79.4%,the standard deviation was 12.1%, the transfer rate of 4'-O-β-D-glucosyl-5-O-methylvisamminol was 70.4%-98.2%,the average transfer rate was 83.40%,the standard deviation of 10.8%. The results of various methodological studies were consistent with the requirements of RSD. The fingerprinting analysis was performed using similarity evaluation software for traditional Chinese medicine fingerprints,and 9 principal common peaks were identified. The similarity was higher than 0.9. Conclusion:The quality evaluation method established in this study has a good stability,precision and repeatability. The fingerprints have a high similarity,which is suitable for the quality evaluation of standard decoctions of Saposhnikoviae Radix pieces, and can provide the scientific basis for the quality evaluation of other related preparations.
10.Effects of Tongxinluo on MMP-3, 9 and PPARgamma expressions in atherosclerotic rabbits.

OBJECTIVETo study the effects of Tongxinluo on the expressions of matrix metalloproteinase-3 (MMP-3), MMP-9 and peroxisome proliferators-activated receptor gamma (PPARgamma) in atherosclerotic rabbits and explore the mechanism of its anti-atherosclerotic effect.
METHODSTwenty-four rabbits were randomized equally into control group, atherosclerotic model group (fed with high-fat diet for 14 weeks) and Tongxinluo group. The expressions of MMP-3, 9 and PPARgamma in the 3 groups were observed by means of immunohistochemistry.
RESULTSThe expressions of MMP-3, 9 and PPARgamma in the model group and Tongxinluo group were significantly higher than those in the control group (P<0.01). After high-fat diet feeding for 14 weeks, Tongxinluo group showed significantly lower expressions of MMP-3 and 9 but higher expression of PPARgamma than the model group (P<0.01).
CONCLUSIONTongxinluo can inhibit the expression of MMP-3 and 9 and increase the expression of PPARgamma, which might be the mechanism of its anti-atherosclerotic effect.
