1.Preliminary Discuss on Identification and Characterization of One Decolorizing Fungus

According to morphological and microscopic characteristics, a high-efficient decolorizing fungus, the strain Asaw117, was identified as Aspergillus awamori. Selecting eight different dyes from Azo dyes, anthraquinones dyes and oxygen Quinones dyes, the decolorizing assays of various dyes showed that the strain Asaw 117 was the highest decolorizing potential to 0.1 g/L Vat Blue RSN, the discoloration rate up to 100 percent. Comparing to different kinds of medium and several of carbon and nitrogen sources, the strain had the best decolorizing efficient although grew slower in the Czapek medium, otherwise, grew quicker and decolorizing efficient lower in the PDF medium. It could use Vat Blue RSN as a nitrogen source, but not as a carbon source. The medium composing of saccharose and ammonium nitrate as carbon and nitrogen sources was decolorizing potential markedly during different combinations of carbon and nitrogen sources. So the strain has good potential for the dyeing wastewater treatment
2.Study of the antiulcer effect of pantoprazole

Aim To evaluate the effects of pantoprazole on various experimental acute ulcer inrats and mice. Methods The model of a gastric ulcer of rats or mice was caused bystree- induced ulcer and ligatel pylurus-induced ulcer. Results & Conclusions At adose of 5, 10, 20 mg? kg-1 of Pantoprazole can markedly decrease the ulcer index ofstree-induced ulcer. Pantoprazole(4, 8, 16 mg? kg -1 ) significantly decrease the areaof ligated pylorus-induced gastric ulcer. It was also found that pantoprazole caninhibit the output of basic gastric acid.

Sudden cardiac death(SCD) from early myocardial ischemia is often lack of typically morphological findings and clinical manifestation, thus cases of SCD may be suspected as criminal cases. It is necessary to clarify the cause of death, which is significance for medico-legal investigation. This article reviewed the latest advancement in the studies on the application of inorganic ions, CK-MB, cTn, ANP and BNP for certification of death from SCD in order to provide a practical way for diagnosis of SCD in forensic pathology.

4.Characteristics and distribution of ERP by different field stimulation.

OBJECTIVE:
To study the variation of latency and amplitude of the event related potential (ERP) and its distribution in human scalp when the normal subjects were stimulated with different visual fields.
METHODS:
The ERP recorded in scalp with the stimulation of 10 degrees visual field and 60 degrees visual field respectively in 20 healthy volunteers with normal visual function.
RESULTS:
Two different visual field stimulation may evoke the different exogenous components P1 (70-125 ms), N1 (90-170 ms), P2 (140-220 ms) and endogenous components N2 (190-280 ms) and P3 (290-430 ms). The latencies of all the components evoked by 10 degrees visual field were shorter than that of the 60 degrees visual field while the amplitudes of N1 and N2 were lower and appeared over the extensive encephalic region; and the amplitudes of the P1, P2 and P3 were higher and appeared in occipitotemporal, prefrontal and occipital region, respectively.
CONCLUSION
Two different visual field stimulation may evoke all the ERP components with significant differences in the latency, amplitude and distribution. The differences may reflect the different visual information integration and processing in human brain during the different visual field stimulation.

5.Clinical observation of treatment of infantile diarrhea due to spleen deficiency using five-step pediatric tuina of Huxiang school

Objective:To observe the clinical efficacy of treating infantile diarrhea due to spleen deficiency with five-step pediatric tuina of Huxiang school. Methods:Using a randomized controlled trial design, sixty eligible kids with diarrhea due to spleen deficiency were randomized into an observation group and a control group, with 30 cases in each group. The observation group was intervened by the five-step pediatric tuina method of Huxiang school, and the control group received conventional tuina treatment. The intervention was conducted once a day, consecutive 5-day treatment as 1 course, at a 2-day interval between courses, successively for a total of 4 courses. Changes in the primary and secondary symptoms of diarrhea due to spleen deficiency were observed, and the clinical efficacy was evaluated. Results: After treatment, the scores of primary and secondary symptoms and the general score of diarrhea due to spleen deficiency were improved; the improvements in fecal form and frequency, decreased appetite, bloating after meals and fatigue and sluggishness were more significant in the observation group than in the control group. Conclusion: The five-step pediatric tuina method of Huxiang school and conventional tuina both can improve the primary and secondary symptoms in infantile diarrhea due to spleen deficiency, while the former one can produce more significant efficacy.
6.Quick LC-MS/MS method for the determination of daptomycin in human plasma

Objective To develop a LC-MS/MS method for determination of daptomycinin human plasma.Methods The plasma samples were precipitated by acetonitrile,then the supernatant were separated on Eclipse Plus C18column (2.1 mm × 100 mm,3.5 μm) and eluted with acetonitrile-10 mmol · L-1 ammonium acetate (80∶ 20,containing 0.1% formic acid),and the column temperature was 25 ℃.Detection of the analyte was achieved using positive ion electrospray ionization (ESI) in the multiple reaction monitoring (MRM) mode.The MS/MS ion transitions monitored were m/z 811.0→m/z 159.1 and m/z 237.1→m/z 194.1 for daptomycin and internal standard,respectively.The specificity,standard curve and lower limitation of quantitation,precision and recovery rate and stability as well as the matrix effect were investigated.Results The linear regression equation was y =1.49 × 10-2x-2.06 × 10-3 (r =0.9980) The linear range of daptomycin was 0.10-100 μg · mL-1,with the lower limit of quantitation (LLOQ) of 0.10 μg · mL-1.Accuracy was 97.1%-104.8%,Intra-day and inter -dayrelative standard deviations were both below 15%.The absolute recovery in plasma was81.9%-91.4%.The matrix effects were 90.01%-109.28%.Conclusion The established method israpid,sensitive,accurate,specific and reliable,and suitable for the determination ofdaptomycin in human plasma and pharmacokinetic study.
7.Cloning and expression of zot gene of Vibrio cholerae and its biological activity

The aim of this study is to construct the expression vector of zot gene of Vibrio cholerae and to realize the expression of zot gene of Vibrio cholerae in E.coli and study the biological activity of its recombinant expression product.In order to express zot protein in E.coli,the full-length open reading frame of zot was amplified by PCR from the standard strains of Vibrio cholerae MO45 genome DNA.The PCR product was cloned into prokaryotic expression vector pET-32a(+) with restriction enaymes Bam H I and XhoI.The recombinant vector pET-32a(+)-zot was transformed into E.coli BL21 (DE3) and expressed by IPTG induction.The zot fusion protein was detected by SDS-PAGE and Western blotting and purified by Ni-NTA affinity chromatography.After expression and purification,the recombinant expression protein played as a target for human small intestinal epithelial cells.Restriction endonuclease digestion,PCR and DNA sequencing analysis showed that the zot gene of 1 200 bp was amplified from Vibrio cholerae DNA,and the recombinant plasmid pET-32a(+)-zot was constructed and detected its expression in prokaryotic cell successfully with SDS-PAGE and Western blot techniques.The zot recombinant protein was successfully expressed and purified.The purified zot recombinant protein can cause human intestinal epithelial cells apoptosis.
8.Comparison of processed and crude Polygoni Multiflori Radix induced rat liver injury and screening for sensitive indicators.

To investigate the difference of liver injury in rats gavaged with crude and processed Polygoni Multiflori Radix. The 75% ethanol extract of crude and processed Polygoni Multiflori Radix (50 g · kg(-1) crude medicine weight/body weight) were continuous oral administered to rats for 6 weeks. Serum biochemical indicators were dynamically detected, the change of liver histopathology was assessed 6 weeks later. Principal component analysis (PCA) was adopted to screen sensitive indicator of the liver damage induced by polygoni multiflori radix. Biochemical tests showed that the crude Polygoni Multiflori Radix group had significant increase of serum ALT, AST, ALP, DBIL and TBIL (P < 0.01 or P < 0.05) and significant decreases of serum IBIL and TBA (P < 0.01 or P < 0.05), while the processed Polygoni Multiflori Radix group showed no obvious changes, compared to the untreated normal group. Histopathologic analysis revealed that crude Polygoni Multiflori Radix group exhibited significant inflammatory cells infiltration in portal area around the blood vessels, tissue destruction and local necrosis of liver cells. There were not obvious pathological changes in processed Polygoni Multiflori Radix group. The results demonstrated that the injury effect of processed Polygoni Multiflori Radix on liver injury of rats was significantly lower than that of unprocessed, and that processing can effectively reduce the hepatotoxicity of Polygoni Multiflori Radix. Traditional transaminase liver function indicators were not sensitive for crude Polygoni Multiflori Radix induced liver damage. The serum content of DBIL and TBIL can reflect the liver damage induced by crude Polygoni Multiflori Radix early and can be sensitive indicators for clinical monitoring the usage of it.

9.Clinical features of children with incomplete Kawasaki disease.

OBJECTIVETo investigate the clinical features of children with incomplete Kawasaki disease (IKD), and to provide reference for the early diagnosis of IKD.
METHODSThe clinical data of 22 hospitalized children with IKD were analyzed retrospectively and compared with the data of 63 children with Kawasaki disease (KD) who were hospitalized during the same period of time. Another 20 children with pyrexia were enrolled as the control group.
RESULTSPyrexia was observed in all children. Compared with the KD group, the IKD group had significantly lower proportions of children with changes in the limbs, conjunctival hyperaemia, and cervical lymphadenectasis (P<0.05), a significantly higher serum level of glutamic-pyruvic transaminase (P<0.05), and significantly lower levels of plasma albumin, serum sodium, and interleukin-6 (P<0.05). There was no significant difference in the rate of γ-globulin application between the IKD and KD groups; however, the IKD group had a significantly higher incidence rate of coronary artery lesion than the KD group (P<0.05).
CONCLUSIONSThe symptoms and signs in children with IKD are untypical. The liver function test and serum hyponatremia and IL-6 measurements may be useful for the diagnosis of IKD.

10.Effects of sea cucumber cerebroside and its long-chain base on lipid and glucose metabolism in obese mice.

OBJECTIVETo investigate the effect of sea cucumber cerebroside(SCC) and its long-chain base(LCB) on lipid and glucose metabolism in obese mice.
METHODSThe mouse obese model was established by feeding high fat diet. The mice were randomly assigned to 4 groups: control group, model group, SCC group and LCB group. After 4 weeks, the glucose tolerance test was undertaken. After 5 weeks, the body fat content, organic indexes, serum lipid level, glycemic index and liver lipid level were determined.
RESULTSCompared with the model group, the glucose tolerance in the SCC group and LCB group was ameliorated significantly (P<0.01, P<0.05); glycemic index (P<0.01, P<0.01), the weight of adipose tissue (P<0.05, P<0.01) and the hepatic TG were reduced significantly (P<0.05, P<0.05).
CONCLUSIONSea cucumber cerebroside and its long-chain base can improve the glucose and lipid metabolism in obese mice.
