Objective To study the effect of PPARγ ligand troglitazone (TGZ) on invasion and metastasis of gastric cancer cells, and investigate the relationship of PPARγ ligand with gastric cancer.Methods The expression of PPARγ in gastric cancer cell line MGC803 were detected by immunofluorescence cytochemistry method. The effect of different density TGZ on proliferation activity and adhesion of gastric cancer cell were detected by MTT chromatometry. The effect of different ligands on invasion and metastasis of gastric cancer cell MGC803 were detected by invasion system in vitro. Results The expression of PPARγ mainly located in cell nucleus. TGZ inhibited the proliferation of gastric cancer cell, decreased cell adhesion, locomotory capacity and invasion to matrigel, which had time and dose-dependent relationship.When treatment with 0. 1,1.0 and 10μ mol/ L TGZ, inhibition ratio of invasion and metastasis of cell was 8.79% ,31.31% ,51.42% and 28.29% ,4. 27% ,59. 27% respectively, which had statistical significance compared with control group( P <0. 05). When treatment was 10μ mol/L TGZ, cell adhesion was 0. 32 ±0. 03, it was statistically significant higher than that in control group (0. 52 ± 0. 04, P < 0. 05 ). Conclusion Human gastric cancer cell line MGC803 expressed functional PPARγ protein. TGZ inhibited adhesion and invasion of MGC803 cell on ECM at different degree, the effect of combination of two ligands was evident, which mechanism of action needed to be further investigated.

OBJECTIVE:To study the antitumor effect of phellinus linteus polysaccharide on sarcoma S180 cells in vivo and in vitro. METHODS:Sarcoma S180 cells in logarithmic growth period were selected,adding into 0(blank control),2,4,8 mg/mL phellinus linteus polysaccharide solution and respectively culturing for 12,24,36,48 h. The in vitro proliferation inhibition rate of cells was determined by MTT method;its apoptotic morphology was observed by fluorescence staining and cell apoptosis rate was detected by flow cytometry. S180 tumor-bearing mice models were established and randomly divided into control group,phellinus linteus polysaccharide high-dose,medium-dose,low-dose groups(400,200,100 mg/kg),10 in each group. Model mice were in-tragastrically administrated related medicined,once a day,for 12 d. Mice were executed after 24 h of last administration,tumor weight was determined,tumor inhibition rate was calculated. Immunohistochemistry was conducted to detect the tumor suppressor gene PTEN and oncogene C-myc protein expressions in tumor tissue. RESULTS:Compared with blank control group,phellinus linteus polysaccharide can increase the proliferation inhibition rate of S180 cells and induce the increase of apoptosis rate(P<0.05 or P<0.01),showing a concentration-time manner. Compared with control group,the tumor inhibition rates in phellinus linteus polysaccharide groups were obviously increased (P<0.01),PTEN protein expressions were strengthened (P<0.05 or P<0.01) and C-myc protein expressions were weakened (P<0.05). CONCLUSIONS:Phellinus linteus polysaccharide shows antitumor ef-fect in vivo and in vitro,which can up-regulate the PTEN,down-regulate C-myc protein expressions.

Objective To evaluate the efficacy of arthroscopic debridement combined with microfracture on management of chronic enthesiopathy of Achilles tendon.Methods Fifteen cases of chronic enthesiopathy of Achilles tendon were enrolled in this study from January 2013 to January 2016.There were 11 males and 4 females,with an average age of 55.5±8.9 years.Five were in left side,and 10 in right side.All the patients failed to recover after conservative treatments for 18 months to 10 years.Pathological tissue was eliminated by blade.Then microfracture was made.Arthroscopic awls (1.5 or 2 mm in diameter) were used for making multiple holes at the footprint of achilles tendon.These holes were made far enough (3-4 mm) apart so they do not break into each other.Fat droplets and bleeding from the marrow cavity were seen when the appropriate depth (approximately 5 to 10 mm) has been reached.The patients were evaluated preoperatively and postoperatively using American Orthopaedic Foot and Ankle Society (AOFAS) Ankle-Hindfoot scores and visual analogue scale (VAS) scores.Results Average operation time and length of stay were 30.6± 15.4 min and 6.2±2.8 d,respectively.Postoperative incisions were Ⅰ stage healing for all patients.There were no perioperative and postoperative complications related to the procedure,such as rupture of achilles tendon,blood vessel and nerve injury.No infection and recurrence was found in all cases.All patients were followed up for an average time of 13.5 months.No rupture of achilles tendon or relapsing symptom was found.Average VAS scores before surgery,6 month after surgery and at the lattest follow-up were 7.3± 1.6,2.4± 1.3,and 2.3± 1.3,respectively.Average AOFAS Ankle-Hindfoot scores were 38.5± 7.8,92.9±6.9,and 91.6±7.1,respectively.Average postoperative VAS scores and AOFAS Ankle-Hindfoot scores were significantly improved compared with those before surgery.There was no statistically significant difference among all check point after operation in VAS scores and AOFAS Ankle-Hindfoot scores.At the latest follow-up,all patients were satisfied with the degree of painful relief.According to AOFAS Ankle-Hindfoot scores,overall excellent and good rate was 93.3％ (excellent in 10 cases,good in 4 cases,and fair in 1 case).Conclusion Clinical results of arthroscopic debridement combined with microfracture for treatment of chronic enthesiopathy of Achilles tendon are good.This technique is easy and safe to perform.

Objective To explore the relationship between acceptance of disability and hope level in patients with total laryngectomy.Methods Totally 84 patients with total laryngectomy were recruited and investigated with the general information questionnaire,Acceptance of Disability Scale (ADS)and Herth Hope Index (HHI).Results The total score of ADS was (85.82±12.64),which showed medium level acceptance of disability.The total score of Herth Hope Index (HHI) was (35.50±8.12),which showed high level HHI.Significantly positive correlation was found between acceptance of disability and HHI.Conclusions Nurses should pay more attention to the acceptance of disability in patients with total laryngectomy,and provide effective psychological intervention to improve the patients' menial health and quality of life.

Objective: To compare effects of drug and psychotherapy in the treatment of depressive disorder. Methods: 63 depressive patients were randomly divided into two treatment groups: drug or psychotherapy. MMPI, SDSS and HAMD were used for assessing treatment effects on personality and social functions. Results: The effects of psychotherapy and drug were compatible for controlling symptoms of depression. Psychotherapy was superior to drug in preventing relapses, as well as improving cognitive and social functions. Conclusion: Psychotherapy should be applied to depressive patients. Dosage and duration of administering sedative antidepressants should be reduced in the treatment of depression.

Objective To examine the incidence of metabolic syndrome (MS) in incipient female patients with rheumatoid arthritis (RA), and study the relationship between MS and disease activity of RA. Methods The clinical data of 102 incipient female RA patients (RA group) and 125 healthy controls (control group) were retrospectively analyzed. The incidence of MS was compared between 2 groups, and the clinical and laboratory data were analyzed between patients with MS and patients without MS. Results There were no statistical differences in age, body mess index (BMI), waist circumference, systolic blood pressure, diastolic blood pressure, fasting blood glucose (FBG), triglyceride (TG) and high-density lipoprotein cholesterol (HDL-C) between 2 groups (P>0.05). The incidences of waist circumference ≥ 80 cm, TG ≥ 1.70 mmol/L, HDL-C ≤ 1.30 mmol/L, MS according to the National Cholesterol Education Program Adult Treatment Panel Ⅲ (NCEP-ATPⅢ) standard and MS according to the Chinese Diabetes Society (CDS) standard in RA group were significantly higher than those in control group: 39.2% (40/102) vs. 25.6% (32/125), 37.3% (38/102) vs. 24.0%(30/125), 34.3%(35/102) vs. 21.6%(27/125), 34.3%(35/102) vs. 22.4%(28/125) and 27.5%(28/102) vs. 16.0% (20/125)%, and there were statistical differences (P<0.05). There were no statistical differences in the incidences of BMI≥25 kg/m2, systolic blood pressure ≥130 mmHg (1 mmHg = 0.133 kPa), diastolic blood pressure ≥85 mmHg and FBG≥5.6 mmol/L between 2 groups (P>0.05). The RA patients were divided into RA with MS (35 cases) and RA without MS (67 cases) according to the MS diagnostic standard of NCEP-ATPⅢ. The erythrocyte sedimentation rate (ESR), C-reactive protein (CRP) and blood uric acid in RA with MS patients were significantly higher than those in RA without MS patients:(68.1 ± 26.3) mm/h vs. (40.1 ± 30.6) mm/h, 4.260 mg/L vs. 1.505 mg/L and (330.5 ± 69.1)μmol/L vs. (269.0 ± 67.5)μmol/L, and there were statistical differences (P<0.05). There were no statistical differences in age, involved joint number, rheumatoid factor (RF), hemoglobin and incidence of anti-cyclic citrullinated peptide antibody (CCP antibody) between RA with MS patients and RA without MS patients (P>0.05). Conclusions The incidence of MS in incipient female patients with RA is higher than that in healthy controls, and RA patients are more prone to abdominal obesity, TG increasing and HDL-C decreasing. The inflammatory index and blood uric acid levels in RA with MS patients are significantly higher than those in RA without MS patients.

Objective:To investigate the stiffness characteristic of triple-negative human breast cancer at different size in a implantation nude female mice model using shear wave elastography(SWE) and to compare the clinical pathologic features of tumors with elasticity variables.Methods:Human breast cancer MDA-MB-231 cells were injected into 30 nude female mice and 27 transplanted tumors were successfully found in nude female mice. Ultrasound and SWE were longitudinally performed on maximum diameter plane of 21 tumours in 21 nude mice. The elastic parameters of maximal elasticity(Emax), mean elasticity (Emean) and standard deviation of elasticity(Esd) were recorded. The mice were divided into 3 groups according to the tumor size. They were group A with tumor size less than or equal to 5 mm, group B with tumor size greater than 5 mm and less than or equal to 10 mm, group C with tumor size larger than 10 mm and smaller than or equal to 15 mm. Compared with pathology, the relationships between Ki67 of transplanted tumor and elastic parameters were analyzed.Results:As the transplanted tumors increased, the values of Emax, Esd, Ki67 all increased. The lesions maximal size, Emax, Esd, Ki67 were significant higher in group B ( P<0.001, P=0.006, P=0.002, P=0.026) and group C ( P<0.001, P<0.001, P<0.001, P=0.028) than group A. The other parameters were not significantly different among the groups(all P>0.05). The size of transplanted tumors was significantly and positively correlated with Emax ( rs=0.673, P=0.001), Esd ( rs=0.661, P=0.001), and Ki67 ( rs=0.509, P=0.018). Conclusions:SWE Emax and Esd can reflect the tumor tissue stiffness change and biological activity during the tumor growth.

To explore novel coumarin derivatives with more potent anti-proliferative activity, a series of novel compounds were designed and synthesized by linking Schiff base and N, N-bis (2-chloroethyl) amine pharmacophore of nitrogen mustards to the coumarin's framework. Their structures were confirmed by 1H NMR, MS and element analysis techniques. In vitro anti-proliferative activities were evaluated against HepG2, DU145 and MCF7 cell lines by the standard MTT assay. The results showed that some of the target compounds exhibited strong anti-proliferative activities against selected tumor cells, and compounds 7c, 7f, 7g, 7h and 7q were better than or equal to the activities of positive control, they deserved further development.

AIM To prepare diosgenin nanosuspensions.METHODS The nanosuspensions prepared by media milling method were solidificated by freeze drying method.With particle size and polydispersity index (PDI) as evaluation indices,stabilizer kind,ratio of diosgenin to stabilizer,ratio of preliminary nanosuspension volume to grinding bead amount,milling time,lyoprotectant kind and its amount as influencing factors,single factor test was applied to screening preparation and solidification processes.The morphology of nanosuspensions was observed,then the particle sizes and polydispersity indices of nanosuspensions and freeze-dried powder were determined.RESULTS The optimal conditions were determined to be 6:1 for ratio of diosgenin to Pluronic F127 (stabilizer Ⅰ),50:1 for ratio of diogenin to sodium dodecyl sulfate (SDS,stabilizer Ⅱ),1:4 for ratio of preliminary nanosuspension volume to grinding bead amount,120 min for milling time,and 8％ PEG-6000 and 2％ mannitol as lyoprotectants.The average particle size and polydispersity index of rod-like or flaky nanosuspensions were (348.1 ±14.2) nm and 0.244 ± 0.059,respectively,which were lower than those of freeze-dried powder.At room temperature,the particle sizes of nanosuspensions and freeze-dried powder remained stable within 35 d and 3 months,respectively.CONCLUSION The physical stability of diosgenin freeze-dried powder is superior to that of its nanosuspensions,which can be used after being reconstituted.
