1.Effect of ischemic preconditioning on ischemia-reperfusion-injured myocardium and phosphorylated Akt during the disease course of diabetes in rats

Objective To evaluate the effect of ischemic preconditioning(IPC) on ischemia-reperfusion-injury of myocardium and phosphorylated Akt(p-Akt) during the disease course of diabetes in rats.Methods The rat model of diabetes mellitus was reprodused by streptozocin(40mg/kg,i.v.).Myocardial ischemia was then created by temporary ligation of a branch of the left anterior descending(LAD) coronary artery after 2 and 9 weeks,respectively.Thirty six SD rats were randomly divided into six groups(6 each):non-diabetic IPC(NDMIP) group,non-diabetic I/R(NDMIR) group,2-week diabetic IPC(2DMIP) group,2-week diabetic I/R(2DMIR) group,9-week diabetic IPC(9DMIP) group and 9-week diabetic I/R(9DMIR) group.Ischemic/reperfusion was induced by temporary occlusion of LAD coronary artery.After the experiment,creatine kinase-MB(CK-MB) isoenzyme and myocardial infarct size were measured,and the expression of p-Akt was detected by Western-blotting,the arrhythmia score was then evaluated.Results CK-MB level in NDMIP group was significantly lower than that in NDMIR group and 9DMIP group(P0.05);the arrhythmia score in 9DMIP group was significantly higher than that in 2DMIP group(P0.05);the expression of p-Akt in 9DMIP group was significantly lower than that in 2DMIP group(P0.05);the myocardial infarct size in 9DMIP group was larger than that in 2DMIP group.Conclusions IPC can provide significant microvascular protection against prolonged ischemia/reperfusion in acute diabetic rats,but not in chronic diabetic rats.The attenuation of myocardial protection by IPC may be associated with a decrease in p-Akt activation.

3.Effects of Sangju Qingjie Decoction on ventilator-induced lung injury in rats

Objective To investigate the effects of Sangju Qingjie Decoction on the ventilator-induced lung injury in rats.Methods Thirty-six healthy male SD rats of both sexes,weighing 300-350 g,were randomly divided into 3 groups(n =12 each):control group(group C); mechanical ventilation group(group V)and Sangju Qingjie Decoction group(group SJ).The animals were anesthetized with intraperitoneal 3% urethane 1.4 g/kg and tracheostomized.The femoral artery and vein were cannulated for blood sampling and drug administration.The animals were mechanically ventilated for 2.5 h(VT =40 ml/kg,RR 40 bpm,I:E =1:1,FiO2 21%).Ingroup SJ,Sangju Qingjie Decoction 300 g was injected into the stomach through a gastric tube once a day for 10 consecutive days,and mechanical ventilation was performed 2 h after the last injection.While in groups V and C,the equal volume of normal saline was given instead of Sangju Qingjie Decoction.Arterial blood samples were collected before ventilation,at the end of ventilation,and at 30 min after ventilation(T0-2)for blood gas analysis.Respiratory index(RI)and oxygenation index(OI)were calculated.The animals were then sacrificed and the lungs were removed for microscopic examination and determination of the content of TNF-α,IL-6 and IL-10 and W/D lung weight ratio.Results Compared with group C,RI at T1,2,the content of TNF-o,IL-6 and IL-10 and W/D ratio were significantly increased,and OI was significantly decreased at T1.2 in groups V and SJ(P < 0.05).Compared with group V,RI at T1,2,the content of TNF-α and IL-6 and W/D ratio were significantly decreased,and OI at T1,2 and the content of IL-10 were significantly increased in group SJ(P < 0.05).The pathological damage was attenuated in group SJ compared with group V.Conclusion Sangju Qingjie Decoction can attenuate the ventilator-induced lung injury through inhibition of inflammatory response in rats.
4.Clinical values of using platelet rich plasma with autologous fat injection in facial rejuvenation

Objective To explore the clinical value of autologous fat injection with platelet rich plasma in facial rejuvenation.Methods 35 female beauty seekers were collected in this study.They were divided into two groups:sample autologous fat group (FAT group,n=20) and combined autologous fat and platelet-rich plasma group (PRP group,n =15).The average level of injection was 34 ml.The fat was collected at the abdominal or thigh sites using 18 G needle connecting with negative pressure suction-pump.The fat was centrifugated 2 min to get rid of the lower layer of water and the upper layer of oil.Weigao PRP extract kit was used to obtain the PRP.The ratio of PRP to fat was 1:5.The injection technique was according to Colemen method.The survival rate,satisfaction and complication were compared between the 2 groups.Results There were no infection,hematoma,fat liquefaction necrosis occurred in the 2 groups.Compared with the FAT group,the skin texture and facial contour of the PRP group was improved.Conclusions The platelet rich plasma can improve the survival rate of fat and has a stable effect,which is one of the safe and ideal methods of facial rejuvenation.
5.The relationship between acceptance of disability and hope level in patients with total laryngectomy

Objective To explore the relationship between acceptance of disability and hope level in patients with total laryngectomy.Methods Totally 84 patients with total laryngectomy were recruited and investigated with the general information questionnaire,Acceptance of Disability Scale (ADS)and Herth Hope Index (HHI).Results The total score of ADS was (85.82±12.64),which showed medium level acceptance of disability.The total score of Herth Hope Index (HHI) was (35.50±8.12),which showed high level HHI.Significantly positive correlation was found between acceptance of disability and HHI.Conclusions Nurses should pay more attention to the acceptance of disability in patients with total laryngectomy,and provide effective psychological intervention to improve the patients' menial health and quality of life.
6.The Clinical analysis of duodenal trauma

Objective To perform the clinical characferistics of diagnosis and treatment of duodenal trauma and to improve the result of surgical treatment in patients with duodenal injury.Methods The clinical data of 32 cases with duodenal injury were analyzed retrospectively.Results All cases were surgically treated.26 cases were cured 13 cases were with complications,of which 9 cases with duodenal fistula,4 cases with peritoneal infection.6 cases died.Conclusion Early diagnosis and surgical treatment,proper management and intensive postoperative care can improve the result of patients with duodenal trauma and decrease complications.

Ginkgo is one of the most successful cases of botanical drugs developed by modern science and technology during the past fifty years all over the world. At present ginkgo has been applied to the prevention and treatment of cardiovascular disease widely, and has good clinical efficacy. Type 2 diabetes has been proved to be the risk equivalents of cardiovascular disease, therefore it has an important scientific significance for looking for more effective drugs of prevention and control of diabetes. To seek more efficient and safe drug from the plant medicine which has the function of regulate blood sugar and improve insulin resistance becomes a hotspot at home and abroad. Basic and clinical studies have shown the ginkgo preparations of Chinese medicine have certain regulation effect on blood sugar and insulin resistance. In this paper, we review the mechanisms and clinical applications of ginkgo preparations on diabetes and its applications during the past 10 years.

9.A novel immunization strategy to induce strong humoral responses against HIV-1 using combined DNA, recombinant vaccinia virus and protein vaccines.

To optimize the immunization strategy against HIV-1, a DNA vaccine was combined with a recombinant vaccinia virus (rTV) vaccine and a protein vaccine. Immune responses against HIV-1 were detected in 30 female guinea pigs divided into six groups. Three groups of guinea pigs were primed with HIV-1 DNA vaccine three times, boosted with rTV at week 14, and then boosted with gp140 protein at intervals of 4, 8 or 12 weeks. Simultaneously, the other three groups of animals were primed with rTV vaccine once, and then boosted with gp140 after 4, 8 or 12 weeks. The HIV-1 specific binding antibody and neutralizing antibody, in addition to the relative affinity of these antibodies, were detected at different time points after the final administration of vaccine in each group. The DNA-rTV-gp140 immune regimen induced higher titers and affinity levels of HIV-1 gp120/gp140 antibodies and stronger V1V2-gp70 antibodies than the rTV-gp140 regimen. In the guinea pigs that underwent the DNA-rTV-gp140 regimen, the highest V1V2-gp70 antibody was induced in the 12-week-interval group. However, the avidity of antibodies was improved in the 4-week-interval group. Using the rTV-gp140 immunization strategy, guinea pigs boosted at 8 or 12 weeks after rTV priming elicited stronger humoral responses than those boosted at 4 weeks after priming. In conclusion, this study shows that the immunization strategy of HIV-1 DNA vaccine priming, followed by rTV and protein vaccine boosting, could strengthen the humoral response against HIV-1. Longer intervals were better to induce V1V2-gp70-specific antibodies, while shorter intervals were more beneficial to enhance the avidity of antibodies.
