1.Effects of estrogen on neuron structure and expression of estrogen receptor in hypotha-lamus

Objective To explore the changes of ER-IR and the ultra structure in the medial preoptic area, arcuate nuclei of early-aged mice treated with estrogen. Methods Immunohistochemistry assay and electron microscopy were used in this study. Results ER-IR in the medical preoptic area and arcuate nuclei were greatly reduced after estrogen was given. The cell nuclei of neurons in these areas migrated towards the side, the nuclear membrane became folded, synapse became richer, and the number of synapse vesicle increased. Conclusion Estrogen can affect the neuron structure and function through the change of estrogen receptor expression in the medial preoptic area and arcuate nuclei of the hypothalamus cardiovascular center.
2.The experimental study of the protective function of nabumetone on the injury of gastric mucosa caused by duodenogastric reflux

0.05 ). On the contrary, the COX-2 score in nabumetone group was obviously lower than that in the operation group (P 0.05 ). Conclusion Nabumetone has obvious therapeutic effects on bile-reflux gastritis caused by duodenogastric reflux. Nabumetone can significantly decrease the level of COX-2 in the gastric mucus membrane.
4.A clinical study on knee osteonecrosis in patients with systemic lupus erythematosus

Objective To investigate the clinical features of osteonecrosis of the knee (ONK) in patients with systemic lupus erythematosus (SLE).Methods A retrospective chart review was conducted to identify patients of SLE with ONK.The clinical characteristics of these patients were analyzed.Results Between 2008 and 2013,eighty-one patients with SLE were analyzed in the 210th Hospital of People's Liberation Army.Eleven of these patients were diagnosed as ONK.All of these ONK patients were female.The age of onset ranged from 19-57 (36±13) years.The disease duration ranged from 4.5-31.0 (13±6) years.The local clinical symptom included knee swelling with pain in 5 cases,only pain with no swelling in 1 patient,no symptoms in 5 patients.All patients were with aseptic necrosis of the femoral head.For laboratory examination,7 patients had elevated triglyceride lipids in blood.Three of 7 patients accompanied with cholesterol level increased.Total cumulative dosage of oral corticosteroids was (66 824±27 328) mg.Conclusion ONK in patients of SLE is common and the majority of them have impaired renal function and elevated blood lipids,as well as long-term glucocortico-steroid therapy.
5.Type 1 diabetes mellitus and stroke

Many studies have shown that the incidence of type 1 diabetes mellitus (T1DM) is increasing with an annual of 2% to 5%.As the same with type 2 diabetes mellitus,T1DM is also a risk factor for stroke.Although some T1DM studies have taken stroke as a component of the composite endpoint of cardiovascular events,a few studies have focused on the risk of stroke in patients with T1DM.Recent studies have shown that the risk of stroke in patients with T1DM is significantly higher than those without diabetes mellitus,especially in the population under the age of 50.In addition,the T1DM patients died of the risk of stroke are 3 to 4 times higher than the general population.This article reviews the relationship between T1DM and stroke.
6.Roles of statins in the prevention of ischemic stroke

Effective prevention is the best approach for reducing the burden of stroke.The reduction of low-density lipoprotein cholesterol with statins may decrease the risks of patients with the first ever stroke and ischemic stroke or recurrent stroke in patients with transient ischemic attack.In stroke prevention,the treatment of statins has become the most important advance following aspirin and antihypertensive treatment.This article reviews the roles of statins in the prevention of stroke.

9.Combination of carboxyamidotriazole and 1-Methyl-L-tryptophan has synergistic inhibtory effects on programmed death 1 expression

OBJECTIVE To evaluate whether the IDO1 inhibitor 1- methyl- L- tryptophan (1- MT) combine calcium influx inhibitor carboxyamidotriazole (CAI) could further enhance the suppression of programmed death 1 (PD-1) in CD8 + T cells and investigate the curative effect of the combined use. METHODS CD8 +T cells were isolated from normal mice spleen by negative selection using magnetic cell separation. The isolated CD8 +T cells were cultured in RPMI 1640 medium containing 10% FBS and 100 U·mL- 1 IL-2 and activated by the addition of anti-CD3 and anti-CD28 (1 g·L- 1 each mabs). CD8 + T cells were pretreated for 48 h with drug and the fluo- 3 as a marker of intracellular calcium concentration was detected by flow cytometry. The calcineurin (CaN) levels were assayed with ELISA in CD8+T cells after 48 h incubation with 10 μm CAI. The nuclear translocations of NFAT and AHR were detected by immunofluorescent staining after 48 h of drug treatment. The expression of PD-1 in CD8+T cells was analyzed by flow cytometry. RESULTS Intracellular fluorescent intensity was markedly debase due to CAI treatment(P<0.01). Meanwhile, the changes of CaN content had a resembled correlation (P<0.01). Immunofluorescence experiment showed that after combination therapy the transfer of NFAT and AHR in nuclear substantially reduced. Flow cytometry revealed that after the combination caused a significant decrease in PD-1 expression in CD8+T cells. CONCLUSION CAI and 1-MT could inhibit markedly the expression of PD-1 in CD8 +T cells by inhibiting the nuclear translocation of NFAT and AHR, respectively and the combination of them has synergetic effect.
10.Detection of?-catenin and c-myc expression in gastric adenocarcinoma by tissue microarray and its clinical significance

Objective To study the expression and clinical significance of?-catenin and c-myc in gastric adenocarcinoma and corresponding para cancer gastric mueosa.Methods Two hundreds and ninety seven samples were collected from 102 cases of gastric adenocarcinoma,including 102 cancerous tissues and 195 para-cancerous tissues.Each sample was made into 282,156 and 156 dots tissue microarrays.Expressions of?-catenin and c-myc proteins were detected by immunohistochemical stain- ing.Results The expressions of?-catenin and c-mye were increased gradually in the process of gastric carcinogenesis.The rates of?-catenin and c myc expression were higher in carcinoma than that in intesti- nal metaplasia( P<0.05,P<0.001).The aberrant expression of?-catenin was closely related to the depth of invasion(P<0.05),and the expression of c-myc was related to histological grade( P<0.05). The aberrant expression of?-catenin was significantly correlated with the expression of c-myc in gastric adenocarcinoma(P<0.05).Conclusions The aberrant expression of?-catenin may activate the expres sion of target gene c-myc,which plays a pivotal role in the carcinogenesis and progression of gastric adenocarcinoma.
