Objective PRKAG2 G100S is a novel mutation recently found in Chinese patients with PRKAG2 cardiac syndrome. The present study is to investigate the mechanism by which PRKAG2 G100S induces cardiac hypertrophy. Methods The recombinant adenovirus containing human PRKAG2 gene was constructed and used to infect H9c2 cells and neonatal rat cardiomyocytes using EGFP as the reporter gene, and Western blotting analysis was used to determine PRKAG2 protein expression. The intracellular free Ca2+ was determined in H9c2 cells before and 48 h after infected with Ad-EGFP, Ad-PRKAG2 or Ad-PRKAG2 G100S by incubating with Rohd2/AM. The glycogen contents in neonatal rat cardiomyocytes were determined by PAS 48-72 h after infection. Results Bright green fluorescence was observed in the cultured H9c2 cells and neonatal rat cardiomyocytes 48-72 h after infected with Ad-PRKAG2 GlOOSEGFP and Ad-PRKAG2-EGFP. Over-expressed PRKAG2 protein was identified with PRKAG2 monoclonal antibody. Intercellular free Ca2+ concentrations in H9c2 cells were similar among various groups 48-72 h after infection, and the concentrations were not significantly different before and 48 h after infection in each group. PAS revealed more glycogen in the neonatal rat cardiomyocytes in PRKAG2 G100S group 48 h after infection compared with the other two groups. Conclusion Our findings indicate that myocardial glycogenosis might contribute to the pathogenesis of cardiac hypertrophy in patients with PRKAG2 G100S mutation, and the imbalance of calcium homeostasis in cardiomyocytes might not be involved in the pathogenesis.

Myeloid differentiation protein-2(MD-2)can separately and simultaneously bind lipopolysaccharide(LPS)and toll-like receptor 4(TLR4)has been shown to play critical roles in mediated recognition responses to LPS by TLR4 and signal transduction induced by LPS.MD-2 can be bound by LPS,not TLR4.The cells have no responsiveness or weak responsiveness to LPS without MD-2.MD-2 can be secreted into blood plasma,formed soluble MD-2 and remotely regulated cells that contained TLR4 without MD-2.MD-2 has been shown to play important roles in endotoxin signal transduction.MD-2 is a small molecular,short nucleic acid fragment,easily regulated should become a new potential anti-inflammatory target.

Twelve populations of Inula lineariifolia were used as materials to measure morphological characteristics, photosynthetic parameters and chemical constituents. It aims to provide a theoretical basis for germplasm resources evaluation. The results showed that I. lineariifolia had relatively rich morphological diversity, there were significant differences of morphological characteristics, photosynthetic parameters and chemical constituents among populations. There was positive correlation on morphological characteristics and P(n). Twelve populations were divided into three-type. The three populations of Xuyi, Mingguang and Fengyang were of narrower-longer leaf, bigger biomass,better photosynthetic and higher chemical constituents. Then they were classified for a similar group. It proved that the three populations were more suitable for cultivation and promotion.

Objective To observe the effects of porous polylactic acid/polyglycolic acid copolymer(PLGA) filling into extraction socket on the regeneration of alveolar bone in rats.Methods Sixty male Wistar rats were divided into experimental group and control group(n=30).PLGA scaffold was immediately implanted in the mandibular incisor root sockets after removal of incisor teeth.Soft X-ray photography,structural observation,light microscopy were used to evaluate the effects of composite on bone healing in a rat tooth extraction socket.Results The relative lengths of residual alveolar ridge in the experimental group 4,8 weeks after operation were shorter than that in the control group,and there were significant differences between the experimental group and the control group(P

Aim: To compare the stability of Endostar~(TM) and endostatin under different temperatures and pH using polyacrylamide gel electrophoresis( PAGE) and Western blot and to compare the activity of Endostar kept at 4 ℃ and 37 ℃ by inhibition of endothelial cell proliferation. Methods: Endostar and endostatin expressed by Phicha pastoris were kept at 4 ℃, and 37 ℃ for 96 hours, respectively. The electrophoresis of the samples was detected by reduced and non-reduced PAGE. The results were further confirmed by Western blot with rabbit anti-Endostar polyclonal antibody. The inhibitory effect of Endostar stored at different temperatures on HUVEC was also exam-ined by cell-based assay. Results: Single band at 20 kD was detected in all lanes of SDS-PAGE gel loaded with endostatin and Endostar samples under reducing condition. In acidic native PAGE with three different pH values, endostatin showed a smear characteristic, whereas Endostar showed a unique band in acidic non-continuous native PAGE. Although the smear phenomenon was also observed under two conditions of constant native elec-trophoresis, the major band of Endostar could be detected. Similar electrophoretic behavior was found for endosta-tin and Endostar stored at both 37 ℃ and 4 ℃ . Western blot showed similar results to those by PAGE. Furthermore, Endostar stored at these two temperatures also had identical inhibitory effect on proliferation of HUVEC. Conclusion: Endostar and endostatin exhibit similar thermostability at regular conditions, but Endostar was more stable than endostatin expressed in P. pastoris under acidic condition.

On account of the dense cuticles of the fresh stem and the light, hard and pliable texture of the dried stem, Dendrobii Caulis is difficult to dry or pulverize. So, it is very important to the ancient doctors that Dendrobii Caulis should be properly treated and applied to keep or evoke its medicinal effects. The current textual research results about the preliminary processing, processing and usage methods of Dendrobii Caulis showed that: (1) In history the clinical use of fresh or processed Dendrobii Caulis as teas and tinctures were very common. (2) Its roots and rhizomes would be removed before using. (3) Some ancillary approaches were applied to shorten drying times, such as rinsing with boiling mulberry-ash soup, washing or soaking with liquor, mixing with rice pulp and then basking, etc. (4) According to the ancients knowledge, the sufficient pulverization, by means of slicing, rasping, hitting or pestling techniques, was necessary for Dendrobii Caulis to take its effects. (5) The heat processing methods for Dendrobii Caulis included stir-baking, stir-frying, steaming, decocting and stewing techniques, usually with liquor as an auxiliary material. Among above mentioned, steaming by pretreating with liquor was most commonly used, and this scheme was colorfully drawn in Bu Yi Lei Gong Pao Zhi Bian Lan (Ming Dynasty, 1591 CE) ; moreover, decocting in advance or long-time simmering so as to prepare paste products were recommended in the Qing Dynasty. (6) Some different processing programs involving stir-baking with grit, air-tightly baking with ondol (Kangs), fumigating with sulfur, which appeared in modern times and brought attractive outward appearance of the drug, went against ancients original intentions of ensuring drug efficacy.
