Objective: To discuss the effects of different joint line heights on patellofemoral joint (PFJ) contact pressure at different knee flexion angels in human cadaveric total knee arthroplasty. Methods: Seven normal fresh-frozen human cadaveric knees with intact joint cartilage were tested on an Instron 8501 testing machine with a load of 30 kg. The load of quadriceps tendon was adjusted to balance the specimen at the desired angles. Total knee arthroplasty were performed with femurs of increased lengths (i. e. the heights of joint line were +2 mm, +4 mm, and +6 mm) and tibial plateaus of different heights. The peak PFJ contact pressure was measured with Fuji pressure-sensitive film before and after total knee arthroplasty. Results: After total knee arthroplasty, at knee flexion angles of 60°, 90° and 120°, the peak contact pressures of lateral PFJ facet were significantly higher when the heights of joint line were +4 mm and +6 mm than those when the heights of joint line were 0 and +2 mm (P<0.05); at knee flexion angles of 30°, 60°, 90° and 120° the peak contact pressure of medial PFJ facet were significantly higher when the heights of joint line were +6 mm than those when the heights of joint line were 0, +2 mm, and +4 mm (P<0.05). Conclusion: The peak PFJ contact pressure increases if the height of joint line is elevated by more than 4 mm in total knee arthroplasty, so the variation of joint line should be controled within 4 mm in total knee arthroplasty.

AIM: To investigate the expressions of the cyclin dependent kinase inhibitor (CKI) in human corneal epithelium.METHODS: The expressions of CKI, P27, P21 and proliferating cell nuclear antigen (PCNA) were tested in different regions of corneal epithelium by SP immunohistochemistry.RESULTS: Limbal basal cells stained positively for PCNA while central corneal epithelium cells stained negatively for PCNA, their difference was statistically significant (P＜0.01).Positive staining for P27 and P21 were observed in central epithelium, but there was no positive staining in limbal epithelium. Their difference were also statistically significant (P＜0.01).CONCLUSION: The different expressions of CKI P27, P21and PCNA in different corneal epithelial regions suggest that in limbal basal layer there are a group of cells that have higher proliferative capacity staying in G1 status, namely stem cell.

Objective To analyze the mechanism of fentanyl combines midazolam,by comparison with some of the commonly used anesthetic method to determine in the ethical advantages of anesthesia.Methods 45 rats were randomly divided into three groups,Group A:1％ anesthetized by intraperitoneal injection of sodium pentobarbital 40 mg/kg; Group B:5 ％ ketamine 120 mg/kg intraperitoneal injection; Group C:midazolam 5 mg/kg + fentanyl 0.05 mg/kg Last intravenous anesthesia.Observe the effects of anesthesia.Results The rats which are used of fentanyl conbines midazolam anesthesia,the onset of anesthesia time:0.02 ± 0.03 min,maintenance of anesthesia time:89.73 ± 22.59 min,the the narcotic operation time:1.51 ± 0.30 min,the success of anesthesia cases:15 cases.Conclusions Fentanyl combines midazolam compared with the currently used method of anesthesia,the advantages of the more prominent,it has significant improvement and protection about the security,utilization and recovery ethics in experimental rats.

AIM: To discuss the effect of wearing customized curvature soft corneal contact lens to dry eye degree of flight attendants.
METHODS:Eighty cases (160 eyes) of flight attendants from China Southern were divided into two groups:control group 40 cases ( 80 eyes ) wearing ready-made Bausch soft corneal contact lens ( curvature 8. 4 ); the experiment group 40 cases ( 80 eyes ) , wearing Bausch soft corneal contact lens with customized curvature. Tear break-up time ( BUT ) , Schirmer Ⅰ test ( SⅠt ) and fluorescein ( FL ) staining were as dry eye evaluation index. The results was statistically analyzed.
RESULTS: BUT, SⅠt average shortening value of the experimental group were less than that of the control group, there was statistical significance (P<0. 01). FL staining positive increase, the number of experimental group was fewer than that of control group, with statistical significance (P<0. 05).
CONCLUSION: Wearing customized curvature soft corneal contact lens can prevent the flight dry eye more effectively than fixed curvature product.

Compared with C3 plant,C4 plant had evident growth advantage,higher rates of water and nutrition using,and higher bio-yield than C3 plant,Sugarcane was one of the typical C4 plants.DNA was extracted from sugarcane leaves,primers were designed by the cDNA sequence of PPDK gene from GenBank.Then DNA was amplified by LA-PCR(Long Acute PCR) method,ligated into pMD18-T vectors,and transformed into E.coli.JM109.Full-length PPDK gene sequence of sugarcane was obtained,the sequence was 13.5kb in length.For convenient of the next experiment,two digest sites(XhoI and NotI) were introduced into primers,the full-length PPDK gene was splitted to two parts,and was ligated into pMD18-T simple vectors,respectively.Whole PPDK gene clone of sugarcane was finished.The strains were storaged in the lab.

As one of the ten characteristics of cancer, inflammatory indicators have certain guiding significance for the prognosis and treatment of patients with malignant tumors of the digestive system. As a new type of inflammation index, the systemic inflammatory response index is significantly better than other traditional inflammatory indexes in evaluating the prognosis of patients with digestive system malignant tumors. This article reviews the related studies on the systemic inflammatory response index in evaluating the prognosis of digestive system malignant tumors.

A high active soybean isoflavone glucoside hydrolase-producing mould strain was isolated from spirit qu. Its optimal hydrolase-producing conditions were as follows: 2.5% wheat bran as carbon source, 1% NaNO3 as nitrogen source, initial pH7. 0, culture medium volume 40mL/250mL, inoculating quantity 8% , culture temperature 30℃, revolutions 160r/min and culture time 84h. The enzyme activity reached 82 U/mL. Cu2+ can inhibit Absidia sp. R strain from producing the hydrolase, the influence of other metal ions was not remarkable on it.

Objective To establish the anti-infective tissue engineered bones (TEBs) and evaluate the anti-infective and osteogenic effects of the infection-prevention TEBs on femoral large bone defects in goats.Methods Based on the controlledrelease antibiotic system fibrin gel-coated vancomycin alginate beads (FG-Vanco-AB),the infection-prevention TEBs were established and evaluated.They were transplanted into the critical-size defects in the right femurs of goats.TEBs without the controlled-release antibiotic system were used as controls and transplanted into the left femoral defects.The breakpoint sensitivity of vancomycin (5 mg/mL) for S.aureus was used as a standard concentration.Postoperatively,the vancomycin concentrations in the lesion site,in the adjacent site and in the circulation,as well as the anti-infective effects of the infection-prevention TEBs were evaluated by High-performance liquid chromatography (HPLC).Bone hcaling was assessed by histology,CT and ECT.The results were used to evaluate the osteogenic effect of the infection-prevention TEBs.Results Results from ESM,CLSM and in vivo tracing showed that the in vitro and in vivo survival conditions of seeded cells were analogous to those of TEBs.The effective concentration (over the bactericidal concentration) of vancomycin in bilateral defects and in blood lasted for 28 days,2 days and 7 days,respectively.The concentration of vancomycin in the femur decreased gradually from the grafted site to both ends.At 28 and 56 days postoperatively,the ECT results showed no significant difference between the right and left femurs.CT and histology demonstrated that at 14,28 and 112 days after surgery,bone defects in the bilateral femurs were repaired synchronously,and were completely covered by new bone tissue after 112 days.Conclusion The anti-infective TEBs were successfully established.FG-Vanco -AB in the transplanted sites provided the local bone tissues with anti-infective capability whilst not interfered the process of bone reconstnction and wound healing.

Through scale-up cultivation of Eriobotrya japonica suspension cells using WAVE bioreactor, the cell growth and ursolic acid (UA) accumulation were studied. The comparison test was carried out in the flask and the reactor with cell dry weight (DW) and UA content as evaluation indexes. The culture medium, DW and UA content were compared in 1 L and 5 L working volumes of bioreactor. The orthogonal test with main actors of inoculation amount, speed and angle of rotation was developed to find the optimal combination, in 1 L working volume of bioreactor. DW of the cell growth and the UA content in bioreactor were higher than those of the shaker by 105.5% and 27.65% respectively. In bioreactor, the dynamic changes of elements in the fluid culture, the dry weight of the cell growth and the UA content in 1 L and 5 L working volumes were similar. Inoculation of 80 g, rotational speed of 26 r · min(-1), and angle of 6 ° was the optimal combination, and the cell biomass of 19.01 g · L(-1) and the UA content of 27.750 mg · g(-1) were achieved after 100 h cultivation in 1 L working volume of bioreactor. WAVE Bioreactor is more suitable than flasks for the E. japonica cell suspension culture, and culture parameters can be achieved from 1 L to 5 L amplification.
