1.Effects of vasopressin on shuttle-box passive avoidance behavior in rats

Arginine-8-vasopressin (AVP) could promote the acquisition of shuttle-box passive avoidance behavior and delay the extinction of the behavior when injected into the hippocampus of rats.Intrahippocampal injection of 6-hydroxydopamine (6-OHDA) prevented AVP from enhancing memory.The authors had also observed that AVP could accelerate the degradation of cate-cholamine in the hippocampus when the contents of monoamine transmitters and their metabolites were analyzed with high-performance liquid chromatography and electrochemical detection. These facts suggest that vasopressin promotes the shuttle-box passive avoidance behavior in rats,strcnthens long-term memory as well as short-term memory,and exerts its effect on memory through its enhancing the degradation of norepinephrine and/or dopamine in the hippocampus.
2.Terahertz spectroscopy for lung cancer detection: a preliminary study

Objective To preliminarily explore the application of terahertz spectroscopy in the early detection of lung cancer by comparing the differences among lung cancer tissues,paracancerous tissues,and normal lung tissues with terahertz spectroscopy and screen the suitable terahertz frequency for specific diagnosis of lung cancer.Methods Thirty groups of lung tissue samples from non-small cell lung cancer patients at stage of Ⅰ to Ⅲ A were collected,and each group contained lung cancer tissue,paracancerous tissues,and normal lung tissue from the same patient.These lung cancer specimens included 17 cases of adenocarcinoma,10 cases of squamous cell carcinoma,2 cases of large cell lung cancer,and 1 case of sarcomatoid carcinoma.Terahertz spectroscopy from 0.2 THz to 2.0 THz was used to detect these tissue samples,and the absorption coefficient was compared.Results ① The absorption coefficient was significantly higher in the lung cancer tissues than the normal lung tissues.② Terahertz spectroscopy at a frequency of 0.8 to 1.4 THz may have the specific diagnostic value of lung cancer.Conclusion Since the absorption rate is higher in the lung cancer than the normal lung tissue,which is suggested that terahertz spectroscopy may be regarded as a novel non-invasive diagnostic approach for lung cancer.The specific diagnostic value of the terahertz frequency may be 1.4-2.0 THz for lung cancer.

4.Children's iodine nutrition status before and after implementation of a new standard of iodized salt in Rizhao City of Shandong Province

Objective To analyze the impact of a new salt iodine standard on children's iodine nutrition status.Methods A survey on iodine nutritional status was carried out in Rizhao before and after salt iodine content adjustment (2011,2012-2014).A town was selected from 5 areas (east,west,south,north,middle) in every county of Rizhao,4 villages were selected from each town,and then edible salt samples were collected in 15 households from each village.A central primary school was selected in some sampled town,at least 20 of these children's instant urine samples were collected for detection of urinary iodine,and at least 40 children aged 8-10 years old were selected for examination of thyroid volume.Results Totally 1 164 salt samples were collected before the adjustment,93.7% (1 091/1 164) of salt iodine concentration was in 18-33 mg/kg;totally 400 urine samples of 8-10 children were collected and the median urinary iodine (MUI) was 250.7 μg/L;the thyroid glands of 200 children were inspected and the goiter rate with palpation was 3.0% (6/200).Totally 3 600 salt samples were collected after the adjustment,88.5% (3 186/3 600) of salt iodine concentration was in 18-33 mg/kg;totally 1 122 urine samples of 8-10 children were collected and the MUI was 140.4 μg/L;the thyroid glands of 1 336 children were inspected,the goiter rate with palpation was 1.0% (13/1 336).Conclusions After implementation of the new standard,children's iodine nutrition has down to appropriate levels.So the salt iodine concentration after adjustment may be safer and the salt iodization prevention strategies should be carried out for a long time in Rizhao City.
5.Study on Adjuvant Immunoactivity of Polysaccharides from Panax Japonicus

Objective To study the adjuvant immunoactivity of polysaccharides from Panax japonicus by alcohol of different concentrations;To discuss its part with the strongest adjuvant immunoactivity. Methods Polysaccharides from Panax japonicus was sunk with alcohol of different concentrations, and 30%alcohol compound, 60%and 90%alcohol polysaccharide were obtained. Different segments of polysaccharide and OVA protein were injected to mice once a week for three times for immunity. Five days after the last immunity, the mice were executed to collect blood, and the antibody titer was determined. The three parts of alcohol compound were scanned by infrared spectrum to determine the type of polysaccharide preliminarily. Results Compared with the control group, the antibody titer of different segments of polysaccharide obviously increased, especially the polysaccharide sunk by 60%alcohol. Infrared spectrum analysis showed that polysaccharides from Panax japonicus contained pyranose ring structure. Conclusion Polysaccharides from Panax japonicus has significant adjuvant immunoactivity, and polysaccharide sunk by 60%alcohol has the strongest adjuvant effects.
6.Effects of TotalPanax Japonicus Extract onD-galactose Induced Aging Mice

Objective To research the protective effects of totalPanax Japonicus extract on learning memory, antioxidation, and anti-apoptosis of aging mice,and explore the mechanism. Methods Male mice were randomly divided into normal group, model group, Vitamin E (VE) group, Panax Japonicus extract low and high dose group. Except for the normal group, the other groups were injected withD-gal on the back of the neck subcutaneously to establish aging model. Normal group and model group were given a gavage with saline and each treatment group was given a gavage with totalPanax Japonicus extract and VE once a day for 7 weeks after the aging model established. All mice were be measured their learning and memory ability in the eighth week. After the test, the morphological changes of CA1 neurons were observed by HE stain. SOD, GSH-Px, MDA levels in brain tissue were measured by biochemical method, the expressions of Bcl-2 and Bax mRNA were evaluated by RT-PCR.Results Mice inPanax Japonicus extract low and high dose group could spend less time in searching for the platform, improve the learning and memory ability. TotalPanax Japonicus extract increased the activity of SOD and GSH-Px, while decreased the content of MDA. In addition, it could increase the expression level of Bcl-2 mRNA and reduce the expression level of Bax mRNA as well.Conclusion TotalPanax Japonicus extract has anti-aging effect.
7.Treatment and prognosis analysis of acute leukemia patients during pregnancy

Objective:To explore the clinical characteristics, diagnosis and treatment of acute leukemia patients during pregnancy.Methods:The clinical data of 16 cases with acute leukemia during pregnancy from January 2009 to December 2018 in the First Affiliated Hospital of USTC were retrospectively analyzed. The diagnosis and treatment regimens, pregnancy outcome, the early fetus and survival status of patients were also analyzed.Results:All 16 leukemia cases were confirmedly diagnosed and classified by bone marrow puncture, including 13 cases of acute myeloid leukemia (5 cases of non-acute promyelocytic leukemia and 8 cases of acute promyelocytic leukemia) and 3 cases of acute lymphoblastic leukemia. At the time of confirmed diagnosis, 6 patients were in first trimester, 6 cases in second trimester and 4 cases in late trimester. As for pregnancy outcome, 1 patient had natural birthing, 5 patients underwent cesarean operation, 9 patients underwent artificial abortion and 1 patient had spontaneous abortion. Chemotherapy was performed in 15 patients during pregnancy, 11 patients received chemotherapy for treatment of primary disease after pregnancy, 3 patients died during the treatment. During the follow-up of 13 cases, 8 patients survived and 5 patients lost follow-up.Conclusions:Early diagnosis of acute leukemia during pregnancy is very important. Bone marrow puncture should be performed timely to make clear diagnosis when blood routine result is abnormal during antenatal care. Multidisciplinary consultation should be initiated in time, and the best treatment plan should be worked out to guard against serious complications during pregnancy.

Aim To investigate SIRT1 activity and an-ti-inflammatory effects of Chikusetsu oleanane saponin ( COS) on the RAW264. 7 macrophage cells induced by lipopolysaccharide ( LPS ) . Methods The nitric oxide ( NO) release was detected with Griess method. Western blot was used to detect the expression of tumor necrosis factor-α( TNF-α) , interleukin 1β( IL-1β) . Nuclear factor-κB ( NF-κB) and silent information ad-justment factor 1 ( SIRT1 ) were tested by immunofluo-rescence. Results 1 mg · L-1 LPS co-cultured with COS at 25~300 mg·L-1 had no significant effect on the growth of RAW264. 7 cells. Compared with the LPS group, COS effectively inhibited the NO release and suppressed the expression of TNF-α and IL-1β, and also inhibited the translocation of NF-κB and up-regulation of SIRT1 . Conclusion COS has protective effects on RAW264. 7 cells stimulated by LPS, which may be related to up-regulating the expression of SIRT1 and promoting the deacetylation of NF-κB, thereby in-hibiting the translocation of NF-κB and reducing the expression of TNF-α and IL-1β.
9.Synergistic Induction of Apoptosis by Chemotherapeutic Drugs and Cytokines in Mouse T-Lymphoma Cell Line

The objective of the study is to find out the synergistic effect on apoptosis resulting from the combination of chemotherapeutic drugs and some cytokines. Dexamethasone (DEX), etoposide (VP16), arsenic trioxide (As(2)O(3)) and alltrans-retinoic acid (ATRA) were added to the murine T lymphoma cell line RMA as well as to the cells preincubated with IL-2, IL-6 or GM-CSF, respectively. The effect on apoptosis was observed. All four chemotherapeutic drugs inhibited the cell proliferation. DEX, VP16 or As(2)O(3), except ATRA, singly induced apoptosis of RMA cells. The DEX concentration of inducing apoptosis was reduced when it was added together with ATRA. IL-2, IL-6 and GM-CSF did not induce apoptosis when the cytokines were added to RMA separately, however, apoptosis could be induced by combination of IL-2 and IL-6. The cytokines facilitated the apoptotic action of chemotherapeutic drugs, the drug concentration for inducing apoptosis decreased and the time period of starting apoptosis shortened. The results demonstrated that there was synergistic effect of chemotherapeutic drugs and some cytokines for induction of apoptosis. It raised an experimental basis for combination of chemotherapeutic drugs and some cytokines, especially IL-2, in the treatment of malignant lymphoma.
10.Erythropoietin mobilizes renal progenitor cells to reduce ischemic reperfusion injury in rats with nephron-sparing surgery

Objective To investigate the effects of the erythropoietin (EPO) on ischemia reperfusion injury (IRI) in rats with nephron-sparing surgery (NSS). Methods Fifty-four Sprague Dawley rats were divided into 3 groups randomly after right kidney nephrectomy: Sham group, NSS group (PBS+NSS) and EPO group (EPO+NSS). During NSS, renal artery was clamped for 40 min to induce IRI. Sham group just adopted exposure renal artery without vascular clamped. Rats in NSS group were injected intraperitoneally with PBS for 3 days before NSS. Rats in EPO group were injected intraperitoneally with EPO for 3 days before NSS. After 12 h, 24 h, 72 h, blood sample and renal tissues were collected. The serum creatinine (Scr) and urea nitrogen (BUN) were evaluated. The pathology injury was evaluated by HE staining. The CD24/CD133 double-positived renal progenitor cells (RPCs) were tested by flow cytometry. The CD133 and PCNA protein were quantified by immunohistochemical staining. The expressions of Wnt7b and β-catenin protein were detected by Western blotting. Results Rats in NSS group had more elevated Scr, BUN and pathology injury scores 12 h, 24 h and 72 h after operation than those in Sham group (all P<0.05). Compared with those in the NSS group, the Scr and BUN in the EPO group were significantly lower 24 h after the surgery (all P<0.05), and the pathology injury score also decreased (P<0.05). The proportion of RPCs, expressions of CD133 and PCNA, and expressions of Wnt7b and β-catenin protein were significantly higher after 24 h of the surgery in NSS group than those in the Sham group (all P<0.05). While compared with those in the NSS group, the proportion of RPCs and expressions of CD133, PCNA, Wnt7b and β-catenin increased at the EPO group (all P<0.05). Conclusions EPO can reduce the IRI after NSS, and its mechanism may be related to the mobilization of the RPCs by the Wnt7b/β-catenin signal pathway.
