1.Assessment of tooth bleaching efficacy with spectrophotometer.

OBJECTIVETo analyze the changes in CIE L*, a*, and b* at cervical, body, and incisal sites after tooth bleaching by using a spectrophotometer.
METHODSSixty-seven intact and healthy maxillary central incisors were in-vestigated. These incisors were darker than A3 according to the Vita Classical shade guide. The CIE tooth shade parameters L*, a*, and b* were simultaneously recorded at three tooth areas (cervical, body, and incisal) with a spectrophotometer before and after tooth bleaching (35%H2O2 coordinating with Beyond whitening accelerator irradiating). The shade dif-ferential (DeltaE) was calculated. ANOVA, paired t-test, and Pearson correlation analysis were used for data analysis.
RESULTSThe efficacy rates of tooth bleaching were satisfactory, with 86.6%, 86.6%, and 85.1% in the cervical, body, and incisal sites, respectively. The average values of DeltaE were 5.09, 4.44, and 4.40 in the cervical, body, and incisal sites. Tooth bleaching significantly increased L* and significantly decreased a* and b* in all tooth areas (P < 0.01). The decreasing range of Deltab* was more than the increasing range of DeltaL* at the cervical site; opposite results were observed at the incisal site. A positive correlation was detected between baseline b* and DeltaE.
CONCLUSIONThe spectrophotometer could objectively evaluate the whitening effect of tooth bleaching at the different tooth sites. The tooth bleaching system (35%H202 coordinating with Beyond whitening accelerator irradiating) exerts powerful bleaching actions in most of the tooth areas investigated. The order of tooth bleaching effectiveness is cervicalbody>incisal. Yellow coloration is decreased mainly at the cervical site, and brightness was increased mostly at theincisal site. The effectiveness of tooth bleaching increases as the baseline b* value increases.

2.Optimization of vancomycin and linezolid dosage regimen for treatment of Gram-positive cocci infections

Objective To estimate and optimize the dosage regimen of vancomycin and linezolid for treatment in different Gram-positive cocci infections. Methods The pharmacokinetic data of vancomycin and linezolid were collected, and the pharmacodynamics in vitro of these drugs for staphylococcus epidermidis, staphylococcus aureus, enterococcus faecalis and enterococcus faecium were analysed. The cumulative response fraction (CFR) was evaluated in different dosage regimens of two drugs against four types of bacteria. Results The regimen of 3 500 mg/d vancomycin was recommended for patients with staphylococcus epidermidis infection. The regimen of 2 500 mg/d vancomycin was recommended for patients with staphylococcus aureus infection. The regimens of 3 000 mg/d vancomycin and 400 mg linezolid given 2 times/day were recommended for patients with enterococcus faecalis infection. The regimens of 2 500 mg/d vancomycin and 400 mg linezolid given 2 times/day were recommended for patients with enterococcus faecium infection. Conclusion In application of vancomycin and linezolid for treatment of Gram-positive cocci infections, different dosage regimens should be used in different types of infections.
3.Effects of Sangju Qingjie Decoction on ventilator-induced lung injury in rats

Objective To investigate the effects of Sangju Qingjie Decoction on the ventilator-induced lung injury in rats.Methods Thirty-six healthy male SD rats of both sexes,weighing 300-350 g,were randomly divided into 3 groups(n =12 each):control group(group C); mechanical ventilation group(group V)and Sangju Qingjie Decoction group(group SJ).The animals were anesthetized with intraperitoneal 3% urethane 1.4 g/kg and tracheostomized.The femoral artery and vein were cannulated for blood sampling and drug administration.The animals were mechanically ventilated for 2.5 h(VT =40 ml/kg,RR 40 bpm,I:E =1:1,FiO2 21%).Ingroup SJ,Sangju Qingjie Decoction 300 g was injected into the stomach through a gastric tube once a day for 10 consecutive days,and mechanical ventilation was performed 2 h after the last injection.While in groups V and C,the equal volume of normal saline was given instead of Sangju Qingjie Decoction.Arterial blood samples were collected before ventilation,at the end of ventilation,and at 30 min after ventilation(T0-2)for blood gas analysis.Respiratory index(RI)and oxygenation index(OI)were calculated.The animals were then sacrificed and the lungs were removed for microscopic examination and determination of the content of TNF-α,IL-6 and IL-10 and W/D lung weight ratio.Results Compared with group C,RI at T1,2,the content of TNF-o,IL-6 and IL-10 and W/D ratio were significantly increased,and OI was significantly decreased at T1.2 in groups V and SJ(P < 0.05).Compared with group V,RI at T1,2,the content of TNF-α and IL-6 and W/D ratio were significantly decreased,and OI at T1,2 and the content of IL-10 were significantly increased in group SJ(P < 0.05).The pathological damage was attenuated in group SJ compared with group V.Conclusion Sangju Qingjie Decoction can attenuate the ventilator-induced lung injury through inhibition of inflammatory response in rats.
4.Role of NF-κB in apoptosis of immortalized neural progenitor cells

Objective To investigate the role of NF-κB in apoptosis of immortalized neural progenitor cells (INPCs) . Methods INPCs were cultured in 6-well plates and were randomly divided into 5 groups ( n = 6 each) : group I was not transfected with any plasmid (group INPC); group Ⅱ was transfected with control plasmid (group INPC/CMV); group Ⅲ was transfected with plasmid RcCMV-p50 (group INPC/p50); group Ⅳ was transfected with plasmid RcCMV-p65 (group INPC/p65) and group V was transfected with plasmid RcCMV-p50 and RcCMV-p65 (group INPC/p50p65). Group INPC/CMV ( H ), INPC/p50 (Ⅲ) and INPC/p65 (Ⅳ) were screened by G418, and the positive clones were then cultured for 3-4 weeks. The transcription of p50 mRNA or p6S mRNA was detected by RT-PCR. The NF-κB activity was measured by luciferase reporter gene assay. The cell apoptosis was measured by annexin V/PI staining. In group INPC/p50p65 and group INPC/p65, the cultured positive clone was transiently transfected with plasmid RcCMV-p50. Two days after transfection, the same measurement was performed in group INPC/pS0p65 and the other groups. Results The expression of p50 mRNA was significantly increased in group INPC/p50 and INPC/p50p65 as compared with the other groups ( P < 0.05) . The expression of p65 mRNA, the NF-κB activity and the apoptotic rate were significantly increased in group INPC/p65 and INPC/p50p65 as compared with the other groups ( P < 0.05). Conclusion Enhanced NF-κB activity can increase immortalized neural progenitor cell apoptosis.
5.A preliminary study of the application of Montreal Cognitive Assessment Beijing version in community dwelling older adults residing in Shenyang

ObjectiveTo assess the value of the Beijing version of Montreal Cognitive Assessment ( MoCA) in community dwelling older adults residing in Shenyang,China.MethodsThe stratified random sampling method was used to investigate the population over 60 years old in 4 communities of Shenyang in the year 2011.Mini-Mental State Examination (MMSE) and the Beijing version of MoCA were administered to all participants.258 old people finished the assessment.ResultsThe internal consistency of the MoCA Beijing version was good,yielding a Cronbach alpha of 0.836.The correlation between the MoCA Beijing version and the MMSE was good(r=0.623,P<0.001 ).Only 15.1% participants had an education of over 12 years,but 26.3% participants had an education of 6 years or less.Only 3 items of MoCA Beijing version ( naming lion,forward digit span,recall daisy)showednosignificantdifferencebetweenpersonswithandwithoutover 6yearsofeducation.ConclusionsThe results indicates that the Beijing version of MoCA have good reliability and validity.This study shows that an education of 6 years or less might be the proper population to add the one point in China,and the cutoff-point of 26 for normal is too high for Chinese population.
6.Study of macrophage uptake drug and oxygen loaded lipid microbubbles in vitro

Objective To investigate the feasibility and parameters for macrophage uptake oxygen and paclitaxel loaded lipid microbubbles (OPLMBs) in vitro.Methods OPLMBs were synthesized by mechanical vibration method and incubated with macrophages in different conditions in vitro.The ratio of macrophages uptake OPLMBs was observed by bright field microscope.Results The ratio of macrophages uptake OPLMBs was closely related to the size,concentration as well as the incubation time of OPLMBs with the macrophage.The ratio of macrophages uptake OPLMBs had a reverse relationship with the size of microbubbles.The highest rate of macrophages uptake OPLMBs was (18.9 ± 0.69)% when the ratio of macrophages to microbubbles was 1 to 10,the time for macrophages co-incubation with microbubbles was 30 min,which was the suitable condition for macrophages uptake OPLMBs.Conclusions OPLMBs can be phagocytized successfully by macrophages in vitro.The size,concentration as well as the incubation time of OPLMBs with the macrophages are important parameters contributed to the efficiency of macrophage phagocytosis.
7.Effect of different irrigating solutions on bonding strength of fiber post to root canal.

OBJECTIVEThe purpose of this study is to find some appropriate treatments on the dentin of root canal wall to obtain better bonding strength of fiber posts.
METHODS36 new extracted non-caries single root canal anterior teeth were collected and post spaces were prepared. The teeth were divided into six groups randomly and processed by different post-space treatments: Sodium chloride irrigation (group I , control group); irrigation with 2% chlorhexidine solution followed by 17% EDTA solution (group II ); irrigation with 3% hydrogen peroxide followed by 17% EDTA solution (group II); irrigation with 5.25% sodium hypochlorite (NaClO) followed by 17% EDTA solution (group IV); etching with 35% phosphoric acid-gel for 30 s (group V); 17% EDTA-gel for 1 min (group VI). After each post-space treatment, sodium chloride irrigation was applied in II-VI group. Fiber posts were then luted in the treated roots using 3M ESPE RelyX Unicem and the thin-slice push-out test was performed. The dentin surfaces and bonding interface were examined under scanning electron microscope. Fracture mode of fiber posts were observed by stereo microscope.
RESULTSGroup II-VI were effective in getting rid of smear layer. Resin rags of dentin bonding interface were observed in group II and group V. The majority fracture mode of specimens in each group were mixed mode which contained dentin destruction. Compared with control group, the bonding strength of group II, V increased obviously (P < 0.05).
CONCLUSIONIrrigation with 2% chlorhexidine solution followed by 17% EDTA solution and 35% phosphoric acid-gel etching improved the bonding strength of fiber posts. The depth of post space has no influence of bond strength.

10.Dynamic changes of pulmonary lesions on CT in patients with severe acute respiratory syndrome treated by non-invasivc positive pressure ventilation

Objective To study the dynamic changes of pulmonary lesions of severe acute respiratory syndrome(SARS)treated by non-invasive positive pressure ventilation(NIPPV)on CT. Methods Serial chest CT images in 18 patients with SARS were retrospectively analyzed before and after NIPPV(4—12 days),and a long-term follow-up(3—36 months)examination by CT was carried out.And 27 SARS cases without NIPPV were enrolled as the control group.Results(1)There were dynamic changes in 15 cases of 18 cases:partial multilobar consolidations of bilateral lungs changed into ground-glass opacities in 7 of 18 cases,a circular pulmonary emphysema around bronchial vascular strains of the lower lobes was seen in 5 of 18,subpleural pulmonary emphysema was seen in 8 of 18,pulmonary barotraumas were showed in 2 of 18.Residual of subpleural curvilinear shadow related to NIPPV was found in 2 cases on the follow-up scan.(2)Above changes on CT were not found in 27 cases without NIPPV.Conclusion There were changes of pulmonary lesions of SARS on CT related to NIPPV treatment,It is beneficial to assess the CT images in the NIPPV treatment of the related pneumonia in SARS.
