1.Contents Determination of 2 Components in Hadro-chlorine Gutta by HPLC

OBJECTIVE:To establish the HPLC method for the simultaneous content determination of chloramphenicol and prednisolone acetum in hadro-chlorine gutta.METHODS:The chromatographic column was HiQ sil C 18 ,the methanol-water-glacial acetic acid(70∶30∶0.04)served as mobile phase,and the detecting wavelength was278nm,the flow rate was0.8ml/min and the column temperature was30℃,the sample size was10?l.RESULTS:A satisfactory linear relationship be?tween was found when the concentration determination ranges of chloramphenicol and prednisolone acetum at22.36~89.44?g/ml and25.00~100.00?g/ml respectively;The average recoveries for chloramphenicol and prednisolone acetum were99.65%(RSD=0.88%)and99.57%(RSD=1.02)respectively.CONCLUSION:The method was simple,accurate and good in repro-ducibility,which can be used for the simultaneous contents determination of 2 components in hadro-chlorine gutta.
2.Clinical analysis of bacterial culture and susceptibility test for chronic prostatitis

Objective To study the effect of bacterial culture and susceptibility test for chronic prostatitis.Methods 108 cases of chronic prostatitis patients with prostatic fluid were separated culture and sensitivity test.Resuits The bacteria masculine rate was 67.4% in the group,it was in 71.3% in Gram-positive cocci,it was 17.6% in Gram-negative bacilli,it was 3.7% in Gram-positive baciUi and it was 7.4% in Fungi; Staphylococcus anreus,Coagulase negative staphylococcus,Streptococcus viridans,Enterococcus faecalis and Bacillus coil were drug fast to frequently used antibiotic on the wide-bound.Conclusion The bacterial infection has an important role in the invasion of chronic prostatitis and the incidence rate was high in the bacterial multidrug resistant.
3.Study on Relationship between Life Style and Chronic Diseases

Objective To analyze the relationship between life style and chronic diseases.Method 2 886 people were selected randomly from three districts of Tianjin and surveyed with a questionnaire on their healthy condition and physical examination.2 622 people were taken a diet survey by 3-days foods intakes weighing and 24-hours diet recall,then the relative risk factors between life style and coronary heart disease,cerebrovascular disease,diabetes,hypertension,overweight and obesity were analyzed.Result The standardized prevalence rates of coronary heart disease,cerebrevascular disease,diabetes,hypertension, overweight and obesity of residents were 9.24%,4.33%,6.74%,34.05%,and 42.62%,respectively.The more the intake of energy, the more the risk for people to suffer from overweight and obesity,OR value being 1.00-1.70.With intake of more than 20 g dietary fiber per day,the risk for suffering from overweight and obesity could be reduced,OR value being 0.68.Logistic analysis showed that heavy salty taste could increase the risk for suffering from hypertension by 42%.Watching TV more than 3 hours per day increased the risk for suffering from cerebrovascular disease and diabetes,OR value being 1.802 and 1.703 respectively.Sleep less than 7 hours per day was a dangerous factor for hypertension,coronary heart disease and cerebrovasular disease,OR value being 1.517, 1.833 and 2.041 respectively.Conclusion The life style such as dietary pattern,sedentary occupation and sleep hours have strong connection with chronic diseases,such as hypertension,diabetes etc.Reasonable diet,suitable exercises,reducing sedentary position and increasing sleep hours will be good for health,and also for the prevention and control of chronic diseases.
4.Theoretical research and clinical application of anterior cervical plating system

Anterior cervical plating has been widely used to treat cervical spine trauma, degeneration, deformity and tumor due to the advantages such as augmentation of cervical stability and promotion of bone graft fusion. This paper introduced the classification and evolution of plate-screw internal fixation system, analyzed the biomechanics of anterior cervical plate, and discussed its clinical application. Although the therapeutic effect of anterior cervical plating is favorable, there are still various complications postoperatively. Therefore, it is necessary to improve the anterior cervical plate from different aspects to expand its clinical application.
5.Effects of Early Intervention of Didang Decoction on AMP-activated Protein Kinase Signaling Pathway in Diabetic Rats

Objective To observe the effects of the intervention of Didang Decoction at different times on changes of AMPK signaling pathway related factors in macrovascular endotheliocytes of diabetic rats; To discuss the mechanism of mitochondria energy metabolism regulating the AMPK signaling pathway for macrovascular endothelial defense function. Methods Injection of STZ into the caudal vein and administration of high fat diet wer used to generate diabetic rat model. All rats were randomly divided into the following 7 groups: control, model, metformin, simvastatin, early-, middle-, and late-stage Didang Decoction group. Western blot was used to detect the expressions of APMKα1 and PGC-1α in rat aortic endothelial cells. Changes in the intracellular AMP and ATP levels were detected by ELISA. Real time fluorescence quantitative PCR was used to detected mRNA expressions of Caspase-3, eNOS, and Bcl-2 in tissue of thoracic aorta. Results Compared with the model group, the expressions of AMPKα1 and PGC-1α in the early-stage and middle-stage Didang Decoction group and simvastatin group increased (P<0.05); the gene expressions of Bcl-2, and eNOS significantly increased in the early-stage Didang Decoction group and simvastatin group (P<0.05), while the expressions of Caspase-3 decreased significantly (P<0.05). The expression of ATP increased significantly and the expression of AMP decreased significantly in the early-stage Didang Decoction group and simvastatin group (P<0.05), and the best effects were shown in the early-stage Didang Decoction group. Conclusion Early intervention of Didang Decoction can enhance energy metabolism in the mitochondria of macrovascular endothelial cells by regulating the AMPK signaling pathway, and then plays a role in strengthening the defense function of macrovascular endothelial cells.
6.Expression of miR-195 and its clinicopathologic significance in gastric canc er

Objective To investigate the expression of miR-195 and its association with clinicopathologic features in gastric cancer. Methods The expression level of miR-195 in 30 cases of gastric cancer and matched non-tumor adjacent tissue specimens were examined by using reverse transcription real-time PCR.The relationship between the expression of miR-195 and its clinicopathologic features of gastric cancer was analyzed.At the same time, the expression of miR-195 in four gastric cancer cell lines and normal human gastric epithelial cells were detected. Results The expression level of miR-195 was significantly lower in the tumor tissues than that in the adjacent tissues (P<0.05). Down-regulated miR-195 expression was associated with lymph node metastasis (P<0.05) in gastric cancer patients. Clinical and pathological analysis showed that miR-195 in gastric carcinoma tissues had no relationship with sex, age, the degree of tumor differentiation and tumor metastasis stage. Conclusion It is possible that miR-195 plays an important role in the generation and progression of gastric cancer, and it was associated with lymph node metastasis in gastric cancer patients.

Objective To explore the correlation between global end diastolic volume index(GEDI)and central venous pressure(CVP)of severe sepsis patients during fluid resuscitation by pulse induced continuous cardiac output(PiCCO )test. Methods Sixty-four severe sepsis patients had been treated by fluid resuscitation strictly in accordance with the guidelines for treatment of sepsis in 2008. During the treatment,GEDI and corresponding CVP data were measured and recorded to observe the correctness of volume reaction under different CVP ranges,and the relativity between GEDI and CVP was analyzed. Results In the whole process of monitoring circulatory blood volume,no significant correlation between CVP and GEDI(r=-0.012,P>0.05)was found in severe patients with severe sepsis. When the CVP was in 0-8 mm Hg(1 mm Hg=0.133 kPa)or greater than 12 mm Hg,there was no correlation with the GEDI(r=-0.009,-0.020,respectively,both P>0.05). When the CVP was in 8-12 mm Hg at the 6 hours resuscitation target required by the guidelines,there was an obvious positive correlation between CVP and GEDI(r=0.653,P<0.01). Conclusion CVP is not suitable to be an exclusive target indicator for fluid resuscitation volume.

BACKGROUND: Hundreds of mouse embryonic stem cell lines are established presently. Most of these cell lines are collected from 129, C57BL/6J and BALB/C mice. Success rate of creating embryonic stem cell lines with Kunming mice is very low. OBJECTIVE: To explore the method of isolation and culture of murine embryonic stem cells of kunming species to increase the efficiency of establishment of embryonic stem cell lines from Kunming species mouse. DESIGN, TIME AND SETTING: The cell experiment was performed at the Chongqing Key Laboratory of Neurology from April 2006 to June 2007. MATERIALS: Blastocysts of 3.5 days and 4 days from Kunming species mouse were collected. METHODS: The blastocysts of 3.5 days and 4 days from Kunming species mouse were cultured on the fibroblast cell feeder layers for 4-5 days, and the cells from inner cell mass were isolated and subsequently cultured in vitro in 2.5 g/L trypase-0.4 g/L ethylenediamine tetraacetic acid (EDTA) and 1 g/L trypase-0.2 g/L EDTA at room temperature for 2-4 minutes. Embryonic stem cell colony was mechanically straggled. MAIN OUTCOME MEASURES: Colony growth was observed and determined by alkaline phosphatase staining, OCT-4 staining and karyotype analysis. 10.Value of magnetic resonance cholangiopancreatography for diagnosis of pancreas divisum

Objective:To investigate the value of magnetic resonance cholangiopancreatography(MRCP)in the diagnosis of pancreas divisum by comparing with endoscopic retrograde cholangiopancreatography(ERCP).Methods:The MRCP and ERCP images of 8 patients with pancreas divisum were retrospectively analyzed.The diagnostic accuracy and findings by MRCP were compared with those by ERCP.Results:MRCP had a diagnostic accuracy of 87.5%(7/8)based on the result of ERCP.ERCP displayed the dominant dorsal pancreatic ducts in all 8 cases and ventral pancreatic ducts in 6 cases;MRCP also displayed the dominant dorsal pancreatic ducts in all 8 cases,but the ventral pancreatic ducts only in 3 cases.Conclusion:As a non-invasive technique,MRCP has important clinical value in the diagnosis of pancreas divisum.
