Objective To investigate the relationship between IGF2BP3 hypomethylation with colon tumor differentia?tion. Methods Tissue samples from 41 colorectal cancer were collected from March 2011 to August 2011 in our hospital, among which 19 cases were well-differentiated adenocarcinoma, 12 cases were of moderately differentiated adenocarcinoma and 10 cases were of poorly differentiated adenocarcinoma. Meanwhile biopsy samples from 12 cases of colonic colitis were collected as a control. The expression of IGF2BP3 was assessed by immunohistochemistry and Western blot. An innovate of ELISA technique was used to examine global methylation levels while IGF2BP3 methylation level was tested by methylation specific PCR technique. Results The expressions of IGFBP3 are higher in all colon cancer groups than that in colitis (P<0.05). The expression of IGFBP3 in poorly differentiated adenocarcinoma is higher than that in all the other groups, but there is no significant difference between its expressions in the well differentiated group and in the moderately differentiated adeno?carcinoma group. The global DNA level and IGFBP3 methylation level of every colon cancer group is lower than those in coli?tis (P<0.05). Also, a tendency of decreasing global DNA and IGFBP3 methylation status was observed comparing well differ?entiated towards poorly differentiated carcinomas (P<0.05). Conclusion IGF2BP3 expression in colorectal cancer is asso?ciated with differentiation of colon cancer tissue. A lower global and IGF2BP3 DNA methylation suggest a worse differentia?tion of colon cancer. DNA hypomethylation may therefore play a regulatory role in the expression of IGF2BP3 in colon cancer tissue.

Objective To investigate the expression of p38 mitogen actirated protein kinase (p38MAPK) in renal fibrosis in diabetic mice. Methods Male homozygous C57 BL/6 mice were divided at random into control group intraperitoneally(i. p.) injected with citrate buffer and diabetes group received 5 consecutive daily intraperitoneal inserum glucose level exceeding 16.7 mmol/L. Mice were killed at 0,4weeks ,8weeks, 12weeks and 16weeks respectively after STZ injected. The kidney tissues were obtained from diabetic and control mice. Serum glucose, extracellular matrix, and 24 h urinary albumin excretion rate(UAE)and the serum creatinine(Scr) were measured. The kidney tissue was used for histological and morphometric studies of glomerular matrix expansion (PAS), and the expression of phosphorylated p38MAPK and TGFβ1 were measured by immunohisteehemical staining. p38MAPK and TGFβ1mRNA were analyzed by reverse transeriptase-PCR. Results The serum level of glucose in diabetic mice increased significantly. Scr and 24 h UAE, and glomerular matrix expansion in diabetic mice were obviously higher than that in control mice. Phosphorylated p38MAPK and TGFβ1 levels were obviously increased in the kidney of diabetic mice compared with those in control mice(P <0. 01) ;TGFβ1 expression was positively related to the expression of phosphorylated p38MAPK. Conclusion The overexpression of phosphorylated p38MAPK in kidney shoud play an important role in the development of renal fibrosis associated with diabetic nephropathy in mice.

OBJECTIVE Cistanche deserticola Y. C. Ma is an official plant that grows in arid or semi-arid areas. Our early work demonstrated that Cistanche extracts protect against sperm damage in mice under bisphenol A induced reproductive damage. Echinacoside (ECH)is one of the major active components. The present study investigated the mechanisms behind the possible protective effects of ECH against oligoasthenospermia in rat and identified the interaction between ECH and AR. METH-ODS The distribution of ECH was assayed by HPLC,the quantity and quality of sperm was evaluated and hormone levels were determined by radioimmunosorbent assay. The levels of androgen receptor (AR)and key steroidogenic-related genes were reduced as determined by Western blotting and qPCR analysis. The interaction between ECH and AR were evaluated by fluorescence localization assay,indi-rect ELISA and molecular docking. RESULTS ECH significantly increased the quantity of sperm and secretions of luteinizing hormone and testosterone.ECH was distributed to the hypothalamus but not in the testes.ECH combined with hypothalamic AR in the pocket of Met-894 and Val-713 to inhibit transfer of AR from the cytoplasm to nuclei in the hypothalamus. While negative feedback of sex hormone regula-tion was inhibited, positive feedback was stimulated to increase the secretion of luteinizing hormone and testosterone subsequently enhancing the quantity of sperm. C. militaris significantly alleviated the BPA-induced reproductive damage by increasing testicular superoxide dismutase (SOD), glutathione peroxidase (GSH-PX) and glutathione (GSH); as well as by reducing serum malondialdehyde (MDA). C. militaris not only obviously enhanced the levels of serum LH and T, but it also improved the sperm count and motility compared to the BPA-treated group.CONCLUSION C.militaris and ECH protect the BPA induced reproductive damage.ECH blocks AR activity in the hypothalamus to increase the quantity of sperm and protect against oligoasthenospermia in rats.

Journal of Clinical Hepatology 2019;35(4):921-924
The liver is the key place for most metabolic pathways, so it is often involved earliest and has the most severe injury, and liver injury of some inherited metabolic diseases may develop into liver cirrhosis and even malignant tumors of the liver and tended to have poor prognosis. Inherited metabolic liver disease often has complicated clinical manifestations, and the limitations of conventional diagnostic and therapeutic methods are difficult issues for clinical physicians. With the development of cellular and molecular biology techniques, new diagnostic and therapeutic methods, such as gene detection, gene therapy, and stem cell transplantation, have been gradually applied in clinical practice, bringing hope to patients with refractory diseases. This article reviews the recent advances in the diagnosis and treatment of inherited metabolic liver diseases commonly seen in clinical practice, such as hepatolenticular degeneration, hereditary hemochromatosis, glycogen storage disease, ɑ1-antitrypsin deficiency, hereditary hyperbilirubinemia, and hereditary cholestasis.
