1.Cloning Expression and Functional Verification of recF and recO Gene from Deinococcus radiodurans R1 in Escherichia coli

Objective To construct expressing recombinant of recF and recO from Deinococcus radiodurans R1and express the target protein in E.coli BL21(DE)3.Methods recF and recO gene were amplified by PCR from genomic DNA of Deinococcus radiodurans R1 and inserted into expression plasmid vector pET30b(+) to construct pET30b(+)-recF and pET30b(+)-recO.The recombinant plasmids were transformed into E.coli BL21(DE)3 and the recombinant proteins were expressed by the isopropyl-?-D-thiogalactopyranoside(IPTG) and were analyzed with SDS-PAGE.The changes in the survival rate of bacteria in each group before-and after UV-irradiation were calculated.Results The recombinant plasmid pET30b(+)-recO and pET30b(+)-recF was obtained and the recombinant protein could be highly expressed in E.coli BL21(DE)3.Conclusion This study has provided a foundation for further studies and applications of the recombinant RecF and RecO,and initial detection shows that recO and recF gene can increase the resistant ability of E.coli.
2.Cloning for Full-length Pyruvate Orthophosphate Dikinase Gene of Sugarcane

Compared with C3 plant,C4 plant had evident growth advantage,higher rates of water and nutrition using,and higher bio-yield than C3 plant,Sugarcane was one of the typical C4 plants.DNA was extracted from sugarcane leaves,primers were designed by the cDNA sequence of PPDK gene from GenBank.Then DNA was amplified by LA-PCR(Long Acute PCR) method,ligated into pMD18-T vectors,and transformed into E.coli.JM109.Full-length PPDK gene sequence of sugarcane was obtained,the sequence was 13.5kb in length.For convenient of the next experiment,two digest sites(XhoI and NotI) were introduced into primers,the full-length PPDK gene was splitted to two parts,and was ligated into pMD18-T simple vectors,respectively.Whole PPDK gene clone of sugarcane was finished.The strains were storaged in the lab.
3.Experimental study on diagnosing the brachial plexus injury through quickly detecting the ChAT activity

Objective To explore a better method to diagnose the brachial plexus injury as pre-or post-ganglionic,which is quick,simple,quantitative,cheap and accurate.Methods Forty-five SD rats were divided randomly ino pre-or post-ganglionic brachial plexus injury models group,and the choline acetyl- transferase(CHAT)activity was detected by using the radioisotope after different days.Results The ChAT activity of two groups both fall day by day:in the pre-agnglionic group,it falls quickly,and reach 1/10 of nor- mal(675?258)cpm;in the post-ganglionic group,it falls slowly,and reach 1/2 of normal even after 90 days (4906?1119)cpm.Conclusion Intra-operative measure of ChAT can be used to differentiate pre-or post- ganglionic brachial plexus injury.
4.Pain in human skin caused by laser stimulation

Laser techniques are widely applied in medical research and military affairs. The characters of laser make it the best way to evoke pain.Pain induced by laser stimuli is influenced by laser parameters such as wavelength, pulse duration and stimulus area in addition to the properties of skin such as the distance from the brain, type and color of skin. In this review,both laser evoked pain and factors influencing it are discussed.
5.Clinical efficacy of ALA-PDT in the treatment of condyloma acuminatum

Objective To evaluate the clinical efficacy of 5-aminolevulinic acid-induced photodynamic therapy (ALA-PDT) in the treatment of condyloma acuminatum. Methods Thirty-six cases of condyloma acuminatum were treated with ALA-PDT. Results After the treatment, 33 (91.67%) patients were cured, 35 (97.22%) improved. Relapse occurred in 3 (8.33%) patients. Side effects were noted in 2 (5.56%) patients. Conclusions ALA-PDT is a relatively simple, effective and safe treatment with a lower recurrence rate for condyloma acuminatum, especially for small skin lesions or lesions at specific area. However, the high expense required for multiple treatments with ALA-PDT may restrict its application.
7.Feasibility study of Shuyisha as hemostasis and repair material for liver wound

Objective To discuss the feasibility of Shuyisha as hemostasis and repair material for liver wound. Methods Hemolysis rate, acute toxicity and eytotoxicity of Shuyisha were measured. A hemorrhage model was established by making an open wound (5 mm× 3 nun ×2 mm) on the left liver lobe of mice. Hemostasis was performed with Shuyisha in experimental group and with Surgicel in control group, when the hemostatic time and total blood loss (TBL) were accurately recorded and regular macro-scopic and histological observation carried out. Results The hemolysis rate of Shuyisha was 2.33%, with maximum tolerance does of over 0.48 g/kg and the eytotoxicity at zero. The hemostatie time of Shuy-isha was (5.00 ±0.00) s, with total blood loss of (0.88±0.18) g/kg, better than Surgicel (P< 0.05). Shuyisha was degraded completely within 14 days, with the wound healed within 21 days in ex-perimental group, much better than Surgieel. Conclusions The hemolysis rate, acute toxicity and cy-totoxicity of Shuyisha are up to the requirement of biomedical materials. Shuyisha has effective hemosta-sis, which may be related to its molecular structure and adhesion.

Objective To evaluate the eradication rate and long-term therapeutic effect of a triple therapy consisted of cla-(rithromycin) (CLA), amoxicillin (AMO)and omeperazole on Hp infection,and explore the alternative therapeutic programs and their effects after first therapeutic failure.Methods A total of 92 children with Hp infection were divided into two groups: 70 children were given the triple therapy for one week (CLA group);Twenty-two children were given another triple therapy composed of metronida-(zoole) (MET), AMO and omeperazole for two weeks (MET group).All of the children were followed up for 1-30 months after the therapies ended.Children of the two groups who were therapeutic failure were given retreatment as follows.CLA triple therapy were given for one week to the children who were failure after MET triple therapy;increased doses of CLA with longer treatment course was given to the children who were failure after CLA triple therapy . A tetra therapy consisted of colloidal bismuth subcitrate (CBS), furazolidone (FUR) ,omeperazole and AMO was given to children in whom the retreatment failed.Results The Hp eradication rate of CLA group was 91.4%(64/70),and the Hp eradication rate of MET group was 72.7%(16/22).There was significant difference between eradication rate of the two groups(?~2=5.16 P
9.Clinical analysis of 36 cases of pernicious placenta previa

Objective To investigate the risk factors,management and prophylaxis of dangerous placenta previa.Methods The clinical data of 407 cases of placenta previa were reviewed,36 cases of them with dangerous placenta previa.Comparison was made between group A (dangerous placenta previa,36 cases) and group B (non dangerous placenta previa,371 cases) in terms of age,number of pregnancy and number of ceasarean delivery.Results The incidence rates of pregnant women in group A age ≥ 35years,number of pregnancy ≥3times,and cesarean delivery≥2times were 55.6%,61.1%,25.0%,which were significantly higher than those of group B (25.6%,27.7%,2.4%),the differences were statistically significant (x2 =7.71,8.99,21.97,all P < 0.01).Among the 36 cases of dangerous placenta previa,12 cases were diagnosed with concomitant placenta increta.The specificity and sensitivity for detection of placenta increta were 90.5% (19/21) and 83.9% (10/12) of ultrasonography respectively,and those were 100.0% (3/3) and 87.5% (7/8) of MRI.Of the 36 dangerous placenta previa patients,two patients were treated with induced labour due to unexpected pregnancy under 28 weeks,while the others were refer to surgical operation.5 surgical patients experienced uncontrollable postpartum haemorrhage which necessitated histectomy,while the other 31 cases received conservative operation.Rate of histectomy was 13.9(5/36).Conclusion Dangerous placenta previa is associated with advanced age,multiple pregnancy and multiple cesarean delivery.Accurate preoperational evaluation of the placenta previa should be acquired with ultrasonography and MRI to detect placenta increta,provide information for treatment selection,and prevent postpartum haemorrhage.
10.Clinical analysis of elderly type 2 diabetes mellitus patients with skin complications

Objective To analyze the prevalence and characteristics of skin diseases in elderly patients with type 2 diabetes mellitus (T2DM).Methods Clinical out patient records for both elderly and non-elderly individuals with T2DM were collected from the dermatology department of Beijing Hospital.Statistical comparisons were performed between the elderly group and the non-elderly group on the prevalence of common skin complications and disease characteristics.Analysis was performed in the elderly group to identify any association of common skin disorders with gender,age,duration of T2DM,or level of glycosylated hemoglobin (HbA1c).Results Among 679 elderly T2DM cases,pruritus was the most common complication (26.1%),followed by eczema (21.t %) and tinea pedis/onychomycosis (18.3%).Pruritus was more prevalent in the elderly group (177 cases,26.1%) compared to the non-elderly group (34 cases,9.8%) (x2 =6.006,P<0.01).On the other hand,tinea pedis/onychomycosis was less prevalent in the senior group (124 cases,18.3 %) than in the non-senior group (112 cases,32.2 %) (x2 =37.437,P<0.01).In the senior group,pruritus was more prevalent among male patients (105 out of 358 cases,29.3%) than among female patients(72 out of 321 cases,22.4%) (x2 =4.181,P =0.041).On average,patients with eczema were older[(72.3 ± 2.2)years] and had a longer history of T2DM [(8.3 ± 0.3)years] compared with patients without eczema[age:(65.2±1.9)years,history of T2DM:(5.2±0.1)years] (t=1.250,P=0.023 for age;t=1.680,P=0.033 for disease duration).Patients with tinea pedis/onychomycosis had higher HbA1c levels (6.7 ± 0.4) % than patients without tinea pedis/onychomycosis (6.1 ± 0.3) % (t =0.790,P<0.01).Conclusions Pruritus is the most common skin complication in elderly patients with type 2 diabetes and is more common in elderly patients and male patients.Risk factors for developing eczema in elderly patients with type 2 diabetes may include older age and long disease duration.High levels of glycosylated hemoglobin in elderly patients with type 2 diabetes indicates increased risk for foot fungi infections.
