1.Breeding of Arachidonic Acid Producting Strain with Mortierella Sabellina by Ultraviolet Mutation

In order to gain the strain which has high growth vigor and whose biomass concentration satisfy further fermentation to produce arachidonic acid, we took the yield of microbes olein and arachidonic acid as evaluative index, adopted twice ultraviolet mutation, determined the time of ultraviolet irradiation by single factor expriment, and then the content of the arachidonic acid was measured by GC. The results of this experiment showed that the power of viltalight lamp was 20 W, exposure distance was 30 cm and exposure time was 80 s, lethality of spores of Mortierella isabellina was 76.4%. After twice ultraviolet mutation and repeatedly screen, a mutation of Mortierella isabellina Z80s2-109 whose arachidonic acid concentration in biomass were 3.77 times of the control strains was obtained and genetic stability.
2.Serum level of chemerin and bone mineral density in patients with Graves disease

Objective:To investigate relationships between serum chemerin and bone mineral density (BMD) in patients with newly diagnosed Graves disease (GD).Methods:A total of 120 newly diagnosed GD patients with a course more than 3 months were enrolled from the Department of Endocrinology between June 2013 and June 2015.Sixty age-and sex-matched healthy people served as a normal control.Serum levels of chemerin,β-crosslaps (β-CTX),and N-MID-osteocalcin (N-MID-OT) were measured by ELISA.Fat mass and BMD were evaluated by dual energy X-ray absorptiometry (DEXA).Results:Compared with the normal control,the fat mass,lean weight,fat mass index (FMI) and body mass index (BMI) in the GD group were decreased,and BMD in all skeletal sites was decreased.There was a positive correlation between them (all P<0.05).Serum level of chemerin was increased and it was positively correlated with β-CTX or N-MID-OT level and negatively correlated with fat mass,FMI or BMI in the GD group.There was a negative correlation between chemerin level and BMD in femoral neck,total hip,lumbar or right forearm distal 1/3 (rs=-0.352,-0.279,-0.379,-0.289,-0.394;P<0.05).After adjusting for age,fat mass or BMI,the correlation of chemerin with total hip or bone mineral density remained significant (rs=-0.273,-0.378;P<0.05).Multiple linear regression analysis revealed that chemerin or BMI was correlated with BMD (P<0.05).Conclusion:The decrease of bone mineral density in patients with GD is not only related to the direct or indirect effect of excessive thyroid hormones on systemic and osteoblastic cells,but it is also related to the negative regulation of bone metabolism due to the elevated chemerin level.

4.The influence of all-round intervention on the quality of life and the emotion of cancer patients who have accepted the gamma knife therapy

Objective To investigate the influence of all-round intervention on the quality of life and the emotion of cancer patients who have accepted the gamma knife therapy. Method Using health education, psychotherapy and the behavior treatment for 260 patients with malignant tumor who had accepted the gamma knife therapy. Evaluated the quality of life and the emotion of these patients before and after the intervention respectively by using questionnaire of QOL, SCL-90, SDS and SAS. Results The values of indicated the QOL, SCL-90, SDS and SAS before the all-round intervention were 23.12?4.68, 68.12?33.26, 40.12?10.28 and 42.48?12.16 respectively. The values which had obtained after the all-round intervention were 46.26?3.28, 36.12?26.23, 28.16?5.23 and 26.12 ?10.66 respectively, there were significant differences between them,P
5.ON ANCIENT CHINESE DIET IN HUANG TI'S“NEI CHING SU WEN”

Huang Ti's "Nei Ching Su Wen" is the earliest classical work on medical science in ancient China. It summed up the results of observations made in long years through practice by the ancient Chinese labouring people, accumulating thereby lots of valuable knowledge and experience in different lines of medicine.Some two thousand years ago, Chinese medical scientists in the Chou and the Chin Dynasties already conceived the advanced idea of laying special emphasis on prophylactic measures, and began to adopt diet therapy as a means of medical treatment. Principles of preparing complete diet were worked out by them. And in this connection, it is mentioned in the book "Nei Ching Su Wen" that "five kinds of cereals are the means of subsistence, five kinds of cattle provide beneficial food in the form of meat, five kinds of vegetables can be used for food enrichment, and five kinds of fruits may serve as supplement." These principles point out the necessary sorts of food to constitute a complete diet and their respective positions in it. That is to say, cereals including beans and peas are the staple food and animal meat is of secondary importance, with vegetables to enrich and fruits to supplement the first two. A diet thus prepared will not only consist of all the nutritive elements needed by the human body but also represent an appropriate arrangement of the different kinds of nutrients. Such a diet is, no doubt, in accord with the dietetic theory of modern science.
6.NOK Activates STAT3 Signaling by a JAK2-Dependent Mechanism

Novel oneogene with kinase-domain (NOK) can activate multiple mitogenic signaling pathways including the janus kinases(JAK) and signal transducer and activator of transcription proteins (STAT). It was showed that NOK specifically and physicallyinteracted with STAT3 in human embryo kidney 293T (HEK293T) cells. In addition, NOK could directly interact with most of the STAT3 subdomains except coiled-coil and C-terminal domains. Removing ectodomain and transmembrane domain of NOK markedly enhanced its intermolecular interaction with STAT3. Also, NOK could co-immtmoprecipitate with JAK2. in vivo. Importantly, co-expression of NOK and JAK2 produced a synergistic effect on NOK-mediated STAT3 activation, while inactivating the kinase domain of JAK2 completely prevented this synergistic effect. Overall, the results indicated that NOK might complex with both STAT3 and JAK2 and activate STAT3 signaling by a JAK2-dependent mechanism.
7.Canine femoral artery displacement with the chitosan-sulfated silk fibroin artificial blood vessel:the formation of vascular endothelial cells, vascular smooth muscle cells and fibroblasts

BACKGROUND:The chitosan-sulfated silk fibroin artificial blood vessel was obtained in our previous study. OBJECTIVE:To investigate the biocompatibility and function of the chitosan-sulfated silk fibroin artificial blood vessel replacement into the canine femoral artery. METHODS:Eight Beagles were randomly allotted to two groups, and chitosan-sulfated silk fibroin artificial blood vessel was implanted into the femoral artery (experimental group), but animalsin control group received no intervention. Six months after implantation, the coagulation function was detected, the vascular formation, including endothelial, smooth muscle and fibroblast layers were observed through hematoxylin-eosin staining under electron microscope. RESULTS AND CONCLUSION:(1) Hematoxylin-eosin staining showed that vascular endothelial cells, vascular smooth muscle cells and fibroblasts were found in the experimental group, which were similar with the normal vascular structures. (2) Immunohistochemistry:in the experimental group, there was a deeply stained band at the tissue edege through Factor Ⅷ staining, suggesting the formation of the endothelium;there were abundant brown particles precipitated in á-SMA staining, suggesting the vascular smooth muscle cel formation;Vimentin staining showed various brown particle precipitations indicating the fibroblast formation, and all were close to the normal vascular structures. (3) Electron microscope observed that the inner surface and section of the artificial vessel were similar with the normal one, and the three-layer structures formed in the section arranged irregularly. (4) There were no differences in the prothrombin time, activated partial thromboplastin time, thrombin time and fibrinogen between groups. To conclude, these results suggest that the chitosan-sulfated silk fibroin artificial blood vessel holds good biocompatibility and performances.
8.Protective Effects of Insulin on Acute Global Cerebral Ischemia Reperfusion Injury in Rats

Objective To observe effects and mechanisms of insulin on reperfusion injury after cerebral ischemia.Methods Sixty-six male Wistar rats were used in this study.All rats were divided into 3 groups as treated group(A),control group(B) and random sham-operated group(C).Four-vessel occlusion was used to establish global cerebral ischemia reperfusion model in study groups.The treated group were divided into 5 groups(A1-A5) and intraperineally injected with biosynthetic human insulin 2 IU/(kg?d) and 50%glucose 2 g/(kg?d) for 7 days,the blood glucose was monitored in preoperative and postoperative 3,6,12,24 h,and the blood glucose was maintained between 3.5-6.5 mmol/L.These animals of control group were given with saline 2 mL/(kg?d) for 7 days in abdominal cavity.All the rats were killed in the seventh day,brain homogenate was collected for detection of neuron specific enolase(NSE)and nitric oxide(NO).The hippocampus was separated for observation of electronic microscope.Results Concentration of NSE in brain tissue in group C was significantly higher than that of group A and group B,while the level in group A was higher than that of group B.Concentration of NO in group C was lower than that of group A and group B while the level of NO in group A was significantly lower than that of group B.Electron microscope showed that the ultrastructure of sham-operated group was nearly normal,damage degree of hippocampal neuron and gliacyte and capillary was gradually worse from group A1,A2 to A4,the damage degree of group B1,B2 and B4 was serious and there was no difference among them.Conclusion Insulin can really promote recovery of the cerebral injury after ischemia reperfusion.
9.Transumbilical breast augmentation with insertion of saline-filled breast implants

Objective To evaluate the surgical technique and procedures of transumbilieal breast augmentation with an inverse U-shape incision and insertion of saline-filled breast implants. Methods With specialized instruments, the subcutaneous tunnel was undermined to the inframammary crease after inverse U incision made along the umbilical border. The subpectoral space was dissected with a dissector after the advancement of obturator across the inframammary crease. The expander was inserted into the subpectoral pocket to be filled to obtain the high degree of symmetry of the breasts. Finally, the previous-ly prepared implants were inserted after the expander deflated and removed. Results All patients gained satisfactory results with no complications, such as hematoma, infection, implant deflation and Baker Ⅲ or Ⅳ capsular contracture. Conclusions Breast augmentation could be performed through umbilical inci-sion with the usage of saline-filled breast implant. This is an alternative of incisions in breast augmenta-tion.
10.Clinical significance and outcomes of fetal ventriculomegaly

Objective To evaluate the clinical significance and prognosis of tetus with lateral ventriculomegaly.Methods We retrospectively analyzed 92 singleton pregnant women who were antenatally diagnosed with fetal ventriculomegaly by ultrasound in genetic conselling clinics,Department of Gynaecology and Obstetrics,Nanfang Hospital,Southern Medical University between August 2007 and August 2010.All participants were divided into three groups according to the width of the lateral ventricles:group A (10.0-12.0 mm,n=50),group B (12.1-14.9 mm,n=38) and severe ventriculomegaly group (≥15.0 mm,n=4).All fetuses were followed up.Chi-square test(or Fisher's exact test),Bonfferoni method,Mann Whitney or Kruskal-Wallis test were used for statistics.Results In group A,B and severe ventriculomegaly group,18% (9/50),65.8%(25/38)and 3/4 of fetuses were complicated with structural malformation,respectively (x2 =22.934,P =0.000),and statistical significance were found only between group A and B (x2 =20.798,P=0.000).The incidences of fetal chromosomal aberration were 4.0% (2/50),7.9% (3/38)and 0/4 in the three groups,respectively (x2=0.878,P = 0.645).Eventually,all four cases with severe cerebral ventriculomegaly were terminated.Among cases of mild ventriculomegaly,63 women continued the pregnancy (48 in group A and 15 in group B).The intrauterine improvement of group B was poorerthan that of group A (Z=-3.317,P =0.001).Respectively,three,ten and two cases of ventriculomegaly deteriorated,stabilized and regressed in group B,and the corresponding figures were 3,15 and 30 in group A.In group A,the prognosis of fetus with non-isolated ventriculomegaly was poorer than that of fetus with isolated ventriculomegaly (Z=-2.631,P=0.009).For neonates 14 days after birth (n=62),the rates of normal neonatal behavioral neurological assessment (NBNA) scoring were respectively 93.8% (45/48) and 71.4% (10/14) for groups A and B with statistical difference (Fisher's exact test,P =0.040).Bayley scales of infant development (BSID) used to evaluate infants at 12-month-old revealed that the psychomotor developmental index (PDI) between group A and group B had statistical difference (8,26 and 1 case of good,moderate and poor development in group A; one,seven and three cases in group B; Z=-2.203,P=0.043).However,the mental developmental index (MDI) between group A and B had no statistical difference.Twenty babies among the survived ones received magnetic resonance imaging (MRI) examination.The results showed that lateral ventricle width regressed in nine babies (45.0%) and progressed in one baby (5.0%).Ten cases (50.0%) did not change.The change of ventriculomegaly after birth was related to BSID evaluation.Better prognosis of fetuses were found in those with regressed ventriculomegaly (P=0.033).Conclusions Fetuses with mild ventriculomegaly (10.0 12.0 mm) have a favorable outcome.Further investigations,such as fetal development,chromosomal examination,intrauterine infection screening and MRI if necessary,are suggested for fetus with ventriculomegaly.Moreover,intensive intrauterine and postnatal follow-up is recommended.
