3.Effect of CIP2 A over-expression on proliferation and apoptosis of gastric epithelial cells

AIM:To investigate the effects of cancerous inhibitor of protein phosphatase 2A (CIP2A) over-ex-pression on the proliferation and apoptosis of gastric epithelial cells .METHODS:The mRNA expression of CIP2A and cy-clin D1 in the tissues of normal gastric mucosa and gastric polyps was detected by RT -qPCR.The GES-1 cells were divided into control group, Ad-emp group and Ad-CIP2A group.The cell proliferation ability was detected by MTT assay and BrdU assay , and the cell apoptosis was analyzed by flow cytometry .The expression of apoptosis-related molecules was determined by Western blot and RT-qPCR.The levels of inflammatory cytokines were measured by ELISA after GES-1 cells were infec-ted with Ad-emp and Ad-CIP2A.Furthermore, the protein levels of p-Rb, E2F1 and cyclin D1 in the GES-1 cells was de-termined by Western blot after transfected with E2F1 siRNA.RESULTS:The expression of CIP2A and cyclin D1 in ade-nomatous gastric polyps tissues was significantly higher than that in normal gastric mucosa tissues , and no significant change of that between hyperplastic gastric polyps tissues and normal gastric mucosa was observed .After transfected with CIP2A, the proliferation ability of GES-1 cells was increased , the cell apoptosis was inhibited , the concentrations of IL-1βand IL-10 was up-regulated and the protein levels of p-Rb, E2F1 and cyclin D1 were increased, while the protein levels of p-Rb, E2F1 and cyclin D1 were significantly decreased after transfected with E2F1 siRNA.CONCLUSION: CIP2A promotes the proliferation and inhibits the apoptosis of GES-1 cells by activating Rb/E2F1.
4.Low protein diet for patients with chronic kidney disease

Diet therapy is considered as the basic treatment of chronic kidney disease (CKD),and its primary goals are improving metabolic disorder,relieving symptoms and prevent complications,hence to retard renal function lost of CKD patients.However,there have been controversies about the results of clinical studies on nutritional treatment for CKD.This article reviews the effectiveness,safety and compliance of low-protein diet (LPD) for CKD patients,and related clinical management strategies.The article also recommends the feasible diet scheme to provide a reference of clinical LPD therapy for patients with CKD.
5.Enlightenment from the Y2K World Health Report

To attain the systematic goals of the health cause, WHO designed a complete assessment system for measuring the performance of health organizations and conducted an assessment and ranking of various countries using the system. The paper gives a brief account of the report and discusses the enlightenment we can gain from it.
6.The immunotherapy of lung cancer

Lung cancer is characterized by high malignancy and the majority of patients are diagnosed at the advanced or late stage.Chemoradiotherapy has insufficient effect on this malignant tumor.At present the immune therapy has become a new choice for lung cancer treatment.Ipilimumab,antibody to programmed death-1 ,immune cells,cytokines,melanoma-associated antigen A3 vaccine,liposomal BLP25,belagen-pumatucel-L and polypeptide vaccine have been proved effective for lung cancer through various clinical trials. Futhermore,most of them have been moved forward to phase Ⅲ clinical trials in order to get more strong evidence to support the immunotherapy incorporated into the multidisciplinary treatment of lung cancer.
7.Problems and Solutions to Bilingual Teaching in Local Colleges

Allogeneic hematopoietic stem cell transplantation (allo-HSCT) remains a curable method for hematological malignancies.Nearly all transplant candidates have donor with the successful application of haploidentical SCT.This article summarized some reports on donor selection,prophylaxis and treatment of graft-versus-host disease,and graft failure in the 58th American Society of Hematology Annual Meeting to present the progress of allo-HSCT in treating hematological malignancies.
9.Clinical analysis of nine patients with presumed tubercular retinal vasculitis

Objective To observe the clinical features and treatment outcomes of presumed tubercular retinal vasculitis.Methods This is a retrospective non-comparative interventional clinical research.A total of nine patients (11 eyes) with major presentation of retinal vasculitis were included in this study.Patients first consulted the eye clinic and were diagnosed presumed tubercular retinal vasculitis.The patients,seven males and two females,aged from 19 to 66 years,with an average of 43.89 years.The time interval from symptoms to diagnosis ranged from two weeks to six months with an average of 76.27 days.Visual acuity,slit lamp ophthalmoscopy,fundus fluorescein angiography (FFA),optical coherence tomography (OCT),hematological and tuberculosis related investigations were examined and analyzed.All patients had standard anti-tuberculosis treatment.Treatment outcomes were followed for six to 37 months with an average of 14.11 months.Results Baseline visual acuity ranged from hand movement to 0.8 with an average of 0.28.Among 11 eyes,six presented mild to moderate vitritis,five presented as retinal vein occlusion with no obvious vitirits.Fundus examination showed six cases with retinal hemorrhage,four cases with macular edema,two with macular epiretinal membrane,and two with vitreous hemorrhage.FFA revealed 11 cases with leakage of vessels,11 with nonperfusion area,four with macular edema,three with retinal neovascularization,and two with choroidal lesions.OCT of nine eyes suggested six eyes with retinal edema,three with macular edema,three with macular epiretinal membrane.TST of seven patients were all strong positive.T-SPOT.TB of four patients were all positive.Three of eight patients who had chest X-ray or chest CT were suggested tuberculosis infection.Four to six weeks after the start of anti-tuberculosis treatment,vitritis,exudates,retinal and macular edema subsided.During follow up,inflammation was stable with no recurrence observed.The visual acuity of last follow-up ranged from 0.15 to 0.8 with an average of 0.51.Conclusions The main presentations of presumed tubercular retinal vasculitis are vitritis,retinal vein occlusion,and retinal hemorrhage.Standard anti-tuberculosis treatment can improve inflammation and retinal hemorrhage.
10.Expressions and significance of bcl-6 and Ki-67 in diffuse large B-cell lymphoma

Objective To study the expressions and significance of bcl-6 and Ki-67 in diffuse large B-cell lymphoma(DLBCL).Methods Immunohistochemistry technique was used to detect the expressions of bcl-6 and Ki-67 in the tissues of 90 DLBCL patients. 20 cases of reactive hyperplasia of lymph node (RH) were used as control. Results Positive expression rate of bcl-6 are 54.44 %(49/90), 15.00 %(3/20) in DLBCL or RH tissues respectively (x2=10.214,P=0.001). There were correlations between bcl-6 expression and Ann Arbor clinical stage, LDH, B symptoms, or Hans classification (x2=5.257,5.257,4.704,16.024 respectively,all P<0.05).The high expression rate of Ki-67 were 80.00 %(72/90),20.00 %(4/20)in DLBCL or RH tissues respectively (x2=27.585,P=0.000). There were correlations between Ki-67 expression and Ann Arbor clinical stage,IPI or recent effect (x2=5.889,6.451,6.024 respectively,all P<0.05).Conclusion There are significant correlations between the aberrant expression of bcl-6 or Ki-67 and Ann Arbor clinical stage,IPI or Hans classification. The expressions of bcl-6 and Ki-67 may provide important information for the clinical therapy and prognosis of DLBCL.
