1.Arthroscopic reconstruction of double-bundle anterior cruciate ligament with semitendinosus and gracilis tendon

Objective The investigation was to evaluate the method and the efficacy of double-bun-dle anterior cruciate ligament reconstruction with semitendinosus and gracilis tendon through two bone tun-nels.Methods From April1998to May2000,12patients underwent double-bundle anterior cruciate liga-ment reconstruction were reviewed and followed up more than18months.There were12male and2female patients aging from17to38years old(mean,25.5years).5cases were associated with simple anterior cru-ciate ligament injury,and the other7cases suffered from anterior cruciate ligament injury with concomitant torn meniscus.Two bone tunnels of4.5mm diameter were made in the tibial and the femoral attachment site of ACL anteromedial bundle and posterolateral bundle respectively.The anteromedial bundle was recon-structed using semitendinosus tendon,and the posterolateral bundle was reconstructed using the gracilis ten-don.The attachments of the semitendinosus and gracilis tendon outside tibial tunnels were reserved,the re-paired tendons were tied as a knot outside femoral tunnels.There was not need to do any internal fixation.Anterior drawer test was performed in30,60and90degrees of flexion of the knee joint,and Lysholm rating system was used to determine the function of the knee and the level of activity of the patients.Results Be-fore operation,anterior drawer test in30,60and90degrees of flexion of the knee joint was positive in all patients,the mean Lysholm scores were50.5points(range,40to58points).18months after operation,ante-rior drawer test in30,60and90degrees was negative in9cases,anterior drawer test in60,90degrees of flexion of the knee joint was negative in2cases,in90degrees negative in1case;The mean Lysholm scores was85points(range,62to92points).The good and excellent results were obtained in91.7%.Conclusion The combination of double-bundle anterior cruciate ligament reconstruction makes a great progress to anatomic reconstruction,provides better dynamic stability of the knee joint,and predisposes the patients sat-isfactory clinical effects.
2.Influence of cationic cyclopeptide on microstructure and permeability of Caco-2 cell membrane.

The microstructure of cationic cyclopeptide (TD-34) treated Caco-2 cell membrane was observed, and we discussed the relationship between membrane structure and insulin transmembrane permeability. Atomic force microscope (AFM) was used to observe living cell membrane in air condition and tapping mode. Results showed that the surface of Caco-2 cell membrane treated with TD-34 lost its smoothness and nearly doubled its roughness. Apparent permeability coefficients (P(app)) of insulin in Caco-2 cell monolayers increased 2.5 times. In conclusion, AFM can be used to observe microstructure of cationic cyclopeptide treated cell membrane and cationic cyclopeptide enhanced insulin delivery across Caco-2 cell membrane by increasing membrane fluidity.
3.Changes of ANF mRNA expression in the myocardial tissue of rat embryo and suckling induced by 2,3,7,8-tetrachlorodibenzo-p-dioxin

Objective To investigate the changes of ANF mRNA expression in the myocardial tissues of rat embryo and neonate induced by 2,3,7,8-TCDD so as to determine if TCDD can cause injuries of cardiac functions. Methods SD rat embryos were divided into 4 groups respectively exposed to TCDD at the dose of 0.05,0.5,5,10 ?g/kg. The mRNA expression of ANF/ANP in the myocardial tissues of rat embryos of day 15,19 and suckling rats aged 5 days were detected by semiquantitative RTPCR analysis. Results TCDD could upregulate ANF mRNA expression in the myocardial tissues of rat embryos and suckling rats,as compared with control group. Conclusion TCDD can upregulate ANF mRNA expression in myocardial tissues,suggesting that TCDD may cause injuries of cardiac functions of rat embryos and suckling rats.
4.Significance of fiberoptic bronchoscopic brush cytology in lung carcinoma diagnosis

Objective To evaluate the significance of fiberoptic bronchoscopic brush cytology in the diagnosis and histological classification of lung carcinoma.Methods Data of 309 patients with lung carcinoma were retrospectively analyzed.Both bronchoscopic cytology and histology diagnosis were available.The positive rate of bronchoscopic cytology and tissue biopsy were calculated respectively.The classification accuracy of cytological diagnosis for lung carcinoma was evaluated.In tissue biopsy standard,evaluated the significance of bronchoscopic cytology in diagnosis and histological diagnosis.Results The positive rate of bronchoscopic cytology and tissue biopsy were 86.1% (266/309) and 83.8% (259/309),respectively.Bronchoscopic cytology combined with bronchial biopsy could obviously improve the positive rate to 94.2% (291/309) in lung carcinoma diagnosis.Taking the tissue biopsy histological type as a standard,the cytotyping accuracy for brush method was 85.1%(74/87) in squamous carcinoma,82.4%(108/131) in adenocarcinoma and 100%(11/11) in small cell carcinoma for higher.However,the accuracy in diagnosing poorly differentiated carcinomas was only 12.2% (5/ 41).Conclusion Fiberoptic bronchoscopic brush cytology plays an stable and important role in diagnosing lung carcinomas and histological type determination.However,it has limited use in diagnosing poorly differentiated carcinomas.
5.An investigation on the in vitro release behavior of TCM components in different pressure sensitive adhesives

AIM: To examine the effect of acrylic ester and thermosol respectively as pressure sensitive adhesive matrix of zhitong huoxue extract ( Rhizoma Corydalis,Radix aconiti,Radix Paeoniae alba,Radix et notoginseng ginseng,etc) on the dissolution of effective constituent from zhitong huoxue extract.METHODS: The in vitro release experiment was carried out on Permcell diffusion cell,HPLC method was applied to determining the cumulative elease rate of tetrahydropalmatine,aconitine,paeoniflorin and ginsenoside Rg_1 from zhitong huoxue pressure sensitive adhensive.RESULTS: The acrylic ester matrix had sound controlled release profile in vitro of alkaloid components.In KT-A thermosol matrix,the in vitro release rate proved to be inversely proportional to the molecular weight (R = 0.91 ) of the effective constituents,while the 2501 s matrix presented a steadily cumulative release rate over 12 h (30% -43% ).CONCLUSION: Good release rates are observed with effective constituent with small molecular weight,adhesive composition of matrix has an obvious effect on the in vitro release behavior of zhitong huoxue extract.

9.Sampling Analysis of the Use of Precursor Drugs in the Outpatients of Our Hospital

Objective:To understand the application status of precursor drugs in the outpatients of our hospital. Methods:Totally 12 kinds of precursor drugs were included in the management. The data of prescriptions in HIS were collected from July 1 to September 30 in 2015. The defined daily dose (DDD), frequency of drug use (DDDs), drug utilization index (DUI) and the other parameters were calculated and the influencing factors were analyzed by the Pareto method. Results: Among the twelve precursor drugs, DUI of seven kinds was less than or equal to 1. Paracetamol tablets, pseudoephedrine hydrochloride tablets, dextromethorphan hydrobromide and chlorphenamine maleate tablets, nicergoline tablets and compound dextromethorphan hydrobromide tablets II displayed lower DUI, indicating their clinic application was more reasonable. Conclusion:The data showed that the use of precursor drugs in our hospital is basically reasonable.
10.Significance of miR-155, miR-34a and miR-30a expression in diffuse large B-cell lymphoma.

OBJECTIVETo determine the expressions of miR-155, miR-34a and miR-30a in diffuse large B-cell lymphoma (DLBCL) and to explore their potential correlation with clinicopathological characteristics.
METHODSThe expression level of miR-155, miR-34a and miR-30a in 46 DLBCL samples were determined with TaqMan real-time polymerase chain reaction. Interphase fluorescence in situ hybridization (I-FISH) was performed to detect MYC and p53 genes' status, and immunohistochemistry (Envision method) was used to evaluate the expression of CD3, CD10, CD20, BCL-6 and MUM-1 in DLBCL. The DLBCLs were classified into germinal center B cell-like (GCB) and non germinal center B cell-like (non-GCB) subtypes according to Hans' criteria.
RESULTSCompared with normal controls, miR-155 expression level was significantly higher in DLBCL. The expression level of miR-155 in non-GCB type was higher than that in GCB type. It was shown that the patients with MYC rearrangement had lower expression level of miR-155 than the negative controls. Compared with p53 normal group, the expression level of miR-34a was significantly lower in p53 deletion group. It was also shown that the patients with BCL-6 protein expression had lower expression of miR-30a compared with the negative group.
CONCLUSIONmiR-155 expression level is different in normal controls, DLBCL and patients with subtype DLBCL. It therefore has a diagnosis value for DLBCL. miR-34a is of great prognostic significance. miR-155, miR-34a and miR-30a may be potential therapy targets for DLBCL.
