2.EFFECTS OF GENISTEIN ON THE EXPRESSION OF COLLAGEN I AND TRANSFORMING GROWTH FACTOR-?_1 OF OSTEOBLAST

Objective To investigate the effects of genistein(GEN) on the expression of collagen I and transforming growth factor-?1(TGF-?1 ) of osteoblast.Method The secondary generation of skull osteoblast of newborn SD rat was incubated with GEN.The cells were divided into six groups:control group,different dose of GEN(10-8,10-7,10-6,10-5mol/L,respectively) groups and E2 group( 10-10mol/L).MTT(OD),the contents of cell protein,the activity of alkaline phosphatase(ALP),the expression of collagen I and the content of TGF-?1 were detected.Results After 48h and 72h,the MTT(OD) of all GEN group and E2 group were significantly higher than those in control group.The MTT(OD) of control group and 10-8,10-7,10-6mol/L GEN groups in 72h were significantly higher than those in 48 h.The protein of 10-5,10-6 mol/L GEN group and E2 group were significantly higher than those in control group.The ALP activity of all GEN groups and E2 group were significantly higher than those in control group.The level of above indices were correlated with the dose of GEN.The expression of collagen I and the content of TGF-?1 in 10-7,10-6,10-5mol/L GEN group and E2 groups were higher than those in control group.They werecorrelated with the dose of GEN and TGF-?1.Conclusion GEN could stimulate proliferation and differentiation of osteoblast,and enhance the expression of collagen I and content of TGF?-1.Compared with E2,,there were similar effects with the higher dosage of GEN.
3.Study of sentinel node biopsy with methylene blue and patent blue violet injection in patients with breast cancer

Objective To study the identification rate of sentinel lymph node (SLN) in patients with breast cancer (BC) and the accuracy in predicting axillary lymph node(ALN) metastasis using methylene blue (MB) and patent blue violet (PBV) injection. Methods From October, 1999 to April, 2001, 94 patients with BC were selected for this study. Of them, 32 patients were injected with 1% MB and 62 patients with 1% PBV to identify SLN. All 94 patients underwent the axillary lymph node dissection. Results In MB group and PBV group , the SLN identification rate were 65.6% (21/32), 88.7% (55/62); the accuracy rate to predict axillary lymph node status were 90.5% (19/21), 98.2% (54/55) respectively. Conclusion Compared with MB ,PVB is the more ideal vital blue dye in identification of SNB.

Heterogeneous nuclear ribonulcleoprotein(hnRNP)plays a variety of roles in pre-mRNA splicing, nuclear export of mRNA and turnover. It is reported that hnRNP not only takes part in regulating the development of neuron and glial cells, but is closely related to various central nervous system diseases. This paper reviewed the structure, function and the role of hnRNP in the central nervous system in order to provide new insight into the molecule mechanism of nervous system diseases.
5.Antioxidation of soybean isoflavone in vascular endothelial cells with oxidative damage

BACKGROUND: Soybean isoflavone has a variety of bioactivities and its antioxidation becomes a hot spot of research in recent years. At present,the research of soybean isoflavone places more emphasis on animal experiment and clinical observation,but lacks research on cellular level of human body.OBJECTIVE: To observe the influence of soybean isoflavone in vascular endothelial cells with oxidative damage.DESIGN: Controlled trial and observation.SETTING: Central Laboratory, Institute of Public Health,Tianjin Medical University.MATERIALS: The experiment was completed in the Central Laboratory,Institute of Public Health,Tianjin Medical University from January to July 2002.The experimental materials included vascular endothelial cell strain in human umbilical vein,low density lipoprotein,soybean isoflavone and vitamin E,etc.METHODS: The vascular endothelial cells were cultured in vitro.The experiment was divided into 6 groups: blank control group,oxidative damage control group (malondialdehyde content was 1 μmol/L),oxidative damage+vitamin E control group(vitamin E was 50 μmol/L) and oxidative damage+soybean isoflavone 10,50,100 μ mol/L control group. The endothelial cells,which were joined with vitamin E and soybean isoflavone of different concentrations in advance to be incubated for 24 hours,were affected by oxidized low density lipoproteins and then cultured continually for 24 hours.All the indexes of antioxidation were determined in both extra-cell and intra-cell.MAIN OUTCOME MEASURES: Malondialdehyde content,activity of superoxide dismutase and glutathione peroxidase,the release condition of lactate dehydrogenase and productive quantity of nitrogen monoxide inside and outside the endothelial cells of each group.RESULTS: ①Comparison of malondialdehyde content,the activity of superoxide dismutase and glutathione peroxidase in endothelial cells of each group: The malondialdehyde content was higher significantly in oxidative damage control group than in blank control group (P < 0.01),but the activity of superoxide dismutase and glutathione peroxidase was lower significantly in oxidative damage control group than in blank control group(P < 0.01).The malondialdehyde content was lower significantly in oxidative damage+vitamin E control group,oxidative damage+soybean isoflavone 10,50,100 μmol/L control group than in oxidative damage control group(P < 0.01),but the activity of superoxide dismutase and glutathione peroxidase was higher significantly in oxidative damage+vitamin E control group,oxidative damage+soybean isoflavone 10,50,100 μ mol/L control group than in oxidative damage control group (P < 0.01). ②Comparison of the release condition of lactate dehydrogenase and the productive quantity of nitrogen monoxide in endothelial cells of each group: The release percentage of lactate dehydrogenase was higher significantly in oxidative damage control group than in blank control group (P < 0.01),but the productive quantity of nitrogen monoxide was lower significantly in oxidative damage control group than in blank control group(P < 0.01).The release percentage of lactate dehydrogenase was lower significantly in oxidative damage+vitamin E control group,oxidative damage+soybean isoflavone 10,50,100 μmol/L control group than in oxidative damage control group (P < 0.01),but the productive quantity of nitrogen monoxide was higher significantly in oxidative damage+vitamin E control group,oxidative damage+soybean isoflavone 10,50,100 μmol/L control group than in oxidative damage control group(P < 0.01).CONCLUSION: Soybean isoflavone can alleviate the oxidative damage in vascular endothelial cells,caused by oxidized low density lipoprotein,possibly through such antioxidization indexes as malondialdehyde content,the activity of superoxide dismutase and glutathione peroxidase,the release condition of lactate dehydrogenase and the productive quantity of nitrogen monoxide,etc.

7.Association of Gonadal Receptor Genes′ Microsatellites and Dysfunctional Attitudes in Adolescent with Major Depressive Disorder

Objective To explore the associations between microsatellites in gonadal receptor genes and dysfunctional attitudes in adolescent with major depressive disorder(MDD).Methods Polymerase chain reaction(PCR),capillary electrophoresis and genetic scanning were performed in testing the length of microsatellites in first-onset adolescent depressive patients.Dysfunctional attitudes scale(DAS) was used in rating the dysfunctional cognition of adolescent depressive sample.These results were tested by correlative analysis and comparison analysis.Results 1.There existed significantly negative correlation between microsatellite′s length in estrogen receptor ?(ER?) gene and total score of DAS in female adolescent patients with first-onset depressive disorder.2.DAS′ total score of shorter alleles′ group was significantly higher than that of longer alleles′ group on female′ estrogen receptor ?(ER?) Gene.Conclusion The microsatellite′s length of ER? and ER? gene may have associations with dysfunctional attitudes of female adolescent with MDD.
8.Effects of nuclear factor-?B on cell apoptosis and left ventricular segmental function in acute myocardial ischemia-reperfusion

Objective To probe the effects of nuclear factor kappa B (NF-?B) on cell apoptosis and left ventricular segmental function in acute ischemia repeffusion in dogs.Method Twenty-four dogs were randomly divided into three groups:without left anterior artery (lAD) ligation group (C group),LAD was occluded 30 min following reperfusion 120 minutes in isehemical reperfusion group (IR group),and dogs were administered with PDTC before LAD ligation in ischemical reperfusion plus pyorrole dithitocarbamate group (PDTC group).The left ventricular segmental function was detected by echo cardiography using strain rate anlysis software.EF measured by Simpson's method.Cardiac myocyte apoptosis numbers were determined by terminal deoxynudeotidy transferease-mediated biotinylated deoxyuridine triphosphate nick end labeling (TUNEL).lmmunohistochemistry and western-blot anylysis of NF-?B protein expression.Results NF-?B was obviously expression on injury myocardium of IR group,and increased significantly in contrast to control group (P0.05)Conclusions NF-?B might play an important role in acute myocardial ischemia reperfusion.PDTC reduces myocardial iscbemia/repeffasion injury by preventing expression of factor NF-?B.
9.Experimental study of the specific immunoresponse induced by p53 modified dendritic cells.

OBJECTIVETo explore the specific cytotoxic T lymphocyte (CTL) induced by dendritic cells (DC), which were transfected by the plasmid pC53-SN3 encoding p53 gene.
METHODSDC derived from HLA-A2(+) mononuclear cells of the 24-lung cancer patients was transfected with the plasmid pC53-SN3 by lipofectamine and then co-cultured with auto-unpurified T cells to induce potent CTL (T-pC53-SN3). The cytolysis of specific CTL against Calu-6, a HLA-A2(+) human lung cancer cell line, was measured by using lactate dehydrogenase (LDH) releasing assay.
RESULTSThe expression of CD(1a) and CD(83), the correlative markers of DC, increased apparently after transfected with plasmid pC53-SN3, the expression rate was (5.45 +/- 0.89)% and (3.26 +/- 0.47)% versus (52.15 +/- 11.56)% and (25.78 +/- 12.35)%. CD(14) decreased apparently, but other DC correlative markers of CD(1a), CD(40), CD(86), and HLA-DR remained almost the same as that before transfection. Compared with T-IL-2, the CTL derived from PBMNC stimulated by IL-2 (100 U/ml), the cytolytic activity of T-pC53-SN3 against Calu-6 cell line showed a significant increase, but cytolytic activity was 56.79 +/- 15.67 and 39.33 +/- 9.88, respectively, when effect cells: target cells was 10:1. The expression of the CD(8), CD(69), and CD(45)RO/CD(8) of T-pC53-SN3 cells increased significantly, but that of CD(3), CD(4), CD(86), ect, was not significantly different from those of T-pCMV-neo.
CONCLUSIONSIt showed that DC transfected by p53 gene could induce potent HLA-A(2) restrictive CTL to kill tumor cell efficiently.

10.Modified total pelvic floor reconstruction for repair of severe pelvic organ prolapse.

OBJECTIVETo evaluate the clinical outcome of a novel approach for pelvic floor reconstruction using synthetic mesh (modified total pelvic floor reconstruction) for treatment of pelvic organ prolapse.
METHODSModified total pelvic floor reconstruction was performed in 30 patients with severe pelvic organ prolapse (including vault prolapse). The clinical outcome of each patient was assessed.
RESULTSThe mean operation time was (74.2 +/- 21.5) minutes, and the mean blood loss was (103.3 +/- 40.1) ml. Among them, 23 patients (76.7%) were able to micturate spontaneously the next morning after surgery, with residual urine less than 100 ml. The mean post-operative hospital stay was (4.2 +/- 1.8) days. All patients were followed up for a medium of 6 months. Totally 93.3% and 96.7% of patients were objectively (according to Pelvic Organ Prolapse Quantitive Examination score) and subjectively (according to Prolapse Quality of Life) cured, respectively. During follow-up, only one patient was found to have asymptomatic erosion, and de novo urgent urinary incontinence was seen in 6.7% of patients. The most prominent complication was dyspareunia (66.7%). Although the post-operative sexual function was reported to be worse, no significant difference between patients' pre- and post-operative Pelvic Organ Prolapse/ Urinary Incontinence Sexual Function Questionnaire-12 scores was noted.
CONCLUSIONModified total pelvic floor reconstruction is a safe, effective, and micro-invasive approach for severe pelvic organ prolapse repair; however, dyspareunia remains a main concern.
