Objective To study the biomarker panel of superficial bladder transitional cell carcinoma(SBTCC)and analyze the biological pathway in tumorigenesis by Shotgun proteomics strategy.Methods Normal urothelium cells and cancer cells were harvested by laser capture microdissection from clinical specimen and the proteomic expression profile was identified by two-dimensional liquid chromatography tandem mass spectrometry.The isoelectric point,molecular weight,grand average of hydropathicity,transmembrane helices were analyzed by using proteomics tools.Gene ontology was used to comment the identified proteins.The pathway analysis was performed by ArrayTrack software,and visualized by GenMAPP.Results There were 440 and 218 proteins expressed in cancer cells and normal cells respectively,among them 388 proteins were differerntially expressed.All the database about identified proteins was deposited in an accessible form to researchers at http://www.Proteome-SBTCC.org.cn and http://www.Proteome-NHTE.org.cn.There were 267(68.8%)differentially expressed proteins which had GO biological process comments.The biological pathwavs of these proteins included MAPK signaling pathway,focal adhesion,oxidative phosphorylation,ECMreceptor interaction,etc.Conclusion Shotgun strategy proteomies database of normal transitional epithelium and SBTCC is successfully constructed.And the basis for the understanding of cell biology and discovery of biomarker panel for SBTCC iS provided.
4.Construction of lentiviral vector containing siRNA sequence of Siglec-1 and verification of inhibition efficiency

Objective To construct lentiviral vectors containing small interfering RNA (siRNA) sequence of Siglec-1 and to screen the effective vector.Methods Three fragments of Siglec-1 siRNA were designed and cloned into pGCSIL-GFP lentiviral plasmid.And then the plasmid was cotransfected into 293T cells with pHelper 1.0 and pHelper 2.0 plasmids.Forty-eight hours later,culture supernatant with virus particles was collected and concentrated.Virus titer was determined by 10-fold serial dilution method and virus was transduced into primary cultured mouse bone marrow-derived macrophages (BMM).Flow cytometry and QRT-PCR were used to screen effective vector with inhibition ability.Results Three vshRNA lentiviral plasmids and a control plasmid were constructed successfully and verified by DNA sequencing.Virus titer was between 1×10s TU/ml and 1×109 TU/ml,which was suitable for in vitro and in vivo experiments.The Lv-1 could inhibit Siglec-1 expression effectively in vitro transduction of BMM.Conclusion Lentiviral vectors containing siRNA sequence of Siglec-1 were constructed successfully and an effective vector was screened,which may lay the foundation for using the vector in gene knockdown experiment in vivo.
6.The role of Siglec-1 in chemokines secretion and phagocytosis of oxidized low-density lipoprotein on macrophages

Objective By in vitro culture of mouse macrophage cell line RAW264. 7 and primary mouse bone marrow macrophages, the expression of Siglec-1 when stimulated by ox-LDL was observed. Meanwhile, Siglec-1 was up-regulated by M-CSF and down-regulated by small interference RNA targeting Siglec-1 ( si-RNA-Siglec-1) , and the expression of chemokines and lipid uptake ability by macrophages were observed, to explore the role of Siglec-1 on macrophages in atherosclerosis. Methods LDL was oxidized by copper. According to preliminary experiment results, ox-LDL 100 μg/ml was selected as a stimulus. There were 6 experimental groups:normal control group,ox-LDL 100 μg/ml group, ox-LDL 100 μg/ml + si-RNA 2509 2 ng/ml group,ox-LDL 100 μg/ml + si-RNA 3618 2 ng/ml group,ox-LDL 100 μg/ml + M-CSF 5 ng/ml group and ox-LDL 100 μg/ml + M-CSF 10 ng/ml group. si-RNA-Siglec-1 was transfected into macrophage to inhibit the expression of Siglec-1, whereas M-CSF 10 ng/ml or 5 ng/ml were added into the culture medium to enhance the expression of Siglec-1. Quantitative real-time polymerase chain reaction ( qRT-PCR) was used to determine the interfere efficiency of si-RNA-Siglec-1 or M-CSF. After stimulation with ox-LDL for 48 h, cell culture supernatants were collected to determine MIP-1 alpha, MCP-1 and IL-8 concentration by ELISA (n =3 for each group) to evaluate the activation of macrophages. Internalization of lipid particles by macrophages was analyzed by oil red 0 staining. Results Observed by fluorescence microscope, si-RNA-Siglec-1 could be effectively transfected into macrophages with a transfection efficiency about 90% ;PCR results showed that si-RNA 2509 and si-RNA 3618 in a concentration of 40 pmol/L had an inhibition rate of 0. 54 ±0. 11 or 0. 52 ±0. 16 vs 1. 00 ±0. 24 (control group) , t =5. 227 and 4. 992, respectively, all P < 0.01, while M-CSF 10 ng/ml could increase Siglec-1 mRNA expression approximately 4-fold (4. 16 ± 1. 25 vs 1.00 ±0. 24, t =7. 448, P<0. 01). The secretion of MCP-1, MIP-1 alpha, and MIP-2 in si-RNA3618-Siglec-1 group [(359. 28±47. 80) pg/ml, (33. 76 ± 14. 28) ng/ml and (7.87±1.55) ng/ml for MCP-1,MIP-1 alpha, and MIP-2, respectively] was significantly reduced in compare with ox-LDL 100 μg/ml group [ (577. 89 ± 35. 95 ) pg/ml, (69. 17 ± 11. 82) ng/ml and (12.28 ± 1.19) ng/ml for MCP-1, MIP-1 alpha, and MIP-2, respectively], with P value of 0.01, 0.05 and 0.01. In contrary, ox-LDL 100 μg/ml plus M-CSF 10 ng/ml group could significantly promote macrophage chemokine secretion [ (672. 89 ± 43.80) pg/ml, (101.31 ±24.17) ng/ml and (14.81 ±0.54) ng/ml for MCP-1, MIP-1 alpha, and MIP-2, respectively], with P < 0.05 compared with ox-LDL 100 μg/ml group. Meanwhile, lipid intemalization and foam cell formation was inhibited in si-RNA3618-Siglec-l group while ox-LDL 100 μg/ml plus M-CSF 10 ng/ml group could enhance the phagocytosis of ox-LDL by macrophage. Conclusions Siglec-1 may served as a potential phagocytic receptor for ox-LDL involving in macrophage uptake of lipid and turn into foam cells. Furthermore, it can active macrophages and enhance the secretion of MIP-1 alpha, MCP-1 and IL-8, attracting more macrophages and lymphocytes to the site of inflammatory plaque. Targeted inhibition of Siglec-1 reduces macrophage uptake of lipid and secretion of chemokines. Siglec-1 may possibly serve as a potential target of treatment or delay the development of atherosclerosis.
7.Optimization of ?-Lactam Treatment Regimens for Extended-spectrum ?-Lactamases-producing Strains Infections Based on Pharmacokinetics/pharmacodynamics

MIC.RESULTS The bacteriostatic/bactericidal CFR of 0.5g q8h imipenem/cilastatin and 0.5g q8h meropenem were the highest(100%);that of 3.375g q4h sodium piperacillin/tazobactam sodium against ESBLs-producing E.coli was above 90%;the CFRs of ceftriaxone(1g q24h,2g q24h),ceftazidime(1g q8h,2g q8h),cefepime(1g q8h,1g q12h,2g q8h) and cefoperazone sodium/sulbactam(0.5g q8h,1g q8h,2g q8h) against ESBLs-producing strains were clearly less than those of non-ESBLs-producing ones,and the CFRs could not be effectively improved with the dose and frequency increased.CONCLUSIONS The PK/PD simulation is useful to optimize the regimen of anti-infective treatment,and guide its dosing accurately.
8.Endogenous protective mechanism of intestinal barrier injury induced by cardiopulmonary bypass in rats: the relationship with enteric glia cells

Objective To evaluate the relationship between endogenous protective mechanism of intestinal barrier injury induced by cardiopulmonary bypass ( CPB) and enteric glia cells in rats. Methods Forty-eight clean-grade adult male Sprague-Dawley rats, weighing 400-500 g, were divided into 2 groups (n=24 each) using a random number table method: sham operation group (group S) and CPB group. Rats were sacrificed at the beginning of CPB, 60 min of CPB, and 2 and 6 h after CPB, and the intestinal tissues were removed for examination of pathological changes ( by HE staining) and for determina-tion of the expression of ZO-1, occludin, glial fibrillary acidic protein ( GFAP ) and calcium-binding pro-tein ( S-100) and the positive expression of GFAP ( by immunohistochemical method) . Results Compared with group S, the expression of GFAP and S-100 was significantly up-regulated at 60 min of CPB and 2 and 6 h after CPB, the expression of ZO-1 and occludin was down-regulated (P<0. 05), the positive expres-sion of GFAP was enhanced, and the intestinal mucosal injury was marked in group CPB. Conclusion The enhanced activation of enteric glia cells may be involved in the endogenous protective mechanism of in-testinal barrier injury induced by CPB in rats.
9.Responses of Patients and Guardians to Phlebotomists' Wearing Gloves and Disinfecting Their Hands.

BACKGROUND: A hand hygiene policy has been introduced by the Joint Commission International Accreditation Standards for Hospitals (JCI); the purpose of this policy is to ensure that systematic standards are continuously practiced at hospitals in order to improve the quality of medical care and the safety of patients and health care workers. This policy requires that phlebotomists wear new gloves and disinfect their hands before attending a patient to prevent nosocomial infections and to protect the phlebotomists. This study aimed to assess the responses of patients and guardians regarding the hand hygiene procedures. METHODS: Between January and February 2010, a questionnaire survey was performed at our hospital for 310 outpatients, 93 inpatients, and 189 guardians. RESULTS: In all, 70.8% of respondents answered that phlebotomists did not require considerable time to remove and wear gloves, and 69.9% responded that phlebotomists could readily detect veins even with their gloves on. Besides, 81.9% respondents thought that it was sanitarily important for phlebotomists to remove their gloves and disinfect their hands after a venipuncture, whereas only 2.8% thought that this practice caused discomfort. CONCLUSION: The patients and guardians recognized that the hand hygiene procedures were important for their own safety and encouraged their application rather than considering them uncomfortable or inappropriate. Introduction and maintenance of the hand hygiene policy of JCI would not be difficult because patients or guardians recognized its importance.

10.Effect of lysophosphatidylcholine on whole cell K+ current in rabbit coronary smooth muscle cells..

BACKGROUND: Impairment of relaxing response and augmentation of contractile response to vasoactive substances have been reported in atherosclerotic arteries. These alterations in vascular reactivity are considered as an underlying mechanism for the development of acute vasospasm in atherosclerotic coronary artery. Recently, it has been reported that lysophophatidylcholine (LPC), an oxidative metabolite of low density lipoprotein causes this functional abnormality. However, the precise mechanism of LPC induced change of vascular reactivity is still uncertain. METHOD: In this study, to elucidate the underlying mechanisms of abnormal vascular reactivity in atherosclerotic coronary artery, we examined the effect of LPC on whole cell K+current using patch clamping technique in rabbit coronary smooth muscle cells. RESULTS: Application of LPC(1microM) showed dual effect on whole cell outward current which depends on the magnitude of test potentials. At relatively high depolarizing test potentials (> 10 mV), LPC increased amplitude of outward current which was blocked by Gd3+ not by iberiotoxin (100 nM) and TEA (1 mM). Reversal potential of this Gd3+sensitive, LPC-induced current was -9.7 +/- 0.6 mV. At less depolarizing test potentials (< 10 mV), LPC decreased whole cell K+currents in a dose dependent manner (from 0.01 to 10 microM) in the range of -30 mV to +0 mV. Half maximal inhibition of K+current was 1.509 microM at 0 mV test potential (n =5). Depolarizing holding potential (0 mV) prevented this LPC-induced inhibition of K+current. Steady state activation and inactivation parameters of K+current were significantly shifted to the positive direction by application of LPC (p < 0.01, n =8). Pretreatment of staurosporine (100 nM), a blocker of protein kinase C partially blocked LPC-induced decrease of K+currents. CONCLUSION: LPC-induced inhibition of voltage dependent K+current may explain abnormal vascular reactivity in atherosclerotic coronary artery.
